In continuing search for bioactive compounds from Korean marine algae, we found cholinesterase-inhibitory activity in the methanolic extract of brown alga Sargassum sagamianum. After partitioning between CHCl$_3$ and 30% MeOH, the former layer was purified by a series of ODS flash, silica column, gel-filtration on Sephadex LH-20, and HPLC to give two farnesylacetone derivatives. Their structures were identified by comparison with the literature data. Compounds 1 and 2 showed moderate acetylcholinesterase and butyrylcholinesterase inhibitory activities with IC$\sub$50/ values of 65.0∼48.0 ${\mu}$M and 34.0∼23.0 ${\mu}$M, respectively. (omitted)

In our search for bioactive phenolics from plants, the culms of Phyllostachys bambusoides has been selected for investigation of anti-cariogenic and 1,1-diphenylpicrylhydrazyl (DPPH) radical scavenging agents based on the initial screening results. Fractionation process of n-hexane and $CHCl_3$ extracts afforded four phenolic constituents, ferulic acid (1), vanillin (2), coniferaldehyde (3), and coniferyl alcohol (4) as guided by their DPPH free radical scavenging activities. Additionally, activity-guided fractionation of EtOAc extract with anti-cariogenic activity has resulted in the isolation of coniferaldehyde (3), 2,6-dimethoxy-p-benzoquinone (5), p-methoxycinnamic acid (6), (${\pm}$)-balanophonin (7), and 6-methoxychromanone (8). The structures of 1 - 8 were determined by spectroscopic data interpretation, and also by comparison of their data with the published values. Phenolic compounds 1 - 4 exhibited similar DPPH radical scavenging activities compared with the synthetic antioxidant, butylated hydroxytoluene (BHT), and compounds 3 and 5 - 8 showed significant antibacterial activity against cariogenic oral streptococci, Streptococcus mutans and S. sobrinus.
Activation of hepatic stellate cells (HSCs) characterized by increased proliferation and extracellular matrix deposition is identified as the major pathological feature of hepatic cirrhosis. Therefore, suppression of HSC activation has been proposed as an important antifibrotic therapeutic strategy. In the present study, we investigated the antiproliferative activity of root barks of Ulmus davidiana var. japonica (Ulmaceae) by employing HSC-T6 hepatic stellate cells as an in vitro assay system. Further investigation of the n-hexane and $CHCl_3$ fractions of root barks of U. davidiana var japonica led to the isolation of six triterpenoids: friedelin (1), epifridelanol (2), oleanolic acid (3), maslinic acid (4), ${\beta}$-amyrin (5) and ${\alpha}$-amyrin (6), together with ${\beta}$-sitosterol (7) and daucosterol (8). Among these compounds, 2, 3 and 4 significantly inhibited HSC proliferation. In addition, 4 inhibited HSC proliferation in time- and concentration-related manners, via a partially direct toxic effect, as assessed by morphological changes and release of lactate dehydrogenase.
In search for plant-derived cytotoxic compounds, it was found that the $CHCl_3$ and EtOAC extracts obtained from the flowers of Paulownia coreana Uyeki (Scrophulariaceae) exhibited significant cytotoxic activity against human tumor cell lines, A549, SK-OV-3, SK-MEL-2, XF498, and HCT15. Activity-guided fractionation on the basis of the inhibitory activity against the growth of human tumor cell lines, in vitro, and repeated column chromatography afforded several cytotoxic compounds from P. coreana. The structures and stereochemistry of these compounds were established, on the basis of analysis of spectra including IR, UV, EI-MS, $^{1}H-NMR,\;^{13}C-NMR$ and some chemical transformations, as Compound PCCl $(2-hydroxy-4(15),11(13)-eudesmadien-8{\beta},12-olide)$, Compound $PCC2(2,3-dihydro-4-hydroxy-1(15),11(13)-xanthadien-8{\beta},12-olide)$, Compound PCE1 (chrysophanol), Compound PCE2 (emodin), Compound PCE3 (physcion). Cytotoxic activity of compounds obtained from P. coreana. on five tumor cells lines was evaluated by procedure of SRB methods.
