Holstein 종(種) 유우(乳牛)의 유단백질(乳蛋白質)의 유전적다형(遺傳的多型)과 유조성분간(乳組成分間)의 연관성 (Association between Genetic Polymorphisms of Milk Proteins and Milk Compositions in Holstein Cows)

  • 본(本) 연구(硏究)는 유단백질(乳蛋白質)의 유전적표식(遺傳的標識)(genetic marker)를 유우(乳牛)의 유전적(遺傳的) 개량(改良)을 위한 유우등록 및 선발(選拔) 자료로 활용하고자 1992년 국립 종축원에서 사육증인 Holstein종 159두에서 각각 유즙 시료를 채취하여 Polyacrylarnide gel(PAGE)에 의한 전기영동방법에 의하여 ${\alpha}S1$-casein(${\alpha}S1$-CN) 및 ${\beta}$-casein(${\beta}$-CN), ${\kappa}$-casein(${\kappa}$-CN) 및 ${\beta}$-lactoglobulin(${\beta}$-LG)의 유전적(遺傳的) 다형(多型)을 분석(分析)하여 유전적(遺傳的) 변이체의 표현형(表現型), 유전자형(遺傳子型) 및 유전자빈도(遺傳子頻度)를 추정하고 이들 단백질의 유전적(遺傳的) 다형(genetic polyrnorphisms)과 유조성분과의 연관성을 분석(分析) 검토(檢討)하였다. 유단백질(乳蛋白質)인 ${\alpha}S1$-CN, ${\beta}$-CN, ${\kappa}$-CN 및 ${\beta}$-LG 유전좌위(遺傳座位)의 유전적(遺傳的) 변이체의 표현형(表現型) 분포의 관찰치와 기대치간에 유의적(有意的)인 차이가 인정 되지않아 이들 유전자(遺傳子)들이 유전적(遺傳的) 평형을 이루고 있는 것으로 사료되었다. 유단백질(乳蛋白質) 좌위(座位)의 유전자형(遺傳子型)을 분석(分析)한 결과(結果) ${\alpha}S1$-CN BB유전자형(遺傳子型), ${\beta}$-CN AA유전자형(遺傳子型), ${\kappa}$-CN AA유전자형(遺傳子型) 및 ${\beta}$-LG AB 유전자형(遺傳子型)의 출현빈도(出現頻度)가 각각 79.87%, 84.28%, 71.70% 및 49.10%로 높게 나타났다. 유단백질(乳蛋白質)의 유전자빈도(遺傳子頻度)에 있어서는 ${\alpha}S1-CN^B$${\alpha}S1-CN^C$가 각각 0.899 및 0.101이었고, ${\beta}-CN^A$${\beta}-CN^B$은 각각 0.921 및 0.079이었으며, ${\kappa}-CN^A$${\kappa}-CN^B$은 각각 0.837 및 0.163이었고, ${\beta}-LG^A$${\beta}-LG^B$은 각각 0.378 및 0.622이었다. 분산분석(分散分析) 결과(結果) 유단백질(乳蛋白質)의 유전자형(遺傳子型)들은 유조성분인 유지율, 단백질 함량 및 총 고형분 함량에서 유의적(有意的)인 연관성이 인정되었다. 유조성분 함량에 있어서는 ${\kappa}$-CN좌위(座位)의 BB유전자형(遺傳子型)은 유지율 및 유단백질(乳蛋白質) 함량에서 ${\kappa}$-CN좌위(座位)의 AA 및 AB 유전자형(遺傳子型)보다 유의적(有意的)으로 높게 나타났으며, ${\beta}$-LG좌위(座位)의 AA유전자형(遺傳子型)은 유지율에서 ${\beta}$-LG좌위(座位)의 AB 및 BB 유전자형(遺傳子型)보다 유의적(有意的)으로 높게 추정되었다. 이상의 연구결과(硏究結果)를 종합하여 볼때 유조 성분인 유지율 및 단백질함량을 높히기 위하여서는 ${\kappa}$-CN좌위(座位)의 BB 유전자형(遺傳子型)과 ${\beta}$-LG좌위(座位)의 AA 유전자형(遺傳子型)을 선발(選拔)하는것이 보다 유리할 것으로 생각되었다.

