노년기 인지기능 및 건강상태를 반영하는 지표로써 Alpha 고유리듬과 뇌파 및 HRV 지표와의 관계 연구 (A Study of Intrinsic Alpha Rhythm, Electroencephalography, and Heart Rate Variability Index as Indicators of Cognitive Function and Health in Elderly Adults)

  • 이 연구는 노년기 인지기능 및 생리적 건강상태와 관련 있는 정량화 뇌파의 Alpha 고유리듬 피크 위치 및 진폭의 출현 유무가 정신적·육체적 스트레스, 주의집중 등의 뇌파 지표들과 심박변이도 지표 간에 어떠한 관련성과 임상적 의미가 있는지 알아보고자 하였다. 이를 위하여 평균 73세 남녀 노인 76명을 대상으로 전두엽 2채널 뇌파와 1채널 심전도를 동시에 측정 가능한 장치를 이용하여 폐안시 EEG, HRV를 측정하였다. 의미있는 분석 결과는 다음과 같다. 첫 번째, Alpha 고유리듬피크 위치와 상관없이 고유리듬 진폭이 높게 잘 나타난 집단은 낮은 집단에 비해 정신적 스트레스에 대한 저항도는 높고, 육체적 스트레스는 낮게 나타나는 상관성을 보였다. 두 번째, 심박변이도의 HRV index, SDNN, RMSSD 지표는 집단 구분에 상관없이 공통적으로 서로 간 높은 양의 상관성을 보였다. 세 번째, Alpha 고유리듬 진폭이 잘 나타나지 않은 집단에서 좌·우뇌 Alpha 비대칭이 커질수록 육체적 스트레스를 반영하는 Delta/Alpha 비율과 주의집중 저하를 반영하는 Theta/SMR 비율도 커지는 상관성을 보였다. 네 번째, Alpha 고유리듬피크가 느린 리듬쪽에 위치한 집단은 빠른 리듬 쪽에 위치한 집단에 비해 Theta/SMR 비율이 높게 나타나 주의집중도 저하와 관련이 있음을 의미하였다. 따라서 정량화 뇌파의 Alpha 고유리듬 피크 위치 및 진폭의 출현 유무는 노년기 인지기능 및 생리적 건강상태를 반영하는 지표로 활용 가능성이 있음을 시사하였다.

폐암 환자의 혈청과 조직 표본상에서 Alpha 1-Proteinase Inhibitor의 조사 연구 (Investigation of Alpha 1-Proteinase Inhibitor in Serum and Specimen of Lung Cancer Patients)

  • Alpha 1-Proteinase inhibitor[PI] was known as a major protective enzyme against to excessive hydrolytic and proteolytic reaction. So, it was suggested that Alpha 1-PI may implicated in growth of bronchogenic cancer. This study was undertaken to investigate the role of Alpha 1-PI in local invasion of bronchogenic cancer. Three groups of patients were studied; Preliminary research group of 15 bronchogenic cancer patients, Main research group of 13 bronchogenic cancer patients and Normal control group of 10 nephrectomy donor. Serum Alpha 1-PI level was observed in each group of patients during pre-and postoperative days. Pre-operative serum Alpha 1-PI level in preliminary research group [329.2$\pm$14.21mg/dl]and main research group[406.2$\pm$39.30mg/dl] were higher than in normal control group[236.2$\pm$19.55mg/dl] significantly[p<0.005]. Serial Alpha 1-PI level in each group during pre-and postoperative days shows peaked at 3rd. postoperative day in preliminary and main research group, thereafter decreased gradually. Immunohistochemical study for Alpha 1-antitrypsin[A1AT] was carried out by ABC[avidin-biotin peroxidase complex] method using Alpha-1 antitrypsin DAKOR to tumor tissues of 13 lung cancer patients in main research group. 6 cases[46.2%, squamous cell ca.;5, adenocarcinoma;1] of above 13 cases show positive immunoreactivity for A1AT. In conclusion, alpha 1-PI and elastase are disclosed that have defined actions for lung cancer growing or spreading.

