GROWTH AND COMPOSITION OF THE OMANI DHOFARI CATTLE 2. DISTRIBUTION OF CARCASS TISSUES

  • Distribution of wholesale carcass cuts and tissues was studied in Omani Dhofari bulls and steers raised under intensive management and slaughtered over a range of 110 to 210 kg body weight. The fore quarter of Dhofari cattle carcasses was heavier than the hind quarter with the chuck being the heaviest cut in the half carcass followed by the round whereas the flank was the lightest cut. Proportions of the fore quarter and its cuts increased whereas that of the hind quarter and its cuts decreased with increasing carcass weight. The fore quarter contained higher proportions of carcass tissues especially intermuscular fat than the hind quarter. The chuck and round contained the highest proportions of lean and bone and the flank the least. There was a general trend of increasing proportions of fat and decreasing proportions of lean and bone in carcass cuts and fore and hind quarters with increasing slaughter weight and age. As % total body fat (TBF), total carcass fat (TCF) increased whereas total non-carcass fat (TNCF) decreased. The largest proportion of TBF was deposited in the intermuscular site. Among the TNCF depots, the kidney and omental contributed the highest proportions whereas the pelvic and channel were the lowest. Proportions of M. rhomboideus and M. splenius increased in the half carcass whereas that of M. semitendinosus decreased as the cattle increased in size. The axial skeleton contributed 47.4-51.1, the fore limb 21.6-22.6 and the hind limb 23.9-26.2% of the total carcass bone. Proportions of axial skeleton increased whereas that of fore and hind limbs decreased with increasing slaughter weight and age. There were no major effects of castration on the distribution of weight of carcass cuts or carcass tissues. Steers had higher total body fat at 160 kg body weight and higher proportions of mesenteric, scrotal, pelvic, kidney and total non-carcass fat at 210 kg weight than bulls. As % of total body fat, steers fad significantly higher kidney and total non-carcass fat. There was little effects of castration on proportions of dimensions of individual muscles or bones.

  • Glucosinolates(GSLs) are major secondary products($\beta$-thioglucoside N-hydroxysulfates) containing sulfates that are found mainly in Cruciferae family such as cabbage, broccoli, radish, turnip, kale, mustard and rapeseed as the important crops in agronomic and economic aspects. Especially, isothiocyanates(ITCs) have been shown to inhibit carcinogenesis with regulation cancer cell development followed by regulating target enzymes, inducing cell cycle arrest and apoptosis against various human cancer cell lines. Total desulfo(DS)-GSLs of nine type in 210 Korean leaf mustard accessions were isolated and confirmed using electrospray ionization mass spectrometric(ESI-MS/MS) method and DS-GSLs powerful library maked from several reference data. The isolated individual DS-GSLs were identified by removed $\beta$-D-glucopyranose residue ($C_6H_{11}O_5$, MW 163) from MS/MS data. Among them sinigrin(41.7%), glucoiberverin(21.7%) and gluconasturtiin(12.6%) were major components, Especially, despite gluconapin was minor component, accession K046197-1 and K046197-2 showed higher content of 4.11 and 3.31 mg/g(DW), respectively. The total GSLs contents in 210 accessions were ranged from 5.3 to 23.2 mg/g(DW) with a mean value of 13.0 mg/g(DW). As a result of principal component analysis(PCA), the individual GSLs loading plots were composed of three groups and components belonged to each group showed correlationship in quantitative pattern.

Fingerprint of Marker Substances in Gami-Honghwa-Tang(KH-19) by HPLC-DAD (High Performance Liquid Chromatography-Diode Array Detector(HPLC-DAD)에 의한 가미홍화탕 (KH-19)의 지문 분석)

