대황황련해독탕의 사염화탄소 유발 간장해 보호효과 및 급성독성 (Effect of Daewhang-whangryunhaedok-Tang on Carbon Tetrachloride-induced Hepatotoxicity and Acute Toxicity)

  • This study was performed to evaluate hepatoprotective effect of Daewhang-whangryunhaedok-Tang(DWT) on liver injured rats induced by $CCl_4$ and the acute oral toxicity of it in mice. The activities of serum transaminase(ALT/AST), alkaline phosphatase(ALP) and lactic dehydrogenase (LDH), the levels of serum total cholesterol(TC) and triglyceride(TG), change of liver enlargement, and inhibitory activities of lipid peroxidation, catalase and glutathione-S-transferase(GST) in liver microsome were determined in hepatotoxic rats induced by $CCl_4$ DWT DWT was significantly reduced the serum ALT, AST, ALP, LDH, TC and TG levels. And, the increase of lipid peroxidation, decrease of catalase and GST activities in the liver microsome of $CCl_4$-intoxicated rat were significantly improved by the treatment of DWT. Male and female mice were administered maximum dosages of 5,000 mg/kg b.w. of DWT. After single oral administration of DWT to mice, we observed them daily for 2 weeks. DWT did not induce any toxic signs in the mortalities, clinical signs, body weight changes, and gross necropsy findings of mice. Based on these results, it is concluded that DWT may have the hepatoprotective effect on $CCl_4$ induced hepatotoxicity in rats. Also, DWT may have no side effect and its $LD_{50}$ value may be over 5,000 mg/kg b.w. in mice.

Synergetic Hepatoprotective Effects of Korean Red Ginseng and Pueraria Radix on the Liver Damaged-Induced by Carbon Tetrachloride (CCl4) in Mice

  • This study was designed to investigate the synergetic hepatoprotective effects from a mixture of Korean Red Ginseng and Pueraria Radix on carbon tetrachloride ($CCl_4$)-induced hepatotoxicity in mice. Liver toxicity was induced by intraperitoneal administration of $CCl_4$ (0.6 mg/kg) in 12 groups of ICR mice. The negative control group was given $CCl_4$ without test samples and the normal group was given no treatment. Among treatment groups, the RGAP treatment (Korean Red ginseng acetic acid extract : Pueraria radix water extract, w/w, 38.4:57.6) decreased $CCl_4$-elevated ALT (101.60 IU/L), AST (833.89 IU/L), and LDH (365.02 IU/L) levels in the serum, and increased the SOD (11.03 unit/mg protein) and CAT (0.37 unit/mg protein) levels and the LPO levels ($59.09{\mu}M/g$ tissue) more than that in the mice group with $CCl_4$-induced control group hepatotoxicity. These results suggest that administration of a mixture of Korean Red ginseng and Pueraria radix decreases $CCl_4$-induced liver damage and enhances antioxidant activity in mice and imply that administration of the mixture in a certain ratio is more effective than single administration of either Korean Red ginseng or Pueraria radix alone.

Hepatoprotective Effect of Grifola frondosa Water Extract on Carbon Tetrachloride-induced Liver Injury in Rats

  • The present study aimed at assessing the protective effect of water extract from fruit body of the Grifola frondosa (GFW) on carbon tetrachloride ($CCl_4$)-induced hepatotoxicity. Rats orally administered with GFW 0.5, 1.0, 2.0 g/kg for 14 days were treated with $CCl_4$ to induce hepatotoxicity. Pretreatment with GFW remarkably prevented the elevation of serum AST, ALT, ALP, LDH, $\gamma$-GTP, and liver lipid peroxides in $CCl_4$-treated rat and GFW administration in liver injured rats by $CCl_4$ showed significant (p<0.05) protection of liver as evidenced from normal serum enzymes and malondialdehyde (MDA) levels. In the ultrastructural changes, administration of $CCl_4$-induced damage of hepatocytes with vacuolation, a highly damaged endoplasmic reticulum, and degenerating nuclei. However, pre-administration with GFW preserved normal ultrastructure of hepatocytes. These results suggest that GFW had an effect to inhibit $CCl_4$-induced liver injury in rat, and that it could be used as an effective hepatoprotective agent against chemical-induced liver damage.

