The Comparison of Color and Physiological Properties of Hizikia fusiformis Cooking Juice Ethanol Extract Irradiated with Gamma Ray and Electron Beam (감마선 및 전자선 조사에 따른 톳 자숙액 에탄올 추출물의 색상 및 생물학적 활성 변화 비교)

  • This study was carried out to assess the effect of radiation on the changes of Hizikia fusiformis cooking juice ethanol extract and to compare the effect of gamma ray and electron beam. On the applying radiation, the dark color of cooking juice became changed with higher brightness and lower redness and yellowness. But, there was no difference between gamma ray radiation and electron beam radiation. 1,1-Diphenyl-2-picryl-hydrazyl radical scavenging activity and tyrosinase inhibitory activity of cooking juice were shown to be increased by radiation independent on the radiation source types. The reason for the increased biological activities was caused by higher content of total phenolic compounds. The results could be applied to investigate the effect of radiation source on the color and antioxidant activity of biomaterials, and it was thought that irradiation could be an promising method for enhancing the biological activity of biomaterials.

  • Glycyrrhizae Radix is an herbal medicine vulnerable to fungi. So there are regulations on mycotoxins. Therefore, in this study, the affect of various drying methods and additional gamma irradiation (10 kGy) was studied to suppress the microbial generation of Glycyrrhizae Radix. As a result of detecting and comparing microorganisms using the dry film method for samples prepared by each of natural, hot air, freezing, microwave drying and gamma irradiation, the affect of reducing microorganisms by microwave drying and gamma irradiation was confirmed. There was no statistically significant change in the content of the active compounds between the treatment groups.

  • In tissue engineering application, a fibrous structure of scaffolds has been issued as an alternative system to regulate cell survival and tissue regeneration, and electrospinning technique has been popularly used to generate fibrous meshes or sheets mimicking the structure of native extracellular matrix (ECM). However, recent strategy in the scaffold development is expanded to provide the structural property as well as a biological property of native ECM, a variety of surface modification techniques have been used to introduce biological property. In this study, we developed biomimetic poly(L-lactide-co-${\varepsilon}$-caprolactone) (PLCL) nano- and micro-fibrous scaffolds as a unique platform with structural and biological properties with native ECM using electrospinning method and gamma-ray irradiation. Surface morphology of the scaffolds was observed by scanning electron microscopy, and alteration of surface property was evaluated with toluidine blue O staining, water contact angle measurement and ATR-FTIR analysis.

  • Gold nanoparticles (GNPs) have led to the development of a new field in the diagnosis and treatment of diseases such as cancer. An efficient synthesis of gold nanoparticles within the range of 8~57 nm was established by ${\gamma}-ray$ irradiation. The good point of a radiation-based method is the production of gold nanoparticles with a higher concentration and narrower size distribution compared with conventional methods. The size of gold nanoparticles was controlled using two methods. : (i) varying the ${\gamma}-ray$ irradiation dose of 10 to 25 kGy and (ii) varying the concentration of $HAuCl_4$ solution from 4 to 40 mM. In addition, the GNPs were radiolabeled using $[^{125}I]NaI$ in a simple and fast manner with high yields. The produced gold nanoparticles were characterized using a transmission electron microscopy (TEM), a UV-visible spectrophotometer, and a radio-TLC imaging scanner. From these results, these radiolabeled GNPs can be applicable for a radioisotope tag of biomolecules.

  • Studies on the carbohydrate metabolism of yeast as influenced by gamma-irradiation from cobalt-60 have been carried, then the mechanisms of radiation effect on respiration and fermentation were discussed under considerations of permeable changes of irradiated cell membrane. The cells of baker's yeast (Saccharomyces cerevisiae) which had been gamma-irradiated of 240 k.r. doses for an hour, then were put into aerobic oxidation and anaerobic fermentation without substrate. Total and fractionated carbohydrates of irradiated yeast cells were determined by calorimetric method with anthrone and orcinol reagents, the amounts of total carbohydrate, trehalose, RNA-ribose, PCA-soluble glycogen, alkali-soluble glycogen, acetic acid-soluble glycogen, mannan and glucan were determined according to the course of aerobic oxidation and anaerobic fermentation. It is found that the carbohydrates of irradiated cells leak out and amount of the losses teaches eleven times more than that of control, the volume of losses are seems to be replaced by water, it can be suggested the damage of gamma-irradiation occurs in the site of passive transport of cell membrane. The endogeneous aerobic respiration of irradiated cells are increased much more than control, the synthesis of reserve glycogen, glucan and RNA-ribose promoted much more than control. The anaerobic fermentation of irradiated cells are also increased than that of control, but the breakdown of carbohydrate is less than endogeneous respiration of irradiated cells. The synthetic rate is also less than that of aerobic oxidation. In irradiated yeast cells, trehalose is revealed to be primary substrate for endogeneous carbohydrate metabolism, so it is proved that the enzymic patterns are not changed but the activities of enzymes relating endogeneous respiration and autofermentation is activated. It is to be considerable to distiguish endogeneous respiration and autofermentation from exogeneous respiration and fermentation on irradiation, for membrane permeability changes and loses out carbohydrate by ionizing radiation.

