Synthetic Studies on Fused Nitrogen-heterocycles from N-Amino-N,N '-dihydrodiazinediones (II). Condensation of N-Amino-N,N '-dihydrodiazinediones with ${\alpha},{\beta}$-Unsaturated Carbonyl Compounds

  • The condensation of 1-amino-1,2-dihydro-3,6-pyridazinedione (1) and 2-amino-2,3-dihydro-1,4-phthalazinedione (2) with mesityl oxide or 3-penten-2-one in acetic acid-ethanol (1:1) gave 3,4,6,9-tetrahydro-6,9-dioxopyridazino[1,2-a][1,2, 3]triazines (9,11) and 3,4,6,11-tetrahydro-6,11-dioxo[1,2,3]triazino[1,2- b]phthalazines (10,12), respectively. The condensation of 1 and 2 with crotonaldehyde, cinnamaldehyde or acrylaldehyde under the same reaction condition gave only N-alkylidene derivatlives (3-8). When the N-alkylidene derivatives isolated from the reaction of 1 and 2 with crotonaldehyde and cinnamaldehyde (3-6) were refluxed in acetic acid, the corresponding heterocyclic compounds (13-16) were obtained.

  • Four prostate-type triterpenes were isolated from a methanol extract of Alismatis Rhizoma by bioassay-guided isolation using in vitro cytotoxic assay. The compounds were identified as alisol B 23-acetate (1), alisol C 23-acetate (2), alisol B (3), alisol A 24-acetate (4) by spectroscopic methods. Amongst the compounds, alisol B (3) showed significant cytotoxicity against SK-OV3, B16-F10, and HT1080 cancer cell lines with $ED_50$ values of 7.5, 7.5, $4.9\mu\textrm{g}/ml$, respectively.

Stability of Representative Active Compounds on Commercial Biopesticides Based on Neem or Sophora flavescens Extract Under Controlled Temperature (님 또는 고삼추출물을 주원료로 하는 작물병해충 관리용 유기농업자재의 유효성분 열 안정성)

  • This study investigated the thermal stabilities of the four limonoidal active compounds like azadirachtin A, azadirachtin B, deacetylsalannin and salannin and of the two alkaloidal active compounds like matrine and oxymatrine in commercial biopesticides at incubated conditions ($30^{\circ}C$, $35^{\circ}C$, $40^{\circ}C$, $45^{\circ}C$, and $54^{\circ}C$). Half-lives of the limonoidal compounds in biopesticide ranged from 25.6 to 220 days. And the total limonoid contents changed over 15% after 14 days at all the conditions in the tested biopesticide containing neem extract. On the contrary, half-lives of the alkaloidal compounds in biopesticide showed the ranges from 231 to 346 days. And total alkaloidal compounds in all the tested biopesticides containing Sophora flavescens extracts decomposed below 15% after 14 days incubation at $54^{\circ}C$.

  • A benzofuran substance was isolated from the methanol extract of Pauownia tomentosa stem which has been used to treat against gonorrhoea and contusion etc. in the oriental traditional medicine. Its structure was elucidated as methyl-5-hydroxy-[1, 4]naphthoquino-[2, 3-b]benzo-[I, 2-g] benzofuran-6-carboxylate by X-ray crystallography and various spectroscopic evidences and also other three known compounds, paulownin, sesamin, $\beta$-sitosteryl-3-O-$\beta$-D-glucopyranoside, were obtained.

