미토콘드리아에서 생성하는 ATP는 미토콘드리아의 속막에 존재하는 전자전달계 효소(electron transferase)에 의해 생성되며, 이러한 전자전달계 효소는 복합체 I, II, III, IV, V로 구성되어 있다고 알려져 있다. ATP는 ATPase에 의해 생성되며, ATPase는 $F_0$와 $F_1$ 소복합체로 구성되어 있다. 미토콘드리아의 외막에는 Porin 또는 VDAC(voltage-dependent anion-selective channel)이라고 알려져 있는 미세한 구멍 형태의 단백질이 존재하며, 세포질에 존재하는 succinate, malate, ATP와 같은 음전하용질 또는 전자를 선택적으로 통과시키는 역할을 수행하는 것으로 보고된 바 있다. 본 연구에서는 소의 심근 미토콘드리아에 존재하고 있는 porin과 ATPase의 기능과 분포의 관계를 알아보기 위하여, porin과 ATPase Ⅴ-${\beta}$ 항체를 면역반응법을 이용한 광학현미경과 이중면역반응법을 이용한 형광현미경으로 확인하고, 심근 미토콘드리아의 두 단백질 분포를 면역황금표지법을 이용한 전자현미경으로 관찰하였다. 미토콘드리아에서 porin 항체에 대한 미토콘드리아 조직항원의 발색은 조직내에서 전반적으로 관찰할 수 있었으며, ATPase 항체에 대한 조직항원의 발색은 세로면에서 관찰되었다. 이중면역응법에서 porin 항체와 ATPase는 각각 다른 조직에서 발색이 관찰되거나, 같은 조직 내에서 관찰되었다. 면역황금표지법에서 porin 항체는 미토콘드리아의 바깥막에서 황금입자가 표지된 것을 확인할 수 있었으며, ATPase는 미토콘드리아의 속막에서 황금입자가 표지된 것을 확인할 수 있었다. 그러나 ATPase 항체가 황금입자로 표지되지 않은 미토콘드리아도 확인되었다. 이러한 결과로 porin 항체와 ATPase 항체는 미토콘드리아의 바깥막과 속막에 각각 분포양상을 확인하였다. porin 항체의 발색으로 인한 조직 내의 미토콘드리아가 존재하고 있음을 확인할 수 있었으며, ATPase 항체의 발색으로 인한 ATP를 생성하는 미토콘드리아를 확인할 수 있었다. 하지만 porin 항체의 반응으로 확인된 미토콘드리아가 반드시 ATP를 생성하는 것은 아니라는 것을 추측할 수 있었다.
The present investigation was initially undertaken to see if there exists $Na^+-K^+$ activated ATPase in the microsome fraction of the kidney. Having confirmed the presence of such an enzyme, further attempts have been made to characterize its nature and the following conclusions were obtained: (1) The ATPase activity was greatest at the $Na^+$ concentration of 100 mM as well as at $K^+$ concentration of 10 mM. Moreover, the ATPase activity was found to be depressed by $Ca^{++}$ in the presence of $Mg^{++}$. (2) While the ATPase activity was depressed by Ouabain, the magnitude of inhibition was greater in the Na medium than in the K medium. (3) NaCN augmented the ATPase activity whereas NaF and IAA depressed it. On the other hand, DNP had little influence on the ATPase activity. (4) Diamox, vasopressin and aldosterone had no effect while $HgCl_2$ markedly depressed the ATPase activity These findings indicate that the nature of ATPase isolated from the microsome fraction of the rabbit kidney is quite similar to that from other organs such as the heart and the muscle, although there are certain features specific to the type of organs.
표고버섯 중의 광감응성 mitochondrial ATP synthase는 0.1 mM $Fe^{2+}$ 단독 이온에 의하여 그 활성이 대조구에 비해 102%, 증가되었으며, 반면 $Fe^{3+}$ 및 $Mg^{2+}$ 이온은 효소의 활성을 억제시켰다. 0.5 mM $Mg^{2+}$ 존재하에서 0.1 mM $Fe^{2+}$ 이온에 의한 이 효소의 활성은 32% 증가되었으며 0.5 mM $Mg^{2+}$ 존재하에서 $Fe^{3+}$ 이온효과는 단독 $Fe^{3+}$ 이온의 효과와 유사한 경향으로 효소의 활성을 저해하였다. 0.5 mM $Mg^{2+}$과 0.1 mM, 0.5 mM 및 1.0 mM $Fe^{3+}$ 이온의 공존하에서$Fe^{2+}$ 이온에 의한 효소의 활성은 모두 억제되었으며, 특히 0.5 mM $Mg^{2+}$과 0.1 mM $Fe^{3+}$ 이온의 공존하에서 5.0 mM $Fe^{2+}$ 이온에 의하여 53%의 억제현상을 나타내었다. 따라서 표고버섯 중의 광감응성 mitochondrial ATP synthase의 활성은 $Fe^{2+}$ 이온에 의하여 특이적으로 크게 증가되며, 이 효소에 대한 $Fe^{2+}$ 이온의 활성화 효과가 $Mg^{2+}$ 이온에 의하여 크게 영항을 받지 않으나, $Fe^{3+}$ 이온의 공존하에서는 억제됨을 알았다. 활성화 금속이온인 $Fe^{2+}$ 존재하에서 이 효소의 최적 pH는 7.6이며, 최적 온도는 $63^{\circ}C$이었다. 또한 이 효소는 금속 chelating agent인 EDTA에 의하여 효소의 활성이 상실됨으로써 metalloenzyme의 가능성을 제시하였다.
