The purpose of this study was to determine optimum value of aeration, acetate dosage, and $CO_2$ input for the cultivation of Scenedesmus acuminatus. Highest specific growth rate and maximum biomass productivity was obtained by the aeration of 0.72 vvm and lower specific growth rates and maximum biomass productivity were obtained for other aeration tests. When putting 0.3 M of ammonium acetate in JM medium, the highest specific growth rate and maximum biomass productivity were obtained. $CO_2$ input tests were performed during semi-continuous culturing tests. The highest specific growth rate ($0.460d^{-1}$) and maximum biomass productivity ($0.936gL^{-1}d^{-1}$) were obtained after replacing 50% of solution with 0.3 M of acetate solution for $CO_2$ input tests. However, more dilutions after the first dilution resulted in lower specific growth rate and maximum biomass productivity. In aeration tests, the highest specific growth rate ($0.381d^{-1}$) and maximum biomass productivity ($0.253gL^{-1}d^{-1}$) were obtained when cultivating it with JM medium, but the specific growth rate and maximum biomass producitivty were significantly decreased when 50% of solution was replaced by acetate containing solution.
Korean wastewaters have higher nitrogen concentrations than typical wastewaters of other countries. Most treatment processes such as activated sludge processes will need to supplement extra carbon sources for a complete removal of remaining nitrogen after the initial wastewater treatment, Because of these difficult matters, we have searched wastewater treatment methods that require no additional carbon sources. Wastewater treatment by microalgae in photobioreactors, using a green eukaryotic microalgae, Chlorella kessleri, showed a promising results and thus was selected to study further. This system is not intended to replace the conventional system but is to assist the existing biological treatment systems as a supplemental nitrogen removal process. Thus the secondary treated livestock wastewater was tested. Column type photobioreactors developed in our laboratory were used. When aerated with 5% CO$_2$ balanced with air at 1 vvm and illuminated at 100 ${\mu}$mol/㎡/s under 25$^{\circ}C$ and PH 7-8 by CO$_2$ buffering effect, the maximum nitrogen removal rate was 2.6 mg/L/hr. The results confirmed a possibility of microalgal wastewater treatment system as a secondary system to remove extra nitrogen sources. Based on these experimental results, the size of the optimal microalgal wastewater system was calculated. For the wastewater whose initial nitrogen concentration of 150 mg/L, the optimal batch system was found to be a 2 stage system with a combined retention time of 4.6 day. From the continuous experiments, nitrogen removal rates were examined under different dilution rates and 2 stage system was also found to be the optimal system. The combined retention time for the continuous system was 3.5 days. It is expected that conventional biological wastewater treatment systems followed by microalgal systems would reliably decrease the nitrogen concentration below the government criteria even for the livestock wastewater with low C/N ratio.
This study was conducted to determine the serum antibodies against toxoplasma in swine from breeding-pig farm, pig farm and abattoir by latex agglutination(LA) test. LA test was carried out with commercial Toxo-MT kit (Eiken chemical co.). The results obtained were summerized as follows : 1. The cut-off titer of positive and negative reactions by Toxo-MT antigen used in this experiment was determined as the serum dilution of 1 ; 32. 2. positive rates of toxoplasma antibodies in 823 swine sera were 17.0%(140 cases) by LA test. 3. The toxoplasma antibody detection rates against 194 swine sera in breeding-pig farm, 273 swine sera in pig farm and 356 swine in abattoir were 46.9%(91 cases), 8.4%(23 cases) and 7.3% (26 cases) , respectively. 4. In LA test serum antibody titers in 823 test sera were shown as 51 cases (36.4%) in 1 : 32, 40(28.6%) in 1;64, 17(12.1%) in 1:128, 14(10.0%) in 1:256, 10(7.1%) in 1:512, 5(3.6%) in 1:1,024, and 3(2.1%) in 1 : 2,048. 5. Positive rates of toxoplasma antibodies in swine sera from each breeding-pig farm were 20.0∼61.9%.

