Effect of Chlorine Dioxide and Commercial Chlorine Sanitizer on Inhibiting Foodborne Pathogens and on Preventing the Formation of Chemically Injured Cells on Radish Sprouts

  • This study assessed the efficacy of aqueous chlorine dioxide ($ClO_2$) and commercial chlorine sanitizer in terms of its ability to eliminate Listeria monocytogenes, Salmonella Typhimurium, and Escherichia coli O157:H7 on radish sprouts (Raphanus sativus L.). Radish sprouts were inoculated with a cocktail containing one each of three strains of three different foodborne pathogens, then treated with distilled water (control) or chemical sanitizers (100 ppm commercial chlorine, and 50, 100, 200 ppm $C1O_2$) for 1, 5, and 10 min at room temperature ($22{\pm}2^{\circ}C$). Populations of S. Typhimurium, E. coli O157:H7 and L. monocytogenes were counted at 4.64, 6.05, and 4.29 log CFU/g, respectively, after inoculation. Treatment with water did not significantly reduce the levels of any of the three foodborne pathogens. The levels of all three pathogens were reduced by treatment with chemical sanitizers; however, the observed levels of reduction of E. coli O157:H7 and L. monocytogenes were not significant as compared with the controls. The levels of the three pathogens were reduced most profoundly when treated for 10 min with 200 ppm of $C1O_2$, and the reduction levels of S. Typhimurium, E. coli O157:H7, and L. monocytogenes were 1.17, 1.63, and 0.96 log CFU/g, respectively. When chemically injured cells were investigated using SPRAB for E. coli O157 :H7 and by selective overlay methods for S. Typhimurium and L. monocytogenes, respectively, it was noted that commercial chlorine sanitizer generated more numbers of injured pathogens than did $C1O_2$. These data indicate that $C1O_2$ treatment may prove useful in reducing the numbers of pathogenic bacteria in radish sprouts.

Biodegradation Characteristics of Toluene in a Soil-Bioreactor (토양생물반응기내 Toluene의 분해 특성)

  • To investigate the optimal conditions for biodegradation of toluene by Pseudomonas fluorescens KCTC 1767 in a batch soil-bioreactor, the effects of rpm change from 60 to 180, and temperature change from $15^{\circ}C$ to $30^{\circ}C$ in a batch culture and the flow rate change from 55 mL/min to 85 mL/Min in soil-bioreactor on the biodegradation of toluene were studied. In a batch culture the optimal operating conditons were 60 rpm, and $30^{\circ}C$ at initial pH 7, In a soil-bioreactor the optimal flow rate was 55 mL/min in the flow rate of circulation. The lower flow rate of circulation may help to biodegrade toluene adsorped in soil and dissolved in underground water.

  • We used $^1H$ nuclear magnetic resonance (NMR) to study the phase transitions and molecular dynamics in a characteristic ferroelectric liquid crystal with a carbon number n = 7, S-2-methylbutyl 4-n-heptyloxybiphenyl-4'-carboxylate (C7). The results were compared with those of our recent work on S-2-methylbutyl 4-n-octanoyloxybiphenyl-4'-carboxylate (C8), with a carbon number n = 8. While the recrystallization and isotropic phase transitions exhibited a first-order nature in the $^1H$ NMR spin-lattice and spin-spin relaxation measurements, a second-order nature was shown at the Sm-A - Sm-$C^*$ liquid crystalline phase transition. A soft-mode anomaly arising from the tilt angle amplitude fluctuation of the director, of which only a hint had been noticed in the C8 system, was manifested in the C7 system at this transition.

  • $NdFe_{10.7}Ti_{1.3}$ has been studied with X-ray diffraction, Mossbauer spectroscopy and vibrating sample magnet-ometer(VSM). The alloys were prepared by arc-melting under an argon atmosphere. The $NdFe_{10.7}Ti_{1.3}$ contains some $\alpha-Fe$, from X-ray and Mossbauer measurements. The $NdFe_{10.7}Ti_{1.3}$ has the $ThMn_{12}$-type tetragonal struc-ture with $a_{0}=8.607{\AA}\;and\;c_{0}=4.790{\AA}$. The Curie temperature ($T_c$) of the $NdFe_{10.7}Ti_{1.3}$ is 590 K from $M\"{o}ssbauer$ spectroscopy performed at various temperatures ranging from 13 to 800 K. Each spectrum below $T_c$ was fitted with six subspectra of Fe sites in the structure$(8i_{1},\;8i_{2},\;8j_{2},\;8j_{1},\;8f\;and\;{\alpha}-Fe)$. The area fractions of the subspectra at room temperature are 13.8%, 15.4%, 17%, 16.4%, 34.1% and 3.3%, respectively. Magenetic hyperfine fields for the Fe sites decrease in the order, $H_{hf}(8i)>H_{hf}(8j)>H_{hf}(8f)$. The abrupt changes in the magnetic hyperfine field, isomer shift and magnetic moment observed at about 180 K in $NdFe_{10.7}Ti_{1.3}$ are attributed to spin reorientation.

