수리취와 참취 엽의 휘발성 향기성분 비교 (Comparison of Volatile Aroma Compounds between Synurus deltoides and Aster scaber Leaves)

  • This study was investigated to compare the volatile aroma compounds of Synurus deltoides and Aster scaber. The volatile aroma compounds from Synurus deltoides and Aster scaber were extracted by soild-phase microextraction (SPME) methods. S. deltoides had 97 volatile aroma compounds such as including 5-acetyl-1,2-dihydro acenaphtylene (14.63%), ${\beta}$-cubebene (9.31%), caryophyllene (8.97%), ${\beta}$-chamigrene (7.14%), ${\beta}$-selinene (2.71), ${\alpha}$-farnesene (2.47%), ${\alpha}$-bergamotene (2.26%), ${\beta}$-elemene (1.94%), etc. A. scaber had 84 volatile aroma compounds such as (+)-epi-bicyclosesquiphellandrene (10.38%), terpinolen (10.09%), caryophyllene (6.04%), 8-isopropenyl-1,5-dimethyl-1,5-cyclodeca diene (5.42%), ${\alpha}$-himachalene (5.04%), ${\beta}$-thujene (4.37%), ${\beta}$-pinene (4.28%), ${\beta}$-cubebene (3.99%), etc. Conclusively, the main common volatile aroma compounds in S. deltoides and A. scaber leaves were 19 volatile aroma compounds such as caryophyllene, terpinolen, ${\beta}$-cubebene. But the composition and amount of volatile aroma compounds were very different between the two species.

  • One of the most common forms of dementia, Alzheimer's disease (AD) is a progressive neurodegenerative disorder symptomatically characterized by impairment in memory and cognitive abilities. AD is characterized pathologically by the presence of intracellular neurofibrillary tangles and deposition of ${\beta}$-amyloid ($A{\beta}$) peptides, believed to be neurotoxic and now is also considered to have a role on the mechanism of memory dysfunction. In this study, we tested that MeOH extract of Impatiens balsamina L. (IBM) affects on the processing of APP from the APPswe over-expressing Neuro2a cell line. We found that IBM increased over 2 folds of the $sAPP{\alpha}$ secretion level, a main metabolite of ${\alpha}$-secretase. We shown that IBM reduced the secretion level of $A{\beta}42$ and $A{\beta}40$ without cytotoxicity. BACE (${\beta}$-site APP cleaving enzyme) FRET assay shown that BACE activity was specifically decreased in the presence of IBM. We suggest that Impatiens balsamina L. may be an useful source to develop a herbal medicine of BACE inhibitor for Alzheimer's disease.

  • This stduy was conducted to find out the changes of estradiol-17$\beta$ and progesterone levels in the plasma of Cheju mares during gestatation period and before and after parturition with 12 heads of Cheju ponies. The results are as follows : 1. The estradiol-17$\beta$ and progesterone levels on the 1~4 months showed 61.6~134.2pg/ml and 7.74~9.20ng/ml respectively, but the estradiol-17$\beta$ levels rapidly increased to 426.4~1772.9pg/ml, and the progesterone levels decreased to 1.42~5.43ng/ml on the 5~10 months of gestation period. 2. The progesterone levels of the pregnant and the non-pregnant mares showed 4.80ng/ml and 0.04ng/ml in winter, respectively, but both of the two appeared 1,211.5pg/ml and 99.4pg/ml of estradiol-17$\beta$, respectively. Therefore the diagnosis of pregnancy might be more accurate with the levels of estrdiol-17$\beta$ in autumn. 3. The progesterone levels showed 3.20ng/ml the day before parturition and less than 1ng/ml on 2 days after parturition. The estradiol-17$\beta$ levels, showing the rising trend, and 90.1~162.4pg/ml up to 10 days.

