Interaction of Stomatin with Hepatitis C Virus RNA Polymerase Stabilizes the Viral RNA Replicase Complexes on Detergent-Resistant Membranes

  • The hepatitis C virus (HCV) RNA genome is replicated by an RNA replicase complex (RC) consisting of cellular proteins and viral nonstructural (NS) proteins, including NS5B, an RNA-dependent RNA polymerase (RdRp) and key enzyme for viral RNA genome replication. The HCV RC is known to be associated with an intracellular membrane structure, but the cellular components of the RC and their roles in the formation of the HCV RC have not been well characterized. In this study, we took a proteomic approach to identify stomatin, a member of the integral proteins of lipid rafts, as a cellular protein interacting with HCV NS5B. Co-immunoprecipitation and co-localization studies confirmed the interaction between stomatin and NS5B. We demonstrated that the subcellular fraction containing viral NS proteins and stomatin displays RdRp activity. Membrane flotation assays with the HCV genome replication-competent subcellular fraction revealed that the HCV RdRp and stomatin are associated with the lipid raft-like domain of membranous structures. Stomatin silencing by RNA interference led to the release of NS5B from the detergent-resistant membrane, thereby inhibiting HCV replication in both HCV subgenomic replicon-harboring cells and HCV-infected cells. Our results identify stomatin as a cellular protein that plays a role in the formation of an enzymatically active HCV RC on a detergent-resistant membrane structure.

Cloning and Molecular Characterization of ${\beta}$-1,3-Glucan Synthase from Sparassis crispa

  • A ${\beta}$-glucan synthase gene was isolated from the genomic DNA of polypore mushroom Sparassis crispa, which reportedly produces unusually high amount of soluble ${\beta}$-1,3-glucan (${\beta}$-glucan). Sequencing and subsequent open reading frame analysis of the isolated gene revealed that the gene (5,502 bp) consisted of 10 exons separated by nine introns. The predicted mRNA encoded a ${\beta}$-glucan synthase protein, consisting of 1,576 amino acid residues. Comparison of the predicted protein sequence with multiple fungal ${\beta}$-glucan synthases estimated that the isolated gene contained a complete N-terminus but was lacking approximately 70 amino acid residues in the C-terminus. Fungal ${\beta}$-glucan synthases are integral membrane proteins, containing the two catalytic and two transmembrane domains. The lacking C-terminal part of S. crispa ${\beta}$-glucan synthase was estimated to include catalytically insignificant transmembrane ${\alpha}$-helices and loops. Sequence analysis of 101 fungal ${\beta}$-glucan synthases, obtained from public databases, revealed that the ${\beta}$-glucan synthases with various fungal origins were categorized into corresponding fungal groups in the classification system. Interestingly, mushrooms belonging to the class Agaricomycetes were found to contain two distinct types (Type I and II) of ${\beta}$-glucan synthases with the type-specific sequence signatures in the loop regions. S. crispa ${\beta}$-glucan synthase in this study belonged to Type II family, meaning Type I ${\beta}$-glucan synthase is expected to be discovered in S. crispa. The high productivity of soluble ${\beta}$-glucan was not explained but detailed biochemical studies on the catalytic loop domain in the S. crispa ${\beta}$-glucan synthase will provide better explanations.

  • This study was carried out to construct accurate and scientific system of assessment indicators in selection of National Trust conservation areas, which was new concept of domestic environment movement and offer the raw data of new analytic method by introducing the fuzzy theory and weight for overcoming the uncertainty of ranking decision. To transform the Likert's scale granted to assessment indicators into the type of triangular fuzzy number(a, b, c), there was conversion to each minimum(a), median(b), and maximum(c) in applying membership function, and in using the center of gravity and eigenvalue, there was to decide the ranking. The rankings of converted values applied a mean importance and weight were confirmed that they were generally changed. Therefore, the ranking decision was better to accomplish objective and rational ranking decision by applying weight that was calculated in grouping of indicator than to judge the singular concept and to be useful in assessment of diverse National Trust site. In the future, because AHP, which was general method of calculating weight, was lacked, there was to understand the critical point to fix a pertinent weight, and to carry out the study applying engineering concept like fuzzy integral using $\lambda-measure$.

  • In this paper, a method of on-line PI gain-tuninng is proposed for the speed control of brushless D.C. motor by investigating the pattern of input and output without estimating parameter. Proportional gain is tuned in the process to obtain a fast speed response by supplying the maximum constant input. And integral gain is appropriately tuned in the process of proportional control so that the response may be stably converged and the overshoot may be prevented. Therefore because both control and gain-tuning are executed concurrently, additional works that estimate parameters and so on aren't required in the proposed method. In the proposed method, both fast-response and overshoot problem are well solved, and it is more useful and convenient than existing auto-tuning methods in the speed control of D.C. motor. It is illustrated by simulations and experimental results that the proposed method is useful and stable.