Lipopolysaccharide가 파골세포 전구세포의 이동에 미치는 영향 (The effect of lipopolysaccharide on the migration of osteoclast precursors)

  • 파골세포에 의한 골흡수는 1) 혈관을 통한 파골세포 전구세포의 골표면 이동 및 2) 골표면에서 파골세포 전구세포로부터 파골세포 분화 두 단계를 거쳐 일어난다. Stromal cell derived factor $(SDF)-1{\alpha}$ 는 파골세포 전구세포의 화학주성인자이며 matrix metalloproteinase (MMP)-9는 파골세포 전구세포의 이동에 관여하는 단백 분해효소이다. 파골세포 전구세포의 골표면 이동에 있어서 LPS의 역할을 규명하기 위하여 E. coli 및 Actinobacillus actinomycetecomitans LPS의 1) 파골세포 전구세포 유도능, 2) LPS에 의한 파골세포 전구세포의 이동에 있어서 MMP 및 $SDF-1{\alpha}$ 의 관련성을 평가하였다. LPS에 의한 차골세포 전구세포의 RAW 세포의 이동은 matrigel 또는 type I collagen을 도포한 transwell을 이용하여 평가하였으며 MMP-9 및 $SDF-1{\alpha}$ 의 발현은 RT=PCR 또는 ELISA로 평가하였다. 각 세균의 LPS는 matrigel 또는 type I collagen을 통한 파골세포 전구세포의 이동을 증가시켰다. MMP 억제제는 각 세균의 LPS에 의한 파골세포 전구세포의 이동을 억제하였다. LPS는 파골세포 전구세포의 MMP-9의 발현을 증가시켰다. 각 세균의 LPS는 마우스 두개골에서 분리한 조골세포의 $SDF-1{\alpha}$ 의 발현을 증가시켰다. $SDF-1{\alpha}$ 을 함유한 LPS 처리 조골세포 배양상층액은 파골세포 전구세포의 이동을 증가시켰으며 anti $SDF-1{\alpha}$ Ab는 LPS처리 세포 배양상층액에 의한 파골세포 전구세포의 이동을 억제하였다. 이들 결과는 LPS가 파골세포 전구세포에서는 MMP-9을 조골세포에서는 $SDF-1{\alpha}$ 의 발현을 증가시켜 파골세포 전구세포의 이동을 촉진 시킬 수 있음을 시사한다.

영지버섯, 여주, 쓴메밀 및 혼합 시료의 메탄올 추출물의 생리활성 (Biological Activity of Methanolic Extract from Ganoderma lucidum, Momordica charantia, Fagopyrum tataricum, and Their Mixtures)

  • 영지버섯, 여주, 쓴메밀 및 이들 혼합 시료의 70% 메탄올 추출물을 이용하여 생리활성물질(폴리페놀 화합물 및 플라보노이드) 농도와 항산화(DPPH radical 소거 활성, 생체막 지질과산화, Fe/Cu 환원력), ${\alpha}$-glucosidase 저해, tyrosinase 저해 및 항혈전 활성에 대하여 검토하였다. 추출 수율과 폴리페놀 화합물 및 플라보노이드의 함량은 혼합 추출물에서 가장 높았다. 또한 혼합 추출물에서 ${\alpha}$-glucosidase 저해, tyrosinase 저해, 항산화, 환원력 및 항혈전 활성은 혼합 추출물에서 가장 높았으며, 이러한 효과는 처리 농도 의존적 이였다. 이상의 실험 결과에서 영지버섯, 여주, 쓴메밀의 혼합 메탄올 추출물은 폴리페놀 화합물과 플라보노이드를 많이 함유함으로서 높은 ${\alpha}$-glucosidase 저해 및 항산화 활성과 관련성이 높아 보여 항당뇨 건강 기능성식품의 소재 개발에 활용될 가능성이 높아 보인다.

치면세균막 분해효소인 $\alpha$-1,3 glucanase를 생산하는 미생물의 분리 및 효소 특성 (Isolation of $\alpha$-1,3 Glucanase from Microorganism and the Prodution of High Activity $\alpha$-1,3 Glucanase for Hydrolysis of Dental Plaque)

  • Seventeen strains were isolated from soil, cattle rumen, cereal sewage dregs, insect on agar plate containing insoluble glucan as a sole carbon source from immobilized Streptococcus mutans, which produced alpha-1,3 glucanase for lysis of dental plaque. Among these strains isolated from soil, SW-522 and SW-713 that had appeared to produce the high level of alpha-1,3 glucanase, degraded insoluble glucan from S. mutans 97.6% and 49.4%, respectively in 5 hours. The activity of crude alpha-1,3 glucanase from SW-522 was 1.3mg insoluble glucan/min.mg protein. This enzyme was entirely degraded insoluble glucan on glass tube which produced by S. mutans in TH medium with 5% sucrose.