  • Objectives : This study was aimed to evaluate marker substances in Gami-Honghwa-Tang (KH-19) by high performance liquid chromatography-photodiode array detector (HPLC-DAD). Gami-Honghwa-Tang is composed of nine crude herbs, Rehmanniae Radix Preparata, Angelicae Gigantis Radix, Cnidii Rhizoma, Paeoniae Radix, Corni Fructus, Moutan Cortex Radicis, Lycii Fructus, Carthami Flos and Glycyrrhizae Radix. Methods : The separation was performed on an Aquasil C18 (4.6×250mm) column by gradient elution with 0.05% TFA in H2O - 0.05% TFA in acetonitrile (0 min 100:0, 20 min 90:10, 40 min 70:30, 60 min 50:50, 80 min 0:100, 90 min 100:0) as the mobile phase at a flow-rate of 1.0 ml/min with detection at 190-800nm. Also we examined the contents for bacteria, pesticide residue and harmful heavy metals. Results : HPLC-DAD was employed to determine the quantities and the qualities of several marker substances such as 5­hydroxymethyl-2-furaldehyde (5-HMF), paeonol, loganin, paeoniflorin, glycyrrhizin, and decursin in the KH-19. There were no bacterial contents, pesticide residues, or harmful heavy metals. Conclusions : We suggest these results could be a useful evidence for quality control of KH-19. This method permits fingerprints of selected individual marker substances from herbal prescriptions without derivatization, multiple purification steps, or lengthy separation times.

Associations between Feed Efficiency, Body Growth and Serum Insulin-like Growth Factor-I Level for Korean Native Ogol Chickens

  • Increasing of body weight has been one of the important economic factors in the poultry industry. Insulin-like growth factor (IGF)-I is a polypeptide that serves to regulate muscle development and body growth. Moreover, IGF-I is related to feed efficiency. However, there are few studies regarding the regulatory roles of chicken IGF-I/-II compared with that of mammals. Especially, the Korean Native Ogol Chicken (KNOC) has a lean body growth and its body weight is generally lighter than the broiler chicken. Therefore, this study was conducted to investigate associations among serum IGF-I/-II concentration, feed efficiency, and body growth in KNOC. The body weight and feed intake of KNOC were recorded from 20 to 36 weeks at 2 weeks intervals, and blood was taken every 2 weeks. Serum IGF-I/-II were measured by RIA. Chickens were divided into two groups, high and low serum IGF-I concentration. Generally, feed efficiency and growth performance (body weight and weight gain) in the high serum IGF-I group were higher than those of the low group during the experimental period. In particular, the body weight of the IGF-I high group were significantly different from those of the IGF-I low group at 34 and 36 weeks, respectively (p<0.05). Moreover, body weight, weight gain, and feed efficiency had a significant correlation with serum IGF-I at several weeks (p<0.05 and p<0.01). These results show that IGF-I plays an important role in body growth and suggests a possibility that serum IGF-I could be used as a selection marker for body growth in KNOC.

Anaerobic codigestion of urban solid waste fresh leachate and domestic wastewaters: Biogas production potential and kinetic

  • The Biochemical Methane Potential (BMP) of fresh leachate and domestic wastewaters codigestion was determined by laboratory Bach Tests at $35^{\circ}C$ over a period of 90 d using a wide range of leachates volumetric ratios from 0% to 100%. To simulate wastewaters plant treatment step, all the ratios were first air stripped for 48 h before anaerobic incubation. The kinetic of biogas production was assessed using modified Gompertz model and exponential equation. The results obtained showed that cumulative biogas production was insignificant in the case of wastewaters monodigestion while the codigestion significantly improves the BMP. Air stripping pretreatment had positive effect on both ammonium concentration and volatiles fatty acids with reduction up to 75% and 42%, respectively. According to the Modified Gompertz model, the optimal anaerobic co-digestion conditions both in terms of maximal biogas potential, start-up period and maximum daily biogas production rate, could be achieved within large leachate volumetric ratios from 25% to 75% with a maximum BMP value of 438.42 mL/g volatile solid at 50% leachate ratio. The positive effect of codigestion was attributed to a dilution effect of chemical oxygen demand and volatile fatty acid concentrations to optimal range that was between 11.7 to $32.3gO_2/L$ and 2.1 to 7.4 g/L, respectively. These results suggested that the treatment of fresh leachate by their dilution and co digestion at wastewaters treatment plants could be a promising alternative for both energetic and treatment purposes.

