Placental Transfer and Pharmacokinetics of a Single Oral Dose of the Fluoroquinolone Antibacterial DW-116 in Rats (랫드에서 fluoroquinolone 항균제 DW-116의 단회 경구투여에 의한 태반통과와 약물동태연구)

  • The present study was conducted to investigate the placental transfer and pharmacokinetics of the flu-oroquinolone antibacterial DW-116 in pregnant rats. The placental transfer and pharmacokinetics of DW-116 were examined after a single oral dose of 500 mg $^{14}C$ DW-116/kg on gestational day 18. Maternal and fetal tissues were collected at 0.17 0.5,1,2,4,8, and 24 h after dosing. Maximum radioactivity was detected in maternal plasma, placenta, and whole fetus at 1 h, and in amniotic plasma at 4 h after dosing. Thereafter, radioactivity gradually disappeared from these tissues and was 16~28% of maximum levels at 24 h after dosing. Radioactivity in whole fetus were higher than those in the maternal plasma and placenta. The $T_{1/2,abs}$, $T_{1/2,{\beta}},$ AUC, $T_{max},$ and $C_{max}$ in the maternal plasma were approximately 6 min, 13.3 h, 1620 $ug^*hr/ml,$ 0.5 h, and 136 ug/ml, respectively. Those in the placenta were approximately 20 min, 12.3 h, 2150 $ug^*h/$m\ell$,$ 1.0 h, and 172 ug/ml, respectively. Those in the whole fetus were 13 min, 12.8 h,2549 $ug^*h/$m\ell$,$ 1 h, and 191 ug/ml, respectively. In the amniotic fluid of maternal uterus, the 4T_1/2,abs}$, $T1/2,{\beta},$ AUC, $T_{max},$ and $C_{max}$ were approximately 1.3 h,9.3 h,2508 $ug^*h/$m\ell$,$ 4.4 h, and 135 ug/ml, respectively. While DW-116 disappeared biphasically from maternal plasma, whole fetus and placenta, it was eliminated monophasically from amniotic fluid. In conclusion, this study demonstrated that the absorption and distribution of DW-116 in maternal plasma and placenta were extensively rapid, and that the test chemical well passed the blood-placenta barrier and was transferred to the fetus.

Bioequivalence of Lisihexal® tablet to Zestril® tablet(Lisinopril 10 mg) (제스트릴®정(리시노프릴, 10 mg)에 대한 리시헥살®정의 생물학적동등성)

  • Lisinopril is one of the angiotensin-converting enzyme inhibitors, which have been used for treatment of hypertension and heart failure. The aim of this study was to evaluate the bioequivalence of two lisinopril tablet, $Lisihexal^{\circledR}$ and $Zestril^{\circledR}$ as a test and reference, respectively. The study was came out on 28 healthy male Korean volunteers in $2{\times}2$ crossover design. An analytical method with LC-MS-MS was developed for the quantification of lisinopril and enalapril(IS) using SPE method. The condition was selective, sensitive and precise in human plasma, that was enough for the pharmacokinetic study of lisinopril. The pharmacokinetic parameters such as $AUC_t,\;AUC_{inf},\;C_{max},\;T_{max}\;and\;t_{1/2}$ were calculated and ANOVA test was used for the statistical analysis of the parameters using log transformed $AUC_t,\;AUC_{inf}\;and\;C_{max}$. $t_{1/2}$ of test and reference drugs were calculated $11.4{\pm}5.1\;and\;16.1{\pm}9.9\;hr$, respectively. The 90% confidence intervals of $AUC_t,\;AUC_{inf}\;and\;C_{max}$ were log 0.9245$\sim$log 1.0603, log 0.9270$\sim$log 1.0601 and log 0.9548$\sim$log 1.1009, within the acceptable range of log 0.8 to log 1.25 by KFDA bioequivalence criteria. Two medications of lisinopril were evaluated bioequivalent and thus may be prescribed interchangeably.