The ultimate objective of this study is to develop a reliable oxygen-enriched combustion techniques especially for the case of the flue gas recycling in order to reduce the $CO_2$ emissions from practical industrial boilers. To this end a systematic numerical investigation has been performed, as a first step, for the resolution of the combusting flame characteristics of lab-scale LNG combustor. One of the important parameters considered in this study is the level of flue gas recycling calculated in oxygen enriched environment. As a summary of flame characteristics, for the condition of 100% pure $O_2$ as oxidizer without any flue gas recycling, the flame appears as long and thin laminar-like shape with relatively high flame temperature. The feature of high peak of flame temperature is explained by the absence of dilution and heat loss effects due to the presence of $N_2$ inert gas. The same reasoning is also applicable to the laminarized thin flame one, which is attributed to the decrease of the turbulent mixing. These results are physically acceptable and consistent and further generally in good agreement with experimental results appeared in open literature. As the level of $CO_2$ recycling increases in the mixture of $O_2/CO_2$, the peak flame temperature moves near the burner region due to the enhanced turbulent mixing by the increased amount of flow rate of oxidizer stream. However, as might be expected, the flue gas temperature decreases due to presence of $CO_2$ gas together with the inherent feature of large specific heat of this gas. If the recycling ratio more than 80%, gas temperatures drop so significantly that a steady combustion flame can no longer sustain within the furnace. However, combustion in the condition of 30% $O_2/70% $$CO_2$ can produce similar gas temperature profiles to those of conventional combustion in air oxidizer. An indepth analyses have been made for the change of flame characteristics in the aspect of turbulent intensity and heat balance.
The effects of seeding method and optimum time for freezing embryos according to the developmental stages on embryo survival rates after rapid freezing were determined using the FDA-test. The summarized results are as follows : 1. In the rapid freezing of embryos, the sucrose added medium together with Co-seeding or non-seeding showed the FDA scores of 4.67 and 4.20, respectively, but, raffinose addition obtained FDA scores of 4.27 and 3.97. 2. The developmental stage of embryos at freezing was most critical on the survival of embryos after thawing. Higher FDA scores were obtained in the order of blastocyst stage(4.94), morula stage(3.82) and ealy stage(2.65) in sucrose added medium. The same trend was observed in the raffinose added medium with an order or 4.91, 4.47 and 2.32. 3. Microscopic study of embryo before freezing and post-thawing indicated that the embryo showed shrinkage within 5 minutes after the embryo was transfer to the freezing medium. When thawed embryo was tranfered to the dilution medium, swelling of the embryo was observed and there after it reshrank indicating the removal of cryoprotectant from the embryo. The size of the embryo recovered to the original state when it was moved into a PBS-solution.
Inductively Coupled Plasma Dynamic Reaction Cell Quadrupole Mass Spectrometry (ICP-DRC-QMS) was characterized for the detection of the six naturally occurring calcium isotopes. The effect of the operating conditions of the DRC system was studied to get the best signal-to-noise ratio. This experiment shows that the potentially interfering ions such as $Ar^+$, ${CO_2}^+$, ${NO_2}^+$, $CNO^+$ at the calcium masses m/z 40, 42, 43, 44 and 48 were removed by flowing $NH_3$ gas at the rate of 0.7 mL/min $NH_3$ as reactive cell gas in the DRC with a RPq value (rejection parameter) of 0.6. The limits of detection for $^{40}Ca$, $^{42}Ca$, $^{43}Ca$, $^{44}Ca$, and $^{48}Ca$ were 1, 29, 169, 34, and 15 pg/mL, respectively. This method was applied to the determination of calcium in synthetic food digest samples (CCQM-P13) provided by LGC for international comparison. The isotope dilution method was used for the determination of calcium in the samples. The uncertainty evaluation was performed according to the ISO/GUM and EURACHEM guidelines. The determined mean concentration and its expanded uncertainty of calcium was ($66.4{\pm}1.2$) mg/kg. In order to assess our method, two reference samples, Riverine Water reference sample (NRCC SLRS-3) and Trace Elements in Water reference sample (NIST SRM 1643d), were analyzed.
