  • The hop alpha-acid content prediction model developed with meteorological elements in Hoeongseong was Y=28.369-0.003X$_1$+1.558X$_2$-1.953X$_3$-0.335X$_4$-0.003X$\sub$5/-0.119X$\sub$6/, with MSEp of 0.004, Rp$^2$ of 0.9987, Rap$_2$ of 0.9949 and Cp of 7.00. The total sunshine hours (X$_1$), the maximum temperature (X$_3$) and the total precipitation (X$\sub$5/) at flowering stage. the maximum temperature at flower bud differentiation stage (X$_4$) and the maximum temperature at cone ripening stage (X$\sub$6/) influenced on hop alpha .acid content as decrement weather elements. The maximum temperature at cone development stage(X$_2$) effected on ${\alpha}$-acid content as increment weather element.

  • Acid-stable ${\alpha}-amylase$ was partially purified from Paecilomyces subglobosum by Sephadex G-150 gel filtration. About 7.7-fold purification was obtained and the partially purified preparation has 5.0 U of ${\alpha}-amylase$ activity per mg of protein. Using this partially purified ${\alpha}-amylase$, general properties were studied and it showed the maximal activities at the conditions of pH 4.0 and $38^{\circ}C$. High stability of the acid-stable ${\alpha}-amylase$ in acidic condition was observed, whereas thermal stability was similar to the conventional ${\alpha}-amylase$.

  • The effect of antioxidant activity of soyasaponin on the liposomal phospholipid membrane were investigated by spectrophotometry. The oxidation index and oxidation rate of ${\alpha}-tocopherol$ containing egg phosphatidylcholine (EPC) liposomes were markedly decreased in the presence of soyasaponin relative to those of purephospholipid liposmes. ${\alpha}-tocopherol$ containing liposomes delayed the oxidation of liposomes. Especially soyasaponin stimulated the antioxidant activity of ${\alpha}-tocopheroal$ in liposomes. These results indicates that soyasaponin in liposomes had some additive effect on antioxidant of ${\alpha}-tocopherol$ toward liposomes.

  • The regulatory effect of the tumor-suppressor protein p53 on the apoptogenic activity of $17{\alpha}$-estradiol ($17{\alpha}-E_2$) was compared between HCT116 ($p53^{+/+}$) and HCT116 ($p53^{-/-}$) cells. When the HCT116 ($p53^{+/+}$) and HCT116 ($p53^{-/-}$) cells were treated with $2.5{\sim}10{\mu}M$ $17{\alpha}-E_2$ for 48 h or with $10{\mu}M$for various time periods, cytotoxicity and an apoptotic sub-$G_1$ peak were induced in the HCT116 ($p53^{+/+}$) cells in a dose- and time-dependent manner. However, the HCT116 ($p53^{-/-}$) cells were much less sensitive to the apoptotic effect of $17{\alpha}-E_2$. Although $17{\alpha}-E_2$ induced aberrant mitotic spindle organization and incomplete chromosome congregation at the equatorial plate, $G_2/M$ arrest was induced to a similar extent in both cell types. In addition, $17{\alpha}-E_2$-induced activation of Bak and Bax, ${\Delta}{\Psi}m$ loss, and PARP degradation were more dominant in the HCT116 ($p53^{+/+}$) than in the HCT116 ($p53^{-/-}$) cells. In accordance with enhancement of p53 phosphorylation (Ser-15) and p53 levels, p21 and Bax levels were elevated in the HCT116 ($p53^{+/+}$) cells treated with $17{\alpha}-E_2$. The HCT116 ($p53^{-/-}$) cells exhibited barely or undetectable levels of p21 and Bax, regardless of $17{\alpha}-E_2$ treatment. On the other hand, although the level of Bcl-2 was slightly lower in the HCT116 ($p53^{+/+}$) than in the HCT116 ($p53^{-/-}$) cells, it remained relatively constant after the $17{\alpha}-E_2$ treatment. Together, these results show that among the components of the $17{\alpha}-E_2$-induced apoptotic-signaling pathway, which proceeds through mitotic spindle defects causing mitotic arrest, subsequent activation of Bak and Bax and the mitochondria-dependent caspase cascade, leading to PARP degradation, $17{\alpha}-E_2$-induced activation of Bak and Bax is the upstream target of proapoptotic action of p53.

GC/MS Analysis of Volatile Constituents from Acanthopanax senticosus (가시오갈피나무의 정유성분 GC/MS 분석)

  • The chemical composition of the volatile constituents from the leaves, stems, and roots of Acanthopanax senticosus growing wild in Mt. Deok-Yu was determined by GC and GC/MS spectrometric analysis with the aid of NBS, Wiley Library and RI indice searches. The major constituents identified were ${\delta}-elemene\;(23.14%),\;{\beta}-elemene\;(5.59%),\;{\gamma}-cadinene\;(5.43%),\;and\;{\alpha}-bergamotene\;(28.54%)$ in the leaves, ${\alpha}-pinene\;(11.50%),\;{\delta}-elemene\;(17.62%),\;{\alpha}-bergamotene\;(27.21%)$, and bicycloheptane derivative (6.38%) in the stems, and ${\alpha}-pinene\;(21.90%)$, and (+)-aromadendrene (3.77%) in the roots.

