Changes of Chlorophyll and SOD-like Activities of Chinese Chives Dehydrated at Different Heat Treatments (부추의 건조 온도 조건별 클로로필, Superoxide Dismutase 유사활성의 변화)

  • Allium tuberosum Rotter (Liliaceae, Chinese chives) is a perennial herb of which leaves are used for food. This study investigated the effect of pretreatment on quality of dehydrated Chinese chives. Chinese chives was blanched at $80^{\circ}C$ for 20 sec, followed by drying at $70^{\circ}C$, $80^{\circ}C$, $100^{\circ}C$, or drying at $100^{\circ}C$ for 30 min and subsequent drying at $70^{\circ}C$, or $100^{\circ}C$ for 60 min and subsequent drying at $70^{\circ}C$. Optimum drying temperature and time was $100^{\circ}C$ for 30 min and subsequent drying at $70^{\circ}C$, or $100^{\circ}C$ for 60 min and subsequent drying at $70^{\circ}C$. These conditions were shortened time for dehydration and showed smaller decrease than others in Hunter color L, a, b. Dehydrated Chinese chives showed a constant decrease in greenness with storage, probably due to destruction of chlorophyll by light. In the measurement of Hunter color L, a, b, these conditions showed smaller decrease than others in Hunter color for 15 week storage. Chlorophyll content and SOD (superoxide dismutase)-like activity in that condition was higher than others. It was assumed that a phenolic compound that forms its thermostable activity. The fitness of drying models was conducted in order to explain reducing chlorophyll loss and SOD (superoxide dismutase)-like activity loss. Based upon the chlorophyll content, SOD-like activity, and retention of green color of the vegetable, optimum drying conditions was $100^{\circ}C$ for 30 min followed by $100^{\circ}C$ for 30 min and subsequent drying at $70^{\circ}C$, or $100^{\circ}C$ for 60 min and subsequent drying at $70^{\circ}C$.

Effect of Additive, Storage Temperature and Time on the Texture Properties of Baikseolgi (첨가물, 저장온도 및 저장시간에 따른 백설기의 텍스쳐 특성)

  • The effects of sucrose fatty acid ester (SE, 1% w/w) and glycerin (GL, 1% v/w) additions, storage temperature$(0,\;20\;and\;70^{\circ}C)$, and time $(0{\sim}6\;day)$ on texture properties, hardness(H), cohesiveness(O), chewiness(C) and rheological property(R) of Baikseolgi were studied. The H of Baikseolgi increased sharply in the early stage of storage at 0 and $20^{\circ}C$, while increased gently at $70^{\circ}C$ with increasing storage time. After 6 days of storage, the H of Baikseolgi at $20^{\circ}C$ had a little lower than that at $0^{\circ}C$. However, the H of Baikseolgi at $70^{\circ}C$ was 10.7% of that at $0^{\circ}C$. The addition of GL had greater effect on the reduction of H than that of SE. The H of control, SE and GL additions were 336, 216 and $$174\;g_f, respectively, after 6 days at $70^{\circ}C$. The O of Baikseolgi at $70^{\circ}C$ were higher than those at $0^{\circ}C$. The O of GL added Baikseolgi had the highest value and the second and the third were SE added and control, respectively. The O of Baikseolgi decreased with increasing storage time. The C of Baikseolgi of increased with increasing storage time, which had similar curve patterns to the H of Baikseolgi. Instantaneous stress and equilibrium stress of Baikseolgi decreased with increasing storage temperature. The affection of viscous element increased and that of elastic element decreased with increasing storage temperature.

Studies on the Changes in Chemical Composition and Microbiological Aspects of Raw Milk by Microwave Heating (Microwave 열처리에 의한 원유의 화학적 및 미생물학적 성상의 변화에 관한 연구)