Growth Kinetics of Phytoplankton in Shallow Eutrophic Reservoir (얕은 부영양 저수지에서의 식물플랑크톤 성장 역학)

  • This study was conducted to assess the growth characteristics of phytoplankton and to understand seasonal dynamics of phytoplankton in response to limiting nutrients in an agricultural reservoir from November 2002 to December 2003. Marked increase of chl.a concentration observed in July ($99.0{\mu}g/L$) and November ($109.7{\mu}g/L$) after heavy rainfall. TP concentration ranged $48.0{\sim}126.6{\mu}g/L$, and its the temporal variation was similar to that of chl.a concentration. Microcystis spp., dominant phytoplankton species were used for the growth kinetics experiment, except for the season when Aulacoseira spp. (March, April) and Aphanocapsa sp. (May) dominated. In the temperature range between $10{\sim}25^{\circ}C$, the rate of growth increase per $10^{\circ}C$ was almost two folds. The highest maximum growth rate (${\mu}_{max}=1.09day^{-1}$) of phytoplankton observed September, and ${\mu}_{max}$ was lowest ($0.34day^{-1}$) in March when Aulacoseira spp. dominated. The ${\mu}_{max}$ ($0.78{\pm}0.20day^{-1}$) was relatively high in the summer season when water temperature is above $20^{\circ}C$ and cyanobacteria dominated compared to the spring when diatoms dominated ($0.43{\pm}0.08day^{-1}$). The maximum growth rate ($0.55{\pm}0.12day^{-1}$) and the half saturation concentration ($K_s=0.73{\pm}0.15{\mu}M$) of cyanobacteria during winter season (November, December) was higher than those of diatoms. However, the ${\mu}_{max}$ and $K_s$ of cyanobacteria in December was similar to those of diatom, reflecting that diatom cell quota (Mean 48.4 pgP/cell) was greater than cyanobacteria (34.0 pgP/cell) during this time.

  • Canine cataract is developed with aging and can cause the blindness or surgical treatment if not treated timely. In this paper, we propose a method for extracting cataract suspicious areas automatically with FCM(Fuzzy C_Means) algorithm to overcome the weakness of previously attempted ART2 based method. The proposed method applies the fuzzy stretching technique and the Max-Min based average binarization technique to the dog eye images photographed by simple devices such as mobile phones. After applying the FCM algorithm in quantization, we apply the brightness average binarization method in the quantized region. The two binarization images - Max-Min basis and brightness average binarization - are ANDed, and small noises are removed to extract the final cataract suspicious areas. In the experiment with 45 dog eye images with canine cataract, the proposed method shows better performance in correct extraction rate than the ART2 based method.

A Case Study of Human Thermal Sensation (Comfort) in Plastic Houses (온실시설내 인간 열환경지수(열쾌적성)에 대한 사례연구)

  • To analyze human thermal environments in protected horticultural houses (plastic houses), human thermal sensations estimated using measured microclimatic data (air temperature, humidity, wind speed, and solar and terrestrial radiation) were compared between an outdoor area and two indoor plastic houses, a polyethylene (PE) house and a polycarbonate (PC) house. Measurements were carried out during the daytime in autumn, a transient season that exhibits human thermal environments ranging from neutral to very hot. The mean air temperature and absolute humidity of the houses were $14.6-16.8^{\circ}C$ (max. 22. $3^{\circ}C$) and $7.0-12.0g{\cdot}m^{-3}$ higher than those of the outdoor area, respectively. Solar (K) and terrestrial (L) radiation were compared directionally from the sky hemisphere (${\downarrow}$) and the ground hemisphere (${\uparrow}$). The mean $K{\downarrow}$ and $K{\uparrow}$ values for the houses were respectively $232.5-367.8W{\cdot}m^{-2}$ and $44.9-55.7W;{\cdot}m^{-2}$ lower than those in the outdoor area; the mean $L{\downarrow}$ and $L{\uparrow}$ values were respectively $150.4-182.3W{\cdot}m^{-2}$ and $30.5-33.9W{\cdot}m^{-2}$ higher than those in the outdoor area. Thus, L was revealed to be more influential on the greenhouse effect in the houses than K. Consequently, mean radiant temperature in the houses was higher than the outdoor area during the daytime from 10:45 to 14:15. As a result, mean human thermal sensation values in the PMV, PET, and UTCI of the houses were respectively $3.2-3.4^{\circ}C$ (max. $4.7^{\circ}C$), $15.2-16.4^{\circ}C$ (max. $23.7^{\circ}C$) and $13.6-15.4^{\circ}C$ (max. $22.3^{\circ}C$) higher than those in the outdoor area. The heat stress levels that were influenced by human thermal sensation were much higher in the houses (between hot and very hot) than in the outdoor (between neutral and warm). Further, the microclimatic component that most affected the human thermal sensation in the houses was air temperature that was primarily influenced by $L{\downarrow}$. Therefore, workers in the plastic houses could experience strong heat stresses, equal to hot or higher, when air temperature rose over $22^{\circ}C$ on clear autumn days.