저혈당을 경험한 당뇨병 환자의 치료지시 불이행, 가족지지가 우울에 미치는 영향 (Influence of Non-compliance of Treatment and Family Support on Depression in Diabetic Patients with Hypoglycemia)

  • 본 연구는 저혈당을 경험한 당뇨병 환자의 우울에 미치는 영향요인을 파악하여 대상자의 우울을 감소시키기 위한 기초자료를 마련하고자 2015년 10월 1일부터 2016년 3월 15일까지 B광역시 소재 일 병원의 내분비내과 외래를 방문한 119명을 대상으로 설문지를 배부하여 자료를 수집하였다. 수집된 자료는 SPSS 20.0 프로그램을 이용하여 빈도와 백분율, t-검증, 일원분산분석, 사후검증(Scheffe's-test), Pearson's correlation, 단계적 다중회귀분석을 실시하였다. 본 연구 결과 대상자의 우울은 치료지시불이행(r=.42, p<.001)과는 양의 상관관계를, 가족지지(r=-.38, p<.001)와는 음의 상관관계를 보였다. 대상자의 우울에 영향을 미치는 요인은 가족지지(${\beta}=-.29$, p<.001), 치료지시불이행의 하위영역인 약물요법(${\beta}=.21$, p=.020), 식이요법(${\beta}=.23$, p=.014), 한 달 평균 수입 300만원 이상(${\beta}=-.20$, p=.014), 한 달 평균 수입 200만원 이상-300만원 미만(${\beta}=-.17$, p=.033)으로 확인되었으며, 총 30.0%를 설명하였다(Adj. R2=.30, F=20.06, p<.001). 이를 바탕으로 대상자의 우울을 감소시키기 위하여 가족지지 향상 프로그램을 개발하여 적용할 것을 제안한다.

Expression of Cytokines in Radiation Injured Brain at Acute Phase

  • Objective : Radiation therapy is an important treatment for brain tumor. However, serious complications such as radiation necrosis can occur and it may be secondary to the expression of acute phase genes, like cytokines. In particular, inflammatory cytokines (IL-$1{\beta}$, TNF-${\alpha}$) and other immunomodulatory cytokines (TNF-${\alpha}$, TGF-${\beta}1$) might be changed after irradiation (high single dose irradiation). Although it has been reported that IL-1 level is remarkably elevated within 8 week after the irradiation to the rat brain. the change of cytokines levels at acute phase (within 24 hours) has not been reported. In the present study, we examined TNF-${\alpha}$, TGF-${\beta}1$, and IL-$1{\beta}$ levels in acute phase to clarify the early effect of cytokines on the radiation-induced brain damage. Methods : Fifty Sprague-Dawley rats were used and these were divided into irradiation group and control group. After a burr-hole trephination on the right parietal area using a drill, a single 10Gy was irradiated at the trephined site. Their forebrains were extirpated at 30 min, 2 hr, 8 hr, 12 hr and 24 hr, respectively and examined for the expression of TNF-${\alpha}$, TGF-${\beta}1$, and IL-$1{\beta}$. Results : The expression of TNF-${\alpha}$ and TGF-${\beta}1$ were decreased until 12 hr after irradiation but elevated thereafter. The expression of IL-1 was peak at 8 hr and then decreased until 12 hr but elevated after this time window. The present study indicated that expression of cytokines (TNF-${\alpha}$, TGF-${\beta}1$ and IL-$1{\beta}$) were increased at 24 hr after the irradiation to the rat brain. IL-$1{\beta}$ level, on the other hand. reached peak at 8 hr after radiation injury. Conclusion : These findings indicate that IL-1, among various cytokines, may have a more important role in the inflammatory reaction by radiation injury at acute phase and provide some clues for better understanding of the pathogenesis of radiation injury.

