Studies on the Preparation of Fruit Bases for Mixed Yoghurt -Changes in Texture of Fruit Fresh during Heat Treatment- (혼합(混合) 요구르트 과실기제(果實基製)의 가공(加工)에 관한 연구(硏究) -열처리과정중(熱處理過程中) 과실(果實)의 조질변화(組質變化)에 대(對)하여-)

  • This experiment was carried out to study on methods of processing the fruit base which is to be used for preparing the fruit yoghurts. Common fruits were compared and peach was chosen among them for this purpose by determing the rate of decrease in hardness during the process of cooking. The maturity, temperature of cooking and methods of peeling were tested to maintain the desirable hardness during the process, and the effects of metallic ion addition on the hardness of the flesh were also studied. The results obtained were as follows. 1. The peaches were keeping the most stable texture during cooking, among common fruits, however the hardness of apples were decreased markedly. Rate of decreases in the flesh hardness were high during the initial stages of cooking then slowered in all fruits tested. 2. Flesh hardness of the peaches were largely affected by the temperature of cooking. And the texture of freeze-stored fruits was severly damaged during thawing, so that could be used only for the limited purposes such as for stirred-type yoghurt. 3. Divalent matallic ions, especially calcium ion, were effective for supporting the flesh hardness of peaches during cooking. 4. The optimal concentration of calcium chloride addition was 0.3%, and this resulted in its final concentration in the product under the legal dosage without any subsquent changes in colour and flavor. 5. The effects of calcium chloride addition were greater in the froms of dices than in those of slices, and in overmature fleshs than in immature ones. 6. Treatment of calcium chloride by soaking the slices or dices of lye-peeled peaches in 0.3% solution was found to be adaptable for the practical process.

Isolation and Characterization of Complement System Activating Polysaccharides from the Hot Water Extract of the Leaves of Capsicum annuum L. (고추잎 열수추출물로부터 보체계 활성화 다당의 분리 및 특성)

  • It was observed that the hot water extract of the leaves of Capsicum annuum L., a Korean edible plant, had a potent anti-complementary activity. Crude polysaccharide fraction(CAL-0) was obtained by methanol reflux, ethanol precipitation, dialysis and lyophilization. CAL-0 contained 51.8% of total sugar, 8.2% of uronic acid and 16.8% of protein, and consisted of mainly arabinose, galactose and glucose as neutral sugars and galacturonic acid as uronic acid. The anti-complementary activity of CAL-0 decreased greatly by periodate oxidation, but was not changed by pronase treatment. Also, the anti-complementary activity of CAL-0 was reduced partially in the absence of the $Ca^{2+}$ ion. The crude polysaccharide CAL-0 was found to activate the C3 component both in the presence and in the absence of $Ca^{2+}$ through the crossed-immunoelectrophoresis suggesting that those involved in both classical and alternative complement pathway CAL-0 was further separated to an unabsorbed fraction(CAL-1) and six absorbed fractions(CAL-2longrightarrowCAL-7) on DEAE Sepharose CL-6B ion exchange column. Among them four major fractions in activity and yield were obtained, and consisted mainly of arabinose, galactose and glucose with various molar ratios. The major fraction, CAL-2, was purified to give a high molecular fraction(CAL-2-I) and a low molecular fraction(CAL-2-II) on Sepharose CL-6B column. The anti-complementary activity of CAL-2-I, a molecular weight of about 61,000, was higher than it of CAL-2-II.-II.

A Preponderant Enhancement of Conversion Efficiency by Surface Coating of $SnO_2$ Nanoparticles in Organic MK-2 Dye Sensitized Solar Cell