  • In order to determine the optimum pasteurization conditions by microwave heating(MWH) at $50^{\circ}C{\sim}70^{\circ}C$ for 30 minute compared with water bath heating(WBH) at $65^{\circ}C$ for 30minute during storage at $5^{\circ}C$, the chemical composition, microbiological changes and keeping quality were examined and the results were as follows: 1. The fat protein lactose, total solid contents of raw milk, at $50{\sim}70^{\circ}C$ for 30 min. in MWH and at 65 for $30^{\circ}C$ min. in WBH did not changed significantly during the storage at $5^{\circ}C$. 2. The pH and acidity for the raw milk untreated were 6.75 and 0.16%, and those of MWH heated and WBH milk wee 6.75~6.50 and 0.16%~0.19%, phosphatase test were negative at $61^{\circ}C$ for 20 min. at $62^{\circ}C$ for 15 min. at $63^{\circ}C$ for 10 min. at $64^{\circ}C$ for 5 min. at $65^{\circ}C$ for 5 min. in MWH and at $65^{\circ}C$ for 30 min. in WBH. 3. Whey protein content was $18.53mg/m{\ell}$ in raw milk untreated, however, those were decreased as the heating temperature increased. The proteolytic activity of treated milk by WBH(44%) was lower than that by MWH(94%). 4. Total bacteria counts were $2.8{\times}10^5CFU/m{\ell}$ in raw milk untreated, $2.8{\times}10^3CFU/m{\ell}$ at $65^{\circ}C$ for 30 min. $2.4{\times}10^3CFU/m{\ell}$ at $70^{\circ}C$ for 30 min. in MWH and $3.0{\times}10^3CFU/m{\ell}$ at $65^{\circ}C$ for 30 min. in WBH. Because total bacteria count did not increased in MWH at $65^{\circ}C$, $70^{\circ}C$ for 30 min. and $65^{\circ}C$ for 30 min. in WBH during the 10 days storaging, Also, total bacteria counts for treated milk were a most drastic decrease after $61^{\circ}C$, $62^{\circ}C$, $63^{\circ}C$, $64^{\circ}C$, $65^{\circ}C$ for 5 min. in MWH. 5. Coliform bacteria counts were $2.6{\times}10^3CFU/m{\ell}$ in raw milk untreated. There were not detected at $55^{\circ}C{\sim}70^{\circ}C$ for 30 min. in MWH and at $65^{\circ}C$ for 30 min. in WBH. Coliform bacteria counts were not detected after $61^{\circ}C$, $62^{\circ}C$, $63^{\circ}C$, $64^{\circ}C$, $65^{\circ}C$ for 5 min. in MWH. 6. Thermoduric bacteria counts were $5.2{\times}10^4CFU/m{\ell}$ in raw milk untreated, $2.0{\times}10^3CFU/m{\ell}$ at $65^{\circ}C$ for 30 min. $1.9{\times}10^3CFU/m{\ell}$ at $70^{\circ}C$ for 30min. in MWH and $2.2{\times}10^3CFU/m{\ell}$ at $65^{\circ}C$ for 30 min. in WBH. Because thermoduric bacteria counts did not increased in MWH at $65^{\circ}C$, $70^{\circ}C$ for 30 min. and $65^{\circ}C$ for 30 min. in WBH during the 10days storaging. Also, thermoduric bacteria counts were a most drastic decrease after $61^{\circ}C$, $62^{\circ}C$, $63^{\circ}C$, $64^{\circ}C$, $65^{\circ}C$ for 5 min. in MWH. 7. Psychrotrophic bacteria counts were $2.8{\times}10^5CFU/m{\ell}$ in raw milk untreated, $2.0{\times}10^1CFU/m{\ell}$ at $65^{\circ}C$ for 30 min. $2.0{\times}10^1CFU/m{\ell}$ at $70^{\circ}C$ for 30 min. in MWH and $3.0{\times}10^1CFU/m{\ell}$ at $65^{\circ}C$for 30 min. in WBH. Because psychrotrophic bacteria counts did not increased in MWH at $65^{\circ}C$, $70^{\circ}C$ for 30min. and $65^{\circ}C$ for 30 min. in WBH during the 10 days storaging. Also, psychrotrophic bacteria counts were a most drastic decrease after $61^{\circ}C$, $62^{\circ}C$, $63^{\circ}C$, $64^{\circ}C$, $65^{\circ}C$ for 5 min. in MWH.

Studies on Genetics and Breeding in Rainbow Trout(Oncorhynchus mykiss) VII. Fertilization of Fresh Egg with Co-Preserved Sperm and Ultrastructural Changes (무지개 송어의 유전 육종학적 연구 VII. 동결보존시킨 정자와 신선한 난모세포의 수정 및 미세구조적 변화)

