Progesterone, Estradiol 17 beta 및 Cholesterol Sucrose 층으로부터 정자의 Swim-up 분리에 미치는 영향 (Effect of Progesterone, Estradiol 17 beta and Cholesterol on Sperm Swim-up Separation through Sucrose Layer)

  • 난포액에 함유되어 있는 steroids와 sterol이 수정에 참여하는 정자의 주화성에 미치는 영향을 밝히기 위하여, progesterone, estradiol 17 beta 및 cholesterol이 sucrose 층으로부터 정자의 swim-up 분리에 미치는 영향을 조사하였던 바 얻어진 결과는 다음과 같다. 1. Progesterone은 정자의 이동과 운동성을 억제하였으나 수정능획득한 정자를 유인하였으며, 특히 50$\mu$g/ml 수준의 progesterone은 수정능력을 획득한 정자의 이동을 유의하게 자극하였다. 2. Estradiol 17 beta의 첨가수준이 증가할수록 정자의 이동과 운동성이 억제되었으나, 10$\mu$g/ml 수준의 estradiol은 정자의 이동과 운동성 및 수정능획득정자의 이동에 영향을 미치지 않았다. 3. Cholesterol은 정자의 이동과 운동성을 자극하였으나, 수정능획득정자의 이동에 영향을 미치지 않았으며, 특히 50$\mu$g/ml 수준의 cholesterol은 정자의 이동과 운동성을 유의하게 자극하였다. 4. Progesterone, estradiol 및 cholesterol의 병용처리에서, cholesterol은 정자의 이동과 운동을 유의하게 자극하였으나, progesterone과 estradiol은 이러한 cholesterol의 효과를 감소시켰다. Progesterone은 수정획득 정자의 이동을 자극하였으나 estradiol이나 cholesterol은 progester-one의 이러한 효과를 억제하지 않았다. 결론적으로 progesterone은 50$\mu$g/ml 첨가수준에서 수정능력을 획득한 정자의 swim-up이동을 유의하게 자극하였으며, cholesterol은 50$\mu$g/m1 첨가수준에서 정자의 이동을 자극하였으나, estradiol은 10$\mu$g/m1 첨가수준에서 정자의 이동과 운동성에 영향을 미치지 않았다.

  • $\beta$-Glucan 분해효소의 간편하며 예민한 활성도 측정방법으로 $\beta$-glucan에 색소와 cross linking agent를 접합시키는 변형기질 제조시 영향을 미치는 조건을 조사하고 맥아와 세균의 $\beta$-glucan분해효소 측정에 적응시켜 활성도를 측정한 결과는 다음과 같다. 1.0g $\beta$-glucan은 0.1N NaOH용액에서 색소 cibacron blue 3 G-A 1.5g과 cross linking agent인 1,4-butanedioldiglycidyl ether 1.25 $m\ell$을 90분간 끓여서 색소 접합기질로 제조하였을 때 $\beta$-glucanase 활성도 측정에 최적조건이었다. 변형기질은 pH5.3 에서 안정성을 보였으며 Bacillus subtilis K-4-3에서 추출한 효소액에 변형기질을 반응시켰을 때 간편하고 정확한 효소활성 측정이 가능하였다. 또한 색소방법을 DNS방법과 비교한 결과 색소방법이 $\beta$-glucan 분해효소 측정에 적당한 방법이었음이 입증되었다.

Effect of β-Mannanase and α-Galactosidase Supplementation to Soybean Meal Based Diets on Growth, Feed Efficiency and Nutrient Digestibility of Rainbow Trout, Oncorhynchus mykiss (Walbaum)

  • A 12-week feeding trial was conducted with 87 g rainbow trout to evaluate the effects on growth performances, feed efficiency and nutrient digestibility of adding ${\beta}$-mannanase and ${\alpha}$-galactosidase enzymes, solely or in combination. Seven diets were prepared by adding ${\beta}$-mannanase, ${\alpha}$-galactosidase and mixed enzyme at two different levels (1 g/kg and 2 g/kg) to control diet (without enzyme) including soybean meal. Mixed enzymes (1 g/kg, 2 g/kg) were prepared by adding ${\beta}$-mannanase and ${\alpha}$-galactosidase at the same doses (0.5+0.5 g/kg and 1+1 g/kg). At the end of the experiment, addition of ${\beta}$-mannanase, ${\alpha}$-galactosidase and mixed enzyme to diet containing 44% soybean meal had no significant effects on growth performance and gain:feed (p>0.05). In addition, adding ${\beta}$-mannanase, ${\alpha}$-galactosidase and mixed enzyme in different rations to trout diets had no affect on nutrient digestibility and body composition (p>0.05).