Physiological responses on Low Water-temperature Stress of Pacific abalone, Haliotis discus hannai (저수온 스트레스에 의한 북방전복, Haliotis discus hannai의 생리학적 반응)

  • This study was performed to obtain the basic data on physiological responses of low water temperature stress of the cultured Pacific abalone, Haliotis discus hannai. Abalones were exposed at low water temperatures of $7^{\circ}C$ and $4^{\circ}C$. We have investigated survival rate, superoxide dismutase (SOD) activity and total protein (TP) in the abalone by the exposure times (0, 3, 6, 12, 24, 72, 120, 168 and 216 hours). Survival rate of the abalone at $7^{\circ}C$ experiment was 90.8%, whereas at $4^{\circ}C$ experiment was 0% after exposure 10 days. SOD activity was significantly increased until 12 hours after exposure to $4^{\circ}C$, and then was recovered to starting level after 24 hours. However, there was no significant difference between control ($12^{\circ}C$) and $7^{\circ}C$ experiments. TP was significantly increased until 216 hours after 24 hours at $4^{\circ}C$ experiment, but $7^{\circ}C$ experiment showed no significant differences compared to control ($12^{\circ}C$) experiment. Therefore, H. discus hannai was acclimated in low water temperature stress at $7^{\circ}C$, but at $4^{\circ}C$, all abalone died possibly because they exceed the limits of defense ability to too low temperature.

Bioequivalence and Pharmacokinetics of 70 mg Alendronate Sodium Tablets by Measuring Alendronate in Plasma

  • The bioequivalence and pharmacokinetics of alendronate sodium tablets were examined by determining the plasma concentration of alendronate. Two groups, consisting of 24 healthy volunteers, each received a 70 mg reference alendronate sodium tablet and a test tablet in a $2{\times}2$ crossover study. There was a 6-day washout period between doses. The plasma alendronate concentration was monitored for 7 h after the dose, using HPLC-Fluorescence Detector (FD). The area under the plasma concentration-time curve from time 0 to the last sampling time at 7 h $(AUC_{0-7h})$ was calculated using the linear-log trapezoidal rule. The maximum plasma drug concentration $(C_{max})$ and the time to reach $C_{max}(T_{max})$ were derived from the plasma concentration-time data. Analysis of variance was performed using logarithmically transformed $AUC_{0-7h}\;and\;C_{max}$, and untransformed $T_{max}$. For the test medication versus the reference medication, the $AUC_{0-7h}\;were\;87.63{\pm}29.27\;vs.\;102.44{\pm}69.96ng\;h\;mL^{-1}$ and the $C_{max}$ values were $34.29{\pm}13.77\;vs.\;38.47{\pm}24.39ng\;mL^{-1}$ respectively. The $90\%$ confidence intervals of the mean differences of the logarithmic transformed $AUC_{0-7h}$ and $C_{max}$ values were log 0.8234-log 1.1597 and log 0.8222-log 1.1409, respectively, satisfying the bioequivalence criteria guidelines of both the US Food and Drug Administration and the Korea Food and Drug Administration. The other pharmacokinetic parameters for the test drug versus reference drug, respectively, were: $t_{1/2},\;1.87{\pm}0.62\;vs.\;1.77{\pm}0.54\;h;\;V/F,\;2061.30{\pm}986.49\;vs.\;2576.45{\pm}1826.05\;L;\;CL/F,\;835.32{\pm}357.35\;vs.\;889.48{\pm}485.87\;L\;h^{-1}; K_{el},\;0.42{\pm}0.14\;vs.\;0.40{\pm}0.18\;h^{-1};\;Ka,\;4.46{\pm}3.63\;vs.\;3.80{\pm}3.64\;h^{-1};\;and\;T_{lag},\;0.19{\pm}0.09\;vs.\;0.18{\pm}0.06\;h$. These results indicated that two alendronate formulations(70-mg alendronate sodium) were biologically equivalent and can be prescribed interchangeably.

Kinetic Study on Aminolysis of Y-Substituted-Phenyl Picolinates: Effect of H-Bonding Interaction on Reactivity and Transition-State Structure

