$IO_4$-산화전분 변형에 의한 밀 $\beta$-Amylase의 안정성 증가 (Stabilization of Wheat $\beta$-Amylase by Modification with $IO_4$-Oxidized Starch)

  • 과요오드산-산화전분으로 밀 $\beta$-amylase(Himaltosin GL. 일본한큐바이오사)를 변형시켜서 인공당단백질을 만들었다. pH 8.0에서 변형한 효소는 비변형효소의 96%. pH 9.7에서 변형한 효소는 17%의 활성이 남았다. 6$0^{\circ}C$에서의 열 안정성은 $\alpha$-cyclodextrin ( $\alpha$-CD) 존재 시에 변형하여 $\alpha$-CD 존재시에 분석한 효소는 10분 뒤에 비활성의 8%가 남은 반면 변형하지 않은 효소는 5% 밖에 남지 않았다. pH안정성은 변형시켜서 $\alpha$-CD존재 하에 분석한 효소가 가장 높아서 pH 2~5와 6~12에서 안정성이 매우 증가하였다. HPLC분석 결과 효소는 하나의 피크를 나타냈으며 변형시킨 것은 당결합으로 분자량이 커져서 유출시간이 약간 빨라졌다.

$La_{0.5}Ce_{0.5}Co_{1-x}Cu_xO_{3-{\alpha}}$ Perovskite촉매의 선택적 CO 산화반응 및 특성 분석에 관한 연구 (Study on Catalytic Activity of the Selective CO Oxidation and Characterization Using $La_{0.5}Ce_{0.5}Co_{1-x}Cu_xO_{3-{\alpha}}$ Perovskite Catalysts)

  • [ $La_{0.5}Ce_{0.5}Co_{1-x}Cu_xO_{3-{\alpha}}$ ](X=0, 0.1, 0.3, 0.5) perovskites were prepared by coprecipitation method at pH 7 or pH 11 and its catalytic activity of selective CO oxidation was investigated. The characteristics of these catalysts were analyzed by $N_2$ adsorption, X-ray diffraction(XRD), SEM, $O_2$-temperature programmed desorption(TPD). The pH value at a preparation step made effect on particle morphology. The smaller particle was obtained with a condition of pH 7. The better catalytic activity was observed using catalysts prepared at pH 7 than pH 11. The maximum CO conversion of 98% was observed over $La_{0.5}Ce_{0.5}Co_{0.7}Cu_{0.3}O_{3-{\alpha}}$ at $320^{\circ}C$. Below $200^{\circ}C$, the most active catalyst was $La_{0.5}Ce_{0.5}Co_{0.9}Cu_{0.1}O_{3-{\alpha}}$, of which conversion was 92% at $200^{\circ}C$. By the substitution of Cu, the evolution of ${\alpha}$-oxygen was remarkably enhanced regardless of pH value at preparation step according to $O_2$-TPD. Among the different ${\alpha}$-oxygen species, the oxygen species evolved between $400^{\circ}C$ and $500^{\circ}C$, gave the better catalytic performance for selective CO oxidation including $La_{0.5}Ce_{0.5}CoO_3$ in which Cu was absent.

Paenibacillus woosongensis의 만난분해 효소활성 (Mannanolytic Enzyme Activity of Paenibacillus woosongensis)

  • 만난의 분해활성을 갖는 Paenibacillus woosongensis의 배양 상등액으로부터 mannanase, ${\beta}$-mannosidase와 ${\alpha}$-galactosidase 활성이 관찰되었다. 배양상등액 내의 locust bean gum를 분해하는 mannanase는 pH 5.5와 $60^{\circ}C$, para-nitrophenyl-${\beta}$-D-mannopyranoside를 분해하는 ${\beta}$-mannosidase는 pH 6.5, $50^{\circ}C$, para-nitrophenyl-${\alpha}$-D-galactopyranoside를 분해하는 ${\alpha}$-galactosidase는 pH 6.0-6.5와 $50^{\circ}C$에서 각각 최대활성을 보였다. 배양상등액의 만난 분해효소는 mannotriose, mannotetraose, mannopentaose, mannohexaose와 같은 만노올리고당을 분해할 뿐 아니라 mannobiose도 분해하였다. 또한 melibiose, raffinose, stachyose 등의 ${\alpha}$-1,6 결합형 galacto-oligosaccharides를 분해하여 galactose를 생성하였다. 이러한 결과로 보아 P.woosongensis는 galactomannan을 완전히 분해하는데 필요한 3종류 효소를 모두 생산하는 것으로 판단된다.

