Protective Effect of Polygonum Multiflorum on Cell Damage in UVB-irradiated HaCaT Keratinocytes (적하수오(赤何首烏)의 UVB로 자극한 피부 각질세포 보호 작용)

  • Objectives: This study was performed to assess the protective effect of Polygonum multiflorum(PM) on UVB-irradiated HaCaT Keratinocytes damage. Methods: The protective effects of Polygonum multiflorum(PM) were determined by UVB-irradiated HaCaT assay. We assessed protective effects of Polygonum multiflorum(PM) on LDH release and nitrite production from HaCaT. COX-2, Bcl-2, Bax, $TNF{\alpha}$, c-jun, c-fos, NF-${\kappa}B$, iNOS, Bcl-xL gene expression were determined in HaCaT using real-time PCR method. Results: 1. PM inhibited LDH Release in UVB-irradiated HaCaT Keratinocytes. 2. PM inhibited Nitrite Production in UVB-irradiated HaCaT Keratinocytes. 3. PM suppressed the Gene Expression of COX-2 in UVB-irradiated HaCaT Keratinocytes. 4. PM increased the Gene Expression of Bcl-2 in UVB-irradiated HaCaT Keratinocytes. 5. PM didn't increase the Gene Expression of Bax in UVB-irradiated HaCaT Keratinocytes. 6. PM suppressed the Gene Expression of $TNF{\alpha}$ in UVB-irradiated HaCaT Keratinocytes. 7. PM suppressed the Gene Expression of c-jun in UVB-irradiated HaCaT Keratinocytes. 8. PM suppressed the Gene Expression of c-fos in UVB-irradiated HaCaT Keratinocytes. 9. PM suppressed the Gene Expression of NF-${\kappa}B$ in UVB-irradiated HaCaT Keratinocytes. 10. PM suppressed the Gene Expression of i-NOS in UVB-irradiated HaCaT Keratinocytes. 11. PM didn't increase the Gene Expression of Bcl-xL in UVB-irradiated HaCaT Keratinocytes Conclusions: In conclusion, these results suggest that PM inhibited the cell damage in UVB-irradiated HaCaT.

  • The influence of temperature on the rate of egg production and embryonic development of Microcotyle sebastis was investigated to determine the precise time of a second treatment. The survival time of the adults of M. sebastis was inversely proportional to temperature. The number of laid eggs per each replicate during the first 24 h was $39.3{\pm}4.0$ at $10^{\circ}C$, $62.7{\pm}14.2$ at $15^{\circ}C$, $101.0{\pm}5.6$ at $20^{\circ}C$ and $89.0{\pm}11.0$ at $25^{\circ}C$. The time required for egg hatching of M. sebastis was $31.30{\pm}4.88$, $17.52{\pm}3.24$, $11.59{\pm}3.02$ and $10.76{\pm}3.10$ days at 10, 15, 20 and $25^{\circ}C$, respectively. The regression models of the time required for the beginning and 50% point of hatching according to the different temperatures were as follows; Beginning of hatch: D=58.2000-$4.2067{\times}Temp+0.0867{\times}(Temp)^2$ ($P\leq0.01$), 50% of hatch: D=91.3833-$7.5767{\times}Temp+0.1767{\times}(Temp)^2$ ($P\leq0.01$).

Anti-oxidative Activities of Commercial Edible Plant Extracts Distributed in Korea (국내 유통 중인 식용식물 추출물의 항산화효과)