한우와 Holstein종의 κ -casein 유전자의 발현조절부위의 염기서열 분석 (Analysis of Sequence on promoter of κ -casein Genes in Korean Native Cattle and Holstein)

  • 본 연구는 한우와 Holstein종의 $\kappa$ -CN 유전자 발현조절부위, 즉 $\kappa$ -CN gene 5'-flanking region에 대한 특성을 구명하기 위하여 specific primer를 이용하여 PCR(polymerase chain reaction) 방법으로 분석하였다. PCR 증폭 결과, 349bp 크기의 증폭산물을 얻었으며 한우와 Holstein종의 $\kappa$ -CN 유전자의 발현조절부위의 염기서열 분석결과에 있어서는 -82bp에서 -431bp까지 분석되었으며, 종간의 염기서열 비교에서 -386bp에서는 Holstein종에서는 T 염기가 한우에서는 C 염기로 치환되었으며, -241bp와 -192bp에서는 Holstein종에서는 T 및 C 염가가 존재하였으나, 한우에서는 결실되었다. 또한, -183bp에서는 한우에서는 T 염기가 존재하였으나, Holstein종에서는 염기가 결실되어 있음을 알 수 있었다. 또한 한우와 Holstein 종간의 상동성 분석에 있어서는 한우 $\kappa$ -CN 유전자 발현조절부위의 염기서열이 Holstein종과 98.9% 의 상동성을 보였다.

Effects of Genetic Variants of κ-casein and β-lactoglobulin and Heat Treatment on Coagulating Properties of Milk

  • Fifty-two Holstein cows with different phenotypes of $\kappa$-casein ($\kappa$-CN) and $\beta$-lactoglobulin ($\beta$-LG) were selected to provide weekly milk samples for heating at 30, 70, 75 and $80^{\circ}C$ for 2 min. Coagulating properties of heated milk samples measured as rennet clotting time, rate of curd firming and curd firmness at cutting were determined by a Formagraph. Milk samples were analysed for fat and casein. Least squares analyses of data, after adjustments were made for effect of milk casein and fat contents, indicated that although an increase in heating temperatures resulted in less desirable coagulating properties, the effect of milk types was inherent irrespective of heating temperatures. The shortest rennet clotting time (6.06 min), fastest rate of curd firming (5.61 min) and firmest curd (38.05 mm) were obtained from milk with the B variant for $\kappa$-CN and B variant for $\beta$ -LG when preheated at $30^{\circ}C$. It appears that milk bearing $\kappa$-CN B is more resistant to heat perturbation. All milk samples having the $\kappa$-casein AA (milk types AA/AA, AA/AB, AA/BB) did not have a measurable K20 value when preheated at $70^{\circ}C$. This effect was observed for $\kappa$-casein AB (milk types AB/AA, AB/AB, AB/BB) at $75^{\circ}C$ and $\kappa$-casein BB (milk types BB/AA, BB/AB, BB/BB) at $80^{\circ}C$.

DNA Polymorphisms of κ-Casein, β-Lactoglobulin, Growth Hormone and Prolactin Genes in Korean Cattle

  • The gene and genotypic frequencies of ${\kappa}$-casein (${\kappa}$-CN), ${\beta}$-lactoglobulin (${\beta}$-LG), growth hormone (bGH) and prolactin (bPRL) loci in Korean cattle were investigated using PCR-RFLP analyses. Genomic DNA samples were obtained from 290 cows and 30 AI bulls. In both cows and bulls, the most predominant genotypes of ${\kappa}$-CN, ${\beta}$-LG, bGH and bPRL loci were AB, BB, AA and AA, respecitively. The frequencies of A and B alleles for ${\kappa}$-CN locus were .612 and .388 for cows and .567 and .433 for bulls. The respective frequencies of A and B alleles for ${\beta}$-LG locus were .153 and .847 in cows and .217 and .783 in bulls. The frequencies of A and B alleles for bGH locus were .769 and .231 in cows and .784 and .216 in bulls, respectively. The frequencies of A and B alleles for bPRL locus were .678 and .322 for cows and .767 and .233 for bulls. Differences in frequencies of these alleles were not significant between cows and bulls at all loci examined. If the DNA polymorphisms of these candidate genes are associated with economically important traits, they could serve as genetic markers for genetic improvement in future marker-assisted selection programs in Korean cattle.