대두(大豆) 및 Aspergillus niger α-galactosidase의 효소학적(酵素學的) 연구(硏究) (Enzymatic Studies on the α-Galactosidases from Soybean and Aspergillus niger)

  • 발아대두 $\alpha$-galactosidase와 Aspergillus niger가 생산하는 $\alpha$-galactosidase의 효소학적 성질을 비교하기 위하여 대두 발아 중의 $\alpha$-galactosidase활성 및 소당류의 함량변화를 측정하였고, 대두 발아 과정 중 활성이 가장 강할때에 추출한 $\alpha$-galactosidase 및 Aspergillus niger를 밀기울 배양하였을 때 생성되는 $\alpha$-galactosidase를 염석, 이온교환 크로마토그래피 및 겔여과 등을 사용하여 정제한 후 정제효소의 이화학적 및 효소학적 성질을 측정, 비교하여 다음과 같은 결과를 얻었다. 1. 대두 $\alpha$-galactosidase의 활성은 대두를 $25^{\circ}C$에서 120시간 발아시켰을 때 가장 높았으며, 대두 중의 raffinose는 96시간, stachyose는 120시간 발아시켰을 때 완전히 분해되었다. 2. Aspergillus niger를 밀기울배지에서 $30^{\circ}C$, 4일간 배양했을 때 $\alpha$-galactosidase 활성이 최고에 달하였다. 3. 대두 $\alpha$-galactosidase는 황산암모늄 염석, DEAE-Cellulose 및 DEAE-Sephadex A-50 이온교환 크로마토그래피, Sephadex G-l50 겔여과 등에 의하여 6.6배까지 정제되었으며 그의 비활성이 825U/mg protein, 수율 2.5%에 달하였고, Aspergillus niger의 $\alpha$ -galactosidase는 23.7배까지 정제되었으며 그의 비활성이 1,229U/mg protein, 수율 14%에 달하였다. 4. 정제된 대두 및 Aspergillus niger의 $\alpha$-galactosidase는 HPLC, PAGE 및 SDS-PAGE에 의해서 순도가 확인되었다. 5. 정제효소의 이화학적 성질 1) Aspergillus niger의 $\alpha$-galactosidase는 periodic acid schiff 염색에 의하여 당단백질임이 확인되었다. 2) 대두 $\alpha$-galactosidase의 등전점은 pH4.8이었고, 분자량이 30,000인 monomer이었으나, Aspergillus niger의 $\alpha$-galactosidase는 등전점이 pH4.6이었고 분자량은 112,000이었으며 분자량 28,000인 monomer 4개로 구성된 tetramer이었다. 3) 대두 및 Aspergillus niger $\alpha$-galactosidase의 활성에 관여하는 아미노산은 diethyl pyrocarbonate에 의한 화학 수식에 의하여 histidine임이 확인되었다. 4) 대두 $\alpha$-galactosidase의 활성은 2-mercaptoethanol과 L-cysteine에 의하여 약간 저해되었다. 6. 정제효소의 효소학적 성질 1) 대두 $\alpha$-galactosidase의 최적 작용 pH는 pH6.0, 최적 작용온도는 $40^{\circ}C$이었고, Aspergillus niger $\alpha$-galactosidase 각각 pH6.5 및 $40^{\circ}C$이었다. 2) 대두 및 Aspergillus niger의 $\alpha$-galactosidase는 $45^{\circ}C$이하에서 비교적 안정하였으나 $60^{\circ}C$에서 10분 처리시 대두 $\alpha$-galactosidase는 25%, Aspergillus niger $\alpha$-galactosidase는 46%의 잔존활성을 나타내었다. 3) 대두 $\alpha$-galactosidase는 pH5.5~6.5, Aspergillus niger의 $\alpha$-galactosidase는 pH6.0~7.0에서 매우 안정하였다. 4) 대두 및 Aspergillus niger의 $\alpha$-galactosidase간에 기질 특이성상의 차이가 없었으며, stachyose보다 raffinose를 잘 분해하였고, gaIactose는 양효소의 활성을 저해하였다. 5) 대두 $\alpha$-galactosidase의 P-nitrophenyl-$\alpha$-gaIactopyranoside, raffinose 및 stachyose에 대한 Km값은 각각 5.3mM, 50.0mM 및 55.5mM이 었고, Aspergillus niger $\alpha$-galactosidase에 있어서는 각각 5.0mM, 37.0mM 및 55.5mM이었다. 6) 대두 $\alpha$-galactosidase의 p-nitrophenyl-$\alpha$-d-gaIactopyranoside에 대한 활성화 에너지는 13.024Kcal/mole, $Q_{10}$값은 2.0이 었으며, Aspergillus niger의 $\alpha$-galactosidase는 각각 8.515Kcal/mole 및 1.38이었다.

