Biological Control of Fusarium Stalk Rot of Maize Using Bacillus spp. (Bacillus spp.를 이용한 옥수수 밑둥썩음병의 생물학적 방제)

  • Maize (Zea mays L.) is an economically important crop in worldwide. While the consumption of the maize is steadily increasing, the yield is decreasing due to continuous mono-cultivation and infection of soil-borne fungal pathogens such as Fusarium species. Recently, stalk rot disease in maize, caused by F. subglutinans and F. temperatum has been reported in Korea. In this study, we isolated bacterial isolates in rhizosphere soil of maize and subsequently tested for antagonistic activities against F. subglutinans and F. temperatum. A total of 1,357 bacterial strains were isolated from rhizosphere. Among them three bacterial isolates (GC02, GC07, GC08) were selected, based on antagonistic effects against Fusarium species. The isolates GC02 and GC07 were most efficient in inhibiting the mycelium growth of the pathogens. The three isolates GC02, GC07 and GC08 were identified as Bacillus methylotrophicus, B. amyloliquefaciens and B. thuringiensis using 16S rRNA sequence analysis, respectively. GC02 and GC07 bacterial suspensions were able to suppress over 80% conidial germination of the pathogens. GC02, GC07 and GC08 were capable of producing large quantities of protease enzymes, whereas the isolates GC07 and GC08 produced cellulase enzymes. The isolates GC02 and GC07 were more efficient in phosphate solubilization and siderophore production than GC08. Analysis of disease suppression revealed that GC07 was most effective in suppressing the disease development of stalk rot. It was also found that B. methylotrophicus GC02 and B. amyloliquefaciens GC07 have an ability to inhibit the growth of other plant pathogenic fungi. This study indicated B. methylotrophicus GC02 and B. amyloliquefaciens GC07 has potential for being used for the development of a biological control agent.

Nonlinear Color-Metallicity Relations of Globular Clusters: an Observational Approach

  • The origin of globular cluster (GC) color bimodality, which is one of the salient phenomena observed in most large galaxies, has not yet been fully resolved. The phenomenon has conventionally been interpreted as a bimodal metallicity distribution based on an assumption of linear GC color-metallicity relations (CMRs). Recent studies however suggest that nonlinear GC CMRs can cause a bimodal color distribution even from a single-peaked metallicity spread. Using photometric and spectroscopic data on GCs in NGC 5128 (Cen A) and NGC 4594 (Sombrero), we investigate the nonlinearity of GC CMRs and compare the observed GC CMRs with the predictions of stellar population simulation models. Our careful selection of old GCs effectively reduces the scatter and reveals the nonlinear nature of the GC CMRs for various colors. The overall shape of the observed CMRs agrees well with that of the modeled CMRs, while offsets are present for some colors. We discuss the implications of our results in terms of the GC color bimodality and GC formation in NGC 5128 and NGC 4594.

Assessment on the East Asian Summer Monsoon Simulation by Improved Global Coupled (GC) Model (Global Coupled (GC) 모델 개선에 따른 동아시아 여름 몬순 모의성능 평가)

  • The performance of East Asian summer monsoon is assessed for GC2 and GC3.1, which are climate change models of the current and next climate prediction system in the Korea Meteorological Administration (KMA), GloSea5 and GloSea6. The most pronounced characteristics of GC models are strong monsoon trough and the weakening of the Western North Pacific Subtropical High (WNPSH). These are related to the weakening of the southwesterly wind and resulting weak monsoon band toward the Korean Peninsula. The GC3.1 is known to have improved the model configuration version compared to GC2, such as cloud physics and ocean parameters. We can confirm that the overall improvements of GC3.1 against GC2, especially in pressure, 850 hPa wind fields, and vertical wind shear. Also, the precipitation band stagnant in the south of 30°N in late spring is improved, therefore the biases of rainy onset and withdrawal on the Korean Peninsula are reduced by 2~4 pentad. We also investigate the impact of initialization in comparison with GloSea5 hindcast. Compared with GCs, hindcast results show better simulation within 1 month lead time, especially in pressure and 850 hPa wind fields, which can be expected to the improvement of WNPSH. Therefore, it is expected that the simulation performance of WNPSH will be improved in the result of applying the initialization of GloSea6.