For further removal of non-biodegradable CODs and color in biologically treated distillery waster water, we selected a chemical treatment with Fe(III) and cationic polymers and then another chemical treatment with Fenton reagent. We developed Pregenerated Bubble Flotation(PBF) to effectively remove the chemical sludge from each chemical reaction process. The flotation unit was constructed with hydraulic loading rate, 7 ㎥/$m^2$.hr. The CODMn and suspended solids (SS) in biologically treated distillery waste water were reduced by the first PBF from 310-1096 mg/L to 141-303 mg/L and from 160-990 mg/L to 48-385 mg/L, respectively. Again, after the Fenton reaction process, floated SS was skimmed off at the top of the flotation unit and the final effluent was directly discharged without any tap water dilution. The quality of final effluent can be below 40 mg/L-CODMn but IISan Distilery has been maintained effluent quality of 73 mg/L-CODMn and 10-80 mg/L-SS. The chemical cost was saved by more than 30% as compared with that of prior process.

The immune reinforcement of the probiotic lactic acid bacteria Lactobacillus acidophilus, Streptococcus thermophilus and Bifidobacterium bifidum was studied in RAW 264.7 cell line treated with diluted solution (dilution to $2^{5}$) of the supernatnats of lactic acid bacteria. RAW 264.7 cell line was used as a macrophage model to assess the effects of lactic acid bacteria on the production of nitric oxide (NO) and cytokine tumor necrosis factor (TNF)-$\alpha$ and cell morphological changes. The production of NO and TNF-$\alpha$ were largely affected by lactic acid bacteria in dose-dependent manner in 24 or 48 hr cultures and cell morphological changes were also largely affected by lactic acid bacteria. Especially Bifidobacterium bifidum differentially stimulated the production of NO and TNF-$\alpha$. NO production was increased by Bifidobacterium bifidum 25 $\mu$l/ml more than LPS (20 ng/ml) control, and TW-$\alpha$ by Bifidobacterium bifidum 6.25 $\mu$l/ml more than LPS (10 ng/ml) control. The in vitro approaches employed here should be useful in further characterization of the effects of lactic acid bacteria on systemic immunity.
There have been a number of studies of methods for recycling animal wastewater to provide new bioresources. In the present work, a marine algal culture medium, designated KEP II, was prepared by adding swine waste (3% v/v) fermented by soil bacteria to a dilution of f/2 culture medium (CT). When Phaeodactylum tricornutum was grown in batch culture in KEP II, the cells lasted long at the exponential phase producing the specific growth rate and biomass; the production of total amino acids and secondary metabolites rose up to 5-fold. It also substantially enhanced the maximum quantum yield of photo system (PS) II of P. tricornutum, greatly increased the level of thylakoid membranes containing PS, and stimulated the production of pyrenoids, including enzymes for $CO_2$ fixation in chloroplasts. KEP II should improve the cost efficiency of industrial mass batch cultures and the value of microalgae for long-term preservation of fresh aquaculture feed as well as production of anticancer and antioxidant agents. Specifically, a low-cost medium for growing the diatoms of aquaculture feed will be economically advantageous.
For the development of high efficiency and low emission combustion systems, high temperature air combustion technology has been tested by utilizing preheated air over 1100 K and exhaust gas recirculation. In this system, combustion air is diluted with large amount of exhaust gases ($N_2$, $CO_2$), such that the oxygen concentration is relatively low in the reaction zone, leading to low flame temperature. Since, the temperature fluctuations and sound emissions form the flame are small and flame luminosity is low, the combustion mode is expected to be flameless or mild combustion. Experiment was performed to investigate the turbulent flame structure and $NO_X$ emission characteristics in the high temperature air combustion focused on coflowing jet diffusion flames which has a fundamental structure of many practical combustion systems. The effect of turbulence has also been evaluated by installing perforated plate in the oxidizer inlet nozzle. LPG was used as a fuel. Results showed that even though $NO_X$ emission is sensitive to the combustion air temperature, the present high temperature air combustion system produce low $NO_X$ emission because it is operated in low oxygen concentration condition in excess of dilution.