  • A kinetic study is reported on nucleophilic substitution reactions of Y-substituted-phenyl picolinates (7a-7h) with a series of cyclic secondary amines in 80 mol % $H_2O$/20 mol % DMSO at $25.0{\pm}0.1^{\circ}C$. Comparison of the kinetic results with those reported previously for the corresponding reactions of Y-substituted-phenyl benzoates (1a-1f) reveals that 7a-7h are significantly more reactive than 1a-1f. The Br${\o}$nsted-type plot for the aminolysis of 4-nitrophenyl picolinate (7a) is linear with ${\beta}_{nuc}=0.78$, which is typical for reactions proceeding through a stepwise mechanism with expulsion of the leaving group being the rate-determining step. The Br${\o}$nsted-type plots for the piperidinolysis of 7a-7h and 1a-1f are also linear with ${\beta}_{lg}=-1.04$ and -1.39, respectively, indicating that the more reactive 7a-7h are less selective than the less reactive 1a-1f to the leaving-group basicity. One might suggest that the enhanced reactivity of 7a-7h is due to the inductive effect exerted by the electronegative N atom in the picolinyl moiety, while the decreased selectivity of the more reactive substrates is in accord with the reactivity-selectivity principle. However, the nature of intermediate (e.g., a stabilized cyclic intermediate through the intramolecular H-bonding interaction for the reactions of 7a-7h, which is structurally not possible for the reactions of 1a-1f) is also responsible for the enhanced reactivity with a decreased selectivity.

Effect of Heat Treatment Conditions on the Characteristics of Gel Made from Arrowroot Starch in Korea Cultivars (국내산 칡 전분 젤 특성에 미치는 가열처리 조건의 영향)

  • This study was conducted to investigate the effects of starch concentrations and heating conditions on the gel characteristics of arrowroot starch. Arrowroot starch gels with various pHs, and starch concentrations, were prepared using different temperatures and heating times, and then stored for 24 hrs at $4^{\circ}C$. The hardness of sample gels made at pH 2.0 and 4.0 increased as the starch concentration increased from 7% to 10%, with the maximum value of 94 N being obtained when the gel was prepared at pH 4.0 with a starch concentration of 10%. The maximum hardness of samples prepared with concentrations of starch ranging from 7~9% appeared at $80^{\circ}C$, regardless of the heating temperature and time. Furthermore, the hardness of samples prepared at greater than $100^{\circ}C$ was relatively lower than that of samples prepared at other temperatures. When a starch concentration of 8% was used, the degree of gelatinization(DR) increased as the heating temperature increased, with the maximum value of DR being about 76% at $120^{\circ}C$, regardless of heating time. After storage for 24 hrs, the hardness of samples prepared at $70^{\circ}C$, $80^{\circ}C$ and $90^{\circ}C$ appeared to decrease, while that of samples prepared at $100^{\circ}C$, $110^{\circ}C$ and $120^{\circ}C$ increased. The correlation between hardness and the degree of gelatinization or retrogradation was very high when samples were prepared at $80^{\circ}C$ with a starch concentration of 9%, as indicated by a correlation coefficient of greater than 0.95. Overall, the microstructures of freeze-dried arrowroot starch gel were composed of a continuous network of amylose and amylopectin with fragmented ghost structures in an excluded phase, but these ghost structures were more evident after storage and with increased heating temperature.

Studies on the Main Factors Affecting the Mycelial Growth of Phellinus linteus (목질진흙버섯균 Phellinus linteus의 균사체 생육에 미치는 주요 인자에 관한 연구)

  • This study was carried out to obtain the basic data on artificial culture of Phellinus linteus. The optimum condition for the mycelial growth was $25{\sim}30^{\circ}C$ and pH $6.0{\sim}7.0$. The carbon sources such as D-glucose, D-mannose and Dextrose were favorable to mycelial growth. As nitrogen sources, peptone, cassamino acid and glutamic acid appeared to be favorable. The optimum C/N ratio was about 20:1, when 2% of glucose was provided as a carbon source. The better organic acids and vitamin among tested ones were gallic, silicic acids and biotin. The mineral nutrients of $KH_2PO_4,\;FeSO_4{\cdot}7H_2O,\; MgSO_4{\cdot}7H_2O,\;ZnSO_4{\cdot}7H_2O$ were effective and the optimum concentrations were 0.05, 0.001, 0.02 and 0.003%, respectively.

Biodegradation of Diesel with Pseudomonas sp, KDi19 in Liquid Medium (Pseudomonas sp. KDi19를 이용한 액체배지내에서 경유의 생물학적 분해)

  • In this study, we isolated bacteria from petroleum contaminated soil which were near to underground storage tanks(UST). Through the screen test, we selected high efficiency bacterium, KDi19, for biodegradation of diesel. KDi19 was identified as Pseudomonas sp. by 16S rDNA, fatty acid, and morphological physiological characteristics. KDi19 degraded 956.3 mg/L(95.6%) of 1,000 mg/L diesel for 48 hours(incubation condition : temperature; $30^{\circ}C$, cell concentration; 1.0 g/L, pH 7). At low temperature, $20^{\circ}C$, $15^{\circ}C$, $10^{\circ}C$, KDi19 respectively removed 63.9%, 18.5% and 17.0% of 1,000 mg/L diesel for 48 hours(cell concentration 1.0 g/L, pH 7). At low concentration of diesel, 50 mg/L and 100 mg/L, KDi19 degraded 97.9% and 96.2% of diesel for 24 hours(temperature; $30^{\circ}C$, cell concentration: 1.0 g/L, pH 7), respectively.