수유기에 투여된 Butyl Benzyl Phthalate가 랫드 차산자에 미치는 영향 (Effects of Butyl Benzyl Phthalate on Dams and F1 during Lactation Period of Rats)

  • BBP (Butyl benzyl phthalate), a widely used plasticizer. can enter the food and environment as consequence of its manufacture, use, and disposal. BBP was found to be developmental and teratogenic or endocrine disrupting chemical in rats. The effects of BBP were investigated in female rats (P) and second generation (F1) via lactations. Sprague-Dawley were given BBP by oral administration at 0, 5, 10, 100, 1000 mg/kg on day 0 to 21 of lactation period. The results were as follows : At maternal findings, there were some significant changes (p<0.05) in relative organ weight, especially liver and uterus weight by BBP administration. In estrous cycle, high treated group was inclined to be proestrus or estrus compared to control group. BBP indues estrous cycle earlier than the control group. At fetal findings, there were some significant changes in relative liver and spleen weight, especially 100, 1000 mg/kg administered groups. The relative weight of ventral prostate was decreased, so it was represent to dose-response tendency. Parent rats (P) were detected monobenzyl phthalate (MBeP) 3.21~5.81 $\mu\textrm{g}$/ml in 100, 1000 mg/kg dose groups. MBeP of male and female fetuses (F1) were detected at the level of 1.21~2.63 $\mu\textrm{g}$/ml of serum. Male serum concentration oi MBeP was higher than the females'. Estrogen receptor $\alpha$ expression by BBP and bisphenol A in uterus and testis of F1 were studied. The ER$\alpha$ expression were increased in F1 male testis and female uterus. F1 male showed distint ER$\alpha$ expression, especially in the combined exposrue. Synergistic ER$\alpha$ expression was found by combined treatment group of BBP and bisphenol A. From the above results, it could be concluded that the effects of dams and F1 by BBP administration during lactation period were estrogenic, and BBP can transfer to F1 via lactation, and make estrogenic at F1 reproductive organs.

Comparison of Three Different Culture Systems for Establishment and Long-Term Culture of Embryonic Stem-like Cells from In Vitro-Produced Bovine Embryos

  • Although embryonic stem cells (ESCs) or ES-like cells are reported from many mammalian species other than the mouse, the culture system for murine ESCs may not be suitable to the other species. Previously many other research groups have modified either human or mouse ESC culture systems for bovine ESC culture. In this study, we compared three different culture mediums consisting of DMEM, ${\alpha}$-MEM or KnockOut$^{TM}$-DMEM (KO), which are modified from human or mouse ESC culture system, for the generation of bovine ESCs. In this study, some pre-requisite events which are important for establishment and long-term propagation of ESCs such as inner cell mass (ICM) attachment on feeder cells, primary colony formation and sustainability after passaging. Once the ICM clumps attached on feeder cells, this was designated as passage 0. In regards to the rate of ICM attachment, ${\alpha}$-MEM was superior to the other systems. For primary colony formation, there was no difference between DMEM and ${\alpha}$-MEM whereas KO showed lower formation rate than the other groups. For passaging, the colonies were split into 2~4 pieces and passed every 5~6 days. From passage 1 to passage 3, DMEM system seemed to be appropriate for maintaining putative bovine ESCs. On the other hand, ${\alpha}$-MEM tended to be more suitable after passage 6. Although ${\alpha}$-MEM support to maintain a ES-like cell progenies to passage 15, all three culture systems which are modified from human or mouse ESC culture media failed to retain the propagation and long-term culture of putative bovine ESCs. Our findings imply that more optimized alternative culture system is required for establishing bovine ESC lines.

