  • 미생물막 유동층 반응기로 염료공장 폐수의 유기물 처리효율을 고찰하였다. 공장폐수를 2배, 3배 및 6배 희석하여 처리하였던 바, 각 희석배율 모두 F/M비가 0.2에서 0.3으로 증가할 때 처리효율이 급격히 낮아졌다. 한편, 같은 F/M비에서는 희석율이 높을수록 처리효율이 높았으며 수리학적 체류시간이 증가함에 따라서 처리효율도 증가하였다. 6배 희석시 F/M비가 0.2 이하에서 BOD 처리효율이 $90{\sim}97%$로 가장 좋았다. 또 반응기 설계를 위한 통역학적 계수 Y, $k_b$값 및 상관계수 r은 2배 희석시 $Y=0.3365\;k_d=0.03782\;day^{-1}\;r=0.997$, 3배 희석시 $Y=0.3341\;k_d=0.0275\;day^{-1}\;r=0.996$ 및 6배 희석시는 $Y=0.5460\;k_d=0.03434\;day^{-1}\;r=0.998$로 나타났다.

Production and Characterization of a Monoclonal Antibody against Human ${\beta}_2$-adrenergic receptor

  • The purpose of the present study was to produce and characterize a monoclonal antibody against human ${\beta}_2$-adrenergic receptor. Male BALB/c mice were immunized with glutathione S-transferase (GST) fusion protein of the C-terminal portion of the human ${\beta}_2$-adrenergic receptor which was expressed in E.Coli. The immunized splenocytes were fused with myeloma SP2/0-Agl4 cells. The resulting hybridomas were screened for the production of a monoclonal antibody which can recognize human ${\beta}_2$-adrenergic receptor, and then subcloned by limiting dilution. The resulting monoclonal antibody was named as mAb$\beta$CO2. The mono-clonal antibody $\beta$CO2 was determined as IgM subtype and then purified by anti-mouse IgM-agarose affinity chromatography. The results of ELISA, Western blot, and immunocytochemistry showed that mAb$\beta$CO2 recognized human ${\beta}_2$-adrenergic receptor in the ${\beta}_2$-adrenergic receptor-GST fusion protein and human spider-moid carcinoma cell line A431 with highly specific immunoreactivity, The monoclonal antibody $\beta$CO2 may provide useful tools for the study of the $\beta$-adrenergic receptor of human and other species including rats.

메탄연료의 점화특성에 미치는 연소 생성물 희석 및 벽면온도의 영향 (The Effects of Combustion Products Dilution and Wall Temperature on the Ignition of Methane Fuel)

  • The ignition characteristics in a confined axisymmetric coflow $CH_4$ jet were investigated numerically with the Fire Dynamics Simulator(FDS). The $CH_4$ fuel stream was diluted with main combustion product gases, such as $O_2$, $N_2$, CO, $CO_2$, and $H_2O$, and the mixed fuel stream was heated up to the sufficient temperature where a supplying fuel stream can be ignited. For the calculation of chemical reaction in the simulation, a 2-step global finite chemistry model was considered. Boundary condition for confined wall was optimized by investigating the effects of wall temperature on the ignition characteristics of fuel stream. In addition, the effects of composition of diluents in the fuel stream and fuel stream temperature on the ignition of fuel steam were investigated. The ignition characteristics of $CH_4$ stream with diluents were very sensitive to the wall temperature, composition of diluents in the fuel stream and fuel stream temperature.

강원도 옥계 금광상에 관한 광물학적·지화학적 연구 (Mineralogy and Geochemistry of the Ogkye Gold Deposits, Gangwondo Province)