The Bioequivalence of Two Carbamazepine Controlled Release Formulations (카르바마제핀 서방형 제제간의 생물학적 동등성 비교)

  • Carbamazepine is an anticonvulsant drug that has been shown to be as effective as phenytoin or phenobarbital in treatment of grand mal and complex partial seizures and is also approved as the drug of choice for treatment of the pain associated with trigerminal neuralgia. And the therapeutic or toxic effects of carbamazepine are better related to plasma concentration than to dosage, which can be attributed to interindividual variability in the pharmacokinetics. A slow rate of carbamazepine dissolution in the gastrointestinal tract is believed to be the cause of its relatively slow and erratic rate of absorption. For these reasons pharmacokinetic evaluation of newly formulated carbamazepine is neccessary. In this study, the bioequivalence in carbamazepine between the $TegretoI^{TM}$ CR tablet (Geigy Co.) and $Carmazepine^{TM}$ CR tablet (Myung In Co.) was evaluated. 12 normal volunteers (age $21\~27$ years old) was divided into two groups, and a randomized cross-over study was employed. The pharmacokinetic parameters ($C_{max},\;T_{max}$ and AUC) obtained of oral administration of each formulatim of carbamazepine 400 mg were evaluated and ANOVA was utilized for the statistical analysis of parameters. $C_{max}\;is\;8.26{\pm}3.1{\mu}g/ml\;(C.V.\;37.3\%)\;in\;TegretoI^{TM}\;and\;9.39\{pm}2.9{\mu}g/ml\;(C.V.\;30.5\%)$ in $Carmazepine^{TM},\;T_{max}\;is\;28.0{\pm}5.9\;hrs(C.V.\;21.1\%)$ in $Tegretol^{TM}\;and\;24.0{\pm}7.2\;hrs(C.V.\;30.2\%)$ in $Carmazepine^{TM}$ and AUC is $786.4{\pm}360.5{\mu}g{\cdot}hr/ml\;(C.V.\;45.8\%)$ in $TegretoI^{TM}\;and\;792.8{\pm}228.6{\mu}g{\cdot}hr/ml\;(C.V.\;28.8\%)$ in $Carmazepine^{TM}$, respectively. As the result of the data, two formulations are bioequvalent, and the lower C.V. of $Carmazepine^{TM}$ in every individual can be merit.

A Study on Material Degradation Evaluation of 9Cr1MoVNb Steel by Micromechanics Test Method (미소역학 시험기법에 의한 9Cr1MoVNb강의 열화도 평가)

  • The Micromechanics test is new test method which uses comparatively smaller specimen than that required in conventional material tests. There are several methods, such as small-specimen creep test, the continuous indentation test, and small punch(SP) test. Among them, the small punch(SP) test method has been applied to many evaluation fields, such as a ductile-brittle transition temperature, stress corrosion cracking, hydrogen embrittlement, and fracture properties of advanced materials like FGM or MMC. In this study, the small punch(SP) test is performed to evaluate the mechanical properties at high/low temperature from $-196^{\circ}C$ to $650^{\circ}C$ and the material degradation for virgin and aged materials of 9Cr1MoVNb steel which has been recently developed. The ${\Delta}P/{\Delta}{\delta}$ parameter defined a slope in plastic membrane stretching region of SP load-displacement curve decreases according to the increase of specimen temperature, and that of aged materials is higher than the virgin material in all test temperatures. And the material degradation degrees of aged materials with $630^{\circ}C$ -500hrs and $630^{\circ}C$ -1000hrs are $36^{\circ}C$ and $38^{\circ}C$ respectively. These behaviors are good consistent with the results of hardness($H_v$) and maximum displacement(${\delta}_{max}$).