Cytotoxic Triterpenoids from the Fruiting Bodies of Ganoderma lucidum

  • Twelve triterpenoids (1 - 12) were isolated from $CHCl_3$-soluble fraction of fruiting bodies of Ganoderma lucidum. Extensive spectroscopic and chemical studies established the structures of these compounds as butyl lucidenate P (1), butyl lucidenate $E_2$ (2), butyl lucidenate $D_2$ (3), butyl lucidenate Q (4), ganoderiol F (5), methyl ganoderate H (6), methyl ganoderate J (7), lucidumol B (8), ganodermanondiol (9), methyl lucidenate N (10), methyl lucidenate A (11) and butyl lucidenate N (12). All of the compounds were examined for their cytotoxic activity against HL-60, HeLa, and MCF-7 cancer cell lines. Among them, compounds 4 and 8 showed cytotoxic activity with $IC_{50}$ values of 6.6 and 1.6 ${\mu}M$ against HL-60, respectively. In addition, compound 8 also showed cytotoxic activity with $IC_{50}$ values of 2.0 ${\mu}M$ against HeLa cancer cell line, other compounds were moderate or inactive.

  • A series of new liquid crystalline dimesogenic compounds with chiral tails was synthesized, and their thermal and liquid crystalline properties were studied. The chain length of the central polymethylene spacers (x) was varied from dimethylene (2) to decamethylene (12). These compounds were characterized by elemental analysis, IR and NMR spectroscopy, differential scanning calorimetry (DSC), and cross-polarizing microscopy. All compounds were found to be enantiotropically liquid crystalline, and the values of melting ($T_m$) and isotropization temperature ($T_i$) as well as enthalpy change (Δ$H_i$) and entropy change for isotropization (Δ$S_i$) decreased in a zig-zag fashion revealing the so-called odd-even effect as x increases. Their mesomorphic properties fall into three categories depending upon x; (a) compounds with x=2 and 4 formed two different mesophases, smectic and cholesteric phases in that order on heating, and vice versa on cooling, (b) compounds with x=3, 7, 8, 10 and 11 reversibly formed only the cholesteric phase, and (c) compounds with x=5, 6, 9 and 12 exhibited only a cholesteric phase on heating, whereas on cooling they formed two different mesophases, cholesteric and smectic phases, sequentially.

Studies on Whitening and UV Damage Protection Effect of the Pterin Compounds (테린계 화합물의 미백 및 자외선 손상방어 효능에 관한 연구)

  • (6R)-5,6,7,8-tetrahydrobiopterin ($6-BH_4$) cofactor is essential for various process, and is present in probably every cell or tissue of higher organism. $6-BH_4$ is required lot various enzyme activities, and for less defined functions at the cellular level. And it is well known about the antioxidant effects as a non-protein compound. Recently, scientists proposed another roles for $6-BH_4$ in melanogenesis. $6-BH_4$ is a well known tyrosinase inhibitor. In this study, we found that methyl-$BH_4$ and $6-BH_4$ have antioxidant activities and inhibitory activity for melanin synthesis. These pterin compounds were not toxic in HaCaT and B16F10 cells and showed scavenging activity against DPPH radicals. We also showed that pterin compounds decreased protein levels of tyrosinase and TRP-1. In a clinical test, pterin compounds showed the significant skin whiteining effect after treatment for 3 weeks. Furthermore pterin compounds significantly suppressed the UVB-induced expression of $PGE_2$ and IL-6 genes induced UVB In HaCaT and inhibited UVB-induced melanogenesis in B16F10 cells. These results showed the effect of pterin compounds as a cosmeceutical ingredient.

A Novel Synthetic Route to Highly Cross-Linked Poly(vinyl ethers): Ⅲ. Synthesis and Free Radical Polymerization of Aryloxyethyl Vinyl Ethers Having an Electron Acceptor in ortho- or meta-Position