Nrf2 Overexpression Predicts Prognosis and 5-FU Resistance in Gastric Cancer

  • Objective: NF-E2-related factor 2 (Nrf2) is activated in several human malignancies. However, the role of Nrf2 in gastric cancer (GC) remains incompletely understood. In this study, we therefore analyzed associations of Nrf2 expression status with clinical features and chemotherapeutic resistance in GC. Materials and Methods: A total of 186 samples from GC patients who underwent gastrectomy were used for prognostic assessment. A further 142 samples from GC cases who received first-line combination chemotherapy were applied for investigation of chemoresistance. The Nrf2 expression was evaluated by immunohistochemistry in GC samples, and its relationship with clinicopathological parameters and chemotherapy sensitivity was analyzed. The effect of Nrf2 gene silencing on chemotherapy resistance was also examined by cell viability assay in vivo. Results: Of the 186 patients with GC, 104/186 (55.9%) showed high expression for Nrf2. The overexpression of Nrf2 was an independent predictor of overall survival [OS, hazard ratio (HR) 3.9; P=0.011] and disease-free survival (DFS, HR 4.3; P=0.002). The gene silencing of Nrf2 reduced resistance to cell death induced by 5-FU in GC cell lines. Conclusion: Our data show that Nrf2 is an independent prognostic factor in GC. Furthermore, Nrf2 confers resistance to chemotherapeutic drug 5-FU in GC cells. Taken together, Nrf2 is a potential prognostic marker and predictive for 5-FU resistance in GC.

Weaning pig performance can be enhanced by replacing dietary inorganic copper and zinc with glycine or methionine-chelated copper and zinc

  • A total of 180 21-day-old weaning pigs ([Yorkshire × Landrace] × Duroc) with an initial body weight of 6.44 ± 0.01 kg were randomly assigned to 9 treatments for evaluating the effects of replacing dietary inorganic copper (Cu) and zinc (Zn) with glycine (Gly) or methionine (Met)-chelated Cu and Zn on growth performance and nutrient digestibility. The experimental period was 35 days. There were four replicated pens per treatment, with five pigs (three males and two females) per pen. Dietary treatments consisted of a basal diet (CON), in which the sources of Cu and Zn were in inorganic form. The inorganic Cu and Zn in the basal diet were replaced by glycine-chelated (GC) and methionine-chelated (MC) Cu and Zn by 30, 50, 70, or 100% to form the GC1, GC2, GC3, GC4, or MC1, MC2, MC3, MC4 groups. The 100% replacement of dietary inorganic Cu and Zn with GC or MC increased (p < 0.05) average daily gain, average daily feed intake, and gain-to-feed ratio. The complete replacement of dietary inorganic Cu and Zn with GC or MC led to enhanced (p < 0.05) digestibility of dry matter, nitrogen, Cu and Zn. Thus, the replacement of inorganic Cu and Zn with GC or MC can improve the growth efficiency and nutrient utilization of weaning pigs.

Method Development for the Odor-Active Compound Determination by Gas Chromatography/Flame Ionization Detection/Olfactometry (냄새성분 측정을 위한 기체 크로마토그래피/불꽃이온화 검출/후각 검출법의 개발)

  • Oder-active compounds are complex in a sample. These compounds are usually analyzed by GC or GC/MSD while such analytical measurement can quantify specific volatile organic compounds, it has limitations in identifying odor-active compounds. To resolve this problem, GC-Sniffing or GC-Olfactometry method has been attempted. In this study, GC/FID/Olfactometry system was developed. This system can simultaneously sniff and detect GC effluents by traditional GC combined with human olfactory system. The time gap between FID and ODP response was dependent on the kinds and concentrations of chemicals and panels, with more volatile, stronger and shorten breath cycle panel showing narrow time gap. Thus, clear relationship between FID and ODP should be considered to identify the odor-active compounds.

Analysis of the Taste Components and Antioxidant Properties of Cheonggukjang Containing Korean Red Ginseng

  • This study was performed to investigate the taste composition and antioxidant properties of cheonggukjang containing Korean red ginseng (RGC), as compared to either general cheonggukjang (GC) or non-fermented boiled soybeans (BS). Amylase activity was the highest (576.7 unit/g) in RGC, whereas protease activity was the highest (326.0 unit/g) in GC. The total soluble sugar contents of BS, GC, and RGC were 2,027.5, 905.5, and 837.5 mg/100 g, respectively. RGC had the highest amount of total amino acids (2,127.4 mg/100 g) and essential amino acid (50.9%) among the samples. The ratio of sweet to bitter components was higher in RGC than in GC. Although the extracts of RGC had higher radical scavenging activity for 1,1-diphenyl-2-picrylhydrazyl (DPPH) than BS or GC, regardless of the extract concentration, the ethanol extract of RGC showed the highest scavenging ability (92.4%) at 2.0 mg/mL. The chloroform extracts from GC and RGC showed their greatest superoxide dimutase-like activities at 17.2 and 19.7% at a concentration of 2 mg/mL, respectively. Regardless of the samples, the nitrite scavenging ability was positively correlated to the extract concentration, and RGC had highest ability among samples under the same extract concentrations.