  • Gold mineralization of the Ogkye gold mine was deposited mainly in quartz veins up to 150 cm wide which occupy fissures in Cambrian Pungchon limestone. Ore minerals are relatively simple as follows: pyrite, arsenopyrite, pyrrhotite, sphalerite, electrum and galena. On the basis of the Ag/Au ratio on ore grades, mode of occurrence and assoicated mineral assemblages, the Ogkye gold deposit can be classified as pyrite-type gold deposit (Group IIB). Fluid inclusion data indicate that ore minerals were deposited between $400^{\circ}$and $230^{\circ}C$ from relatively dilute fluids (0.2 to 7.3 wt.% eq. NaCl) containing $CO_2$. The ore mineralization resulted from a complex history of $CO_2$ effervescence and local concomitant boiling coupled with cooling and dilution of ore fluids. Gold deposition was likely a result of decrease of sulfur activity caused by sulfide deposition and/or $H_2S$ loss accompanying fluid unmixing. Sulfur isotope compositions of sulfide minerals (${\delta}^{34}S=3.5{\sim}5.9$‰) are consistent with ${\delta}^{34}S_{H_2S}$ value of 4.8 to 6.1‰, suggesting mainly an igneous source of sulfur partially mixed with wall-rock sulfur.

가막간 표층퇴적물 중의 금속 및 비금속 원소의 생지화확적 분포특성 (Biogeochemistry of Metal and Nonmetal Elements in the Surface Sediment of the Gamak Bay)

  • 양식활동이 활발한 반폐쇄적인 내만에서 퇴적물의 지화학적 특성을 파악하기 위하여 2010년 4월 가막만 전역의 19개 정점에서 표층 퇴적물을 채취하여 퇴적물 중의 금속(V, Cr, Mn, Fe, Co, Ni, Cu, Zn, Ag, Cd, Hg, Pb, As), 비금속(P, Se) 및 악티늄족 중 U원소의 분포특성을 파악하였다. 또한, 퇴적물기준 및 농축계수를 이용하여 금속원소의 오염도를 평가하였다. V, Cr, Fe, Co 및 Ni 등은 석영희석 효과, Cd 및 U은 유기물 희석효과, Mn, Ag, As 및 Se는 만의 북부 및 남부해역 정점들에서 표층퇴적물의 강한 환원환경경하에서 이산화망간 및 황산염이 유기물 분해의 산화제로 이용되면서 화학적 재분배에 의해 농도분포가 결정되어지는 것으로 보여진다. 미국 퇴적물 오염 기준(SQGs)에 의한 오염평가는 As의 경우 만의 중앙부를 제외한 대부분의 정점, Ni은 전 조사정점에서 ERL을 초과하였다. 농축계수(EF)를 이용한 평가는 Ni, Ag, Cd 및 As가 평균 EF가 1을 약간 초과하는 단계를 보였으며, 이외의 성분은 1과 유사하거나 혹은 그 이하 양호한 수준을 나타내었다.

  • A joining of dissimilar metal combination faces significant problems such as poor strength and cracking associated with brittle intermetallic compounds(IMC) formed. An application of laser allows low heat input; leading to less dilution and smaller heat affected zone. The $CO_2$ laser overlay was conducted on an AC2B alloy with feeding Fe-based powders. The overlay area was significantly influenced from the travel velocity rather than the powder feeding rate. The interface between the overlay and substrate consisted of the hard and brittle IMC($FeAl_3,\;Fe_3Al,\;Fe_2Al_5$), which initiating and propagating the crack. The reciprocating test for the slide wear was conducted on a multi-pass overlay experiment. Comparing with the multi-pass overlay with no overlap, the overlay with 50% overlap showed better wear resistance.

Highly Porous Tungsten Oxide Nanowires As Resistive Sensor for Reducing Gases

  • Gas sensor properties of $WO_3$ nanowire structures have been studied. The sensing layer was prepared by deposition of tungsten metal on porous single wall carbon nanotubes followed by thermal oxidation. The morphology and crystalline quality of $WO_3$ material was investigated by SEM, TEM, XRD and Raman analysis. A highly porous $WO_3$ nanowire structure with a mean diameter of 82 nm was obtained. Response to CO, $NH_3$ and $H_2$ gases diluted in air were investigated in the temperature range of $100{\sim}340^{\circ}C$ The sensor exhibited low response to CO gas and quite high response to $NH_3$ and $H_2$ gases. The highest sensitivity was observed at $250^{\circ}C$ for $NH_3$ and $300^{\circ}C$ for $H_2$. The effect of the diameters of $WO_3$ nanowires on the sensor performance was also studied. The $WO_3$ nanowires sensor with diameter of 40 nm showed quite high sensitivity, fast response and recovery times to $H_2$ diluted in dry air. The sensitivity as a function of detecting gas concentrations and gas sensing mechanism was discussed. The effect of dilution carrier gases, dry air and nitrogen, was examined.