  • Recent human genetic studies have shown that $G{\beta}5$ is related to various clinical symptoms, such as sinus bradycardia, cognitive disability, and attention deficit hyperactivity disorder. Although the calcium signaling cascade is closely associated with a heterotrimeric G-protein, the function of $G{\beta}5$ in calcium signaling and its relevance to clinical symptoms remain unknown. In this study, we investigated the in vitro changes of store-operated calcium entry (SOCE) with exogenous expression of $G{\beta}5$. The cells expressing $G{\beta}5$ had enhanced SOCE after depletion of calcium ion inside the endoplasmic reticulum. $G{\beta}5$ also augmented Stim1- and Orai1-dependent SOCE. An ORAI1 loss-of-function mutant did not show inhibition of $G{\beta}5$-induced SOCE, and a STIM1-ERM truncation mutant showed no enhancement of SOCE. These results suggested a novel role of GNB5 and Stim1, and provided insight into the regulatory mechanism of SOCE.

Production of Chlorphenesin Galactoside by Whole Cells of ${\beta}$-Galactosidase-Containing Escherichia coli

  • We investigated the transgalactosylation reaction of chlorphenesin (CPN) using ${\beta}$-galactosidase (${\beta}$-gal)-containing Escherichia coli (E. coli) cells, in which galactose from lactose was transferred to CPN. The optimal CPN concentration for CPN galactoside (CPN-G) synthesis was observed at 40 mM under the conditions that lactose and ${\beta}$-gal (as E. coli cells) were 400 g/l and 4.8 U/ml, respectively, and the pH and temperature were 7.0 and $40^{\circ}C$, respectively. The time-course profile of CPN-G synthesis under these optimal conditions showed that CPN-G synthesis from 40 mM CPN reached a maximum of about 27 mM at 12 h. This value corresponded to an about 67% conversion of CPN to CPN-G, which was 4.47-5.36-fold higher than values in previous reports. In addition, we demonstrated by thin-layer chromatography to detect the sugar moiety that galactose was mainly transferred from lactose to CPN. Liquid chromatography-mass spectrometry revealed that CPN-G and CPN-GG (CPN galactoside, which accepted two galactose molecules) were definitively identified as the synthesized products using ${\beta}$-gal-containing E. coli cells. In particular, because we did not use purified ${\beta}$-gal, our ${\beta}$-gal-containing E. coli cells might be practical and cost-effective for enzymatically synthesizing CPN-G. It is expected that the use of ${\beta}$-gal-containing E. coli will be extended to galactose derivatization of other drugs to improve their functionality.

한국인의 식이섭취와 암 유발의 관계에 관한 연구 -제 1 보 특히 ${\beta}-Carotene$ 섭취량과 혈청내 수준을 중심으로 - (A Study of Relation Between Dietary Vitamin A Intake and Serum Vitamin A Levels and Cancer Risk in Korea)