2 - 4 ㎛ Spectroscopy of Red Point Sources in the Galactic Center

  • We present results from our long-term observing campaign, using the NASA IRTF at Maunakea, to obtain 2 - 4 ㎛ spectra of 118 red point sources in the line of sight to the Galactic Center (GC). Our sample is largely composed of point sources selected from near- and mid-infrared photometry, but also includes a number of massive young stellar objects. Many of these sources show high foreground extinction as shown by deep 3.4 ㎛ aliphatic hydrocarbon absorption feature, which is a characteristic of the diffuse ISM and comes from the long line of sight through the diffuse medium toward the Central Molecular Zone (CMZ), the central 300 pc region of the GC. The deep 3.1 ㎛ H2O ice absorption band coming from the local, dense material in the GC CMZ suggests that most sources are likely located in the GC CMZ. A few of these sources show weak CCH3OH ice absorption at 3.535 ㎛, which can provide a strong constraint on the CCH3OH ice formation in the unique environment of the CMZ. From the best-fitting models, the optical depths of these features are determined and used to generate a well-rounded view of the ice composition across the GC CMZ and the spectral characteristics of massive YSOs in the GC.

DPPA2 Protein Expression is Associated with Gastric Cancer Metastasis

  • Gastric cancer (GC) as the fourth most common cause of malignancies shows high rate of morbidity appropriating the second leading cause of cancer-related death worldwide. Developmental pluripotency associated-2 (DPPA2), cancer-testis antigen (CT100), is commonly expressed only in the human germ line and pluripotent embryonic cells but it is also present in a significant subset of malignant tumors. To investigate whether or not DPPA2 expression is recalled in GC, our aim in this study was to elucidate DPPA2 protein expression in gastric cancer. Fifty five GC tumor and their related margin normal tissues were recruited to evaluate DPPA2 protein expression and its probable associations with different clinicopathological features of the patients. DPPA2 was overexpressed in GC cases compared with normal tissues (P < .005). While DPPA2 expression was detected in all GC samples, its high expression was found in 23 of 55 tumor tissues (41.8%). Interestingly, 50 of 55 normal samples (90.9%) were negative for DPPA2 protein expression and remained 5 samples showed very low expression of DPPA2. DPPA2 protein expression in GC was significantly correlated with lymph node metastasis (p = 0.012). The clinical relevance of DPPA2 in GC illustrated that high level expression of this protein was associated with lymph node metastasis supporting this hypothesis that alteration in DPPA2 was associated with aggressiveness of gastric cancer and may be an early event in progression of the disease. DPPA2 may be introduced as a new marker for invasive and metastatic GCs.

Cisplatin-Based Therapy for the Treatment of Elderly Patients with Non-Small-Cell Lung Cancer: a Retrospective Analysis of a Single Institution

  • Background: In spite of the fact that platinum-based doublets are considered the standard therapy for patients with advanced non-small-cell lung cancer (NSCLC), no elderly-specific platinum based prospective phase III regimen has been explored. The aim of this retrospective singlecenter study was to evaluate the efficacy and side effects of cisplatin-based therapy specifically for the elderly. Methods: Patients receiving platinum-based treatment were divided into three groups. In the first group (GC), Gemcitabine was administrated at 1000 $mg/m^2$ on days 1, 8 and cisplatin was added at 75 $mg/m^2$ on day 1. In the second group (DC), 75 $mg/m^2$ docetaxel and cisplatin were administered on day 1. The third group (PC) received 175 mg of paclitaxel and 75 mg of cisplatin on day 1. These treatments were repeated every three weeks. Result: GC arm had 36, the DC arm 42 and the PC arm 29 patients. Grade III-IV thrombocytopenia was higher in the GC arm (21.2% received GC, 2.8% received DC, and 3.8% received PC), while sensory neuropathy was lower in patients with GC arm (3.0%, 22.2%, and 23.1% received GC, DC and PC, respectively). There were no statistically significant difference in the response rates among the three groups (p>0.05). The median Progression-free survival (PFS) was 5.0 months and the median Overall survival (OS) in each group was 7.1, 7.4 and 7.1 months, respectively (p>0.05). Conclusion: The response rate, median PFS and OS were similar among the three treatment arms. Grade III-IV thrombocytopenia was higher in the GC arm, while the GC regimen was more favorable than the other cisplatin-based treatmetns with regard to sensory neuropathy.