  • Many plant extracts are known to have antioxidative effects. However, their activities can be reduced or disappeared during mass production process. The purpose of this study is to compare antioxidative effects of edible plant extracts distributed in Korea. forty three kinds of edible plant extracts commercially available in Korea were selected and investigated for their total phenolics contents and antioxidative potentials(DPPH radical and superoxide anion radical scavenging activities). In contents of total phenolics, the commercial plant extracts from Artemisia annua(whole plant), Ilex paraguariensis(leaf, Silybum marianum(fruit and leaf, Ulmus pumila(bark), Coliolus versicolor(fruit), and Curcuma longa(root and stem) contained over 70 mg/g of powder, DPPH radical scavenging activities($SC_{50}$, 50% scavenging concentration) of A. annua, I. paraguariensis, Pinus densiflora(leaf),S. marianum, U. pumila, and C. longa were $53.96{\pm}0.81\;ppm,\;24.61{\pm}2.12\;ppm,\;35.96{\pm}1.11\;ppm,\;57.46{\pm}2.13\;ppm,\;55.25{\pm}1.65\;ppm\;and\;12.99{\pm}1.67ppm$, respectively, while that of positive control(vitamin C) was $3.86{\pm}0.81\;ppm$. $SC_{50}$ values against superoxide anion radical of A. annua, Cinnamomum zeylanicum(bark), I. paraguariensis, Rubus coreanus(fruit and leaf), Morus alba(leaf), P. densiflora, S. marianum, U. pumila, C. versicolor, C. longa, Perilla frutescens var. acuta(leaf), and H. sabdariffa(leaf and newer) were $53.21{\pm}1.83ppm,\;50.12{\pm}2.12ppm,\;5.59{\pm}0.84ppm,\;41.60{\pm}8.93ppm,\;20.19{\pm}0.97ppm,\;15.19{\pm}1.66ppm,\;21.20{\pm}1.88ppm,\;15.71{\pm}0.91ppm,\;55.48{\pm}2.42ppm,\;52.12{\pm}2.44ppm,\;23.80{\pm}1.98ppm\;and\;11.14{\pm}0.51ppm$, respectively($SC_{50}$ value of vitamin C: $9.61{\pm}0.93ppm$). In particular, both 1 paraguariensis and P. densiflora had high content of phenolics as well as high scavenging activities of DPPH radical and superoxide anion radical. Consequently, above two commercial extracts may be useful as a source of antioxidative nutraceutics.

A Study on Various Trans Fatty Acid Contents (Trans지방산의 함량변화에 관한 연구)

  • In this study, the degree of rancidity and trans fatty acids formation was assessed in Soybean oil(SBO), Corn germ oil (CGO), Canola oil(CNO) and Olive oil(OLO). All samples treated under various conditions were analyzed in order to determine their physicochemical characteristics(RI: Refractive index, Tocopherol, AV: Acid Value, IV: Iodine Value) and total trans fatty acid contents via GC. The results were as follows: The AV of corn germ oil was the highest (0.49$\pm$0.01 and 0.72$\pm$0.04 respectively) among the 4 kinds of oils at $170{\pm}2^{\circ}C$ and $210{\pm}2^{\circ}C$. The IV of olive oil was the lowest(88.7$\pm$0.6 and 89.2$\pm$0.5) among the 4 kinds of oils at $170{\pm}2^{\circ}C$ and $210{\pm}2^{\circ}C$. The trans fatty acid contents of the soybean oil, corn germ oil, canola oil and olive oil, respectively(in g/100) increased from 0.41, 0.60, 0.44 and 0.11 prior to heating to 0.84, 1.36, 0.94 and 0.81 after 7 hours. Catechin and BHT reduced trans fatty acid formation by 0.5-15.5% under all treatment conditions. In particular, Catechin exerted a more profound inhibitory effect on trans fatty acids formation than that did BHT.

Effect of CQ-amine ratio on the degree of conversion in resin monomers with binary and ternary photoinitiation systems