  • Even though the anticarcinogenic effect of dietary factors especially beta - carotene has been reported by various investigators, the mechanism of the action of ${\beta}-carotene$ has not yet been identified. We carried out the present study to determine the possibilities of relative cancer risk related to dietary intake of vitamin A ( both ${\beta}-carotene$ and retinol ) and blood levels of vitamin A among Koreans. The subjects were divided into two groups; cancer patients and controls. Blood levels for ${\beta}-carotene$ and retinol were analyzed by alumina column chromatography and colorimetry. Dietary intake was examined by food profile and convenient method for evaluating nutritional status through recalling 10 years of food habits. The results obtained are as follows : 1 ) Calorie, protein, fat, and carbohydrate intakes of cancer patient were lower than those of control. Calorie and carbohydrate intakes showed no significant difference but protein and fat intakes were significantly lower in cancer patients. According to cancer sites, in stomach cancer only fat intake was significantly lower than that of control. In lung and larynx cancer calorie, protein, fat and carbohydrate intakes showed similar trend as in control. 2 ) Vitamin A intake of cancer patient was significantly lower than that of control. It was estimated that 83.6% of total Vitamin A intake were provided by ${\beta}-carotene$ for control and cancer patient respectively. 3 ) The mean intake of dietary ${\beta}-carotene$ in cancer patient was significantly lower than that in control ( $7002\;\mu}g/day$ versus $10326\;{\mu}g/day$ ) According to cancer sites in mean intake of dietary ${\beta}-carotene$ was significantly lower in all but stomach cancer compared with that of control. Lung and larynx cancer showed lowest ${\beta}-carotene$ intake with mean value of $5855{\mu}g/day$ and $5492{\mu}g/day$ respectively. 4 ) The mean intake of dietary retinol in cancer patient was significantly lower than that in control ( $245{\mu}g/day$ versus $338{\mu}g/day$ ), but the difference was not significant. 5 ) The relative risk of all cancers in the first (lowest) to the forth quartile level of ${\beta}-carotene$ consumption such as $0-5999{\mu}g/day$. $6000-8999{\mu}g/day$, $9000-11999{\mu}g/day$/ day and $12000-20000{\mu}g/day$ was 85 : 1.7 : 20 : 1.0. The relative risk of all cancers in the first (lowest) to the forth quartile level of retinol consumption, such as $0-299{\mu}g/day$, $300-599{\mu}g/day$, 600-899${\mu}g/day$, and $900-1200{\mu}g/day$, was 1.14 : 067 : 0.21 : 1.0. 6 ) The various food group consumption of cancer patient were significantly lower than those of control in green leafy vegetables, fruits, sea weeds, milk and cheese and eggs. But the Kimchie consumption in cancer patient was three fold higher than that of control ( $1840\;{\mu}g/day$ versus $562\;{\mu}g/day$ ) and in the stomach cancer, Kimchie consumption was the highest, ( $1890\;{\mu}g/day$) According to cancer sites, the consumption of green leafy vegetables was significantly lower in all but stomach cancer compared to control and other vegetables showed no difference between two. In lung and larynx cancer, green leafy vegetables consumption was lowest ( $6094{\mu}g/day$ $5921{\mu}g/day$) and milk and cheese consumption was also( $5\;{\mu}g/day$ and $11{\mu}g/day$) 7 ) The recovery of ${\beta}-carotene$ from human serum by alumina column chromatography was $94.4{\pm}2.3%$. 8 ) Cancer patients showed significantly lower serum retinol ($56.4{\pm}18.1\:{\mu}g/100ml$ versus $72.2{\pm}21.8\:{\mu}g/100ml$) and ${\beta}-carotene$ ($48.9{\pm}33.8\:{\mu}g/100ml$ versus $72.2{\pm}42.6\:{\mu}g/100ml$) concentrations than in controls. 9 ) But breast cancer patients were not significantly different from controls in their serum retinol and ${\beta}-carotene$ concentrations.

Characterization of Double Transgenic Mice Harboring Both Goat $\beta$-casein/hGH and Goat $\beta$-casein/hG-CSF Hybrid Genes

  • In an attempt to simultaneously produce two human proteins, hGH and hG-CSF, in the milk of transgenic mice, we constructed goat $\beta$-casein-directed hGH and hG-CSF expression cassettes individually and generated transgenic mice by co-injecting them into mouse zygotes. Out of 33 transgenic mice, 29 were identified as double transgenic harboring both transgenes on their genome. All analyzed double transgenic females secreted both hGH and hG-CSF in their milks. Concentrations ranged from 2.1 to $12.4\;mg/m{\ell}$ for hGH and from 0.04 to $0.13\;mg/m{\ell}$ for hG-CSF. hG-CSF level was much lower than hGH level but very similar to that of single hG-CSF mice, which were introduced with hG-CSF cassette alone. In order to address the causes of concentration difference between hGH and hG-CSF in milk, we examined mRNA level of hGH and hG-CSF in the mammary glands of double transgenic mice and tissue specificity of hG-CSF mRNA expression in both double and single transgenic mice. Likewise protein levels in milk, hGH mRNA level was much higher than hG-CSF mRNA, and hG-CSF mRNA expression was definitely specific to the mammary glands of both double and single transgenic mice. These results demonstrated that two transgenes have distinct transcriptional potentials without interaction each other in double transgenic mice although two transgenes co-integrated into same genomic sites and their expressions were directed by the same goat $\beta$-casein promoter. Therefore goat $\beta$-casein promoter is very useful for the multiple production of human proteins in the milk of transgenic animals.

