$17{\beta}$-estradiol Prevents the Expression of $CEBP{\alpha}$-mediated Adipocyte Marker Genes in Female Ovariectomized C57BL/6 Mice

  • Adipogenesis is a complex sequence of events that culminates in the differentiation of fibroblast-like preadipocytes into specialized lipid-filled adipocytes and also involves a cascade of expression of many transcription factors such as peroxisome proliferator-activated receptor ${\gamma}(PPAR{\gamma})$ and CCAAT/enhancer-binding proteins (C/EBPs). $PPAR{\gamma}$ and C/EBPs transcriptionally transactivate adipocyte specific genes, including fatty acid transport protein (FAT/CD36) and leptin. To determine whether $17{\beta}$-estradiol modulates $C/EBP{\alpha}$ actions on adipogenesis in high fat diet-fed female ovariectomized (OVX) C57BL/6 mice, mice were treated with $17{\beta}$-estradiol for 7 days and the effects of $17{\beta}$-estradiol on adipose tissue mass and expression of adipocyte specific gene as well as $C/EBP{\alpha}$ were measured. Compared to vehicle-treated OVX control mice, OVX mice treated with $17{\beta}$-estradiol for 7 days had lower adipose tissue weights that were similar to weights in high fat diet-fed sham-operated (Sham) mice. OVX mice showed the increased expression of $C/EBP{\alpha}$ mRNA compared with Sham mice. However, $17{\beta}$-estradiol treatment in OVX mice inhibited OVX induced-$C/EBP{\alpha}$ activation, indicating that $17{\beta}$-estradiol may act as an inhibitor of $C/EBP{\alpha}$ action. Moreover, $17{\beta}$-estradiol decreased mRNA levels of adipocyte marker genes, such as lipoprotein lipase, FAT/CD36 and leptin, to levels in Sham mice. These results suggest that down-regulation of adipogenesis by $17{\beta}$-estradiol may be due to reduced adipose $C/EBP{\alpha}$ activities in female OVX C57BL/6 mice.

Inhibitory Effects of Curcuminoids on $17{\beta}$-hydroxysteroid Dehydrogenase Type 1 Activity in Animal Livers

  • 17-${\beta}$-hydroxysteroid dehydrogenase type 1 ($17{\beta}$-HSD type 1) mediates the reaction of $17{\beta}$-estradiol (E2) production from estrone (E1). Inhibitory effects of curcuminoids on $17{\beta}$-HSD type 1 activity were investigated to find a lead compound for treating estrogen-dependent diseases including breast cancer. Among curcuminoids, demethoxycurcumin showed potent inhibitory effect ($IC_{50}=2.7{\mu}M$) on mouse $17{\beta}$-HSD type 1. Curcuminoids also displayed their inhibitory effects on the production of $17{\alpha}$-estradiol which is a carcinogenic metabolite produced by the enzyme. Bisdemethoxycurcumin ($IC_{50}=1.3{\mu}M$) showed potent inhibitory effect on the $17{\alpha}$-estradiol production by chicken $17{\beta}$-HSD type 1. Curcuminoids did not inhibit ERE transcriptional activity with and without E2. Taken together, curcuminoids can be used for treating and preventing E2-dependent diseases via inhibition on $17{\beta}$-HSD type 1 activity.

$17{\beta}$-estradiol Attenuates Renal Fibrosis in Mice with Obstructive Uropathy (폐쇄성 요로병증에서 $17{\beta}$-estradiol에 의한 신섬유화 감소 효과에 대한 연구)

  • Purpose : Men are generally more prone to chronic renal disease and progression to end stage renal disease than women. The purpose of this study is to prove the effect of gender and sex hormone on renal fibrosis in mice with unilateral ureteral obstruction (UUO) and to elucidate the specific underlying mechanisms. Methods :We compared the expression of ${\alpha}$-smooth muscle actin (${\alpha}$-SMA) in female and male mice with complete UUO (day 7). After this, we estimated the changes of renal fibrosis in the female mice with oophorectomy and in the female mice with oophorectomy and replacement of $17{\beta}$-estradiol, respectively. Results : The level of ${\alpha}$-SMA in the female kidney with UUO was significantly lower than that in the male kidney with UUO. oophorectomy and replacement of $17{\beta}$-estradiol did not change the expression of angiotensin II type 1 (AT1) receptor in the female kidney with UUO, whereas the expression of angiotensin II type 2 (AT2) receptor was significantly more elevated in the intact female (IF) and the oophorectomized female with estrogen (OF+E) than that in the oophorectomized female (OF). The expressions of inducible nitric oxide synthase (iNOS) in the IF and OF+E mice were significantly more elevated than that in the OF mice, which was similar to the expression of AT2 receptor. Conclusion : The female gender is associated with resistance to renal fibrosis in obstructive uropathy and this gender difference may originate from the existence of $17{\beta}$-estradiol, which has an anti-fibrotic effect via upregulation of the AT2 receptor and iNOS.

