  • This study was conducted to get basic information for factors affecting protoplast fusion between Petunia hybrida 'Titan red' and Nicotiana sanderae. The experiments such as fusogen, time of PEG treatment, temperature at fusion, $CaCl_2{\cdot}2H_2O$ concentration in fusion solution, and $CaCl_2{\cdot}2H_2O$ concentration and pH in eluting solution were carried out to increase the fusion efficiency. The results obtained were as follows; Fusion between P. hybrida and N. sanderae was accelerated when the mixture of the protoplasts was treated with 30% PEG 6,000 solution containing 5.5 mM $CaCl_2{\cdot}2H_2O$ for 10 minutes at $25^{\circ}C$, and subsequently eluted with a eluting solution containing 50 mM $CaCl_2{\cdot}2H_2O$ adjusted to pH 9.0.

Combined Effect of Agrimonia pilosa Ledebour Extract and NaCl for Control of Escherichia coli O157:H7 (Escherichia coli O157:H7의 제어를 위한 선학초(Agrimonia pilosa Ledebour) 추출물과 NaCl의 병용효과)

  • Gamma irradiated and non-irradiated Agrimonia pilosa Ledebour were extracted by 70% ethanol. The combined effects of the Agrimonia pilosa Ledebour extract and NaCl on survival of Escherichia coli O157:H7 in tryptic soy broth were investigated. E. coli O157:H7 decreased ca 1 log cycle by the addition of 2% sample extract, and the anthbacterial activity was increased as the concentration of sample extract was increased. The irradiation effect of the sample on antibacterial activity was not observed. On the treatment of NaCl alone, E. coli O157:H7 was inactivated (ca 3~4 log cycle reduction within 48 hr) in more than 7% NaCl. The higher inactivation(ca 5 log cycle reduction within 48 hr) occurred in the presence of 2% sample extract and 5% NaCl than in the addition of each alone. The extracted antibacterial substance was stable in the pH range of 4.0 to 7.0, heat treatment at 121$^\circ C$ for 15 min, and freezing at -18$^\circ C$ and thawing at 37$^\circ C$. There fore, the sample extract, would substantially increase the food-safety in terms of E. coli O157:H7.

Removal Characteristic of Acidic Ion in Aqueous Solution by Alumina (알루미나에 의한 수용액 중의 산성이온 제거 특성)

  • The removal characteristics of ionic species, such as $Ca^{2+}$, $Mg^{2+}$, $Sr^{2+}$, $SO{_4}^{2-}$, $NO{_3}^-$, and $Cl^-$ by adsorption on the alumina were investigated. Alumina precusor powders were prepared from $Al(NO_3)_3{\cdot}9H_2O$ and $NH_4OH$. Alumina materials prepared from the heat treatment in a furnace at $450{\sim}750^{\circ}C$ for 5 h were analysed using FT-IR and the Brunauer-Emmett-Teller (BET) method. The specific surface area of the product particles decreased significantly with treatment temperature. The adsorption capacities of $SO{_4}^{2-}$and $NO{_3}^-$ on alumina were 23 mg/g and 12.4 mg/g, respectively. But, removal efficiencies of $Cl^-$ were less than 4 mg/g. In general, the removal efficiencies of the anion species were decreased with increasing treatment temperature. The best anion removal efficiency was obtained when the alumina was treated under $450^{\circ}C$. Removal efficiencies of $Ca^{2+}$, $Mg^{2+}$, and $Sr^{2+}$ were increased with increasing treatment temperature.

Study on the Induction of Antlerogenesis by Calcium Chloride Treatment in Female Elk Deer (염화칼슘 처리에 의한 엘크 암사슴의 뿔 발생 유도에 관한 연구)