Principal Milk Components in Buffalo, Holstein Cross, Indigenous Cattle and Red Chittagong Cattle from Bangladesh

  • The aim of the present study was to get a total physical and chemical characterization and comparison of the principal components in Bangladeshi buffalo (B), Holstein cross (HX), Indigenous cattle (IC) and Red Chittagong Cattle (RCC) milk. Protein and casein (CN) composition and type, casein micellar size (CMS), naturally occurring peptides, free amino acids, fat, milk fat globule size (MFGS), fatty acid composition, carbohydrates, total and individual minerals were analyzed. These components are related to technological and nutritional properties of milk. Consequently, they are important for the dairy industry and in the animal feeding and breeding strategies. Considerable variation in most of the principal components of milk were observed among the animals. The milk of RCC and IC contained higher protein, CN, ${\beta}$-CN, whey protein, lactose, total mineral and P. They were more or less similar in most of the all other components. The B milk was found higher in CN number, in the content of ${\alpha}_{s2}-$, ${\kappa}$-CN and ${\beta}$-lactalbumin, free amino acids, unsaturated fatty acids, Ca and Ca:P. The B milk was also lower in ${\beta}$-lactoglobulin content and had the largest CMS and MFGS. Proportion of CN to whey protein was lower in HX milk and this milk was found higher in ${\beta}$-lactoglobulin and naturally occuring peptides. Considering the results obtained including the ratio of ${\alpha}_{s1}-$, ${\alpha}_{s2}-$, ${\beta}$- and ${\kappa}$-CN, B and RCC milk showed best data both from nutritional and technological aspects.

Association between PCR-RFLP Polymorphisms of Five Gene Loci and Milk Traits in Chinese Holstein

  • The objective of this study was to assess the association of polymorphisms in ${\kappa}$-cn, ${\beta}$-lg, ${\beta}$-lg 5′ flanking region, CSN1S2, and IGFBP-3 genes with milk production traits and mastitis-related traits in Chinese Holstein. Traits analyzed were 305 day standard milk yield, protein percentage, fat percentage, the ratio of fat percentage and protein percentage, pre-somatic cell count, somatic cell count, and somatic cell score, respectively. CSN1S2 locus was uninformative because only one genotype BB was found in Chinese Holstein. Allele frequencies of A and B in IGFBP-3 gene were 0.5738 and 0.4262 in Chinese Holstein population, which was different from reported Qinchuan cattle population. The genotypes of animals at IGFBP-3 locus significantly affected 305 day standard milk yield, protein percentage, and somatic cell score. The ${\beta}$-lg genotypes had a significant effect on protein percentage and the ratio of fat percentage and protein percentage. Polymorphism in ${\beta}$-lg 5′ flanking region was associated with 305 day standard milk yield, protein percentage, fat percentage, pre-somatic cell count, and somatic cell count. No significant associations of the polymorphism in ${\kappa}$-cn gene were observed for any trait.

  • Milk protein polymorphisms were genotyped by polyacrylamide gel electrophoresis (PAGE) from 109 Maiwa and 100 Jiulong yaks. The relationships between milk protein polymorphisms and 3 milking traits were studied. The results showed that $\beta$-CN, $\kappa$-CN and $\alpha$-La were monomorphic, and ${\alpha}_{s1}$-CN and $\beta$-Lg were polymorphic, with ${\alpha}_{s1}$-CN D and $\beta$-Lg E as dominant genes, respectively. The frequencies of ${\alpha}_{s1}$-CN D were 0.8073 and 0.6000 in two populations and $\beta$-Lg E were 0.9770 and 0.9700. The mean heterozygosities were 0.1021 and 0.1867 in the two populations. No significant effects on milking traits and milk protein compositions were observed except for ${\alpha}_{s1}$-CN locus on fat percentage in Jiulong yak.