Bacillus circulans 유래 cellulolytic xylanase 유전자(bglBC2)의 염기서열 결정 및 분석 (Nucleotide Sequence of Cellulolytic Xylanase Gene (bglBC2) from Bacillus circulans)

  • 클로닝된 Bacillus circulans ATCC21367 유래 cellulolytic xylanase 유전자(bglBC2)의 염기서열을 결정 분석하였다. 본 유전자는 1,224 bp의 407개 아미노산을 암호하는 open reading frame (ORF)으로 구성되어 있었으며 염기서열로부터 산출된 유전자의 분자량은 45 kDa으로 효소의 SDS-PAGE로부터 측정된 분자량과 일치하였다. ATG 개시 코돈의 9bp 위쪽에 Shine-Dalgarno (SD) 서열로 추정되는 5'-AAAGGAG-3' 서열이 확인되었고 그 상단에 promoter로 추정되는 -35 서열(TTTACA)과 -10 서열(TATACT)이 위치하고 있었으며, 이는 B. subtilis promoter consensus sequence와 유사하였다. 한편, 이 효소의 아미노산 서열은 이미 보고된 B. circulans KSM-N257의 alkaline $endo-\beta-1,4-glucanase$와는 97%, B. circulans WL-12의 $endo-\beta-1,3-1,4-glucanase$와는 75%, Bacillus sp. KSM-330의 $endo-\beta-1,4-glucanase$ (cellulase)와는 45%의 유사성을 나타내었다. 또한 bglBCS 염기서 열의 정보를 GenBank에 등록하였으며 등록번호는 Ar269256이다.

Prognostic Value of β-catenin Expression in Breast Cancer Patients: a Meta-analysis

  • Background: ${\beta}$-catenin plays a crucial role in the progression of breast cancer (BC) and a prognostic role of in BC patients has been widely reported. However, controversy still remains. Materials and Methods: Identical search strategies were used to search relevant literature in electronic databases updated to July 1, 2014. Individual hazard ratios (HRs) and 95% confidence intervals (CIs) were extracted and pooled HRs with 95%CIs were used to evaluate the strength of association between positive ${\beta}$-catenin expression in different subcellular locations and survival results of BC patients. Subgroup and meta-regression analyses were performed to explore heterogeneity. Funnel plots of Begg's and Egger's linear regression test were used to investigate publication bias. Heterogeneity and sensitivity were also assessed. All the work was completed using STATA. Results: A total of 2,204 patients from 12 evaluative studies were finally included. Pooled HRs and 95%CIs suggested that ${\beta}$-catenin expression in cytoplasm/nucleus had an unfavorable impact on both overall survival (OS) (HR: 1.93, 95%CI: 1.40-2.65) and disease free survival (DFS)/ recurrent free survival (RFS) (HR: 1.60, 95%CI: 1.20-2.13) in BC patients. However, here was no significant association between ${\beta}$-catenin expression in the membranes with OS (HR: 0.65, 95%CI: 0.42-1.02) or DFS/RFS (HR: 0.66, 95%CI: 0.38-1.13). Publication bias was absent in all of the four outcomes. Sensitivity analysis revealed that the results of this meta-analysis were robust. Conclusions: Positive ${\beta}$-catenin expression in cytoplasm/nucleus rather than in membrane is a significant prognostic factor in patients with BC who have been surgically treated.

Synthetic Lead Compounds Modulate Activity of Large-conductance $Ca^{2+}$-activated Potassium Channels Expressed in Xenopus Oocytes