  • In order to maximize spectrum efficiency and data rate MIMO(Multiple Input Multiple Output) is adopted to wireless system. OFDM-based WiMAX and LTE accommodate MIMO as mandatory technology. STC(Space Time Coding) and SM(Spatial Multiplexing) are used in downlink while in uplink C-MIMO(Collaborative MIMO) is used to improve data throughput. In this paper conventional pairing schemes, RPS(Random Pairing Scheduling) and DPS(Determinant Pairing Scheduling) are analyzed. From the analysis the performance of DPS algorithm is better than that of RPS because DPS measures orthogonal factor between paired users. However, there are potential problems such as hardware complexity and performance. To overcome the issues Power-Based Scheduling(PBS) algorithm is proposed for C-MIMO. PBS can provide higher performance compared to RPS and dramatically reduce hardware complexity compared to DPS

Simulation and analysis of DC characteristics in AlGaN/GaN HEMTs on sapphire, SiC and Si substrates (Sapphire SiC, Si 기판에 따른 AlGaN/GaN HEMT의 DC 전기적 특성의 시뮬레이션과 분석)

  • In this paper, we report on the 2D (two-dimensional) simulation result of the DC (direct current) electrical and thermal characteristics of AlGaN/GaN HEMTs (high electron mobility transistors) grown on Si substrate, in comparison with those grown on sapphire and SiC (silicon carbide) substrate, respectively. In general, the electrical properties of HEMT are affected by electron mobility and thermal conductivity, which depend on substrate material. For this reason, the substrates of GaN-based HEMT have been widely studied today. The simulation results are compared and studied by applying general Drift-Diffusion and thermal model altering temperature as 300, 400 and 500 K, respectively. With setting T=300 K and $V_{GS}$=1 V, the $I_{D,max}$ (drain saturation current) were 189 mA/mm for sapphire, 293 mA/mm for SiC, and 258 mA/mm for Si, respectively. In addition, $G_{m,max}$ (maximum transfer conductance) of sapphire, SiC, Si was 38, 50, 31 mS/mm, respectively, at T=500 K.

Development of Kinetic Models Describing Kinetic Behavior of Bacillus cereus and Staphylococcus aureus in Milk

  • This study developed predictive models to evaluate the kinetic behaviors of Bacillus cereus and Staphylococcus aureus in milk during storage at various temperatures. B. cereus and S. aureus (3 Log CFU/mL) were inoculated into milk and stored at $10^{\circ}C$, $15^{\circ}C$, $20^{\circ}C$, and $30^{\circ}C$, as well as $5^{\circ}C$, $15^{\circ}C$, $25^{\circ}C$, and $35^{\circ}C$, respectively, while bacterial populations were enumerated. The growth data were fitted to the modified Gompertz model to estimate kinetic parameters, including the maximum specific growth rate (${\mu}_{max}$; Log CFU/[$mL{\cdot}h$]), lag phase duration (LPD; h), lower asymptote ($N_0$; Log CFU/mL), and upper asymptote ($N_{max}$; Log CFU/mL). To describe the kinetic behavior of B. cereus and S. aureus, the parameters were fitted to the square root model as a function of storage temperature. Finally, the developed models were validated with the observed data, and Bias (B) and Accuracy (A) factors were calculated. Cell counts of both bacteria increased with storage time. Primary modeling yielded the following parameters; ${\mu}_{max}$: 0.14-0.75 and 0.06-0.51 Log CFU/mL/h; LPD: 1.78-14.03 and 0.00-1.44 h, $N_0$: 3.10-3.37 and 2.09-3.07 Log CFU/mL, and $N_{max}$: 7.59-8.87 and 8.60-9.32 Log CFU/mL for B. cereus and S. aureus, respectively. Secondary modeling yielded a determination of coefficient ($R^2$) of 0.926.0.996. B factors were 1.20 and 0.94, and A factors were 1.16 and 1.08 for B. cereus and S. aureus, respectively. Thus, the mathematical models developed here should be useful in describing the kinetic behaviors of B. cereus and S. aureus in milk during storage.