  • Objectives: This study evaluated the effect of camphorquinone (CQ)-amine ratio on the C=C double bond conversion of resins with binary and ternary photoinitiation systems. Materials and Methods: Two monomer mixtures (37.5 Bis-GMA/37.5 Bis- EMA/25 TEGDMA) with binary systems (CQ/DMAEMA in weight ratio, group A [0.5/1.0] and B [1.0/0.5]) and four mixtures with ternary system (CQ/OPPI/DMAEMA, group C [0.1/1.0/0.1], D [0.1/1.0/ 0.2], E [0.2/1.0/0.1] and F [0.2/1.0/0.2]) were tested: 1 : 2 or 2 : 1 CQ-amine ratio in binary system, while 1 : 1 ratio was added in ternary system. The monomer mixture was cured for 5, 20, 40, and 300 sec with a Demetron 400 curing unit (Demetron). After each exposure time, degree of conversion (DC) was estimated using Fourier transform infrared (FTIR) spectrophotometer (Nicolet 520, Nicolet Instrument Corp.). The results were analyzed by ANOVA followed by Scheffe test, with p = 0.05 as the level of significance. Results: DC (%) was expressed in the order of curing time (5, 20, 40, and 300 sec). Group A ($14.63{\pm}10.42$, $25.23{\pm}6.32$, $51.62{\pm}2.69$, $68.52{\pm}2.77$); Group B ($4.04{\pm}6.23$, $16.56{\pm}3.38$, $37.95{\pm}2.79$, $64.48{\pm}1.21$); Group C ($16.87{\pm}5.72$, $55.47{\pm}2.75$, $60.83{\pm}2.07$, $68.32{\pm}3.31$); Group D ($23.77{\pm}1.64$, $61.05{\pm}1.82$, $65.13{\pm}2.09$, $71.87{\pm}1.17$); Group E ($28.66{\pm}2.92$, $56.68{\pm}1.33$, $60.66{\pm}1.17$, $68.78{\pm}1.30$); Group F ($39.74{\pm}6.31$, $61.07{\pm}2.58$, $64.22{\pm}2.29$, $69.94{\pm}2.15$). Conclusion: All the monomers with ternary photoinitiation system showed higher DC than the ones with binary system, until 40 sec. Concerning about the effect of CQ-amine ratio on the DC, group A converted into polymer more than group B in binary system. However, there was no significant difference among groups with ternary system, except group C when cured for 5 sec only.

Thermographic Assessment in Dry Eye Syndrome, Compared with Normal Eyes by Using Thermography (열화상카메라를 이용한 정상안과 건성안의 서모그래피 비교)

  • Purpose: The purpose of this study was to compare and analyze the ocular surface and the palpebral conjunctiva of categorized subjects, which were divided into normal eye group and dry eye group, by using a thermal camera. Methods: Subjects were 144 eyes of 72 normal university students, who didn't have any corneal disease, abnormal lacrimal ducts, medical records regarding ocular surgeries, or experience of using contact lens. Subjects were divided into two groups, which were normal eye group and dry eye group, based on the results of TBUT, Schirmer I test, and McMonnies test. After categorizing the subjects, the temperature of the subjects' ocular surface and the palpebral conjunctiva were measured and analyzed by using a thermal camera (Cox CX series, Answer co., Korea). Results: In the normal eye group's Central Ar.1, Nasal Ar.2, Temporal Ar.3, Superior Ar.4, Inferior Ar.5, the measured amount of temperature change on each area was $-0.13{\pm}0.08$, $-0.14{\pm}0.08$, $-0.12{\pm}0.08$, $-0.14{\pm}0.08$, $-0.10{\pm}0.09(^{\circ}C/sec)$. The dry eye group's results were $-0.17{\pm}0.08$, $-0.16{\pm}0.07$, $-0.16{\pm}0.08$, $-0.17{\pm}0.09$, $-0.15{\pm}0.08(^{\circ}C/sec)$. When compared with the normal eye group, the values of Ar.1, Ar.3, Ar.5 were significantly different in the dry eye group(p<0.05). The amount of temperature change, which was observed on the palpebral conjunctiva(Ar.1:central, Ar.2: nasal, Ar.3: temporal) of the normal eyes, measured by thermography, was $34.36{\pm}1.12$, $34.17{\pm}1.10$, $34.07{\pm}1.12^{\circ}C$ on each area. Same values taken from the dry eye group was $33.55{\pm}0.94$, $33.43{\pm}0.97$, $33.51{\pm}1.06^{\circ}C$ on each area. The values of Ar.1, taken from the dry eye group, had a significant difference, compared to the values of the normal eye group(p=0.05). Conclusion: The temperature of the ocular surface decreased faster on the dry eyes, compared to the normal eyes. The temperature measured on the palpebral conjunctiva of the dry eyes were also lower than the normal eyes. The temperature changes on the ocular surface, observed with a thermal camera, were objective values to assess the stability of tear films, and might provide useful data for studies related to dry eye syndrome.