염화탄화수소의 고온열분해 반응에서 생성물 반응경로 특성을 파악하기 위해 $H_2$ 반응분위기에서 1,1-dichloroethylene($CH_2CCl_2$) 열분해반응 실험을 수행하였다. 열분해반응 실험은 등온관형반응기를 이용하여 반응온도 $650{\sim}900^{\circ}C$, 반응시간 0.3~2.0초에서 진행하였으며, 반응물 mole 분율은 전체 실험에서 $CH_2CCl_2:H_2$ = 4:96 일정하게 유지하였다. 반응물 $CH_2CCl_2$가 완전분해온도는(분해율 99% 이상) $825^{\circ}C$(반응시간 1초 기준)였으며, $H_2$ 반응분위기에서 $CH_2CCl_2$ 주요 분해반응경로는 H원자 추출 및 첨가교체치환 연쇄반응으로 파악되었다. $CH_2CCl_2$가 46% 분해되는 $700^{\circ}C$에서 1차 생성물로 $CH_2CHCl$가 28%로 가장 높은 농도로 검출되었다. $775^{\circ}C$ 이상에서는 탈염소화된 $C_2H_4$가 2차 생성물로 가장 많이 생성되었으며, 반응온도가 증가할수록 염소 원자수가 작은 화합물이 생성되었으며 이들 화합물은 열화학적으로 안정된 물질이다. $825^{\circ}C$ 이상의 고온반응영역에서 탈염소반응의 부산물인 HCl과 $C_2H_4$, $C_2H_6$, $CH_4$$C_2H_2$ 등과 같은 열화학적으로 안정된 탄화수소가 주요생성물로 검출되었다. 본 연구에서 고찰된 반응계에서 분해와 생성물분포 특성을 고려하고 열화학이론 및 반응속도론을 기초로 주요 반응경로를 제시하였다.
삼백초는 항산화 활성을 갖는 플라보노이드 화합물을 포함하고 있는 여러해살이 식물로 알려져 있다. 본 연구에서는 삼백초 물 추출물과 유기용매 분획물의 항산화 활성, 항염증 활성 및 PMA에 의해 유도된 MMP-2 및 MMP-9활성 억제 효과를 조사하였다. 시료들은 삼백초 물 추출을 헥산(hexane), 클로로포름(CHCl3), 에틸 아세테이트(Ethyl acetate), 부탄올(butanol) 및 물(water)과 같은 용매로 분획화하여 사용하였고, 물 분획물의 수득율이 9.25%로 가장 높았다. 항산화 활성은 DPPH assay, 세포 생존율 측정은 MTS assay, 항염증 활성은 마우스 대식세포 Raw 264.7세포에서NO 생성 그리고 MMP-2 및 MMP-9의 mRNA 발현 및 단백질 활성 억제는 인간구강편평세포암종 YD-10B 세포에서 RT-PCR과 zymography방법을 통해 측정하였다. 본 연구의 결과에 의하면 MMP-2/-9 활성은 PMA에 의해 YD-10B세포에서 증가하였다. PMA 처리된 YD-10B 세포에서, 에틸 아세테이트 분획물이 증가된 MMP-2/-9의 mRNA 발현 및 단백질 활성을 유의하게 억제하였다. 그리고 항산화 활성도 에틸아세트 분획물이 73.38%의 가장 높게 나타났다. 또한 세포 독성을 나타내지 않는 농도에서, 에틸아세트 분획물이 Raw 264.7세포에서 농도 의존적으로 유의하게 항염증 활성을 보였다. 그러므로 본 연구에서는 삼백초 물 추출의 에틸아세트 분획물이 구강암의 암 침윤을 억제할 수 있는 효과적인 암 예방 및 치료를 위한 항암제로서의 가능성을 암시하고 있다.