  • Purpose: $17\beta$-estradiol is the most active endogenous estrogen, which is related to favorable changes in the plasma lipid profile, to relaxation of the coronary vessels, and to a decrease in platelet aggregation and vascular smooth muscle cell migration. However, although the beneficial effect of estrogens on plasma lipoproteins (ie, lowering low-density lipoprotein and increasing high-density lipoprotein cholesterol) contributes to cardiovascular protection, it does not fully account for the protective effect, particularly in the application of physical therapy, including low frequency electrical stimulation. Methods: The aim of this study was to demonstrate the inhibition of stressors, such as endothelin-1 (ET-1), serotonin (5-hydroxytryptamine, 5-HT), prostaglandin $F2\alpha$ ($PGF2\alpha$), and a protein kinase C (PKC) activator 12-deoxyphorbol 13-isobutyrate (DPB), induced isometric tension by $17\beta$-estradiol in vascular smooth muscle strips, respectively. In addition, the effects of low frequency electrical stimulation at the meridian points (CV-3, -4, Ki-12, SP-6, LR-3, BL-25, -28, -32, -52) on the indirect antihypertensive effect were examined by monitoring the changes in the serum $17\beta$-estradiol concentration in healthy volunteers. Results: Isometric tension analysis showed that the responses of inhibited tension by $17\beta$-estradiol were similar to the same stressors in rat aortic smooth muscle strips. Furthermore, although the continued amplitude modulation (AM) type of electrical stimulation was not increased significantly by electrical stimulation, the current of the frequency modulation (FM) type of low frequency electrical stimulation increased the serum $17\beta$-estradiol concentration in normal volunteers. Conclusion: These results, in part, suggest that $17\beta$-estradiol has the capacity to supress stressor-induced muscle tension, and electrical stimulation, particularly current of the FM type, has a modulatory effect on the sex steroid hormones, particularly $17\beta$-estradiol, in healthy volunteers.

  • Estrogen receptors (ERs) are steroid receptors located in the cytoplasm and on the nuclear membrane. The sequence similarities of human $ER{\alpha}$, mouse $ER{\alpha}$, rat $ER{\alpha}$, dog $ER{\alpha}$, and cat $ER{\alpha}$ are above 90%, but structures of $ER{\alpha}$ may different among species. Estrogen can be agonist and antagonist depending on its target organs. This hormone play roles in several diseases including breast cancer. There are variety of the relative binding affinity (RBA) of ER and estrogen species in comparison to $17{\beta}-estradiol$ (E2), which is a natural ligand of both $ER{\alpha}$ and $ER{\beta}$. The RBA of the estrogen species are as following: diethyl stilbestrol (DES) > hexestrol > dienestrol > $17{\beta}-estradiol$ (E2) > 17- estradiol > moxestrol > estriol (E3) >4-OH estradiol > estrone-3-sulfate. Estrogen mimetic drugs, selective estrogen receptor modulators (SERMs), have been used as hormonal therapy for ER positive breast cancer and postmenopausal osteoporosis. In the postgenomic era, in silico models have become effective tools for modern drug discovery. These provide three dimensional structures of many transmembrane receptors and enzymes, which are important targets of de novo drug development. The estimated inhibition constants (Ki) from computational model have been used as a screening procedure before in vitro and in vivo studies.

INFLUENCE OF 17-$\alpha$-ESTRADIOL ON CATECHOLAMINE SECRETION FROM THE PERFUSED RAT ADRENAL GLAND

  • It has been known that adrenal corticosteroids influence the expression of adrenomedullary catecholamine-synthetizing enzymes and also suppress the emission of axonal-like processes in cultured chromaffin cells. In the present study, it was attempted ta investigate the effect of 17${\alpha}$-estradiol on catecholamine(CA) secretion evoked by acetylcholine(ACh), DMPP, McN-A-343, excess K$\^$+/ and Bay-K-8644 from the isolated perfused rat adrenal gland. The perfusion of 17${\alpha}$-estradiol (10$\^$-6/ 10$\^$-4/M) me an adrenal vein for 20min produced relatively dose-dependent inhibition in CA secretion evoked by ACh (5.5 ${\times}$ 10$\^$-3/M), DMPP (10$\^$-4/M for 2min), McN-A-343 (10$\^$-4/M for 4min) and Bay-K-8644 (10$\^$-5/M for 4min), while did not affect the CA secretory effect of high K$\^$+/(5.6 x 10$\^$-2/M). Also, in the presence of 17${\beta}$-estradiol, CA secretion of ACh, DMPP and McN-A-343 without any effect on excess K$\^$+/-evoked CA secretion. However, in adrenal glands preloaded with 17${\alpha}$-estradiol (10$\^$-5/M) plus tamoxifen (10$\^$-5/M), which is known to be a selective antagonist of estrogen receptors (for 20min), CA secretory responses evoked by ACh, DMPP and McN-A-343 were considerably recovered as compared to that of 17${\alpha}$-estradiol only, but excess K$\^$+/-induced CA secretion was not affected.