  • In this study, notch defects are evaluated using fracture mechanics. To understand the effects of notch defects, FE analysis is conducted to predict the limit load and J-integral for middle-cracked and single-edge cracked plates with various sizes of notch under tension and bending. As the radius of the notch increases, the energy release rate also increases, although the limit load remains constant. The values of fracture toughness($J_{IC}$) of SM490A are determined for various notch radii through FE simulation instead of conducting an experiment. As the radius of the notch increases, the energy release rate also increases, together with a more significant increase in fracture toughness. To conclude, as the notch radius increases, the resistance to crack propagation also increases.

  • This study was carried out to develop prediction model for fill slope failure of forest road in igneous rock area using fuzzy theory which is non-linear model. The results were summarized as follows. The importance weight of factors on fill slope failure was ranked in the order of fill slope length, fill slope gradient, soil type, aspect, road position and longitudinal slope form. The degree of potential slope failure was high mainly under the such conditions as fill slope length greater than 8m, fill slope gradients steeper than $40^{\circ}$, constituent material with weathered rock, aspect of NE and road on ridge position. The optimal prediction model was developed with 0.15 of optimal coefficient(c) and 3.1165 of ${\lambda}$-value when fuzzy integral value of slope failure possibility is more than 0.5. And the discriminant accuracy was 86.8%, which shows the high availability for discrimination.

Cure Behaviors and Thermal Stabilities of Epoxy Resins Initiated by Latent Thermal Catalyst (열잠재성 촉매 개시제를 이용한 에폭시 수지의 경화거동 및 열안정성)

  • In this work, two thermal latent catalysts, i.e., N-benzylpyrazinium hexafluoroantimonate (BPH) and benzyl-2,5-dimethylpyrazinium hexafluoroantimonate (BDPH), were synthesized. The cure behaviors and thermal stabilities of diglycidylether of bisphenol A (DCEBA) epoxy resins initiated by 1 wt.% of the catalysts were investigated by DSC, NIR, TCA, and DMA Latent properties of the catalysts were examined by conversion of epoxy resins using NIR from $100^{\circ}C$ to $180^{\circ}C$ From the resultes of near-IR, DGEBA/BPH system showed higher conversion than that of DGEBA/BDPH system. The thermal stabilities of DGEBA/BDPH system based on the initial decomposition temperature (IDT) and integral procedural decomposition (IPDT) were relatively lower than those of DCEBA/BPH system. These could be attributed to the hindered structure of BDPH, resulting in decreasing the thermal stability in the DGEBA/BDPH system.

  • A low-cost wireless sensor network (WSN) solution with highly expandable super and simple nodes was developed. The super node was designed as a sensing unit as well as a receiving terminal with low energy consumption. The simple node was designed to serve as a cheaper alternative for large-scale deployment. A 12-bit ADC inputs and DAC outputs were reserved for sensor boards to ease the sensing integration. Vibration and thermal field tests of the Chi-Lu Bridge were conducted to evaluate the WSN's performance. Integral acceleration, temperature and tilt sensing modules were constructed to simplify the task of long-term environmental monitoring on this bridge, while a star topology was used to avoid collisions and reduce power consumption. We showed that, given sufficient power and additional power amplifier, the WSN can successfully be active for more than 7 days and satisfy the half bridge 120-meter transmission requirement. The time and frequency responses of cables shocked by external force and temperature variations around cables in one day were recorded and analyzed. Finally, guidelines on power characterization of the WSN platform and selection of acceleration sensors for structural health monitoring applications were given.

  • A Linear parameterized Sigma-Pi neural network (SPNN) is applied to a tilt-duct unmanned aerial vehicle (UAV) which has a very large longitudinal stability ($C_{L{\alpha}}$). It is uncontrollable by a proportional, integral, derivative (PID) controller due to heavy stability. It is shown that the combined inner loop and outer loop of SPNN controllers could overcome the sluggish longitudinal dynamics using a method of dynamic inversion and pseudo-control to compensate for reference model error. The simulation results of the way point guidance are presented to evaluate the performance of SPNN in comparison to a PID controller.

  • This paper presents a parallel object-oriented programming (OOP) platform for efficient visualization of three-dimensional content in CUDA environments. For this purpose, this paper discusses the features and limitations in implementing C++ object-oriented codes using CUDA and proposes the solutions. Also, it presents how to implement a 3D parallel visualization platform based on the MVC (Model/View/Controller) design pattern. Also, it provides sample implementations for integral MLS (iMLS) and signed distance fields (SDFs) based on the Marching Cubes and Raytracing. The proposed approach enables GPU parallel processing only by implementing simple interfaces. Based on this, developers can expect general benefits that are common in general OOP techniques including abstractization and inheritance. Though I implemented only two specific samples in this paper, I expect my approach can be widely applied to general computer graphics problems.