  • Large-conductance $Ca^{2+}$-activated potassium channels ($BK_{Ca}$ are a widely distributed and play key roles in various cell functions. In nerve cells, $BK_{Ca}$ channels shorten the duration of action potentials and block $Ca^{2+}$ entry thereby repolarizing excitable cells after excitation. $BK_{Ca}$ channel opening has been postulated to confer neuroprotection during stroke, and has attracted attention as a means for therapeutic intervention in asthma, hypertension, convulsions, and traumatic brain injury. Several natural and synthetic compounds including a steroid hormone, $\beta$-estradiol, have been identified as the activators of $BK_{Ca}$ channels. Based on the structural features of the previously reported activators of $BK_{Ca}$ channels, we designed several lead compounds, synthesized chemically, and tested their functional activity on cloned $BK_{Ca}$ channels. The $\alpha$ subunit of rat $BK_{Ca}$ channel was expressed alone or with different $\beta$ subunits in Xenopus oocytes and the effects of the compounds were tested electrophysiological means. One of the lead compounds affected the activity of the $\alpha$ subunit of $BK_{Ca}$ channel in a $\beta$ subunit-specific manner. While the activity of B $K_{ca}$ channel $\alpha$ subunit was Potentiated, the channel composed of $\alpha$ and $\beta$1 subunits were inhibited by this compound. We are currently investigating the mechanism of the $\beta$ subunit-dependent effects and planning to localize the receptor site of the lead compound.f the lead compound.

냉간성형 각형강관 모살용접 T형 접합부의 최대내력(II)-주관웨브 파괴모드- (Ultimate Strength of Fillet-welded T-joints in Cold-formed Square Hollow Sections-chord web failure mode)

  • 본 논문은 냉간성형 각형강관 T형 접합부의 최대내력과 병형제한치에 대한 두 번째 연구이다. 전회의 실험적 연구로부터, 주관폭에 대한 지관폭의 비($\beta$)가 0.8이상인 T형 접합부는 뚜렷한 최대의 하중을 나타내고 접합부의 파괴모드는 주관 웨브의 좌굴파괴이다. 주관 플랜지 파괴모드의 T형 접합부와 유사하게, 일정한 변형량(주관플랜지에서의 변위)에 대응하는 하중을 T형 접합부의 최대내력으로 정의할 수 있을 것이다. Kato와 Zhao에 의해 수행된 실험을 포함한 실험결과로부터 $10.7{\leq}B/T{\leq}42.3$ 이고 $0.8{\leq}{\beta}{\leq}1.0$ 인 범위의 T형 접합부에 대하여, 최대내력을 정의를 위한 변형제한치는 주관폭의 3% 변형량(3%B)으로 제안하였다. CIDECT의 설계식 및 기존의 제안내력식과 실험결과를 비교하였고, 최종적으로 기둥좌굴에 근거한 내력식을 제안하였다.

Bioconversion of Ginsenosides from Red Ginseng Extract Using Candida allociferrii JNO301 Isolated from Meju

  • Red ginseng (Panax ginseng), a Korean traditional medicinal plant, contains a variety of ginsenosides as major functional components. It is necessary to remove sugar moieties from the major ginsenosides, which have a lower absorption rate into the intestine, to obtain the aglycone form. To screen for microorganisms showing bioconversion activity for ginsenosides from red ginseng, 50 yeast strains were isolated from Korean traditional meju (a starter culture made with soybean and wheat flour for the fermentation of soybean paste). Twenty strains in which a black zone formed around the colony on esculin-yeast malt agar plates were screened first, and among them 5 strains having high ${\beta}$-glucosidase activity on p-nitrophenyl-${\beta}$-D-glucopyranoside as a substrate were then selected. Strain JNO301 was finally chosen as a bioconverting strain in this study on the basis of its high bioconversion activity for red ginseng extract as determined by thin-layer chromatography (TLC) analysis. The selected bioconversion strain was identified as Candida allociferrii JNO301 based on the nucleotide sequence analysis of the 18S rRNA gene. The optimum temperature and pH for the cell growth were $20{\sim}30^{\circ}C$ and pH 5~8, respectively. TLC analysis confirmed that C. allociferrii JNO301 converted ginsenoside Rb1 into Rd and then into F2, Rb2 into compound O, Rc into compound Mc1, and Rf into Rh1. Quantitative analysis using high-performance liquid chromatography showed that bioconversion of red ginseng extract resulted in an increase of 2.73, 3.32, 33.87, 16, and 5.48 fold in the concentration of Rd, F2, compound O, compound Mc1, and Rh1, respectively.