  • Nanocrystalline $SnO_2$ colloids are synthesized by hydrolysis of $SnCl_4{\cdot}5H_2O$ in aqueous ammonia solution. The synthesized $SnO_2$ nanoparticles with ca. 15 nm in diameter are coated on a fluorinedoped thin oxide (FTO) conductive substrate and heated at $550^{\circ}C$. The annealed $SnO_2$ film is treated with aqueous $TiCl_4$ solution, which is sensitzied with MK-2 dye (2-cyano-3-[5'''-(9-ethyl- 9H-carbazol-3-yl)-3',3'',3''',4-tetra-n-hexyl-[2,2',5',2'',5'',2''']-quater thiophen-5-yl]). Compared to bare $SnO_2$ film, the conversion efficiency is significantly improved from 0.22% to 3.13% after surface treatment of $SnO_2$ with $TiCl_4$, which is mainly due to the large increases in both photocurrent density from 1.33 to $9.46mA/cm^2$ and voltage from 315 to 634 mV. It is noted that little change in the amount of the adsorbed dye is detected from 1.21 for the bare $SnO_2$ to $1.28{\mu}mol/cm^2$ for the $TiCl_{4-}$ treated $SnO_2$. This indicates that the photocurrent density increased by more than 6 times is not closely related to the dye loading concentration. From the photocurrent and voltage transient spectroscopic studies, electron life time increases by about 13 order of magnitude, whereas electron diffusion coefficient decreases by about 3.6 times after $TiCl_4$ treatment. Slow electron diffusion rate offers sufficient time for regeneration kinetics. As a result, charge collection efficiency of about 40% before $TiCl_4$ treatment is improved to 95% after $TiCl_4$ treatment. The large increase in voltage is due to the significant increase in electron life time, associated with upward shift of fermi energy.

  • Aminoglycosidic antibiotics have multiple effects on muscle. For example, they have been shown to block L-type $Ca^{2+}$ channels in vascular smooth muscle, cardiac muscle and skeletal muscle. Possibly as a consequence of this effect on $Ca^{2+}$ influx, they have been shown to decrease the contractility of cardiac muscle (gentamicin). The present study evaluated the effects of gentamicin on blood pressure, vasorelaxation and left ventricular pressure. Gentamicin(10, 20, 40mg/kg) produced dose-dependent blood pressure lowering in rat. The pretreatment of MgSO$_4$ and imipramine (Na$^{+}$-Mg$^{2+}$ exchange inhibitor) had no effect in gentamicin-induced hypotension. However, the gentamicin-induced hypotension was significantly potentiated in the preincubation of verapamil or nifedipine (L-type $Ca^{2+}$ channel blockers), and was significantly attenuated by CaCl$_2$ and was slightly attenuated by caffeine (phosphodiesterase inhibitor). Gentamicin (10, 30, 100$\mu$g/m1) did not have an effect on relaxation of phenylephrine-precontracted aortic rings but high concentration of gentamicin(100, 300$\mu$g/ml) relaxed KCl-precontracted aortic rings, which relaxation was potentiated by treatment of nifedipine. Whereas gentamicin markedly decreased left ventricular developed pressure (LVDP) in perfused heart. These data suggest that gentamicin has significant blood pressure lowering of the rat, which seems to be mediated by calcium channel-sensitive pathway and blood $Ca^{2+}$ level may be important role in this response.

Increase of Strawberry Fruit Shelf-life through Preharvest Spray of Calcium-chitosan and Post-harvest Treatment with High Pressure CO2 (수확 전후 칼슘-키토산 및 고농도 CO2 단기처리에 의한 '설향' 딸기 과실의 저장성 증진)

  • This experiment examined the effects of preharvest Ca-chitosan spraying and short-term post-harvest treatment with high $pCO_2$ on the shelf-life of 'Seolhyang' strawberry fruit. Fruit firmness decreased when harvested late in the season and was little affected by Ca-chitosan spray treatments. Short-term treatment with high $pCO_2$ after harvest, however, significantly increased fruit firmness regardless of preharvest treatment. The combined treatment of Ca-chitosan spray and high $pCO_2$ led to an additional increase in shelf-life through delay of skin disorder, fruit decay incidence and firmness loss during simulated marketing. The residual effects of both treatments, however, were reduced when fruit were harvested late in the season. Fruit firmness was more affected by high $pCO_2$ treatment whereas decay and visual quality were affected by preharvest spraying with Ca-chitosan. Although more research is required to determine the optimum concentration of calcium and chitosan, and the optimal spray interval, our results show that the combined treatment of preharvest Ca-chitosan spray and postharvest high $pCO_2$ is effective in improving strawberry fruit shelf-life by increasing firmness and delaying decay.