수소/공기/희석제 혼합기의 점화지연과 화학반응 특성연구 (Characteristics of Chemical Reaction and Ignition Delay in Hydrogen/Air/Diluent Mixtures)

  • Hydrogen is considered a cleaner energy source than fossil fuels. As a result, the use of hydrogen in daily life and economic industries is expected to increase. However, the use of hydrogen energy is currently limited because of safety issues. The rate of combustion of the hydrogen mixture is about seven times higher than that of hydrocarbon fuels. The hydrogen mixture is highly flammable and has a low minimum ignition energy. Therefore, it presents considerable risks for fire and explosions in all areas of hydrogen manufacturing, transportation, storage, and use. In this study, the auto-ignition characteristics of hydrogen were investigated numerically for diluted hydrogen mixtures. Auto-ignition temperature, a critical property predicting the fire and explosion risk in hydrogen combustion, was determined in well-stirred reactors. When N2 and CO2 were used to dilute the hydrogen/air mixture, the ignition delay time increased with increasing dilution ratios in both cases. The CO2-diluted mixtures exhibited a longer ignition delay than the N2-diluted mixtures. We also confirmed that lower initial ignition temperatures increased the ignition delay times at 950 K and above. Overall, the auto-ignition characteristics, such as the concentrations of participating species and ignition delay times, were primarily affected by the initial temperature of the mixture.

Susceptibility of various Helicobacter pyloris to New Antiulcer Agents

  • Helicobacter pylori (H. pylori) is a microaerophilic spiral bacterium and infection by it in the human stomach causes gastritis, furthermore, it is considered to be involved in the pathogenesis of peptic ulcers and the development of gastric carcinoma. We assessed the inhibitory activity of new antiulcer drugs against Helicobacter pylori. The activities of new antiulcer agents against Helicobacter pylori strains were determined by the standard agar dilution method with blood agar base #2, supplemented with 5% sheep blood and 4 antibiotics to support growth of these organisms. They were inoculated by multipoint inoculator and incubated at 37$^{\circ}C$ for 3 days under microaerophilic atmosphere. The MIC of antiulcer agents was the lowest concentration that inhibited visible growth of these organisms. According to results of various biochemical tests, these bacteria were identified as Helicobacter pylori strains. And the MIC results showed that the strains were very susceptible to omeprazole and YJA20379s. Some of YJA20379s were more potent than omeprazole. These results suggest that our new antiulcer drugs have potent inhibitory activity against Helicobacter pylori, so that our new antiulcer drugs might be useful for the clinical eradication of gastrointestinal Helicobacter pylori.

  • 서울 인근의 생활하수에서 메탄올을 이용하는 박테리아를 분리하였다. 이의 최적 성장을 위한 온도 및 pH는 각각 $33^{\circ}C$ 및 7.1이었다. 최대비성장율은 $0.42hr^{-1}$을 나타내었다. 최소 배지조성은 기본배지의 양을 안정상테에서 연속발효법으로 구하였으며, 그 조성은 다음과 같다(g / l); Methanol 40, $(NH_4)_2\;SO_42,\;KH_2PO_4\;1.5,\;K_2HPO_4\;0.2,\;H_3PO_4\;0.79,\;Na_2HPO_4{\cdot}12H_2O\;0.15,\;MgSO_4{\cdot}7H_2O\;1.5,\;FeSO_4{\cdot}7H_2O\;0.034,\;MnSO_4{\cdot}4H_2O\;0.005,\;CuSO_4{\cdot}5H_2O\;0.0027,\;CaCl_2{\cdot}2H_2O\;0.25,\;ZnSO_4{\cdot}7H_2O\;0.007,\;(NH_4)_6\;Mo_7O_{24}{\cdot}4H_2O\;0.00048,\;H_3BO_3\;0.00068,\;CoCl_2\; 0.00024$ 최적 배지 조건에서 구한 최대 세포생산성은 3.8g / l / hr였으며, 이때의 회석율은 $0.23hr^{-1}$이었다. 최대 세포농도와 그대의 단백질의 함량은 각각 19.5g / l 및 70%였으며, 이때의 회석율은 $0.1hr^{-1}$이었다.
