Identification of Phosphatidylcholine-Phospholipase D and Activation Mechanisms in Rabbit Kidney Proximal Tubule Cells

  • The present study showed that receptor-mediated activation of rabbit kidney proximal tubule cells by angiotensin II, the $Ca^{2+}$ ionophore A23187, or the protein kinase C activator phorbol myristate acetate (PMA) all stimulated phospholipase D (PLD). This was demonstrated by the increased formation of phosphatidic acid, and in the presence of 0.5% ethanol, phosphatidylethanol (PEt) accumulation. Angiotensin II leads to a rapid increase in phosphatidic acid and diacylglycerol, and phosphatidic acid formation preceeded the formation of diacylglycerol. This result suggests that some phosphatidic acid seems to be formed directly from phosphatidylcholine hydrolyzed by Pill. On the other hand, EGTA substantially attenuated angiotensin II and A23187-induced PEt formation, and when the cells were pretreated with verapamil angiotensin II-induced Pill activation was completely abolished. These results provide the evidence that calcium ion influx is essential for the agonist-induced Pill activation. In addition, staurosporine, an inhibitor of protein kinase C, strongly inhibited PMA-induced PEt formation, but was ineffective on angiotensin II-induced PEt accumulation. $GTP{\gamma}S$ also stimulates PEt formation in digitonin-permeabilized cells, but pretreatment of the cells with pertussis toxin failed to suppress angiotensin II-induced PEt formation. From these results, we conclude that in the rabbit kidney proximal tubule cells the mechanisms of angiotensin II- and PMA-induced Pill activation are different from each other and mediated via a pertussis toxin-insensitive trimeric G protein.

A Case of Advanced Metastatic Gallbladder Cancer Patients Treated with Oriental Medicine for Four Months (4개월간 한방치료를 받은 전이성 담낭암 환자 증례)

  • Objectives : To observe the therapeutic effects of Oriental herbal prescriptions on an advanced metastatic gallbladder cancer patient. Methods : Hang-Am-Plus (HAP), Gun-Chil-Gye-Bok-Jung (GGJ) and Se-Gan-San (SGS) were prescribed three times a day. To observe the therapeutic effects of oriental medical prescriptions, computed tomography (CT) and magnetic resonance imaging (MRI) images, tumor markers and laboratory tests were carried out regularly. Results : The levels of AST, ALT, ALP, ${\gamma}$-GTP, TB, DB, CA19-9, and CEA decreased significantly from November 29th 2011 to March 14th 2012. There was no interval change on CT images taken between November 15th 2011 and January 3rd 2012. Conclusions : In this case, it could be concluded that Oriental medicinal treatment might be considered as a palliative alternative therapy for the advanced metastatic gallbladder patient.

Effects of Moschus, Bovis Calculus, Ursi Fel Aqua-Acupuncture on Liver Damage (사향(麝香).우황(牛黃).웅담(熊膽) 약침(藥鍼)이 간손상(肝損傷)에 미치는 영향(影響)에 관한 연구(硏究))

  • In order to investigate experimentally that effect of Moschus, Bovis Calculus, Ursi Fel aqua-axupuncture on acutely damaged liver of rats induced by radix aconiti. The author admininisterated aqua-acupuncture according to method of manufacture stimulation to corresponding points, Kan-su$(BL_8)$ and Ki-mun($LR_{14}$), and carried out hematological, serological examination and electromicroscopical obseration. WBC level was decreased significantly in the experimental groups at 24 and 48 hours as compared with the control group. ${\gamma}-GTP$ level was decreased significantly in the experimental groups at 24 and 48 hours as compared with the control group. GPT level was decreased significantly in the experimental groups at 24 and 96 hours as compared with the control group, but GOT level did not reeveal any statistical difference. At 24-hours group, the cell organelles of the hepatic cells are destructed. The cisternae of the smooth endoplasmic reticulum are particulary dilated in the 24-hours control group compared with experimental group. At 48-hours group, the destructed cell organelles are recovered in the experimental group compared with control group. At 96-hours group, the cell organelles of hepatic cells are showing feture of normal group. According to the above findings, it is considered that Moschus, Bovis Calculus, Ursi Fel aqua-acupuncture has effects of recovery of acutely damaged liver.

