Differences in Optimal pH and Temperature for Cell Growth and Antibody Production Between Two Chinese Hamster Ovary Clones Derived from the Same Parental Clone

  • To investigate clonal variations of recombinant Chinese hamster ovary(rCHO) clones in response to culture pH and temperature, serum-free suspension cultures of two antibody-producing CHO clones(clones A and B), which were isolated from the same parental clone by the limiting dilution method, were performed in a bioreactor at pH values in the range of 6.8-7.6, and two different temperatures, $33^{\circ}C\;and\;37^{\circ}C$. In regard to cell growth, clone A and clone B displayed similar responses to temperature, although their degree of response differed. In contrast, clones A and B displayed different responses to temperature in regard to antibody production. In the case of clone A, no significant increase in maximum antibody concentration was achieved by lowering the culture temperature. The maximum antibody concentration obtained at $33^{\circ}C$(pH 7.4) and $37^{\circ}C$(pH 7.0) were $82.0{\pm}2.6$ and $73.2{\pm}4.1{\mu}g/ml$, respectively. On the other hand, in the case of clone B, an approximately 2.5-fold increase in maximum antibody concentration was achieved by lowering the culture temperature. The enhanced maximum antibody concentration of clone B at $33^{\circ}C$($132.6{\pm}14.9{\mu}g/ml$ at pH 7.2) was due to not only enhanced specific antibody productivity but also to prolonged culture longevity. At $33^{\circ}C$, the culture longevity of clone A also improved, but not as much as that of clone B. Taken together, CHO clones derived from the same parental clone displayed quite different responses to culture temperature and pH with regards antibody production, suggesting that environmental parameters such as temperature and pH should be optimized for each CHO clone.

  • Mer,trans-Re(≡NC6H5)(PPh3)2Cl3, Ⅰ, reacted with trimethylphosphine to give a mixture of two stereoisomers, mer,trans-Re(≡NC6H5)(PMe3)2Cl3,Ⅱ, and fac,cis-Re(≡NC6H5)(PMe3)2Cl3, Ⅲ. These compounds could also be prepared from the reaction of Re(≡NC6H5)(PMe3)(PPh3)Cl3 with trimethylphosphine. In both reactions the mer,trans-isomer is a major product. The products have been characterized by NMR, elemental analysis, and X-ray crystallography. Crystal data for Ⅱ: monoclinic space group P21, a=10.053(1) Å, b=10.844(1) Å, c=10.058(2) Å, β=113.45(2)°, Z=2, R(wR2)=0.0348 (0.0894). Crystal data for Ⅲ: monoclinic space group P21/n, a=7.183(2) Å, b=16.983(4) Å, c=15.543(4) Å, β=90.38(2)°, Z=4, R(wR2)=0.0603 (0.1484).

The out-of-plane Band Contour of Dichloroborane Molecules

  • The out-of-plane bending, vibrational-rotational FT-IR spectra of dichloroborane molecule were analyzed. The band centers are 785.992, 785.723, 647.217, and 646.997 $cm^{-1}$ and centrifugal distortion constants are $2.2{\times}10^{-6},\;5.5{\times}10^{-6},\;0.2{\times}10^{-6}$, and $0.7{\times}10^{-6}cm^{-1}$ for $H^{11}B^{35}Cl_2$, $H^{11}B^{35}Cl^{37}Cl$, $D^{11}B^{35}Cl_2$, and $D^{11}B^{35}Cl^{37}Cl$, respectively. The theoretical spectrum of $D^{11}B^{36}Cl_2$ was plotted to conform the molecular constants and the rotational constants for the first excited vibrational state of this molecule were A' = 1.165932, B' = 0.106166, and C' = 0.097229 $cm^{-1}$. The calculated spectra were in good agreement with the observed spectra.

  • We report the solid-phase library construction of 222 number of a novel N-[alkyl sulfonamido-spiro(2H-1-benzopyran-2,4-piperidine)-6-yl] substituted amide 1A and amine 1B derivatives. The polymer-bound N-[alkylsulfonamido-spiro(2H-1-benzopyran-2,4-piperidine)-6-yl] substituted amide 9 and amine 10 derivatives were obtained by first diversity generation with various acid chlorides and alkyl halides. Further reactions on the resins 9 and 10 with substituted sulfonyl chlorides produced the desired N-[alkylsulfonamido-spiro(2H-1-benzopyran-2,4-piperidine)-6-yl] substituted amide 1A and amine 1B analogues.

Interactions between Hyaluronic Acid, Lysozyme, Peroxidase, and Glucose Oxidase in Enzymatic Activities at Low pH

  • Purpose: Many substances in saliva or oral health care products interact with each other. The aim of this study was to investigate interactions between hyaluronic acid (HA), lysozyme, peroxidase, and glucose oxidase (GO) in enzymatic activities at low pH levels. Methods: HA (0.5 mg/mL), hen egg-white lysozyme (HEWL, $30{\mu}g/mL$), bovine lactoperoxidase (bLPO, $25{\mu}g/mL$), and GO ($50{\mu}g/mL$) were used. The influences of HA, bLPO, and GO on HEWL activity were determined by measuring the turbidity of a Micrococcus lysodeikticus suspension. The influences of HA and HEWL on bLPO activity were determined by the NbsSCN assay, measuring the rate of oxidation of 5-thio-2-nitrobenzoic acid (Nbs) to 5,5'-dithiobis(2-nitrobenzoic acid) $(Nbs)_2$. The influences of HA and HEWL on GO activity were determined by measuring oxidized o-dianisidine production. All experiments were performed at pH 4, 5, and 6. Results: HA and GO did not affect the enzymatic activity of HEWL at pH 4, 5, and 6. bLPO enhanced the enzymatic activity of HEWL at pH 5 (p<0.05) and pH 6 (p<0.05) significantly. The enzymatic activity of bLPO was not affected by HA and HEWL at pH 4, 5, and 6. HA and HEWL did not affect the enzymatic activity of the GO at pH 4, 5, and 6. Conclusions: Peroxidase enhances lysozyme activity at low pH, otherwise there were no significant interactions in enzymatic activities between HA, lysozyme, peroxidase, and GO at low pH levels.