1,1,1,2-수소불화탄소(HFC-134a)는 에어컨에 주로 사용되는 냉매로, 최근 온실가스로 규제되어 정제를 통한 재사용 방법이 권장되고 있다. 폐냉매의 재사용 기준 평가를 위해서는 폐냉매에 존재하는 미량 성분의 정량분석이 매우 중요하다. 본 연구에서는 표준 물질이 없어서 정량화하기 어려웠던 C, H, Cl, F가 포함된 미량 성분들을 GC/AED (gas chromatograph-atomic emission detector)를 이용하여 정량분석하였다. 이를 위하여 GC/MSD (mass selective detector)를 통한 정성분석을 선행하였다. 또한 성분의 원자 수와 비례하여 반응하는 AED의 특성을 조사하기 위하여, 탄화수소 혼합 표준물질을 이용하여 선형성을 확인하였다. 시료 중 C, H, Cl, F가 포함된 미량 성분의 정성 분석 결과, 주성분인 HFC-134a와 유사 냉매류들을 포함한 총 15 개의 성분이 검출되었다. MSD 결과를 토대로 AED를 이용한 미량 성분들을 정량 분석한 결과, 한 시료는 $CHClF_2$ 성분($45438.38{\mu}mol/mol$), 또 다른 시료는 $C_2H_2ClF_3$ 성분($1311.47{\mu}mol/mol$)이 가장 높은 몰분율을 나타냈다. 본 연구에서는 이 분석법을 기반으로 하여, 표준 물질이 존재하지 않아 정량화하기 어려운 복합 성분들의 정성 및 정량 분석의 확장 적용이 가능할 것으로 보인다.
This study was carried out to examine the behavior of THM formation in water treated with chlorine dioxide where humic acid was used as THM precursor. THM was not detected in bromide-free water, but formed in water containing bromide. When 10 mg/l of chlorine dioxide was added to water containing 5 mg/l of humic acid and bromide respectively, 20.46 ${\mu}$g/l of THM was formed. It is postulated that chlorine dioxide oxidize bromide to hydrobromous acid, which subsequently reacted with humic acids similar to chlorine reaction. The formation of THM could be reduced at low pH. Among THM formed, CHBr$_3$ was the predominant species in the alkaline solution, while CHCl$_3$ in the acidic solution. A sample pretreated with chlorine dioxide for 24h before addition of chlorine showed a reduction of 75.1% in THM formation, compared with a sample not pretreated with chlorine dioxide and a sample treated by chlorine for 24h prior to addition of chlorine dioxide also showed a reduction of 37.8% in THM formation, compared with a sample not added with chlorine dioxide. It may explain that chlorine dioxide oxidizes directly a fraction of THM.
The aerial part of Siegesbeckia pubescens (Compositae) has been used to treat rheumatoid arthritis and hypertension in the Oriental medicine. This crude drug has been used without process (SP-0) or with three times-process of steaming and drying (SP-3) or the nine times of that process (SP-9). To search for the antinociceptive anti-inflammatory components from this crude drug, activity-directed fractionation was performed on this crude drug. Since the $CHCl_3$ extract was shown to have a more potent effect than other extracts, it was subjected to silica gel & ODS column chromatography to yield two diterpene compounds (1). Compound 1 was structurally identified as ent-16 (H, 17-hydroxykauran-19-oic acid, which were tentatively named siegeskaurolic acid A. A main diterpene, siegeskaurolic acid A was tested for the antiiflammatory antinociceptive effects using both hot plate- and writhing anti-nociceptive assays and carrageenan-induced anti-inflammatory assays in mice and rats. Pretreatment with siegeskaurolic acid A (20 and 30mg/kg) significantly reduced the stretching episodes, action time of mice and carrageenan-induced edema. These results support that siegeskaurolic acid is a main diterpene responsible for antinociceptive and antiiflammatory action of S. pubescens. In addition, the assays on SP-0, SP-3 and SP-9 produced the experimental results that SP-9 had more significant effects than other two crude drugs. These results suggest that the processing on the original plant may lead to the higher pharmacological effect.