Physiological Response of Chinese Cabbage to Salt Stress (염 스트레스에 대한 배추의 생리학적 반응)

  • In order to understand the plant responses to salt stress (0, 50, and 100 mM NaCl), Chinese cabbage seedlings grown up to two leaf stages by hydroponic culture were used. Fresh and dry weight, chlorophyll (Chl), antioxidant materials, polyamine content, antioxidant enzyme activities, and inorganic ion level were evaluated. Fresh and dry weights of Chinese cabbage increased with the increase in salinity while the optimal growth occurred at 50 mM NaCl. The Chl a, total Chl, carotenoid content, and Chl a/b ratio increased by the 6 days after treatment with 100 mM NaCI; however, the Chl b content decreased. Glutathione increased in the root of Chinese cabbage for 6 days. Dehydroascorbate increased remarkably by day 6 caused by the salt stress in both leaf and the root. While ascorbate peroxidase increased, the activity of catalase and glutathione reductase decreased gradually in the first leaf for 6 days. The $Na^+$ content increased by 12.5-fold in the 3 days after treatment with 100 mM NaCI in the shoot, whereas the $Ca^{2+}$, $K^+$, and $Mg^{2+}$ content measured in the same treatment decreased by 43 to 57%. Spermidine content decreased as salinity increased, but spermine content increased. The growth promotion, glutathione and ascorbic acid content in Chinese cabbage were increased by low salt stress, and shortening of the cultivation period for growth increase of Chinese cabbage is expected.

The Effects of Hot Water Extraction of Wood Meal and the Addition of CaCl2 on Bending Strength and Swelling Ratio of Wood-Cement Board (목질(木質)의 열수추출(熱水抽出) 및 CaCl2 첨가(添加)가 목질(木質)-세멘트 보드의 휨강도(强度) 및 팽윤율(膨潤率)에 미치는 영향(影響))

  • The effects of pre-treatments, the hot water extraction of wood meal and the addition of chemical ($CaCl_2$) to wood-cement water system on the properties of wood-cement composite such as modulus of rupture (MOR), modulus of elasticity (MOE), water sorption ratio and swelling ratio of resulting boards were studied in this experiment. The wood meals through 0.83mm(20 mesh) and retained on 0.42mm(35 mesh) screen were prepared from Pinus densiflora S. at Z. and Larix leptolepsis G. For hot water extraction, 500 grams of wood meal for each species were heated to boiling with 1,500ml of distilled water in 2-liter beaker for 6 hours. Every 2 hours, the wood meals were washed with boiling distil1ed water and reheated to boiling again. After 6 hours boiling, the boiled wood particles were collected by pouring this particles on 200 mesh screen. The collected particles then washed twice with hot distilled water and dried for 24 hours in an oven at $109{\pm}20^{\circ}C$. A mixture of 663.4 grams of cement with 331.7 grams of wood meal based on oven-dry weight were dry-mixed in a plastic vessel. The mixture was kneaded with 497.6ml of distilled water in the ratio of 1.5ml of water to a gram of wood meal. To add calcium chloride to the mixture as an accelerator, $CaCl_2$ 4% solution by weight per volume, was added to pine-or larch-cement board in the ratio of 3% to cement weight. To set wood-cement board, this mixture was clamped at 30cm ${\times}$ 30cm, in thickness of 1.5cm for 3 days at room temperature, declamped and then placed at open condition for 17 days. The target density was 1.0. The four specimens sized to 5cm in width and 28cm in length were used for MOR and MOE test for each treatment. After MOR test, the tested specimens were cut to the size of 5cm ${\times}$ 5cm for water sorption and swelling test. The twenty specimens used to measure the water sorption ratio (soaking 24 hours) and ten of these were used for swelling ratio measurement The results obtained were as follows: 1) Larch was not suitable for wood-cement boards because larch-cement board developed no strength, but pine showed 97.9kg/$cm^2$ by hot water extraction. 2) To increase MOR, hot water extraction was more effective than the addition of $CaCl_2$ in pine and larch because the $CaCl_2$ addition was seemed to speed up the ratio of cement hydration without reacting with the wood substances. 3) The water sorption ratio was lowered by the addition of $CaCl_2$ to wood-cement system because the chemical additive accelerated the rate of cement hydration. 4) In pine-cement board, the swelling ratio from 0.37 to 0.42 percent was observed in length and the swelling ratio from 0.88 to 2.0 percent in thickness. As a rule, the swelling ratio of wood-cement board was very low and the swelling ratio in thickness was higher than in length.

