Supercritical Fluid Dyeing of Polyester Fiber with Two Different Dispersion Dyes (두 종류의 분산염료를 이용한 폴리에스테르 섬유의 초임계유체 염색)

  • The dyeing of polyester fiber with two different disperse dyes (Disperse Red 60 and Disperse Yellow 54) was carried out using supercritical carbon dioxide ($SCCO_2$) as a dyeing medium at temperatures ranging from $50^{\circ}C$ to $90^{\circ}C$ and at pressures from 15 MPa to 30 MPa. The dye uptake of Disperse Red 60 on polyester fiber was found to increase with temperature at constant pressure and $SCCO_2$ density (700 kg/$m^3$). At $90^{\circ}C$ and 30 MPa, the dye uptake on polyester fiber increased with dyeing time and the saturation concentration of Red 60 was attained within 240 min, while a longer dyeing time was required for Yellow 54 to reach its saturation concentration. When dyestuff mixtures with mixing ratios of 0.01 to 9.0 (Red 60/Yellow 54) were used, the uptake ratio of the two dyes was found to be proportional to 0.26 power of their mixing ratio. Dyed fibers showed an orange color and the depth of the color depended upon the mixing ratio of the dyestuffs.

  • The present study was conducted to investigate results of B. anthracis isolated from Anthrax in the Kyong-Ju of Feb. 12. 1994. 1. In biochemical feature, B. anthracis was a gram-positive rod, non-motility, sporulation, capsulation. It was positive in gelatinase, starch hydrolysis, glucose. But negative in urease, arabinose, mannitol, xylose. 2. B. anthracis grew well on B4 Br A TSA after incubation for 24 hours. The organisim grew well on BA, Br. A, NA, TSA after incubation for 72 hours. The media grew well on Br A instead of BA. 3. On 5% blood agar by laboratory animal, ${\beta}$ -hemolysis was produced from 36 hours to 48 hours incubation. There was perfect ${\beta}$-hemolysis after incubation for 48 hours. On the other side ${\beta}$-hemolysis was begun on 5% goat blood agar after incubation for 60 hours. 4. In the test of antimicrobial susceptibility, B. anthracis was very sensitive to AM, CF, TE, ENR, GM, AN, DFX, S, P, TYLO, N, KM, C, E, Lins+Sp, NN, CC, CFP, CB were sensitive one by one. B. anthracis was no-sensitive to L, XNL, TIA, CL, SXT 5. B. anthracis had never sensitivity to direct inoculation of rat and chicken, after subcutanous inj. It was very sensitive to mouse and goat, hamster, guinea pig, rabbit had a sensibility one by one. 6. The dead laboratory animal which had been inoculated with B. anthracis preserved at $37^{\circ}C$ incubation, B. anthracis didn't cultivate on non-dissected animal after 80 hours but cultivate on dissected animal after 360 hours. 7. The rapidly death could cause high concentration, died from 420 after S. C. 8. The blood smeared samples of hamster from inoculation with B. anthracis, spore germinated In 37$^{\circ}C$ after 5 hours, in $32^{\circ}C$ after 6 hours, in room temperature after 9 hours, in $-4^{\circ}C$ to $-20^{\circ}C$ after 10 hours. 9. B, anthracis inoculated to laboratory animal after SC or PO. Mice and rats feces didn't cultivated with B. anthracis after SC, but did cultivated with B. anthracis after PO. 10. In the test of disinfectant, B. anthracis was high effective to $HgC1_2$, formalin, effect phenol, cresol, but non-effect NaOH, ethanol.