  • To investigate the effects of CaCl2 on artificial induction of antlerogenesis in female deer, various CaCl2 conditions with different concentrations, dosages and times were treated to five female Elk(Cervua Canadensis). After injection of CaCl2 solution to the putative region of antlerogenesis, rate of induction, yield of antler, length of antler, number of point were examined. In regard to the effect of concentration of CaCl2 solution on the induction of antlerogenesis in the female deer, generation of pedicle and antler has not induced under treatment of 2ml and 3ml of 15% solution, but only pedicle has generated with 4ml of 15% solution, otherwise generation of pedicle and antler has induced under treatment of 1.5ml, 2ml and 3ml of 30% solution and 1ml and 2ml of 50% solution. The yields of antler were 319g in 4ml of 15% solution, and 1,290g, 513g and 295g in 1.5ml, 2ml and 3ml of 30% solution, respectively, and 800g and 443g in 1ml and 2ml of 50% solution, respectively. The yields of antler of 30% solution and 50% solution were decreased with increase the dosage volume. The maximum yield of antler was 1,290g at 30% 1.5ml treatment. The length of antler were 25cm in 15% 4ml treatment, and 55cm, 51cm and 35㎝ in 1.5ml, 2ml and 3ml of 30% solution, respectively, and 60cm and 35cm in 1ml and 2ml 50% solution, respectively. There was a tendency that length of antler became longer as yield of antler were grew. The numbers of point were 2 in 15% 4ml treatment, and 5, 2.3 and 1 in 1.5ml, 2ml and 3ml of 30% solution, respectively, and 3 and 1 in 1ml and 2ml of 50% solution, respectively. The number of point was not related to concentration and dosage of CaCl2 solution, but related to the shape of wound due to the time and method of injection. Consequently, the optimum concentration of CaCl2 solution for artificial induction of antlerogenesis in female elk is 30%.

A Study on Synthesis of Ca and Mg Compounds from Dolomite with Salt Additional React (MgCl2·6H2O) (염 첨가 반응(MgCl2·6H2O)을 이용하여 백운석으로부터 Ca 화합물과 Mg 화합물 합성에 관한 연구)

  • In order to utilize dolomite as a calcium/magnesium compound material, it was prepared highly reactive calcined dolomite(CaO·MgO) using a microwave kiln (950 ℃, 60 min). The experiment was performed according to the standard of the hydration test (ASTM C 110) and hydration reactivity was analyzed as medium reactivity (max 74.1 ℃, 5 min). Experiments were performed with calcined dolomite and salt (MgCl2·6H2O) (a) 1:1, (b) 1:1.5, and (c) 1:2 wt% based on the hydration reaction of calcined dolomite. The result of X-ray diffraction analysis confirmed that MgO of calcined dolomite increased to Mg(OH)2 as the salt addition ratio increased. After the separating reaction, calcium was stirred at 80 ℃, 24 hr that produced CaCl2 of white crystal. XRD results, it was confirmed calcium chloride hydrate (CaCl2·(H2O)x) and CaO of calcined dolomite and salt additional reaction was separated into CaCl2. And it was synthesized with Ca(OH)2 99 wt% by NaOH adding reaction to the CaCl2 solution, and the synthesized Ca(OH)2 was manufactured CaO through the heat treatment process. In order to prepare calcium carbonate, CaCO3 was synthesized by adding Na2CO3 to CaCl2 solution, and the shape was analyzed in cubic form with a purity of 99 wt%.

Combined Effects of Postharvest Calcium Chloride and Heat Treatment on the Quality Characteristics of Fresh-Cut Tsugaru Apple

  • To produce fresh-cut fruit products of high quality, the combined effects of postharvest calcium and heat treatment on Tsugaru apple were examined. Whole fruits were treated for 20 min at three different conditions: dipping in 3% $CaCl_2$ solution at $45^{\circ}C$ or ambient temperature, or in heated water at $45^{\circ}C$. The calcium content of the apples dipped in $CaCl_2$ solution at $45^{\circ}C$ was higher than that of the control and the non-heated calcium dipping. The fruits with calcium and heat treatment showed the same pattern of respiration rates as the control did during storage. The browning degree of the apple slices increased to approximately 3% after one-day storage, but no significant difference was observed between the treated and untreated apples. The fruits with $CaCl_2$ solution dipping at $45^{\circ}C$ had higher firmness than those with calcium dipping or heat treatment alone.

Preparation and Photoluminescent Properties of Ca2PO4Cl Activated by Divalent Europium

  • Divalent europium-activated $Ca_2PO_4Cl$ phosphor powders were prepared by a chemical synthetic method followed by heat treatment in reduced atmosphere, and the crystal structures, morphologies and photoluminescence properties of the powders were investigated by x-ray powder diffraction, scanning electron microscope and spectrometer. The effect of Ca/P mole ratio at the starting materials on the final products was evaluated. The optimized synthesis condition obtained in this study was Ca/P mole ratio of 2.0. The present phosphor materials had higher photoluminescence intensity and better color purity than the commercial blue phosphor powders, $(Ca,Ba,Sr)_{10}(PO_4)_6Cl_2:Eu^{2+}$. The result of excitation spectrum measurement indicated that the excitation efficiency of the synthesized powders was higher for the long-wavelength UV region than that of the commercial phosphor. It was thus concluded that the samples prepared in this study can be successfully applied for the light-emitting devices such as LED excited with long-wavelength UV light sources.