Pathogenicity, hemagglutinability, and the effect of physicochemical agents on virus of rabbit hemorrhagic disease (토끼 출혈성 바이러스의 병원성, 적혈구응집성 및 물리화학적 요인에 대한 영향)

  • Rabbits were experimentally infected with rabbit hemorrhagic disease virs and the viral pathogenicity, hemagglutinability, and the effect of physicochemical factors were studied. The experimental results were summariaed as follows: 1. Mean rectal temperature of 11 infected rabbits was $40.0{\pm}0.47^{\circ}C$ prior to the virus inoculation, and $39.9{\pm}0.75^{\circ}C$ after 12hrs., $40.2{\pm}0.65^{\circ}C$ after 24hrs., $40.1{\pm}0.77^{\circ}C$ after 36hurs, and $40.6{\pm}0.56^{\circ}C$ just before the death. 2. Mean death time of infected rabbits was $40.3{\pm}22.0$ honrs and its range was 24 to 93 hours. 3. O, B, AB and A type of human erythrocytes were shown their HA in the order, but rabbit and chicken erythrocytes were not hemagglutinated by the virus. 4. In the hemagglutination, less than 0.25 per cent of a final concentration of erythrocytes and 0.2 per cent of BSA in PBS resulted in the best hemagglutination. Phosphate concentration in a range of 0.01M to 0.10M in PBS was not influenced on the hemagglutination reaction, and its pH 7.0 resulted in a better HA. 5. The hemagglutinating titers, in log 2 scale, of organs and tissues of the virus infected rabbits were $9.3{\pm}3.8$ (liver), $9.1{\pm}3.9$ (blood), $6.2{\pm}2.6$ (spleen) and $5.0{\pm}2.5$ (kidney). 6. The physicochemical factors such as heating ($50^{\circ}C$, 10 min.), trypsin treatment (0.05 pre cent, 5 min.), acid treatment (pH 3.0, 20 min.) and ether extraction (3 times) were not affective to the stability of virus and viral HA activities.

The Chemical Composition of the Nagdong River Downstream Water (낙동강 하류수의 수질조성에 대하여)