  • Several endocrine disrupting chemicals(EDCs) were monitored to evaluate the estrogenic activities and the concentrations by yeast two-hybrid assay and enzyme-linked immunosorbent assay(ELISA) in sewage treatment plant(STP) which consist of industrial and domestic line. In the influent of domestic line, estrone, 17$\beta$-estradiol, 17$\alpha$-ethinylestradiol and alkylphenolethoxylate(APE) were detected up to 167.1, 39.7, 7.3 and 145.4 ng/L, respectively. The average removal efficiency of 17$\beta$-estradiol after the activated sludge process was 77.5% and further removed to 80.8% after the sand filtration-ozonation step. These results suggests that the activated sludge process has limited potential to remove the estrogenic activity effectively. The contributions of the estrogenic chemicals to the estrogenic activities were 70.7, 23.3, 3.7 and 2.3% for estrone, 17$\beta$-estradiol 17$\alpha$-ethinylestradiol and APE, respectively, in the domestic line effluents. Therefore, 17$\beta$-estradiol and estrone contributed most of the estrogenic activity in the domestic line effluents.

  • Application of the spectrophotometer to the analysis of 17 ${\alpha}-ethinyl$ estradiol and $17{\alpha}-ethinyl-19-nortestosterone$ acetate mixture in oral contraceptive has been accomplished. It is used Beckman Du Spectrophotometer as a apparatus. The petroleum ether extract of ethinyl estradiol is determined at $535\;m{\mu}$ and the chloroform extract of norethindrone acetate is determined at $380\;m{\mu}$ respectively. This analytical method is formed Lambert Beer's law. This method can be used to the analysis of ethinyl estradiol aid norethindrone acetate mixture in commercial dosage form of routine assay.

Analysis of Extraction Characteristics of Phytoestrogen Components from Punica granatum L. (석류 phytoestrogen 성분의 추출특성 분석)

  • The optimization of extraction conditions of phytoestrogen from pomegranate by hot water was conducted by analyzing the extraction characteristics. The purpose of this study was effective utilization of bioactive components of pomegranate, and the analyzing was performed with response surface methodology (RSM). This study established 10 sections based on the central composite design with the independent variables of extraction temperature (60, 70, 80, 90, $100^{\circ}C$) and extraction time (1, 2, 3, 4, 5 hr) to predict the optimal conditions for extraction of the effective components. The dependent variables were measured for extracted materials, those were, the major components such as chlorogenic acid, kaempferol, $17-{\alpha}-estradiol\;and\;17-{\beta}-estradiol$ content, and regression analysis was performed by SAS program, and optimal conditions for each characteristics were predicted, and the characteristics of extraction were analyzed by response surface methodology. It was found that chlorogenic acid, kaempferol, and $17-{\alpha}-estradiol$ content were greatly affected by extraction temperature. However, $17-{\beta}-estradiol$ content was affected significantly by extraction time. Regression formulas for each variable were elicited from this study, and the chlorogenic acid, kaempferol, $17-{\alpha}-estradiol\;and\;17-{\beta}-estradiol$ content depending on response surface methodology factor were superimposed. It was shown that optimal temperature and extraction time were $98{\sim}100^{\circ}C\;and\;3{\sim}5$ hrs, respectively.

Effects of Exogenous Oxytocin on Steroid Hormones and Oxytocin Receptor Concentrations in Pregnant Rats (Oxytocin 투여가 임신 Rat의 Steroid Hormones 및 Oxytocin Receptors 농도에 미치는 영향)

  • The present studies were carried out to examine the effects of exogenous oxytocin(OT) on plasma, uterine and placenta of estradiol-17$\beta$, progesterone, prostaglandin F$_2$$_{\alpha}$ (PGF$_2$$_{\alpha}$), Prostaglandin E$_2$(PGE$_2$) and OT receptor concentrations in pregnant rats. Pregnant rats received an injection of exogenous OT on days 14, 16, 18, 20, 22 of pregnancy and day 1 of postpartum. Concentrations of plasma estradiol-17 $\beta$ after OT injection started to increase after day 18 and peaked on day 22 of pregnancy but decreased on day 1 of postpartum. Plasma progesterone concentrations declined gradually from day 18 of pregnancy and decreased more rapidly until postpartum 1 day. Concentrations of estradiol-17$\beta$in uterine tissues after OT injection were sharply increased from day 20 to 22 of pregnancy and progestrone concentrations were peaked on day 16 and decreased rapidly from day 16 to 20 and maintained the same level until day 1 of postpartum. Uterine concentrations of PGF$_2$$_{\alpha}$ and PGE$_2$increased gradually until day 20 and peaked on day 22 of pregnancy but showed a marked decrease on day 1 of postpartum. Concentrations of PGF$_2$$_{\alpha}$ in placental tissues increased rapidly from day 14 of pregnancy and decreased sharply on day 1 of postpartum. Concentrations of PGE$_2$increased gradually after day 14 and peaked on day 20 of pregnancy. The concentration of OT receptor in uterus was significantly elevated from day 20 and rose to maximum on day 22 of pregnancy. These findings show that OT suppress the concentration of progestrone and stimulate productions of estradiol-17 $\beta$, PGF$_2$$_{\alpha}$, PGE$_2$ and oxytocin receptor concentrations in pregnant rats.