Characterizing of Four Obesity Types in Obese Women Based on the Questionnaire of Diseases and Physical Tests (여성 비만의 유발유형별 일반 증상과 검사 특성 연구)

  • Objectives : To characterize four types of obesity and to effectively improve the treatment of obesity through Oriental medicine Methods : At 00 Oriental Medical Center, 203 female subjects who intented to lose weight were requested to complete a questionnaire. These participants were also given physical tests. The Questionnaire consisted of questions both about general life style and obesity in oriental medicine framework. The physical tests were blood tests, a body composition via Inbody 2.0, and physical strength tests. One-way analysis of variance was done to compare the means of items and physical tests between four types of obesity. Duncan was used by post hoc test. Results : 1. Significant differences between obesity type III and obesity type IV in ever tried to lose weight, childhood obesity and excercise times were observed in the questionnaire of general life style(p<0.05). 2. Ducan test showed significant differences between four obesity types in diseases (p<0.05). 3. Significant differences between four obesity types in height, % body fat muscular endurance, soft lean mass, fat mass, Trigliceride, Total cholesterol and ${\gamma}-GTP$ were observed (p<0.05). Conclusions : Further clinical research is necessary in the four types of obesity explored. The diagnosis and treatment based on these types should be further studied.

Development of a Novel Experimental Model for Nephrotoxicity Assessment Using Membrane Vesicles of Rabbit Renal Proximal Tubules (신장근위곡세뇨관 막소포를 이용한 신장독성 실험모델 개발)

  • Basolateral and brush border membrane (BLM and BBM) vesicles of renal proximal tubules were prepared from adult male New Zealand White rabbits to evaluate as a potential model for assessment of nephrotoxicity. PAH uptakes using BLMV, glucose and leucine uptakes using BBMV were measured in the rabbits treated cephaloridine. In addition, urinalysis and histopathological studies were performed to investigate the correlationship with membrane vesicle uptakes. The results were as follows: (1) the activite of Na+, K+ -ATPase was enriched 12.3-fold in vasolateral memvrane vesicles (BLMV) and the specific activity of alkaline phosphatase in purified brush border memvrane vesicles (BBMV) was enriched 10.1-fold compared with each of microsomal homogenate. (2) In the uptake experiments, cephaloridine decreased initial and probenecid-sensitive PAH uptakes in BLMV. (3) Cephaloridine tested decreased initial and phlorizin-sensitive glucose uptakes in BBMV. (4) Cephaloridine tested decreased initial and Na+-dependent leucine uptakes in BBMV. (5) Cephaloridine tested significantly increased the urinary excretion of glucose and activity of ${\gamma}$-GTP. (6) Cephaloridine tested caused moderate necrotic changes in renal tubular cells and formation of urinary cast in the lumina of Henle's loop and collecting tubules besides the swelling of renal tubules.

A Study on Temple Food Intake and Health (사찰음식 섭취와 건강에 관한 연구)

  • The purpose of this study was to examine the effect of temple food intake on health improvement. The survey was conducted from September to October 2013. The subjects in this study were 46 adults who participated in short-term 'Temple Stay' program. Every meal was prepared according to prearranged temple food menu, and it was provided to subjects for 4 weeks. Dietary intake, anthropometric measurements and biochemical parameter were measured and nutritional & health status were analyzed to find the effect of the program. Nutrients content of the temple food was sufficient to dietary reference intake while content of fat, fiber, vitamin A and calcium from the meal was significantly higher than the subjects' usual diet. Mean body weight, BMI and blood pressure (systolic and diastolic) related with obesity and hypertension decreased after the program. The index referring to liver function and cardiovascular disease risk such as serum ALT, ${\gamma}$-GTP, TG and LDL decreased while HDL increased thereby showing positive effect. The above results anticipate that temple foods with an affirmative effect in improving health status have a potential benefit that can be applied to general food service. Further detailed study might provide information in developing healthy diet for a corresponding clinical problem.

The relative risk of major risk factors of ischemic heart disease (주요 위험요인별 허혈성심질환 사망위험도 분석)

  • Due to the dramatic increase of mortality from ischemic heart disease (IHD) during the last decade, it is highly warranted to present the effective prevention strategy. Therefore this study identified the major risk factors of IHD over 10 years of follow-up among 2,268,018 participants of National Health Insurance Exam in 1996 with Cox proportional hazard model. In men, BMI, blood pressure, smoking were significantly associated with IHD, whereas hypertension, perceived health status and ${\gamma}$-GTP were related with IHD in women.