UV-cured Polymer Solid Electrolyte Reinforced using a Ceramic-Polymer Composite Layer for Stable Solid-State Li Metal Batteries

  • In recent years, solid-state Li metal batteries (SSLBs) have attracted significant attention as the next-generation batteries with high energy and power densities. However, uncontrolled dendrite growth and the resulting pulverization of Li during repeated plating/stripping processes must be addressed for practical applications. Herein, we report a plastic-crystal-based polymer/ceramic composite solid electrolyte (PCCE) to resolve these issues. To fabricate the one-side ceramic-incorporated PCCE (CI-PCCE) film, a mixed precursor solution comprising plastic-crystal-based polymer (succinonitrile, SN) with garnet-structured ceramic (Li7La3Zr2O12, LLZO) particles was infused into a thin cellulose membrane, which was used as a mechanical framework, and subsequently solidified by using UV-irradiation. The CI-PCCE exhibited good flexibility and a high room-temperature ionic conductivity of over 10-3 S cm-1. The Li symmetric cell assembled with CI-PCCE provided enhanced durability against Li dendrite penetration through the solid electrolyte (SE) layer than those with LLZO-free PCCEs and exhibited long-term cycling stability (over 200 h) for Li plating/stripping. The enhanced Li+ transference number and lower interfacial resistance of CI-PCCE indicate that the ceramic-polymer composite layer in contact with the Li anode enabled the uniform distribution of Li+ flux at the interface between the Li metal and CI-PCCE, thereby promoting uniform Li plating/stripping. Consequently, the Li//LiFePO4 (LFP) full cell constructed with CI-PCCE demonstrated superior rate capability (~120 mAh g-1 at 2 C) and stable cycle performance (80% after 100 cycles) than those with ceramic-free PCCE.

  • This experiment was made to select the optimum thickness of the polyethylene (P.E) film for Cheongdo Bansi and Sagoksi in the P.E film storage kept at $0^{\circ}C$. The experimental plots were divided into 4 plots by film thickness (0.04, 0.06, 0.08 and 0.10mm) and those were subdivided into 3 plots by fruits number (3, 10 and 50 persimmons) in each film bags. We investigated five experimental items; the change of loss of weight, firmness, titratable acidity, sugar contents and soluble tannin contents. 1. In the changes of loss of weight, the plot of packing in 0.04mm P.E. film bag with 50 persimmons were more retarded than other plots in Cheongdo Bansi, and packing in 0.08mm with 10 persimmons, 0.04 mm with 50 persimmons were more retarded than other plots in Sagoksi. 2. In the change of softening, the plot of packing in 0.04 mm with 50 persimmons were more retarded than other plots in Cheongdo Bansi and Sagkai. 3. In the changes of titratable acidity, the plot of packing in 0.04 mm with 50 persimmons were more slightly decreased than other plots in Cheongdo Bansu also in Sagoksi, packing in 0.06 mm with 10 persimmons were the same results. 4. In the changes of soluble tannin contents, the plots of packing in 0.06 mm with 10 persimmons, 0.04 mm with 50 perimmons were more ratarded in Chenongdo Bansi, also in Sagoksi, packing in 0.04 mm with 10 persimmons 50 persimmons were the same results. 5. In the changes of soluble tannin contents, the plots of packing in 0.04mm with 3 and 10 persimmons were more slowly decreased than other plots in Cheongdo Bansi and Sagoksi, on. the other hand, pcaking in 0.04mm with 50 persimmins in Cheongdo Bansi and Sagoksi, had not astringent taste at 120 days in storage. Judging through the upper results, the most desirable storage conditions for Cheongdo Bansi and Sagoksi were to pack in P.E film bag of 0.04mm with 50 persimmons.

A Set of Anthocyanin Biosynthetic Genes are Differentially Expressed in Strawberry (Fragaria x ananassa cv Maehyang) during the Fruit Development Process (매향 딸기로부터 anthocyanin 합성 유전자의 분리 및 과실발달 과정에서의 발현 분석)

  • Anthocyanin synthesis in strawberry (Fragaria x ananassa cv Maehyang) begins approximately 26 days postflowering and continued throughout fruit ripening. A set of cDNA clones encoding the anthocyanin biosynthetic enzymes were isolated from strawberry. A pair of primers were designed for polymerase chain reaction (PCR) through the comparison of the nucleotide sequences of homologous genes from diverse plants. Reverse transcriptase-PCRs were performed using cDNA synthesized from ripe fruit total RNA and the primers corresponding to each gene. Eight genes of the anthocyanin pathway were cloned and confirmed by sequencing to code for phenylalanine ammonia lyase (PAL), 4-cummarate CoA ligase (4CL), chalcone synthase (CHS), chalcone isomerase (CHI), flavanone-3-hydroxylase (F3H), dihydroflavonol 4-reductase (DFR), anthocyanidine synthase (ANS), UDP-glucose:flavonoid-3-O-glucosyl-transferase (UFGT). Northern analyses showed that the corresponding genes were differentially expressed during the fruit development process. All genes except PAL were predominantly expressed in fruit. Expression of PAL, DFR and ANS was detected 10 days postflowering at the early stage of fruit development, declined for a while and sharply increased 22 days postflowering then showed a peak 34 days postflowering. The other genes, however, were not expressed up to 22 or 30 days postflowering when the initial fruit ripening events occur at the time of initiation of anthocyanin accumulation. The onset of anthocyanin synthesis in ripening strawberry coincides with a coordinated induction of the anthocyanin pathway genes, suggesting the involvement of regulatory genes. We propose that at least two different regulatory mechanisms playa role in the biosynthesis of anthocyanin during color development of strawberry.