Effect of Heat Treatment and Salts Addition on Dongchimi Fermentation (열처리와 염의 첨가가 동치미 발효에 미치는 영향)

  • A short time microwave heat treatment, bringing in hot NaCl solution, addition of KCI, CaCl2, MgCl2 into brining solution and salts mixture of phosphates into half fermented dongchim were investigated for their effects on some quality of dongchimi, a Korean pickle of Chinese radish roots, during fermentation. The reference dongchimi was prepared by brining the radish roots in 7% NaCl added with seasonings at $25^{\circ}C$. The result showed that microwave heat treatment affected little on the pH or total acidity change during fermentation. Soaking the roots in 80-9$0^{\circ}C$ hot salt solution significantly reduced the fermentation and softening rate of dongchimi while a rather rapid fementation was found for those soaked in 7$0^{\circ}C$. Addition of KCI and CaCl2 into brining solution slowed the pH decrease and softening of the roots, respectively. The dangchimi added with the salts mixture of phosphate, citrate and nitrite was significantly extreneded the fermentation time to pH 4.0 by more than two folds.

Effects of Preparation Conditions in the Spray Pyrolysis on the Characteristics of Ca8Mg(SiO4)4Cl2:Eu2+ Phosphor (분무열분해 공정의 제조 조건이 Ca8Mg(SiO4)4Cl2:Eu2+ 형광체 특성에 미치는 영향)

  • In spray pyrolysis, the effects of the preparation temperature, flow rate of the carrier gas and concentration of the spray solution on characteristics such as the morphology, size, and emission intensity of $Ca_8Mg(SiO_4)_4Cl_2:Eu^{2+}$ phosphor powders under long-wavelength ultraviolet light were investigated. The phosphor powders obtained post-treatment had a range of micron sizes with regular morphologies. However, the composition, crystal structure and photoluminescence intensity of the phosphor powders were affected by the preparation conditions of the precursor powders. The $Ca_8Mg(SiO_4)_4Cl_2:Eu^{2+}$ phosphor powders prepared at temperatures that were lower and higher than $700^{\circ}C$ had low photoluminescence intensities due to deficiencies related to the of Cl component. The phosphor powders with the deficient Cl component had impurity peaks of $Ca_2SiO_4$. The optimum flow rates of the carrier gas in the preparation of the $Ca_8Mg(SiO_4)_4Cl_2:Eu^{2+}$ phosphor powders with high photoluminescence intensities and regular morphologies were between 40 and 60 l/minute. Phosphor powders prepared from a spray solution above 0.5 M had regular morphologies and high photoluminescence intensities.

Preparation and Characterization of ClO2 Self-Releasing Smart Sachet (이산화염소 자체 방출 스마트 샤쉐의 제조 및 특성 연구)

  • Chlorine dioxide (ClO2) is widely used for post-harvest sterilization treatment. However, there are limitations in the retail application of ClO2 due to difficulties in handling, expensive facilities, and safety concerns. Therefore, it is necessary to develop a ClO2 technology that can be easily applied and continuously released for a long period. In this study, a series of ClO2 self-releasing sachets were developed. First, poly(ether-block-amide) (PEBAX) and polyethylene-glycol (PEG) composite films containing different ratios of citric acid (CA) were prepared using the solution casting method. The as-prepared PEBAX/PEG-CA composite films were evaluated using FT-IR, DSC, and TGA to confirm chemical structure and thermal properties. Subsequently, PEBAX/PEG-CA composite films were designed in the form of a sachet and NaClO2 powder was transferred into the sachet to achieve a ClO2 self-releasing system. The ClO2-releasing behavior of the sachet was investigated by measuring the release amount of the gas using UV-vis. The release amount of ClO2 increased with increasing CA contents owing to the existence of higher protons (trigger) in the polymer matrix. Further, ClO2 gas was released for a longer time. Therefore, the as-prepared smart sachet can be tuned according to applications and packaging sizes to serve an optimal sterilization effect.