  • RAS guanyl-releasing protein 3 (RasGRP3), a member of the Ras subfamily of GTPases, functions as a guanosine triphosphate (GTP)/guanosine diphosphate (GDP)-regulated switch that cycles between inactive GDP- and active GTP-bound states during signal transduction. Various growth factors enhance hepatocellular carcinoma (HCC) proliferation via activation of the Ras/Raf-1/extracellular signal-regulated kinase (ERK) pathway, which depends on RasGRP3 activation. We investigated the relationship between polymorphisms in RasGRP3 and progression of hepatitis B virus (HBV)-infected HCC in a Korean population. Nineteen RasGRP3 SNPs were genotyped in 206 patients with chronic liver disease (CLD) and 86 patients with HCC. Our results revealed that the T allele of the rs7597095 SNP and the C allele of the rs7592762 SNP increased susceptibility to HCC (OR=1.55, p=0.04 and OR=1.81${\sim}$2.61, p=0.01${\sim}$0.03, respectively). Moreover, patients who possessed the haplotype (ht) 1 (A-T-C-G) or diplotype (dt) 1 (ht1/ht1) variations had increased susceptibility to HCC (OR=1.79${\sim}$2.78, p=0.01${\sim}$0.03). In addition, we identified an association between haplotype1 (ht1) and the age of HCC onset; the age of HCC onset are earlier in ht1 +/+ than ht1 +/- or ht1 -/- (HR=0.42${\sim}$0.66, p=0.006${\sim}$0.015). Thus, our data suggest that RasGRP3 SNPs are significantly associated with an increased risk of developing HCC.

Pharmacological Evidence that Cromakalim Inhibits $Ca^{2+}$ Release from Intracellular Stores in Porcine Coronary Artery

  • In the present study, it was aimed to further indentify the intracellular action mechansm of cromakalim and levcromakalim in the porcine coronary artery. In intact porcine coronary arterial strips loaded with fura-2/AM, acetylcholine caused an increase in intracellular free $Ca^{2+}$ $([Ca^{2+}]_i)$ in association with a contraction in a concentration-dependent manner. Cromakalim (1 ${\mu}M$) caused a reduction in acetylcholine-induced increased $[Ca^{2+}]_i$ not only in the mormal physiological salt solution (PSS) but also in $Ca^{2+}$-free PSS (containing 1 mM EGTA). In the skinned strips prepared by exposure of tissue to 20 .${\mu}M$ B-escin, inositol 1,4,5-trisphosphate ($IP_3$) evoked an increase in $[Ca^{2+}]_i$, but it was without effect on the intact strips. The $IP_3$-induced increase in $[Ca^{2+}]_i$ was inhibited by cromakalim by 78% and levcromakalim by 59% (1 .${\mu}M$, each). Pretreatment with glibenclamide (a blocker of ATP-sensitive $K^+$ channels, 10 .${\mu}M$) and apamin (a blocker of small conductance $Ca^{2+}$-activated $K^+$ channels, 1 .${\mu}M$) strongly blocked the effect of cromakalim and levcromakalim. However, charybdotoxin (a blocker of large conductance $Ca^{2+}$-activated $K^+$ channels, 1 .${\mu}M$) was without effect. In addition, cromakalim inhibited the $GTP{\gamma}S$ (100 .${\mu}M$, non-hydrolysable analogue of GTP)-induced increase in $[Ca^{2+}]_i$. Based on these results, it is suggested that cromakalim and levcromakalim exert a potent vasorelaxation, in part, by acting on the $K^+$ channels of the intracellular sites (e.g., sarcoplasmic reticulum membrane), thereby, resulting in decrease in release of $Ca^{2+}$ from the intracellular storage site.

Effect of Garlic and Medicinal Plants Composites on the Liver Function and Lipid Metabolism of Rats Administered with Ethanol During the Short-term (단기 알코올 투여 시 마늘과 한약재 복합물이 체내 지질 조성 및 간기능 회복에 미치는 영향)

  • This study was performed to observe the effect of hot-water extracts from garlic and 13 kinds of medicinal plants composites (GMP) on hyperlipidemia and hepatoprotective activity in rats administered with alcohol. Male Sprague-Dawly rats were fed an AIN-93 diet (Normal), a normal diet plus ethanol (control, 10 ml of 40% ethanoljkgjday), a control diet plus 0.5% garlic and 1.0% medicinal plants composites extracts (GMP-I), and a control diet plus 1.0% garlic and medicinal plants composites extracts (GMP-II) for 7 days. Blood glucose was higher than the control, but it was markedly decreased in the GMP-II group. Elevation total lipids, cholesterol, triglyceride and phospholipids in serum were markedly decreased in rats fed with GMP-I. GMP-II also inhibited the increase of lipid content in serum. Activities of GOT, GPT, $\gamma$-GTP and ALP in serum elevated by alcohol were significantly inhibited in the GMP group. TBARS content of serum was significantly decreased in GMP groups administered with garlic and medicinal plant extracts. Extracts of garlic and medicinal plants play an important role in recovering liver function in rats from alcohol induced damage.