초임계 이산화탄소를 이용한 돌나물 추출물이 사염화탄소로 유발된 흰쥐의 지질대사, 지질과산화 및 항산화에 미치는 영향 (The Effects of Sedum sarmentosum Bunge Extract using Super Critical Carbon Dioxide on Lipid Metabolism, Lipid Peroxidation and Antioxidation in Carbon Tetrachloride Induced Hepatotoxicity in Rats)

  • Extraction of Sedum sarmentosum Bunge by super critical carbon dioxide was operated under $40-50^{\circ}C$and 200-250 atm, thus, yield of extraction was very low as 4 wt%. Rats were administrated with the extract orally once a day for succesive 6 days, followed by treatment with carbon tetrachloride ($CCl_4$) on the sixth day. The activities of aminotransferase, alkaline phosphatase, ${\gamma}$-glutamyl transpeptidase, lactate dehydrogenase and contents of triglyceride, total cholesterol in the extract-pretreated rats were decreased compared to the $CCl_4$controled rats, whereas content of HDL-cholesterol was increased. Especially content of hepatic malondialdehyde (MDA) and atherogenic index (AI) were decreased and HTR was increased in the extract-pretreated rats, and reduced peroxidative liver damage in the $CCl_4$-induced hepatotoxicity rats. In addition, activities of hepatic superoxide dismutase, catalase, glutathione peroxidase in the extract-pretreated rats were significantly decreased compared to the $CCl_4$ controled rats, but the content of glutathione was significantly increased. These results suggest that extract of Sedum sarmentosum Bunge has hepatoprotective effect in the $CCl_4$-intoxicated rats.

양릉천 저주파 전침자극이 $CCl_4$로 유발된 흰쥐의 간 손상에 미치는 영향 (Hepatoprotective effect of electroacupuncture at GB34 (Yangreungcheon) in $CCl_4$-intoxicated rats)

  • Objectives : To investigate the effect of electro-acupuncture (EA) at GB34 on hepatotoxicity in $CCl_4$-intoxicated rats. Methods : Rats were injected with $CCl_4$ and treated with acupuncture or 2 Hz electro-acupuncture (EA) at left GB34 three times a week for 10 weeks. A non-acupoint in left gluteal area was selected as a sham point. To estimate the effects of EA on hepatotoxicity in rats, body weight, liver weight and liver index were measured, and biochemical assays for serum ALT, AST, ALP and total cholesterol, and hematological analysis for RBC, WBC, PLT, hemoglobin, lymphocytes, neutrophils and monocytes, and histology analysis of liver tissue were performed. Results : 1. Lymphocyte level in blood was significantly decreased by $CCl_4$-intoxication and significantly increased by acupuncture and 2 Hz EA at left GB34. 2. Neutrophill and monocyte level in blood was increased by $CCl_4$-intoxication and significantly reduced by acupuncture and 2 Hz EA at left GB34. 3. Acupuncture and 2 Hz EA at left GB34 significantly reduced serum ALT and AST which were increased by $CCl_4$-intoxication. 4. EA at GB34 significantly reduced serum ALT and AST as compared with EA at sham point in $CCl_4$-intoxicated rat. 5. No significant difference was found between the effects of acupuncture and that of 2 Hz EA on $CCl_4$-induced liver damage in rats. Conclusions : 2 Hz EA at GB34 has hepatoprotective effects on $CCl_4$-induced liver damage in rats and the point-specificity of GB34 may be involved in these effects.

Evaluation of the Efficacy of Kochiae fructus Extract in the Alleviation of Carbon Tetrachloride-induced Hepatotoxicity in Rats

  • Hepatoprotective effects of the extract of Kochiae fructus (KF), a traditional oriental medicinal plant, were evaluated against carbon tetrachloride($CCl_4$)-induced liver damage in rats. Male Sprague-Dawley rats were divided into control, $CCl_4,\;CCl_4$ plus methanol extract of KF (KFM-$CCl_4$), and $CCl_4$ plus butanol extract of KF (KFB-$CCl_4$) groups. KFM and KFB were orally administered once a day (200 mg/kg body weight) for 14 days. A mixture of 0.2 mL/100 g body weight of $CCl_4$ in olive oil was injected at 30 minutes after the final administration of KFM and KFB. The KFB pretreatment resulted in a significant decrease in the serum transaminase and lactic dehydrogenase levels in the $CCl_4$-treated rats. The $CCl_4$ treatment significantly lowered the activities of glutathione, glutathione reductase (GR), glutathione-S-transferase (GST), superoxide dismutase (SOD), catalase and glutathione peroxidase (GSH-Px). However, pretreatment with KFM and KFB resulted in a significant increase in the glutathione, GR and GST levels. KFB increased the activities of SOD, catalase and GSH-Px, but KFM did not alter them. Pretreatment with KFM and KFB resulted in a significant decrease in the production of aminopyrine N-demethylase in the $CCl_4$-treated rats. KF extract would appear to contribute to alleviate the adveISe effect of $CCl_4$ treatment by enhancing the hepatic antioxidant defense system.