치아우식의 주원인균인 Streptococcus mutans (S. mutans)는 산 생성 뿐 아니라 산에 대한 탁월한 저항성을 나타낸다. 본 연구에서는 S. mutans가 pH stress에 노출될 때 형질막 유동성, F-ATPase 활성과 발현 및 양성자 투과성 변화와 그 상관관계를 규명하였다. S. mutans로부터 형질막을 분리한 후 1,6-diphenyl-1,3,5-hexatriene을 사용하여 pH stress가 형질막 유동성 변화에 미치는 영향을 측정하였다. pH 4.8과 pH 8.8에서 배양한 S. mutans는 pH 6.8에서 배양한 S. mutans에 비하여 형질막 유동성이 감소되었다. F-ATPase 활성과 발현은 pH 4.8에서 가장 높았고, pH 8.8에서 가장 낮았다. 양성자 투과성은 pH 4.8과 pH 8.8에서 모두 감소되었으며, 특히 pH 4.8에서의 감소가 컸다. F-ATPase 활성만으로 양성자 투과성이 결정된다면 pH 8.8에서 가장 높아야 하나 pH 6.8보다 감소하는 것은 형질막 유동성 감소에 기인된 양성자 세포내 유입 감소와 관련된 것으로 추정한다. 또한 pH 4.8에서 양성자 투과성이 아주 낮은 것은 높은 F-ATPase 활성에 의한 양성자 세포외 유출 증가 뿐 아니라 형질막 유동성 감소에 의한 양성자 세포내 유입 감소에 기인된 것으로 추정한다. 따라서 pH stress에 의한 형질막 유동성 감소는 S. mutans가 세포내 pH 를 유지하는데 중요한 역할을 하는 것으로 생각되며 에탄올을 포함하여 비특이적으로 세포막 유동성을 증가시키는 약물들은 항우식제에 활용될 수 있을 것으로 추정한다.
It has been reported that the luteal function may be regulated by the intracellular $Ca^{++}$ level which may be adjusted partially by the high affinity $Ca^{++}-ATPase$ in luteal cell membranes. Then, one may expect that luteotropic and/or luteolytic agents, such as gonadotropins, prostaglandin $F_{2{\alpha}}\;(PGF_{2{\alpha}})$ and ouabain, affect the intracellular $Ca^{++}$ level. In this present study, therefore, we examined the effects of luteinizing hormone (LH, or human chorionic gonadotropin, hCG), $PGF_{2{\alpha}}$ and ouabain on the kinetic properties of the high affinity $Ca^{++}-ATPase$ in light membrane, heavy membrane, and microsomal fractions from the highly luteinized ovary. LH (or hCG) increased the affinity and the Vmax for $Ca^{++}$ both in light membrane and heavy membrane. $PGF_{2{\alpha}}$ increased the Vmax in light membrane and decreased the Km in heavy membrane for $Ca^{++}$ at low concentration $(5\;{\mu}g/ml)$. At higher concentration, however, $PGF_{2{\alpha}}$ oppositly affected on kinetic properties, that shown at low concentration. Ouabain, a potent inhibitor of $Na^+-K^+-ATPase$, increased the Km at high concentration $(10^{-4}\;M)$, however, decreased the Vmax for $Ca^{++}$ in light membrane at low concentration $(10^{-6}\;M)$. Also, ouabain increased the Km for $Ca^{++}$ in heavy membrane without changes in the Vmax at both concentrations. It seems that LH and low dose of $PGF_{2{\alpha}}$ increase the intracellular $Ca^{++}$ level and cause in activation of $Ca^{++}-ATPase$, however, higher dose of $PGF_{2{\alpha}}$ and ouabain inhibit directly $Ca^{++}-ATPase$ activity and result in increase in intracellular $Ca^{++}$ level. According to the above results, we suggest that luteotropic and/or luteolytic agents regulate the luteal progesterone $(P_4)$ production through two different pathways; one is cyclic adenosine monophosphate (cAMP)-dependent and another is $Ca^{++}-dependent$. Intracellula. $Ca^{++}$ level regulated by the high affinity $Ca^{++}-ATPase$ may affect both pathways in a time-dependent fashion. LH (or hCG) acts on the luteal $P_4$ production via both pathways. The initial step is $Ca^{++}$ dependent, and the late step is cAMP dependent. $PGF_{2{\alpha}}$ and ouabain increase the intracellular $Ca^{++}$ concentration so that basal luteal $P_4$ production is increased and LH-stimulated $P_4$ production is inhibited by the inhibiting LH-dependent adenylate cyclase activity.