Soil Microbial Flora and Chemical Properties as Influenced by the Application of Pig Manure Compost (돈분퇴비의 시용이 배추재배지 토양의 미생물상 및 화학성에 미치는 영향)

  • Studies were conducted during 2 months from May of 1997 to evaluate the effects of pig manure compost(PMC) on soil microbial flora. To do so, a field experiment of Chinese cabbage(Brassica campestris L.) was conducted in a randomized block design on a sandy loam soil and microbial floral characteristics in soils were analyzed. Treatments to control included the application of PMC at (A) $8Mg\;ha^{-1}$CM-8), (B) $29Mg\;ha^{-1}$(CM-2,9), and (C) $57Mg\;ha^{-1}$(CM-57), and of chemical fertilizer(D) at $320N-80P_2O_5-200K_2O\;kg\;ha^{-1}$(NPK). In each treatment, the rhizosphere and non-rhizosphere soils were tested for the analysis of microbial populations. The populations of bacteria, actinomycetes, and fungi increased in soils with the applications of PMC and chemical fertilizer, but that of Bacillus sp. decreased. However, the population of fluorescent Pseudomonas sp. was reduced in NPK plots only. With increasing application rates of PMC, the number of colony forming units(cfu) of bacteria (Pseudomonas sp. and actinomycetes) and fungi increased. in all PMC-treated plots, the population density peaked at early growth stage for bacteria(including Bacillus sp.), at late growth for fluorscent Pseudomonas sp., and at harvest for fungi and actinomycetes. The rhizosphere effect was greatest for fluorscent Pseudomonas sp. As the application rates of PMC increased, Total N, organic matter, available phosphate, and exchangeable -K, -Ca, and -Mg increased compared to control, but soil pH was lowered. In NPK plots, EC was 3.4-fold and exchangeable K was 5-fold higher than control.

Biliary Atresia -A Survey by the Korean Association of Pediatric Surgeons in 2011- (담도폐쇄증 -대한소아외과학회 회원 대상 전국 조사-)

  • The Korean Association of Pediatric Surgeons (KAPS) performed the second nationwide survey on biliary atresia in 2011. It was a follow-up study to the first survey, which was performed in 2001 for the retrospective analysis of biliary atresia between 1980 and 2000. In the second survey, the authors reviewed and analyzed the clinical data of patients who were treated for biliary atresia by the members of KAPS from 2001 to 2010. A total of 459 patients were registered. Among them, 435 patients primarily underwent the Kasai operation. The mean age of patients who underwent the Kasai operation was $66.2{\pm}28.7$ days, and 89.7% of those patients had type III biliary atresia. Only five patients (1.4%) had complications related to the Kasai operation. After the Kasai operation, 269 (61.8%) of the patients were re-admitted because of cholangitis (79.9%) and varices (20.4%). One hundred and fifty-nine (36.6%) of the patients who underwent the Kasai operation subsequently underwent liver transplantation. The most common cause of subsequent liver transplantation was persistent hyperbilirubinemia. The mean interval between the Kasai operation and liver transplantation was $1.1{\pm}1.3$ years. Overall the 10-year survival rate after the Kasai operation was 92.9% and the 10-year native liver survival rate was 59.8%. We had 23 patients for primary liver transplantation without the Kasai operation. The mean age patients who underwent primary liver transplantation was $8.6{\pm}2.9$ months. In summary, among the 458 Kasai-operation and liver-transplantation patients, 373 lived, 31 died, and 54 were unavailable for follow up. One-third of the patient who survived have had complications correlated with biliary atresia. In comparison with the first survey, this study showed a higher survival rate and a greater number of liver transplantation.