  • Relationships between the electrical conductivity and the contents of the chloride, sulfate, calcium, magnesium, sodium, potassium and total major inorganic ions, and between each, chemical conservative constituents were calculated with the data which sampled at the lesions of Mulgeum and between Namji and Wondong from March 1974 to April 1980. Semilogarithmic relations were found between the electrical conductivity and the contents of monovalent ions, and logarithmic relations were found between the electrical conductivity and the contents of divalent ions at the both regions. The relational equations between the electrical conductivity $\lambda_{25}$and the contents of the major inorganic ions at Mulgeum are as follows: $log\;Cl(ppm)\;=\;2.37{\cdot}\lambda_{25}(m{\mho}/cm)+0.733{\pm}0.141$, $log\;SO_4(ppm)=1.12{\cdot}log\lambda_{25}(m{\mho}/cm)+2.14{\pm}0.18$, $log\;Ca(ppm)=0.615{\cdot}log\lambda_{25}(m{\mho}/cm)+1.67{\pm}0.12$, $log\;Mg(ppm)=0.756{\cdot}log\lambda_{25}(m{\mho}/cm)+1.27{\pm}0.11$, $log\;Na(ppm)=2.82{\cdot}\lambda_{25}(m{\mho}/cm)+0.551{\pm}0.133$, $log\;K(ppm)=1.33{\cdot}\lambda_{25}(m{\mho}/cm)+0.136{\pm}0.095$, and total inorganic ions $C(ppm)=399{\cdot}\lambda_{25}(m{\mho}/cm)-0.9{\pm}14.6$. The relational equations between the electrical conductivity ($\lambda_{25}$) and the contents of the major inorganic ions at the region between Namji and Wondong a.e as follows: $log\;Cl(ppm)=4.27{\cdot}\lambda_{25}(m{\mho}/cm)+0.380{\pm}0.138$, $log\;SO_4(ppm)=0.915{\cdot}log\lambda_{25}(m{\mho}/cm)+1.95{\pm}0.18$, $log\;Ca(ppm)=0.756{\cdot}log\lambda_{25}(m{\mho}/cm)+1.74{\pm}0.12$, $log\;Mg(ppm)=1.00{\cdot}log\lambda_{25}(m{\mho}/cm)+1.41{\pm}0.10$. $log\;Na(ppm)=2.47{\cdot}\lambda_{25}(m{\mho}/cm)+0.614{\pm}0.065$, $log\;K(ppm)=1.62{\cdot}\lambda_{25}(m{\mho}/cm)+0.030{\pm}0.060$, and total inorganic ions $C(ppm)=323{\cdot}\lambda_{25}(m{\mho}/cm)+11.7{\pm}9.3$. Cluster Analysis of Synoptic Scale Meteorological Characteristics on High PM10 Concentration Episodes in the Southeastern Part of Korean Peninsula (한반도 남동 지역에서 발생한 고농도 미세먼지 사례의 종관 기상학적 군집 특성 분석)

  • This study presents the K-means clustering analysis-based classification of the meteorological patterns affecting the occurrence of high PM10 concentration in the southeastern region of the Korean peninsula for the last five years (2014-2018). Regional differences in Busan, Ulsan, and Gyeongnam related to high PM10 episodes, were clarified through the statistical comparison study using synoptic scale meteorological elements using NCEP (National Centers for Environmental Prediction/FNL (Final Operational Global Analysis) re-analysis meteorological data. Meteorological patterns were classified into a total of five categories (C1-C5). The incidence of each cluster was 24.8% (C1), 21.3% (C2), 20.4% (C3), 17.3% (C4), and 16.2% (C5), respectively. The high PM10 concentration in the southeastern region resulted from long and short range transports (C1, C3, C5) from outside of the region, and the emissions (C2, C4) inside the region. In the high PM10 episodes in Busan, Ulsan, and Gyeongnam regions, meteorological characteristics such as different geopotential height and wind speed at 500 hPa in each cluster and the change in the location of high pressure over Korean Peninsula is strongly associated with the dispersion of PM10 around inventories in the region and the tendency of long-range transportation of PM10 emitted from outside of region.

  • Livestock manure is a significant source for greenhouse gas (GHG) emission, and a huge amount of GHG emission is generated during its storage. In the present work, lowering temperature was attempted to mitigate methane ($CH_4$) emission from cattle manure (CM) with high solid content. CM was stored for 60 d at $15-35^{\circ}C$ ($5^{\circ}C$ interval). $CH_4$ emission reached $63.6{\pm}3.6kg\;CO_2\;eq./ton\;CM$ at $35^{\circ}C$, which was reduced to $51.6{\pm}1.8$, $24.1{\pm}4.4$, $14.9{\pm}0.5$, and $3.7{\pm}0.1kg\;CO_2\;eq./ton\;CM$ at 30, 25, 20, and $15^{\circ}C$, respectively. After storage, 30% of COD reduction was observed in the CM stored at $35^{\circ}C$, while the COD removal decreased to only 6% at $15^{\circ}C$. It was found that only 3-11% of COD removal was done by anaerobic process, while the rest of COD removal was done by aerobic biological process. Methanobrevibacter and Methanolobus were found to be the dominant species in the CM, and the dominance of Methanolobus psychrophilus increased at lower storage temperature. Specific methanogenic activity test results showed that the inhibition by low temperature was temporal.