Nanosphere Form of Curcumin Stimulates the Migration of Human Umbilical Cord Blood Derived Mesenchymal Stem Cells

  • Curcumin, a hydrophobic polyphenol derived from turmeric, has been used a food additive and as a herbal medicine for the treatment of various diseases. In the present study, we found the functional role of a nanosphere loaded with curcumin (CN) in the promotion of the motility of human umbilical cord blood derived mesenchymal stem cells (hUCB-MSCs) during the wound closure. We found that the efficacy of hUCB-MSCs migration induced by CN was 1000-fold higher than that of curcumin powder. CN significantly increased the motility of hUCB-MSCs by activating c-Src, which is responsible for the phosphorylation of protein kinase C (PKC) and extracellular signal-regulated kinase (ERK). CN induced the expression levels of α-actinin-1, profilin-1 and filamentous-actin, as regulated by the phosphorylation of nuclear factor-kappa B during its promotion of cell migration. In a mouse skin excisional wound model, we found that transplantation of UCB-MSCs pre-treated with CN enhances wound closure, granulation, and re-epithelialization at mouse skin wound sites. These results indicate that CN is a functional agent that promotes the mobilization of UCB-MSCs for cutaneous wound repair.

케이신의 유전적 변이체에 대한 최근 연구동향과 웹 사이트의 이용 (Current Research Trend of Genetic Variants of Bovine Casein and Application of The Web Site)

  • The nomenclature of genetic variants of casein which is major protein in milk have had a lot of confusion, but now have established. Genetic variants of ${\alpha}_{s1}-CN,\;{\alpha}_{s2}-CN,\;{\beta}-CN,\;{\kappa}-CN$ have reported 8 variants(A, B, C, D, E, F, G, H), 4 variants(A, B, C, D), 13 variants ($A_1,\;A_2,\;A_3,\;A_4$, B, C, D, E, F, G, $H_1,\;H_2$, I), 11 variants(A, B, C, E, $F_1,\;F_2,\;G_1,\;G_2$, H, I, J), respectively. Their data detailed have introduced in several web sites including www.uniprot.org. The studies on genetic variants of casein from Korean native cattle have been reported only ${\beta}-casein\;A_4$ but still not established the protein sequence. The classification and distinct nomenclature of genetic variants of bovine casein were required because the development of milk science and technology have been focused in the region that have to studied biochemically such as functional foods, EMC and GMO et al.

케이크 도넛의 흡유저감에 대한 두부 페이스트와 비전분성 탄수화물 고분자의 영향 (Impact of Tofu Paste and Non-starch Polysaccharides on Oil Uptake Reduction in Cake Doughnuts)

  • 전량 폐기되는 파지두부로부터 제조된 두부 페이스트와 14종의 NSP들을 혼합하여 도넛의 흡유저감에 대한 효과를 조사하였다. 두부 페이스트만을 첨가한 도넛(두부도넛)의 흡유저감율은 10.8%이었으나, 두부 페이스트와 NSP를 함께 사용하는 경우 도넛(NSP-두부도넛)의 흡유저감율은 두부도넛에 비해 향상되었다. 두부 페이스트와 함께 첨가된 NSP들의 흡유저감율에 대한 효과는 NaA가 41.2%로 가장 높았으며, HMP>XAT>CN10T = LMP = ALMP ${\lambda}C$>GG>LBG>${\iota}C$>${\kappa}C$>CN15U>GLG>CN40H 순으로 증가하였다. 또한 NSP 만을 흡유저감 소재로 첨가하였을 때보다 두부 페이스트와 함께 사용하는 것이 도넛의 흡유저감에 더욱 효과적이었고, 그 효과는 음이온성 검류들에서 뚜렷하였다. 유탕처리 시 두부 페이스트의 갈변으로 인해 두부 페이스트 및 두부 페이스트-NSP를 첨가한 도넛들은 대조군에 비해 명도가 감소하고 황색도가 증가하였으나 그 차이는 미미하였다. 두부 페이스트와 함께 NaA, ${\kappa}C$, LBG를 첨가하여 제조된 도넛의 비용적은 대조군과 유사한 수준을 나타내었다. 따라서 도넛의 흡유저감율과 비용적을 고려하면 두부 페이스트와 NaA를 흡유저감 소재로 함께 사용하는 것이 흡유저감 도넛을 제조하는데 가장 적합한 것으로 판단된다. 그럼에도 NaA 보다는 흡유저감율이 적지만 ${\kappa}C$과 LBG을 두부 페이스트와 혼합하여 사용하는 것도 차선책으로서 의미가 있을 것 같다.