착색시간에 따른 칼라렌즈 색좌표 이동 (Color lens CIE L*a*b* coordination transfer by tinted time)

  • 5분 간격으로 착색시간을 증가시켜 R, G, Y, B칼라렌즈를 제작하였다. 색 좌표 분석은 분광측색계와 CIE $L^*a^*b^*$ color system를 사용하였다. 착색 시간에 따른 색좌표 이동 형태는 $b^*(Y-B)={\beta}a^*+{\alpha}$이며, 거의 선형적으로 변하였다. 적색의 경우 착색시간이 길수록 $G{\rightarrow}R$, $B{\rightarrow}Y$ 형태로 이동하고, Parameter ${\alpha}$, ${\beta}$ 값은 각각 -3.49, 0.90을 얻었다. 녹색의 경우 색좌표 이동은 $R{\rightarrow}G$, $B{\rightarrow}Y$ 형태이다. 10분의 착색 시간에서 가장 순수한 녹색 칼라를 얻었다. Parameter ${\alpha}$, ${\beta}$값은 각각 -0.72, -0.55을 얻었다. 황색과 청색의 경우 모두 착색 시간이 길수록 순수한 색이 된다. 황색에서 Parameter ${\alpha}$, ${\beta}$값은 각각 14.11, 1.58이고, 청색에서 Parameter ${\alpha}$, ${\beta}$값은 각각 -11.62, 1.30을 얻었다.

폐광지성(廢鑛地城) 주민(住民)의 요중(尿中) 카드뮴 농도(濃度)와 현기능평가(賢機能評價) (A Study on Urinary Cadmium Concentration and Renal Indices of Inhabitant in an Abandoned Mine Area)

  • 1995년 8월부터 12월 사이에 미량의 카드뮴에 오염된 것으로 밝혀진 폐광 인근 지역에 거주하고 있는 지역주민 184명(남자: 82명, 여자: 102명)을 대상으로 카드뮴에 의한 만성적인 폭로와 신기능장애를 평가하기 위하여 요중 카드뮴과 NAG활성도, $\beta_2MG$배설량 및 총단백질량을 정량하여 비교 분석하였다. 이때 특별히 카드뮴에 폭로된 적이 없는 일반주민 160명(남자 : 64명, 여자 : 96명)을 대조군으로 선정하였다. 남자의 경우 폭로군의 요중 평균 카드뮴농도는 $2.56{\mu}g/\ell,\;2.80{\mu}g/g$ creatinine 및 $2.50{\mu}g/S.G.$, 대조군은 $1.19{\mu}g/\ell,\;1.36{\mu}g/g$ creatinine 및 $1.17{\mu}g/S.G.$ 였고, 여자에서는 폭로군 $2.69{\mu}g/\ell,\;3.94{\mu}g/g$ creatinine 및 $2.63{\mu}g/S.G.$, 대조군 $1.27{\mu}g/\ell,\;1.97{\mu}g/g$ creatinine 및 $1.25{\mu}g/S.G.$로서 폭로군이 대조군에 비해 남 여 모두에서 유의하게 높았다. 연령에 따른 요중 평균 카드뮴 농도는 남자의 경우 $20\sim39$세군에서는 폭로군과 대조군간에 차이가 없었으나, $40\sim59$세군과 60세 이상군에서는 폭로군이 유의하게 높았다. 여자에서는 폭로군이 대조군에 비해 모든 연령층에서 유의하게 높았다. 또한, 폭로군에 있어서 조사대상 지역에 거주한 기간이 길수록 그리고 이 지역에서 생산된 쌀과 채소류를 섭취하는 사람들의 요중 카드뮴 농도가 높게 나타나, 폐광 인근 지역주민들이 미량이지만 상대적으로 많은 양의 카드뮴에 만성적으로 폭로되고 있는 것으로 추정되었다. 폭로군과 대조군의 요중 평균 NAG활성도, $\beta_2-MG$ 배설량 및 총단백질량은 두 군간에 유의한 차이가 없었다. 그러나, 폭로군에서 요중 NAG활성도와 총단백질량은 요중 카드뮴량과 통계적으로 유의한 정상관이 있었으나 $\beta_2-MG$ 배설량은 유의하지 않았다. 또한 2.0 미만군, 2.0이상 10.0미만군, 10.0이상군으로 구분한 요중 카드뮴 농도수준에 따른 분석시 요중 카드뮴 2.0미만군에 비해 10.0이상군의 NAG활성도, $\beta_2-MG$배설량 및 총단백량이 유의하게 증가되었으며, NAG활성도는 2.0이상 10.0미만군에서도 유의하게 증가되었다. 즉, 폭로군에서 요중 카드뮴 농도에 따른 요중 NAG활성도의 증가가 $\beta_2-MG$배설량의 증가에 비해 상대적으로 민감하게 나타나, 향후 카드뮴 폭로군에 대한 색출검사시 $\beta_2-MG$배설량과 함께 NAG활성도를 조사함으로써 조기진단적 가치가 높을 것으로 사료된다.