ESCAPE OF RESONANTLY SCATTERED LYβ AND Hα FROM HOT AND OPTICALLY THICK MEDIA

  • We investigate the escape of $Ly{\beta}$ from emission nebulae with a significant population of excited hydrogen atoms in the level n = 2, rendering them optically thick in $H{\alpha}$. The transfer of $Ly{\beta}$ line photons in these optically thick regions is complicated by the presence of another scattering channel leading to re-emission of $H{\alpha}$, alternating their identities between $Ly{\beta}$ and $H{\alpha}$. In this work, we develop a Monte Carlo code to simulate the transfer of $Ly{\beta}$ line photons incorporating the scattering channel into $H{\alpha}$. Both $H{\alpha}$ and $Ly{\beta}$ lines are formed through diffusion in frequency space, where a line photon enters the wing regime after a fairly large number of resonance scatterings with hydrogen atoms. Various line profiles of $H{\alpha}$ and $Ly{\beta}$ emergent from our model nebulae are presented. It is argued that the electron temperature is a critical parameter which controls the flux ratio of emergent $Ly{\beta}$ and $H{\alpha}$. Specifically for $T\;=\;3{\times}10^4\;K$ and $H{\alpha}$ line center optical depth $\tau{\alpha}\;=\;10$, the number flux ratio of emergent $Ly{\beta}$ and $H{\alpha}$ is ~ 49 percent, which is quite significant. We propose that the leaking $Ly{\beta}$ can be an interesting source for the formation of $H{\alpha}$ wings observed in many symbiotic stars and active galactic nuclei. Similar broad $H{\alpha}$ wings are also expected in $Ly{\alpha}$ emitting halos found in the early universe, which can be potentially probed by the James Webb Telescope in the future.

자외선 조사에 의한 수용액 중에 함유되어 있는 엔도설판 alpha, beta, sulfate의 광분해 (Photodegradation of Endosulfan alpha, beta, and sulfate in Aqueous Solution by UV Irradiation Only)

  • Photodegradation of endosulfan alpha, beta, and sulfate known as the most toxic substance among organochlorine pesticides by UV irradiation was studied at experimental conditions such as different pH aqueous solution and reaction time. The initial concentration of endosulfan alpha, beta, and sulfate in aqueous solution was 500 ppb, respectively. The experiment of photodegradation was conducted in a quartz reactor equipped with a low pressure mercury lamp (100 W, 240 nm). The samples were withdrawn from the photo reactor at intervals of 0, 10 min, 30 min, 1 hr, 2 hr, and 4 hr. Endosulfan sulfate was never hydrolyzed and photodegraded in wide range of pH. At pH 5 and reaction time (240 min), endosulfan alpha was photodegraded up to 67%. Both endosulfan alpha and beta were started to photodegrade at pH 6.5 with the lapse of time, resulting in approximately 99.9% and 87.2% of photodegradation efficiency, respectively. Furthermore, at pH 9, endosulfan alpha and beta was partially hydrolyzed and photodegraded to 99.5% at 120 min of reaction time. During the photolysis, any photo-products of endosulfan alpha, beta, and sulfate were not observed.