Effect of α-Tocopherol Supplementation on Plasma Levels of Antioxidant Vitamins in Anestrus Buffalo Heifers (Bubalus bubalis)

  • The present investigation was undertaken to study the status of plasma antioxidant vitamins in normal cycling and $\alpha$-tocopherol supplemented anestrus buffalo heifers. The pre-supplementation plasma levels ($\mu$mol/L) of vitamin E and $\beta$-carotene were significantly (p<0.05) lower and of vitamin C was significantly (p<0.05) higher in anestrus heifers (4.06$\pm$0.07; 4.56$\pm$0.17; 21.04$\pm$0.21) when compared to normal cycling ones (4.92$\pm$0.05; 6.76$\pm$0.12; 14.24$\pm$0.16). The oral supplementation of$\alpha$-tocopherol at 3,000 mg per week per animal in anestrus heifers resulted in a significant (p<0.01) increase in vitamin E and $\beta$-carotene levels and a significant (p<0.01) decrease in vitamin C concentration. Results indicated that supplementation of $\alpha$-tocopherol to anestrus buffalo heifers improved the antioxidant status by mitigating the harmful effects of free radical induced oxidative stress.

NMR Study on the Preferential Binding of the Zα Domain of Human ADAR1 to CG-repeat DNA Duplex

  • The Z-DNA domain of human ADAR1 ($Z{\alpha}_{ADAR1}$) produces B-Z junction DNA through preferential binding to the CG-repeat segment and destabilizing the neighboring AT-rich region. However, this study could not answer the question of how many base-pairs in AT-rich region are destabilized by binding of $Z{\alpha}_{ADAR1}$. Thus, we have performed NMR experiments of $Z{\alpha}_{ADAR1}$ to the longer DNA duplex containing an 8-base-paired (8-bp) CG-repeat segment and a 12-bp AT-rich region. This study revealed that $Z{\alpha}_{ADAR1}$ preferentially binds to the CG-repeat segment rather than AT-rich region in a long DNA and then destabilizes at least 6 base-pairs in the neighboring AT-rich region for efficient B-Z transition of the CG-repeat segment.

  • It is a convention to use 5% significance level when a statistical test is employed for clinical data. But when a confidence interval is used for testing equivalence, 90% confidence interval has often been used. When $1-{\alpha}$ confidence interval is used for hypothesis testing, its significance level is often ${\alpha}$. So it makes a confusion that the significance level is 10% if 90% confidence interval is employed for testing equivalence. In this paper I will clarify this issue by reviewing relevant papers and conducting simulation studies. I hope that it will be beneficial to statisticians in pharmaceutical companies, CROs, university hospitals.

Studies on Sikhye Wine -3. Commercial Sikhye Wine- (식혜주에 관한 연구 -3보. 시판식혜 올리고당주-)

  • Commercial Sikhye was fermented by Saccharomyces cerevisiae for 10 day at 29$^{\circ}C$. Sucrose was hydrolyzed into glucose and fructose by invertase from the yeast, and the glucose and fructose were converted into ethanol by the yeast. Commercial Sikhye wine was found to contain 6.6% of ethanol, 0.32$\mu$mol/ml of amino acid, 226$\mu\textrm{g}$/ml of protein, and 2.5ml 0.1N NaOH of acidity, respectively, and its pH was 3.21. Limit dextrin in commercial rice Sikhye wine showed both signal of $\alpha$-1, 4- and $\alpha$-1, 6- glucosidic linkage with its estimation ratio of 25:1 by 1H-NMR analysis. The taste of rice Sikhye wine was similar that of wine.

Sugars in Korean and Japanese Sake (한국산 및 일본산 청주의 당에 관한 연구)

  • Sugars in 2 brands of Korean Sake and 5 brands of Japanese Sake were analyzed by HPLC and TLC. Sake was found to contain average 4.6% of total sugar, 2.23% of glucose and 1.46% of maltooligosaccarides from maltose to maltodecaose. Korean two brands were found to 4.21% and 4.93% of total sugar, 3.68% and 3.29% of glucose and 1.25% and 0.92% of maltooligosaccarides from maltose to maltodecaose, respectively. maltooligosaccarides in Sake were digested with $\alpha$-amylase plus Aspergillus awamori $\alpha$-glucosidase and produces glucose. The results suggested that sugars in Sake is not effective as a growth factor of Bifidobacterium.