  • ο-(2-Vinyloxyethoxy)benzylidenemalononitrile (3a) and methyl ο-(2-vinyloxyethoxy)-benzylidenecyanoacetate (3b), m-(2-vinyloxyethoxy)benzylidenemalononitrile (4a), and methyl m-(2-vinyloxyethoxy)benzylidenecyanoacetate (4b) were prepared by the condensation of ο-(2-vinyloxyethoxy)benzaldehyde (1) and m-(2-vinyloxyethoxy)benzaldehyde (2) with malononitrile or methyl cyanoacetate, respectively. Bifunctional vinyl ether monomers 3a and 3b polymerized quantitatively with radical initiators in γ-butyrolactone solution at 65 ℃, while meta-isomers 4a and 4b gave lower yields of polymers under the same conditions. The polymers 5-6 obtained from the monomers 3-4 were insoluble in common solvents due to cross-linking. Under the same polymerization conditions ethyl vinyl ether polymerized well with model compounds ο-methoxybenzylidenemalononitrile 7a, methyl ο-methoxybenzylidenecyanoacetate 7b, m-methoxybenzylidenemalononitrile 8a, and methyl m-methoxybenzylidenecyanoacetate 8b, respectively, to give 1:1 alternating copolymers 9-10 in high yields. Cross-linked polymers 5-6 showed a thermal stability up to 300 ℃, and showed a double phase degradation pattern in their TGA thermograms. Polymers 5-6 showed broad endothermic bands around 75-110 ℃ without any characteristic Tg peaks in DSC thermograms. Alternating copolymers 9-10, except copolymer 9b were soluble in common organic solvents. The inherent viscosities of polymer 9-10 were in the range of 0.35-0.62 dL/g. Polymer films cast from acetone solution were cloudy and tough and Tg values obtained from DSC thermograms were in the range of 118-165 ℃.

Bioactive compounds in food for age-associated cognitive decline: A systematic review (인지기능 개선을 위한 식품유래 생리활성소재에 대한 체계적 문헌고찰)

  • The rapid aging of society has led to a surge in cognitive dysfunction in the elderly. As there is limited evidence for the development of dementia in medicine, a shift in focus on prevention strategies using bioactive compounds in food is required. This systematic review evaluated the effects of various bioactive compounds on age-associated cognitive decline. The literature was searched for terms related to bioactive compounds in cognitive decline and article selection was limited to clinical randomized controlled trials for a single bioactive compound. We identified 21 studies that evaluated the strength of the evidence. ω-3 fatty acids and vitamin B presented a strong evidence level, whereas vitamin D and E, anserine/carnosine, and chromium were defined as having moderate levels of evidence. ω-3 fatty acids relieved cognitive decline and reduced amyloid β-related protein accumulation. Vitamin B decreased homocysteine levels, which is accompanied by alleviation of cognitive function. In conclusion, ω-3 and vitamin B have the potential to improve age-associated cognitive decline.

  • $(NH_4)_{4.5}[H_{3.5}{\alpha}-PtMo_6O_{24}]{\cdot}1.5\;H_2O(A),\;(NH_4)_4[H_4{\beta}-PtMo_6O_{24}]{\cdot}1.5\;H_2O(B),\;and\;K_{3.5}[H_{4.5}{\alpha}-PtMo_6O_{24}]{\cdot}3\;H_2O(C)$ have been synthesized and their molecular structures have been also determined by single-crystal X-ray diffraction technique. Compounds A and C contain the well-known Anderson structure (planar structure) heteropoly oxometalate having approximate $bar{3}_m(D_{3d})$ symmetry, while compound B contains the bent structure heteropoly oxometalate having appproximate $2_{mm}(C2_v)$ symmetry. The bent structure and the planar one are geometrical isomers. These compounds are rot only novel heteroply molybdates containing platinate(IV) but also the first example of geometrical isomerism in the hexamolybdoheteropoly oxometalates. That isomerization surprisingly occurred because of the change of only 0.5 non-acidic hydrogen atom attached to the polyanion such as $[H_{3.5}{\alpha} -PtMo_6O_{24}]^{4.5-}{\to}[H_4{\beta}-PtMo_6O_{24}]^{4-}{\to}[H_{4.5}{\alpha} -PtMo_6O_{24}]^{3.5-}$. It seems that the gradual protonation of the polyanion plays an important role in that isomerism. These heteropolyanions form dimers by strong hydrogen bonds between two heteropolyanions in the respective crystal system.