Mineralogical Characteristics of Tosudites from the Sungsan and Bubsoo Mines, Korea (성산광산과 법수광산에서 산출되는 토수다이트의 광물학적 특성)

  • Mineralogical characteristics of tosudite from the Sungsan and Bubson mines were studied and correlated using X-ray diffraction analysis, chemical analysis and electron microscopy. Tosudite occurs as an alteration product of Cretaceous volcanoclastic rocks in both mines. It is associated with microcrystalline quartz, dickite, illite/smectite or mica/smectite mixed-layer mineral. It forms cryptocrystalline aggregates with flaky habit. XRD analysis suggests that tosudite is an 1:1regularly interstratified dioctahedral smetite/dioctahedral chlorite. Bubsoo tosudite has more(00ℓ ) reflections and more periodice stacking sequence than Syngsan tosudite. Chemical analysis shows that tosudite is a Li-bearing aluminous 1:1 regularly interstrattified mineral composed of K-bedellite and donbassite. Cookeite component may be present in the chlorite layer. Bubsoo tosudite is more Al in tetrahedral site and Ca in interlayer, but less Al in octahedral site than Sugsan tosudite. Tosudite may be formed as the intermediate alteration products, forming after muscovite and before illite/smectite or mica/s$^{\circ}C$mectite, with the range from 100 $^{\circ}C$ to 360 ~ 480 $^{\circ}C$. The hydrothermal solution forming tosudite may be acidic solution with high activities of Si and Al.

Structural Analysis of Volatile Matters and Heavy Oil Fractions from Pyrolysis Fuel Oil by the Heat Treatment Temperature (열처리 온도에 따른 열분해 연료유 내 휘발유분 및 잔류 중질유분의 구조 분석)

  • In order to investigate structural changes of the pyrolysis fuel oil (PFO), the volatile matters and heavy oil fractions were separated from PFO by heat treatment temperature. As a result of $^1H-NMR$ analysis of volatile matters, 1~2 ring aromatic compounds contained in the petroleum residue were mostly removed at a temperature before $340^{\circ}C$. Moreover, new peaks corresponding to aliphatic hydrocarbons were detected at the chemical shift of 2.0~2.4 ppm. It is attributed that the aliphatic hydrocarbon sidechain was cracked from the aromatic compound by the cracking reaction occurred at $320^{\circ}C$. The C/H mole ratio and aromaticity increased with increasing the heat treatment temperature. Therefore, from the structural analysis results of heavy oil fractions and volatile matters from PFO, the decomposition of the aliphatic sidechain by cracking reaction and the separation of volatile matters by boiling point of components were mostly affected structure changes of the PFO.

Quality Assurance of Volumetric Modulated Arc Therapy for Elekta Synergy (Elekta Synergy 선형가속기를 이용한 입체적세기조절회전방사선치료(VMAT) 정도관리)

  • For applying the quality assurance (QA) of volumetric modulated arc therapy (VMAT) introduced in Eulji Hospital, we classify it into three different QA steps, treatment planning QA, pretreatment delivering QA, and treatment verifying QA. These steps are based on the existing intensity modulated radiation therapy (IMRT) QA that is currently used in our hospital. In each QA step, the evaluated items that are from QA program are configured and documented. In this study, QA program is not only applied to actual patient treatment, but also evaluated to establish a reference of clinical acceptance in pretreatment delivering QA. As a result, the confidence limits (CLs) in the measurements for the high-dose and low-dose regions are similar to the conventional IMRT level, and the clinical acceptance references in our hospital are determined to be 3 to 5% for the high-dose and the low-dose regions, respectively. Due to the characteristics of VMAT, evaluation of the intensity map was carried out using an ArcCheck device that was able to measure the intensity map in all directions, $360^{\circ}$. With a couple of dosimetric devices, the gamma index was evaluated and analyzed. The results were similar to the result of individual intensity maps in IMRT. Mapcheck, which is a 2-dimensional (2D) array device, was used to display the isodose distributions and gave very excellent local CL results. Thus, in our hospital, the acceptance references used in practical clinical application for the intensity maps of $360^{\circ}$ directions and the coronal isodose distributions were determined to be 93% and 95%, respectively. To reduce arbitrary uncertainties and system errors, we had to evaluate the local CLs by using a phantom and to cooperate with multiple organizations to participate in this evaluation. In addition, we had to evaluate the local CLs by dividing them into different sections about the patient treatment points in practical clinics.