  • To investigate the stability of ginseng saponin, various physical and chemical treatments for red ginseng alcohol extract (70% ethyl alcohol) were carried out, and then the variations of ginseng saponin in extract were investigated by high performance liquid chromatography (HPLC) method. Irradiation of ${\gamma}$-ray, and ultraviolet ray, sonocatalysis by ultrasonicator, treatment of electronic range, catalytic ozonation did not or slightly affect degradation of ginseng saponins, but they were degraded by heat treatment.

  • Low-salt fermented pollack Theragra chalcogramma roe (Myungran Jeotgal, MJ), a traditional Korean fermented seafood, was prepared using a commercially available method and irradiated using gamma rays (0-10.0 kGy) to investigate the effect of gamma irradiation (GI) on food quality. After irradiation with 2.5 kGy gamma rays, the number of viable cells in MJ was reduced by one log compared to that observed in non-irradiated MJ, and no living cells were detected after 7.5 kGy GI. Though up to 2.5 kGy GI had no effect on color quality, over 7.5 kGy GI resulted in significantly poor quality MJ surface color. Saturated fatty acid contents increased slightly in irradiated MJ, while those of polyenes and monoenes decreased slightly as the GI dose increased. Exposure of MJ to GI had no effect on the free amino acid and mineral composition. These results demonstrated that the appropriate GI dose for MJ ranged from 2.5 to 5.0 kGy, which may be recommended for industrial application.

Physicochemical Characteristics of Gamma Irradiated Changran Jeotkal during Storage at $10^{\circ}C$ (감마선 조사기술을 이용하여 제조된 양념창란젓갈의 이화학적 품질특성)

  • Changran jeotkal, a Korean traditional fermented seafood, was prepared as a pilot scale using a commercial method and irradiated at 0, 2.5, 5.0 and 10 kGy by gamma ray to investigate possibilities for further industrial application. To see the effectiveness and rapid industrialization, hot pepper powder was irradiated at 10 kGy and manufactured the changran jeotkal (HP-10 kGy) as same method since the hot pepper powder was approved legally for gamma irradiation in Korea. The content of volatile basic nitrogen and amino nitrogen was significantly reduced by gamma irradiation in all storage periods. Amino nitrogen contents of 0, 2.5, 5.0, 10 kGy and HP-10 kGy were 98.9, 98.5, 92.4, 88.0 and 93.1mg%, respectively after 12 week of storage at $10^{\circ}C$. In total, 8 kinds of biogenic amines were found from the samples, and the contents in the gamma irradiated changran Jeotkal were lower than those of the control during storage. The sample of HP-10 kGy showed similar physicochemical characteristics to the sample irradiated as $2.5{\sim}5.0\;kGy$. Results indicated that gamma irradiation of fermented seafood products such as seasoned changran jeotkal improved quality stability, thus, we recommend gamma irradiation for industrial application.

  • Neutron activation analysis based on $textsc{k}$$_{o}$-standardization method# ($textsc{k}$o-NAA) is Com as one of the most remarkable progresses of the NAA with advantages of experimental simplicity, high accuracy, excellent flexibility with respect to irradiation and counting conditions, and suitability for computerization. This study was carried out to determine the reactor neutron spectrum parameters, i.e. $\alpha$ and f as the main factors of irradiation quality at NAA #1 irradiation hole on HANARO research reactor, to evaluate peak detection efficiency of the gamma-ray spectrometer for the use in the $textsc{k}$$_{o}$ experiments and to compare the measured concentration results with the certified values of some SRMs applying the experimentally determined to-parameters.ers.

Radiation Sterilization Dose of Biowastes (방사선을 이용한 생물폐자원의 멸균선량 결정)

  • In order to determine the optimal radiation dose for the sterilization of biowastes, the bioburden, frequencies and radiosensitivities of bacteria, mold and fungi in rice straw, chaff, corn stover and sawdust was observed before and after ${\gamma}-ray$ irradiation. Radiation sterilization dose of rice straw, chaff, com stover and sawdust was calculated as 17.7, 17.6, 15.6 and 20.0kGy, respectively, from the mutual compensation of screening dose and derived dose on the basis of $10^{-3}$ SAL. This method could be acceptable for the sterilization of various biowastes including food, pharmaceuticals, etc.