  • This study was carried out to develop new techniques useful for cryopreservation, thawing and artificial insemination, and ultrastructural changes of cryopreserved spermatozoa in rainbow trout(Oncorhynchus mykiss) . Two extenders, such as Tyrode solution and Whittingham's $T_6$ solution, were used to preserve rainbow trout sperm in refrigerator $(-20,\;-40\;and\;-70^{\circ}C)$ or liquid nitrogen $%(-196^{\circ})$. Hand-stripped semen was diluted to 1:16 with two extenders, an then the semen were frozen after mixing semen and each extender containing 1M or 1.5M DMSO solution to 1:1. After 60 days cryopreserved semen was thawed in a $13^{\circ}$ water bath, and subsequently centrifugated. After centrifugation at 1,000 rpm for 5 min thawed semen was washed with extenders, and then fertilized with fresh eggs. The results obtained in these experiments were summarized as follows: After cryopreservation, over 75% of spermatozoa were appeared motile and the survival rate was high. Following cryopreservation by the addition of cryoprotectant such as DMSO, methanol and glycerol, the fertilization rate of the thawed spermatozoa appeared over $99\%$ compared with the control having $99\%$ of fertilization rate. There was no difference between the control and experimental groups such as $(-20^{\circ}C\;-40^{\circ}C\;and\;-70^{\circ}C)$ and $-196^{\circ}$ in fertilization rate. Following cryopreservation at $-196^{\circ}$ by the addition of 1M DMSO of cryoprotectant, each fertilization rate following 24 hours and hatching rate following 24 days showed $96\%$ and $8\%$ by the addition of BSA, but showed $98\%\;and\;10%$ by no addition of BSA. Following 2 months of cryopreservation by the addition of 1M DMSO of cryoprotectant, there were $10%$ of hatching rate at $-196^{\circ}\;and\;10\%\;and\;35\%,\;respectively,\;at\;-40^{\circ}C\;and\;-70^{\circ}C$. Following 2 months of cryopreservation by the addition of 1M methanol of cryoprotectant, there were $22\%$ of fertilization rate at $-20^{\circ}C,\;and\;28\%,\;at\;-70^{\circ}C$ Following 2 months of cryopreservation by the addition of 1M glycerol of cryoprotectant, there were $22\%$ of fertilization rate at $-20^{\circ}C$, and $33\%,\;at\;-70^{\circ}C$. pollowing 2 months of cryopreservation by the addition of 1.5M DMSO of cryoprotectant, there were $27\%$ of fertilization rate at $-20^{\circ}C,\;an\;36\%\;and \;35\%,\;respectively,\;at\;-40^{\circ}C\;and\;-70^{\circ}C$. Following 2 months of cryopreservation by the addition of 1.5M glycerol of cryoprotectant, there were $34\% \;of\;fertilization\;rate\;at\;-20^{\circ}C, \;and\;31\%\;and\;31\%,\;respectively,\;at \;-40^{\circ}C\;and\;-70^{\circ}$. Following 2 months of cryopreservation by the addition of 1.5M methanol of cryoprotectant, there were $28\%$ of fertilization rate at $-20^{\circ}C,\;and\;29\%\;and\;28\%,\;respectively,\;at\;-40^{\circ}C\;and\;-70^{\circ}C.$ From 10 days and 15 days following fertilization at $13^{\circ}C\;and\;10^{\circ}C$, respectively, the mortality rate of fertilized ova was markedly increased. The middle piece of spermatozoa had two set of central doublets, nine set of outer coarse fibres, and mitochondrial sheath. Spermatozoa went through morphological changes during storage, e.g. winding of flagella, detachment of the nuclear envelope and the plasma membrane from the nucleus of the sperm head. There were $1\%$ abnormal spermatozoa in fresh sperm and about $15\%$ during storage.

Prediction of Long-Term Interlaminar Shear Strength of Carbon Fiber/Epoxy Composites Exposed to Environmental Factors (환경인자에 노출된 탄소섬유/에폭시 복합재의 장기 층간전단강도 예측)

  • The purpose of this study was to predict the long-term performance using the interlaminar shear strength of carbon fiber/epoxy composites exposed to environmental factors. Interlaminar shear specimens, manufactured by the filament winding method, were exposed to the conditions of drying at $50^{\circ}C$, $70^{\circ}C$, and $100^{\circ}C$ and of immersion at $25^{\circ}C$, $50^{\circ}C$, and $70^{\circ}C$ for up to 3000 hours, respectively. According to the results, the interlaminar shear strength did not vary significantly with the exposure time for the drying at $50^{\circ}C$ and $70^{\circ}C$, but it increased somewhat for the drying at $100^{\circ}C$ due to the post curing as the exposure time increased. The interlaminar shear strength of the specimens exposed to the immersion at $25^{\circ}C$ did not change significantly at the beginning of exposure, but it decreased with the exposure time and the degree of decrease increased as the environmental temperature increased. The linear regression equations for the environmental temperatures were obtained from the interlaminar shear strength of the specimens exposed to the immersion for up to 3000 hours. Using these linear regression equations, the interlaminar shear strength was estimated to be within 5.5% of the measured value at $25^{\circ}C$ and $50^{\circ}C$, and 2.3% of the measured value at $70^{\circ}C$. Therefore, the proposed performance prediction procedures can predict well the long-term interlaminar shear strength of carbon fiber/epoxy composites exposed to environmental factors.