가공 및 외식식품 중 면류 및 제빵류의 레티놀 및 베타카로틴 함량 조사 (Determination of β-Carotene and Retinol in Korean Noodles and Bread Products)

  • 본 연구에서는 6개의 지역(강원도, 경기도, 경상도, 서울, 전라도, 충청도)에서 채취한 가공 및 외식식품 38종에 함유되어 있는 ${\beta}$-carotene 및 retinol의 함량을 조사하였다. 이들 지용성 비타민들은 알칼리를 이용한 직접 검화법과 용매추출법을 병행하여 수행하였다. 면류 식품 14종 중에서 ${\beta}$-carotene 함량은 비빔국수와 쫄면에서 각각 평균 442.43 ${\mu}g/100g$과 301.39 ${\mu}g/100g$이 검출되며 가장 높은 함량을 나타내었다. 비타민 A(${\mu}gRE$) 함량은 비빔국수와 회냉면, 쫄면, 비빔냉면이 각각 77.5 RE와 61.3 RE, 56.4 RE, 53.2 RE이었다. 만두류 식품 4종에서는 김치만두에서 ${\beta}$-carotene 197.64 ${\mu}g/100g$으로 가장 높게 검출되었으며, 비타민 A(${\mu}gRE$) 함량은 33.3 RE이었다. 떡류 제품 12종에서는 매운 떡볶이와 모듬찰떡에서 ${\beta}$-carotene 함량이 각각 평균 205.11 ${\mu}g/100g$과 41.33 ${\mu}g/100g$으로 검출되었으며, retinol은 매운 떡볶이(1.65~10.45 ${\mu}g/100g$)에서만 검출되었다. 매운 떡볶이와 모듬찰떡의 경우, 비타민 A(${\mu}gRE$)의 함량은 각각 39.67 RE와 6.89 RE로 환산하였다. 이는 하루 필요한 비타민 A(700 ${\mu}gRE$)의 1~6%에 해당하는 함량이다. 제빵류 8종에서는 ${\beta}$-carotene 함량이 야채고로케(110.43~192.29 ${\mu}g/100g$), 햄&치즈샌드위치(33.07~116.57 ${\mu}g/100g$), 페이스트리빵(33.69~104.87 ${\mu}g/100g$), 마늘빵(빵집, 33.88~81.21 ${\mu}g/100g$), 버터크림빵(빵집, 31.61~85.84 ${\mu}g/100g$)에서 높게 검출되었다. 비타민 A(${\mu}gRE$)의 함량은 제빵류 중에서 페이스트리빵과 버터크림빵(빵집), 모닝빵들이 각각 77.3 RE와 51.2 RE, 41.4 RE를 나타내며 가장 높은 함량을 보였다.