Effects of neutron irradiation on superconducting critical temperatures of in situ processed MgB2 superconductors

  • Effects of neutron irradiation on the superconducting properties of the undoped $MgB_2$ and the carbon(C)-doped $MgB_2$ bulk superconductors, prepared by an in situ reaction process using Mg and B powder, were investigated. The prepared $MgB_2$ samples were neutron-irradiated at the neutron fluence of $10^{16}-10^{18}n/cm^2$ in a Hanaro nuclear reactor of KAERI involving both fast and thermal neutron. The magnetic moment-temperature (M-T) and magnetization-magnetic field (M-H) curves before/after irradiation were obtained using magnetic property measurement system (MPMS). The superconducting critical temperature ($T_c$) and transition width were estimated from the M-T curves and critical current density ($J_c$) was estimated from the M-H curves using a Bean's critical model. The $T_cs$ of the undoped $MgB_2$ and C-doped $MgB_2$ before irradiation were 36.9-37.0 K and 36.6-36.8 K, respectively. The $T_cs$ decreased to 33.2 K and 31.6 K, respectively after irradiation at neutron fluence of $7.16{\times}10^{17}n/cm^2$, and decreased to 22.6 K and 24.0 K, respectively, at $3.13{\times}10^{18}n/cm^2$. The $J_c$ cross-over was observed at the high magnetic field of 5.2 T for the undoped $MgB_2$ irradiated at $7.16{\times}10^{17}n/cm^2$. The $T_c$ and $J_c$ variation after the neutron irradiation at various neutron fluences were explained in terms of the defect formation in the superconducting matrix by neutron irradiation.

향미찹쌀전분의 이화학적 특성비교 (Physicochemical Properties of Starches from Flavored Glutinous Rice Varieties)

  • 향미찰벼 전분의 이화학적 특성을 알기 위하여 scanning electron microscope(SEM), 요드정색반응, differential scanning calorimetry(DSC), 그리고 산가수분해도, glucoamylase와 $\alpha$-amylase 가수분해도를 측정하였다. 향미찰벼의 전분입자는 다각형구조이며 직경 4~6$\mu\textrm{m}$였다. 호화특성으로서 호화개시온도는 59.8~62.5$^{\circ}C$였으며 향미찰벼 중 KR92021-B-B-42-3-B와 KR92021-B-B-165-1-B의 호화엔탈피값은 대체로 높았다. 향미찰벼 전분의 아밀로펙틴의 무정형 분획을 품종간 비교하기 위하여 15% H$_2$SO$_4$에 의한 산가수분해도를 측정한 결과 KR92021-B-B-5-2-B와 KR92021-B-B-42-3-B는 산가수분해도가 낮아 아밀로펙틴의 cluster 구조가 다른 품종에 비해 질서있게 나열되어 있음을 알 수 있었다. 향미찰벼 중 KR92021-B-B-5-2-B는 glucoamylase와 $\alpha$-amylase에 의한 전분가수분해도가 높은 특성을 보였다.

Palladium(II) Schiff Base Complexes Derived from Allylamine and Vinylaniline

  • Condensation of salicylaldehyde $(2-HOC_6H_4C(O)H)$ with allylamine afforded the unsaturated salicylaldimine, $2-HOC_6H_4C(H)=NCH_2CH=CH_2$. Similar reactivity was observed with substituted salicylaldehydes. Further reaction of these Schiff bases with palladium acetate or $Na_2PdCl_4$ afforded complexes of the type $PdL_2$, where L = deprotonated Schiff base. The molecular structure of the parent salicylaldimine palladium complex $[trans-(2-OC_6H_4C(H)=NCH_2CH=CH_2)_2Pd]$ (1) was characterized by an X-ray diffraction study. Crystals of 1 were monoclinic, space group $P2_1/n,\;a\;=\;14.0005(9)\;{\AA},\;b\;=7.2964(5)\;{\AA},\;c\;=\;17.5103(12)\;{\AA},\;{\beta}\;=\;100.189(1)^{\circ}$, Z = 4. Analogous chemistry with 4-vinylaniline also gave novel palladium complexes containing a pendant styryl group. Crystals of $[trans-(2-HOC_6H_4C(H)=N-4-C_6H_4CH=CH_2)_2Pd]$ (4) were monoclinic, space group $P2_1/c$, a = 13.7710(14) ${\AA}$, b = 11.0348(11) ${\AA}$, c = 7.8192(8) ${\AA}$, ${\beta}\;=\;98.817(2)^{\circ}$, Z = 2.

  • The michael adducts 2a, b were obtained from the reaction of the phenylacelyl derivative 1 with benzadehyde and p-anisaldehyde and p-anisaldehyde respectively. 2a and 2b were subjected to react with cyanoethanoic acid hydrazide, malononitrile, cyanothioacetamide, cyanoacetamide and 1, 1, 3-tricyano-2-amino propene to yield 4a-h and 5a, b respecitively. Hydrogen peroxide oxidation of 2a, b gave the aurone derivative 6a, b. The pyone derivatives 8a, b were obtained from 2a, b by addition of chioroacetyl chioride followed by dehydrochlorination.