조골세포에서 Aggregatibacter actinomycetemcomitans 생균의 파골세포분화유도 cytokine 발현 유도능 및 침투능 (Induction of osteoclastogenesis-inducing cytokines and invasion by alive Aggregatibacter actinomycetemcomitans in osteoblasts)

  • Osteoblasts regulate osteoclastogenesis by production of various cytokines. Aggregatibacter(A) ac-tinomycetemcomitans is one of periodontopathogens which invades gingival tissue. Therefore, clarifying the effect of alive A. actinomycetemcomitans on osteoblasts is important to understand the mechanism of alveolar bone resorption in periodontitis. We investigated induction of osteoclastogenesis-inducing cytokines, adherence, and invasion by A. actinomycetemcomitans in osteoblasts. Osteoblasts were isolated from mouse calvaria and expression of cytokines was determined by RT-PCR. When the ratio of the number of A. actinomycetemcomtians to the number of osteoblasts was 10:1, 50:1 and 100:1, RANKL mRNA expression was increased. A. actinomycetemcomitans also increased expression of macrophage inflammatory protein (MIP) -1${\alpha}$, interleukin (IL)-1${\beta}$, and tumor necrosis factor (TNF)-${\alpha}$. A. actinomycetemcomitans attached to and invaded osteoblasts at ratio of 1000:1. These results suggest that A. actinomycetemcomitans increases osteoclastogenesis-inducing ability of osteoblasts by stimulating the expression of RANKL, MIP-1${\alpha}$,IL-1${\beta}$, and TNF-${\alpha}$ and that invasion of A. actinomycetemcomitans provides a means by which the bacteria escape from immune system and antibiotic therapy.

Effects of Chromium Supplementation and Lipopolysaccharide Injection on the Immune Responses of Weanling Pigs

  • Sixteen specific pathogen free 4-wk-old crossbred weanling pigs were allotted into a $2{\times}2$ factorial experiment to evaluate the effects of chromium (Cr) on the immune responses after lipopolysaccharide (LPS) injection. Two factors included (1) no Cr or 400 ppb Cr supplementation from chromium picolinate (CrPic) and (2) LPS injection ($200{\mu}g/kg$ BW, intraperitoneally) on day 21 (d 21) and 35 (d 35) as compared with saline application. Plasma samples were obtained from all piglets before (0 h) and at 2 h, 4 h, 8 h, and 24 h after LPS injection. The changes in tumor necrosis $factor-{\alpha}$ ($TNF-{\alpha}$) and leukocyte populations after LPS injection were not significant on d 21. On d 35, the plasma $TNF-{\alpha}$ level was increased at h 2 postinjection, and supplemental Cr reduced the $TNF-{\alpha}$ level. The leukocyte populations had changed profoundly and lymphocyte subsets of $CD2^+$ and $CD8^+$ were reduced at 8 h postinjection. The blood granulocytes were increased and the percentage of $CD2^+$ was reduced in the Cr-fed group on d 35. Furthermore, Cr supplementation decreased the blastogensis of concanavalin A-stimulated peripheral blood mononuclear cells (PBMC) on d 21. These results suggest that 400 ppb Cr supplementation from CrPic in diets may modulate the immune responses in weanling pigs during LPS injection.

The Effects of Customer-appraised Physical Attractiveness of Employees on Emotional Response and Customer Satisfaction in Casual Dining Restaurants: Focused on University and Graduate Students in Seoul

  • 본 연구에서는 대학생 및 대학원생을 대상으로 패밀리레스토랑의 서비스 상황에서 지각된 직원의 신체적 매력도가 감정적 반응 및 고객만족도에 미치는 영향을 고찰하였으며, 이러한 인과관계에서 감정적 반응의 매개역할을 추가적으로 검증하였다. 패밀리레스토랑에서의 서비스 상황에 대한 시나리오 설문을 통해 총 249명의 대학생으로부터 자료를 수집하였다. 분석 결과, 고객에게 인지된 직원의 신체적 매력도는 감정적 반응(${\alpha}_{11}$=.624)과 고객만족도(${\alpha}_{21}$=.495)에 유의한 정(+)의 영향을 주는 것으로 나타났으며, 감정적 반응은 고객만족도(${\beta}_{21}$=.312)에도 유의한 정(+)의 영향을 주는 것으로 조사되었다. 또한 직원의 신체적 매력도가 고객만족도에 미치는 영향은 감정적 반응에 의해 부분적으로 매개되는 것으로 나타났다.