Microwave Dielectric Properties of (Ba1-2xNa2x)(Mg0.5-xZrxW0.5)O3 Ceramics ((Ba1-2xNa2x)(Mg0.5-xZrxW0.5)O3 세라믹스의 마이크로파 유전특성)

  • We investigated the phase evolution, microstructure, and microwave dielectric properties of Na- and Zr-doped $Ba(Mg_{0.5}W_{0.5})O_3$ [i.e., ($Ba_{1-2x}Na_{2x})(Mg_{0.5-x}Zr_xW_{0.5})O_3$] ceramics. $BaWO_4$ as a secondary phase was observed in all compositions, and it increased as the dopant concentration increased. All specimens revealed a dense microstructure. For the composition of x=0.01, polyhedral grains were observed. As the dopant concentration increased, the densification and the grain growth were promoted by a liquid phase. The quality factor($Q{\times}f_0$) decreased remarkably, whereas the dielectric constant (${\varepsilon}_r$) tended to decrease as the dopant concentration increased. The dielectric constant, quality factor, and temperature coefficient of the resonant frequency of the composition of x=0.01 sintered at $1,700^{\circ}C$ for 1 h were 18.6, 216,275 GHz, and $-22.0ppm/^{\circ}C$, respectively.

Development of a thermo-stabel ${\beta}-agarase$ from marine organism

  • Neoagaro-oligosaccharides are produced only by enzymatic degradation of agarose by ${\beta}-agarase.^{1)}$ Neoagaro-oligosaccharides inhibit the growth of bacteria, slow the rate of degradation of starch, are used as low-calorie additives to improve food quality, and have macrophage-stimulating activity. Furthermore, neoagarobiose is a rare reagent that has both moisturizing effect on skin and whitening effect on melanoma $cells.^{2)}$ An agar-degrading marine bacterium was isolated from the sea water at the northeast coast in Cheju island, Korea. The strain was gram negative, aerobic, and motile rod. The 16S rRNA of the strain had the closest match of 98% homology, with that from Agarivorans albus. On the basis of several phenotypic characters and a phylogenetic analysis, this strain was designated Agarivorans sp. JA-1. In solid agar plate, Agarivorans sp. JA-1 produced a diffusible agarase that caused agar softening around the colonies. Agarivorans sp. JA-1 was cultured for 36 hr in marine broth 2216 (Difco, USA) and the supernatant that containing an extracellular ${\beta}-agarase$ was prepared by centrifugation of culture media. The enzyme exhibited relatively strong activity at $40^{\circ}C$ and was stable up to $60^{\circ}C$. Using PCR primers derived from the ${\beta}-agarase$ gene of Vibrio sp., the gene encoding ${\beta}-agarase$ from Agarivorans sp. JA-1 was cloned and sequenced. The structural gene consists of 2931 bp encoding 976 amino acids with a predicted molecular weight of 107,360 Da. The deduced amino acid sequence showed 99% and 34% homology to $agaA^{2)}$ and $agaB^{2)}$ genes for ${\beta}-agarase$ from Vibrio sp., respectively. The expression plasmid for ${\beta}-agarase$ gene of Agarivorans sp. JA-1 is being constructed and the recombinant enzyme will be biochemically characterized.

A Study on the Decompressed Ammonia Stripping from Ammonia Contained Wastewater (폐수의 감압 암모니아 탈기에 관한 연구)