Stabilization of Arsenic in Paddy Soils Using Stabilizers (논토양 내 비소 불용화에 대한 안정화물질의 처리 효과)

  • BACKGROUND: Soil contamination of As is a very sensitive environmental issue due to its adverse impact on human health and different characteristics with other heavy metals. With public awareness of As poisoning, there has been growing interest in developing guideline and remediation technologies for As-contaminated soil. The objective of this research was to evaluate the effectiveness of stabilizing amendments and soil dressing methods on the mobility of As in the contaminated rice paddy soils nearby mining area. METHODS AND RESULTS: Different amendments were mixed with surface and subsurface contaminated soils at a ratio of 3% (w/w) and monitored for five months. Three different extractants including 0.01M $CaCl_2$, TCLP, and PBET were used to examine As bioavailability in the soil and the concentration of As in rice grain was also measured with an inductively coupled plasma (ICP) spectroscopy. The results showed that all amendment treatments decreased As concentration compared to the control. Especially, coal mine drainage sludge (CMDS) treatment showed the highest efficiency of decreasing As concentration in the soil and rice grain. The values of Pearson correlation (r) between As concentrations in the soil and rice grain were 0.782, 0.753, and 0.678 for $CaCl_2$, TCLP, and PBET methods, respectively. Especially, $CaCl_2$ method was highly correlated between As concentrations of the soil and soil solution (r=0.719), followed by TCLP (r=0.706), PBET (r=0.561) methods. CONCLUSION: Stabilizing amendments can effectively reduce available As concentration in the soils as well as soil solution, and thereby potentially mitigating risks of crop contamination by As.

A Field Study on Electrokinetic Removal of Salts from Greenhouse Soil (전기동력학 기술을 이용한 시설재배지 토양 염류제거 실증 연구)

  • A pilot-scale electrokinetic (EK) separation field test ($2{\times}3{\times}0.2m^3$, $W{\times}L{\times}D$) was performed in a greenhouse to remove salts from saline soil. Initially, the greenhouse soil had high electrical conductivity (EC), about 9 dS/m, and contained mainly $Ca^{2+}$, $Cl^-$ and $SO_4^{2-}$ ions. After 2 weeks of EK treatment, the soil EC was reduced to 52% compared with its initial value. The EC reduction was mostly achieved within the first week (47%) due to removal of $Na^+$ and $Cl^-$ ions, but ions with a high adsorption capacity such as $Ca^{2+}$ and $SO_4^{2-}$ ions were difficult to be removed. During the EK test, the soil temperature increased and it reached around $50^{\circ}C$ at some regions. For in situ application to soils in cultivation, the current should be controlled to limit increases in temperature, especially near the cathodes. In conclusion, the in situ EK technique is feasible for the restoration of saline greenhouse soils in or no cultivation and an appropriate strategy is necessary for more effective remediation.

Trends of Thermochemical Technology for the Recovery of Phosphorus from Sewage Sludge Ash (열화학적 방법에 의한 하수 슬러지 소각재에서의 인 회수 기술 동향)

  • Phosphorus (P) is an essential and irreplaceable element for all living organisms, and it is widely used as a fertilizer. Unfortunately, it is estimated that phosphate reservoir is depleted within about 100 years. Sewage sludge ash (SSA) is an alternative resource for P recovery because of its high P content. However, SSA cannot be directly used as a fertilizer due to heavy metals in it and low P bioavailability. Thermochemical treatment with Cl donor is known to reduce heavy metal contents and increase P bioavailability of SSA. Literature review on thermochemical technologies of SSA for the reduction of heavy metals and bioavailability enhancement has been carried out to estimate the status of current P recovery technology and to develop strategic future research plan for P recovery. The review showed that $CaCl_2$ and $MgCl_2$ were the most effective Cl donors and reaction temperature (< $1000^{\circ}C$) was the critical operation condition for the reduction. The removal efficiency depends on the species of heavy metals. Thermochemical technology of SSA for P recovery showed the possibility of commercial application in the near future to overcome the coming crisis of human sustainability by P depletion, but it needs cost effectiveness and more ecofriendly process to reduce energy consumption.