Studies on the suitable temperature for rice drying (미곡의 건조 적정 온도에 관한 연구)

  • In order to establish the optimum drying temperature of the heated air blast-grain circulation dryer, a 2 ton scale of paddy was used in this experiment. The temperatures of heated air used were 45, 50, 55, 60, 65 and $70^{\circ}C$. Moisture content of the paddy was reduced to 15% from 24% by drying. The higher air temperature, the shorter drying time and the less fuel consumed. The portion of cracked kernels was found to be low as $3{\sim}10%\;at\;40{\sim}55^{\circ}C$, but was very high to $28{\sim}30%\;at\;60{\sim}70^{\circ}C$. The germination percent was very high as 98 and 93% at 40 and $45^{\circ}C$, but it was decreased to 86 and 30% at 50 and $70^{\circ}C$, respectively. The recovery yield of milled rice from paddy was decreased by 1.76, 2.63 and 7.52% at 60, 65 and $70^{\circ}C$, respectively, compared with 75.86% as drying at $40^{\circ}C$. The higher heated air temperature, the less head rice of milled rice was recovered, and the decreased ratio was increased $60^{\circ}C$. The higher heated air temperature, the less alkali disintergation value and gel consistency of milled rice and the more water up take, expanded vlume, total solid in residual liquid and intensity of starch iodine blue value of residual liquid, and those tendency was found clear from $60^{\circ}C$ temperature of heated air.

  • In this study it was measured seam strength of T/C which has the different processing methods and density. Also it was measured seam strength by the change of angle in the pattern and the kinds of threads. The results of this study are as follows; The seam strength is stronger in order of $90^{\circ}/90^{\circ}$ > $0^{\circ}/0^{\circ}$ > $70^{\circ}/-70^{\circ}$ > $70^{\circ}/70^{\circ}$ > $60^{\circ}/60^{\circ}$ > $60^{\circ}/-60^{\circ}$ > $30^{\circ}/30^{\circ}$ > $30^{\circ}/-30^{\circ}$ > $45^{\circ}/-45^{\circ}$ > $45^{\circ}/45^{\circ}$ by the cutting directions, in order of T/C3>T/C1>T/C4>T/C2 by the samples and in order of 60's/3>40's/2>60's/2>50's/2 by the threads.

Effect of Drying method on the Fruit and Powder color of Red Pepper (건조방법이 고추 및 고춧가루의 색도에 미치는 영향)

  • This experiment was established to find out the effect of drying methods on the fruit and powder color of red pepper. Hunter's L, a, and b values of the dried fruit were also higher with the cut fruit in comparison with the whole or punched fruit. The Hunter's a value of the pepper powder after drying and grinding was slightly higher for the whole fruit with sun drying, but it was higher for the cut fruit in case of heat drying. The ASTA value was higher when pepper fruits were sun dried. For the heat drying, the value was higher with the drying temperature $60^{\circ}C$.

Effect of hot-air drying temperature on antioxidative activity of sweetpotato leaves (열풍건조 온도에 따른 고구마 잎 메탄올 추출물의 항산화 효과)

  • To provide the basic data to dry vegetate the sweetpotato leaves, the total contents of tannin, flavonoid and polyphenol in the methanol extract from the hot-air dried sweetpotato leaves were analyzed and DPPH radical scavenging activity, ABTS radical scavenging ability, nitrite scavenging ability, and others were comparatively analyzed. The total tannin content was decreased from Shinmi of 10.87 mg/g at $40^{\circ}C$ to 7.28 mg/g at $70^{\circ}C$ and the total flavonoid was decreased from Hayanmi of 55.37 mg/g at $40^{\circ}C$ to 39.63 mg/g at $70^{\circ}C$. That is, the low temperature drying contained more of these substances than in the high temperature drying. The DPPH radical scavenging activity was the highest in Shinmi and Hayanmi of 84.33% and 85.25% at $40^{\circ}C$, and the ABTS radical scavenging ability was a high value of over 80% in the treatment plot at $40^{\circ}C$. The nitrite scavenging ability was highest in Shinmi and Hayanmi of 76.15% and 73.74% at $40^{\circ}C$ but low at $70^{\circ}C$. That is, the antioxidant effect of the hot-air dried sweetpotato leaves was high in the sample of $40^{\circ}C$ and low in the sample of $70^{\circ}C$. Affected by the drying temperature, the high antioxidant effect is resulting from the little decrease of active ingredient when drying at low temperatures.

Physicochemical Properties of Starch from Cow Pea (동부 전분의 이화학적 특성)

  • Starch granules of cow pea were oval with their sizes of $6-22{\mu}$m. X-ray diffraction pattern of starch granules showed some crystallinity at $2{\theta}$ $15^{\circ}$, $17.1^{\circ}$, $18.1^{\circ}$, $22.9^{\circ}$. The blue value of starch was 0.370, amylose contents 28.1% and water binding capacity 70.6%. Swelling power of the starch increased rapidly from $65^{\circ}C$ to $75^{\circ}C$ thereafter increased less rapidly. Solubility showed the same pattern as the swelling power. Optical transmittance of 0.2% starch suspension was increased rapidly from $70^{\circ}C$ and the gelatinization between $70^{\circ}C$ and $90^{\circ}C$ was single stage. Amylogram of 4% starch solution showed no peak viscosity but 6% starch solution showed peak viscosity.