침전법으로 제조한 Alumina 분말의 특성(III) : 소결거동 (Properties of Alumina Powder Prepared by Precipitation Method (III) : Sintering Behavior)

  • Alpha alumina powder with fine particle size and narrow particle size distribution was prepared by precipitation method using Al2(SO4)3.18H2O as a starting material. The alpha alumina powder was prepared by calcining aluminum hydroxide which was formed under various pH values. The sinterabilityof alpha alumina powder and the effect of MgO on the sinterability of alpha alumina powder were investigated. The sinterability of alpha alumina powder was the order of pH=10≒pH11>pH=7≒pH9, and alpha alumina obtained from boehmite which was prepared by precipitation method reached to 97.5% of theoretcal density by the pressureless sintering. The effect of MgO on volume shrinkage of alumina was very slight in the initial sintering stage but remarkable in the final sintering stage. It was also found that MgO controlled effectively the grain growth of alumina.

BC3H-1 분화세포에서의 (Na,K)ATPase ${\alpha}_2$ isoform의 표현증대 (Increased Expression of the ${\alpha}_2$ Isoform of (Na,K)ATPase in the Differentiated Murine Muscle Cell Line BC3H-1)

  • The development of the alpha2 isoform of (Na,K)ATPase which is high affinity ouabain receptors was studied in the differentiating nonfusing muscle cell line BC3H-1. T he differentiation process of BC3H-1 cell line was confirmed by 2-dexy-D-[$^3$H] glucose uptake experiment and the quantity of the expression of ${\alpha}_2$ isoform was measured using a whole cell [$^3$H] ouabain-binding assay. Undifferentiated growing BC3H-1 cells, myoblasts, exhibited low levels of insulin-stimulated glucose uptake and [$^3$H] ouabain-binding sites. In contrast, differentiated BC3H-1 cells, myocytes, had a 5.6-fold increase in insulin-stimulated glucose uptake and 5-fold increase in [$^3$H] ouabain-binding sites. Scatchard analysis showed that myocytes developed more [$^3$H] ouabain-binding sites than myoblasts vath a dissociation constant (kd) of 6${\times}10^{-8}$M and capacity of 6.l${\times}10^{-5}$ sites/cell. Therefore. it seems that myoblasts express low levels of ${\alpha}_2$ subunit and probably the majority of ${\alpha}_1$ subunit, whereas myocytes express high levels of ${\alpha}_2$ isoform. The results indicate that the expression of ${\alpha}_2$ isoform is developmentally regulated during differentiation and that BC3H-1 culture system provides an excellent model for the study of differentiation and mechanism of (Na,K)ATPase action in muscle which requires electrical excitability.

과요오드산 산화전분 변형에 의한 아밀라아제의 안정화 (Stabilization of Amylolytic Enzymes by Modification with Periodate-Oxidized Soluble Starch)

  • The stabilizatio of amaylolytic enzyme such as $\beta$-amylase of barley, $\beta$-amylase of wheat, $\beta$-amylase of sweet potato, $\alpha$-amylase of Bacillus licheniformis, $\alpha$-amylase of Aspergillus sp. and $\alpha$-glucosidase of Aspergillus awamori was attained by modification with periodate-oxidized soluble starch. The pH stability of modified enzyme was increased at pH 9 for $\beta$-amylase of sweet potato, pH 3~5 and 8~11 for $\beta$-amylase of barley, pH 2~3 and 7~12 for $\beta$-amylase of wheat and pH 6 for $\alpha$-glucosidase of Aspergillus awamori. Thermal stability increased 17.6% for $\alpha$-amylase of Aspergillus sp. at 6$0^{\circ}C$ for 10min, 30% for $\alpha$-amylase of Bacillus licheniformis at 10$0^{\circ}C$ for 5min and 4.5% for $\alpha$-amylase of sweet potato at 6$0^{\circ}C$ for 10min compared with those of native enzymes.

  • pH 9.1, 9$0^{\circ}C$, 6시간의 조건에서 $\alpha$-tricalcium phosphate ($\alpha$-Ca$_3$(PO$_4$)$_2$)의 가수분해반응에 의해서 잘 발달된 수산화아파타이트 휘스커상 (길이 5$mu extrm{m}$,지름 0.5$\mu\textrm{m}$)이 합성되었다 수산화아파타이트로 $\alpha$-Ca$_3$(PO$_4$)$_2$의 전환에 미치는 반응조건(온도, 시간, pH)의 영향이 조사되었다. 아울러 XRD, SEM, ICP, TGA와 같은 분석기기를 사용하여 수산화아파트의 미세구조, 조성, 열적 안정성을 검토하였다.