  • This study aims at finding out pertinent reaction conditions for treating high concentration ammonia contained in N-chemical factory wastewater with decompressed ammonia stripping method that was designed. And it also tries to investigate adsorption capability of removed ammonia to soil. The results from experiments are as follows ; 1. The removal rate of N $H_3$-N of synthetic wastewater was under 85% at pH 10 with decompressed ammonia stripping method. The reaction time in pressure 360 mmHg at pH 11 and 12 was shorter than in 460 mmHg, and the removal rate of N $H_3$-N with decompressed ammonia stripping method at 9$0^{\circ}C$ was 11~15% higher than air stripping 2. The optimum conditions for decompressed ammonia stripping with synthetic sample were shown as pH 12, temperature 9$0^{\circ}C$, internal reaction pressure 460 mmHg and reaction time 50 minutes. These conditions were applied to treat the wastewater containing organic-N 290.5mg/$\ell$, N $H_3$-N 168.9mg/$\ell$, N $O_2$-N 23.2mg/$\ell$, N $O_3$-N 252.4mg/$\ell$, T-N 735mg/$\ell$. Organic-N turned out to be removed 60%, the removal rate of N $H_3$-N IS 94%, T-N is 50%. But N $O_2$-N and N $O_3$-N were increased with 7.8% and 14.9% respectively. 3. The CO $D_{Sr}$ removal rate in decompressed ammonia stripping reaction was 42% and S $O_4$$^{2-}$ was removed 8.2%. It was turned out caused with higher pH and thermolysis. 4. In soil adsorption of ammonia desorbed from the decompressed stripping process of wastewater, the recovery rate was 76% in wet soil.

  • Tests were performed on 232 bacterial strains (71 strains of Pseudomonas tolaasii and 161 white line reacting organisms, WLRO) isolated from cultivated mushrooms. As results, P. tolaasii was divided into 5 groups on the basis of the phenotypical characteristics for the strains, and group 3 was the major one including 48 (62%) out of the total 71 strains. WLRO were classified into 23 groups, and group 10 was the major group (65 strains, 30% of the total WLRO tested). A white line was well formed at 22$^{\circ}C$ and at 4 mm distance between P. tolaasii and WLRO colonies in their dual culture on Pseudomonas agar F medium within 36-hr incubation, but not formed at $25^{\circ}C$ even for 72-hr incubation. The morphological, cultural and biological properties of P. tolaasii group 3, and the main group of WLRO, group 10, were different only in the light of pathogenicity. Also group 2 of P. tolaasii had the characteristics similar to group 24 of WLRO which was pathogenic to cultivated mushrooms, suggesting the P. tollaasiii and WLRO strains may be the same species although their white line forming ability and pathogenicity were more or less different from one another. Subculture of the strains in P. tolaasii group 1 induced the variation of their pathogenicity, white line forming ability and utilization of sodium benzoate and sodium tartrate, and thus it can be inferred that they were converted to strains having the characteristics of group 3 or WLRO groups.

Production and Characterization of Acid-stable Pectin Lyase from Bacillus sp. PN33

  • A bacterial strain PN33 producing large amounts of extracellular pectin lyase (PNL, EC 4.2.2.10) was isolated from soil. The isolated bacterium was identified as a strain of Bacillus sp. Production of PNL by the strain was induced only by pectins, with a higher degree of esterification, which had been added to the culture medium as a sole carbon source. The optimal medium for PNL production was determined to consist of 10 g pectin, 2 g yeast extract, 4 g $K_2HPO_4{\cdot}3H_2O$, 0.6 g $MgSO_4$, and 0.11 g $CaCl_2$ per liter (pH 7.0). The PNL activity in the culture supernatant reached the highest level of 132 mU/ml after 32 h cultivation at $37^{\circ}C$ in the optimal medium. The PNL produced was purified to homogeneity by ammonium sulfate fractionation (50~80%), and cation exchange and size exclusion chromatographies. The molecular mass of the enzyme was estimated to be approximately 52 kDa by SDS-PAGE. Almost the same mass was determined by nondenaturing PAGE, indicating that the functional enzyme had a monomeric structure. As expected, the PNL exhibited higher activities on the highly esterified pectins whereas it gave no detectable activity on polygalacturonic acid. The enzyme showed the highest activity at the acidic pH of 6.0, exceptional for a bacterial PNL. Maximum activity was measured at $40^{\circ}C$, although the stability f the purified enzyme was poor at this temperature. alcium (1 mM) was found to activate the PNL activity by $50\%$, and also remarkably increased the thermal stability f the enzyme. Phenylmethylsulfonylfluoride (PMSF) and iethylpyrocarbonate (DEPC) inhibited the PNL activity lmost completely at the concentration of 5 mM. This result ndicates that some serine and histidine residues of the nzyme may play an essential role for catalytic function of he enzyme.