홍화자 분획물의 사염화탄소 유발 간손상 흰쥐에서 지질과산화와 oxygen free radical 제거 효소 활성도에 미치는 영향 (Effect of Subfractions of Carthamus tinctorius L. Semen on the Lipid Peroxidation and Oxygen Free Radical Scavenging Enzyme Activities in CCl_4$-induced Hepatotoxic Rats)

  • Previous studies have shown that methanol extract and its butanol fraction of Carthamus tinctorius L. Semen have the hepatoprotective effect on the CCl4-induced hepatotoxicity. The hepatoprotective effect of the subfractions of butanol fraction has been evaluated by analyzing oxygen free radical scavenging enzyme activities and histopathological examinations. In BS-5 subfraction treated group, the activity of superoxide dismutase has been significantly increased as compared with that of CCl4, treated rats. Antioxidant activity has been evaluated by the examination of the scavenging activity against 1,1-diphenyl-2-picrylhydrazyl radical. BS-5 subfraction has shown strong antioxidant activities. The histopathological examination showed that the treatment of BS-5 subfraction has relieved the ballooning degeneration of hepatocytes which had been generated by CCl4. It appears that the protective effect of BS-5 subfraction would be mediated of the attenuation of lipid peroxidation by acting as a free radical scavenger, which were based on the increase of superoxide dismutase activity.

사염화탄소 유도 급성 간독성 모델에서 치자의 간 보호 효과 (Protective Effect of Gardenia jasminoides Against Carbon Tetrachloride-Induced Acute Hepatotoxicity)

  • Gardenia jasminoides is one of the most widely used herbal preparations for the treatment of liver disorders. This study evaluated the potential beneficial effect of G. jasminoides in a mouse model of carbon tetrachloride ($CCl_4$)-induced liver injury. The mice were treated intraperitoneally with $CCl_4$ (10 ${\mu}l$/kg). They received G. jasminoides (30, 100, 300 mg/kg) 48 h, 24 h and 2 h before and 6 h after administering $CCl_4$. The serum activities of aminotransferase and the hepatic level of malondialdehyde were significantly higher 24 h after the $CCl_4$ treatment, while the concentration of reduced glutathione was lower. These changes were attenuated by G. jasminoides. $CCl_4$ increased the level of circulating tumor necrosis factor-$\alpha$ (TNF-$\alpha$) markedly, which was reduced by G. jasminoides. The levels of hepatic inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) protein expression were markedly higher after the $CCl_4$ treatment. G. jasminoides diminished these alterations. $CCl_4$ increased the level of TNF-$\alpha$, iNOS and COX-2 mRNA expressions, and these increases were attenuated by G. jasminoides. These results suggest that G. jasminoides alleviates $CCl_4$-induced liver injury, and this protection is likely due to the reduced oxidative stress and the downregulation of proinflammatory mediators.

$CCl_4$로 유도된 흰쥐의 간기능 손상에 대한 좌우 양릉천(陽陵泉) 전침 효과 비교 연구 (Comparative study on the effects of 2 Hz electroacupuncture stimulation at left and right GB34's in $CCl_4$-intoxicated rats)

  • Objectives : To compare the effects of low-frequency electro-acupuncture (EA) stimulation at left and right GB34s on hepatotoxicity in $CCl_4$-intoxicated rats. Methods : Rats were injected with $CCl_4$ and treated with 2 Hz electro-acupuncture (EA) at left and right GB34s for 15 minutes 3 times per week, for 10 weeks. Holder group, injected with $CCl_4$ and strained in a cylinder for same period as the EA group, was established to compare the hepatotoxicity against the two electroacupuncture groups. To estimate the effects of EA on hepatotoxicity in rats, body weight, liver weight and liver index were measured. Biochemical assays for serum ALT, AST, ALP and total cholesterol ; hematological analysis for RBC, WBC, PLT, hemoglobin, lymphocytes, neutrophils and monocytes ; and histology analysis of liver tissue were also performed. Results : Lymphocyte level in blood was significantly decreased by $CCl_4$-intoxication, and increased by low-frequency electroacupuncture applied on both left and right GB34s. Low-frequency EA applied on right and left GB34s significantly reduced serum ALT and AST, both of which had been increased by $CCl_4$-intoxication. Conclusion : Low-frequency electroacupunctures at both left and right GB34s have hepatoprotective effects on $CCl_4$-induced liver damage in rats. However, no significant differences were found between the effects of EAs at left and right GB34s.
