Microbial Transformation of Bioactive Diterpenoids from Acanthopanax koreanum by Fusarium oxysporum

  • Microbial transformation of (-)kaur-l6-en-19-oic acid and (-)pimara-9(1l), 15-dien-19-oic acid from A. koreanum was investigated. Throughout the screening of the capability of metabolizing these bioactive diterpenoids, two microorganisms have chosen among various fungi and streptomycetes tested. Scale-up fermentation with Fusarium oxysporum KCTC 6051 produced two metabolites related to the precursor diterpenoids. The two metabolites were isolated by column chromatography and identified by chemical and spectroscopic methods as $2\beta$, $16\alpha$-dihydroxy kauran-19-oic acid and $16\alpha$-hydroxy kauran-19-oic acid. However any microorganisms capable to transform (-) pimara-9(11), 15-dien-19-oic acid was not screened in this condition.

Evaluation of glucosidases of Aspergillus niger strain comparing with other glucosidases in transformation of ginsenoside Rb1 to ginsenosides Rg3

  • The transformation of ginsenoside Rb1 into a specific minor ginsenoside using Aspergillus niger KCCM 11239, as well as the identification of the transformed products and the pathway via thin layer chromatography and high performance liquid chromatography were evaluated to develop a new biologically active material. The conversion of ginsenoside Rb1 generated Rd, Rg3, Rh2, and compound K although the reaction rates were low due to the low concentration. In enzymatic conversion, all of the ginsenoside Rb1 was converted to ginsenoside Rd and ginsenoside Rg3 after 24 h of incubation. The crude enzyme (b-glucosidase) from A. niger KCCM 11239 hydrolyzed the ${\beta}$-($1{\rightarrow}6$)-glucosidic linkage at the C-20 of ginsenoside Rb1 to generate ginsenoside Rd and ginsenoside Rg3. Our experimental demonstration showing that A. niger KCCM 11239 produces the ginsenoside-hydrolyzing b-glucosidase reflects the feasibility of developing a specific bioconversion process to obtain active minor ginsenosides.

  • ${\gamma}-MnO_2$는 示差熱曲線에서 結晶變移의 吸熱 peak가 明白하게 나타났으며 他變態의 轉移溫度 와 相異하였다. 本 peak를 利用하여 半面積法으로 ${\gamma}-MnO_2$만의 定量分析을 檢討하였다. 相對誤差 ${\pm}$5%範圍內에서 試料 50mg∼300mg까지 定量할 수 있었다. ${\alpha},{$beta}-MnO_2$ (pyrolusite)가 妨害物質로서 約 75%까지 混合될경우에도 相對誤差는 ${\pm}$10%程度였으며 約 50%以內가 混合될 경우에는 相對誤差 ${\pm}$5%以內로서 거의 영향이 없었다.

  • Reichstein's substance S의 $11\beta$-hydroxylation 활성이 있는 균주를 선택하고자 21 균주를 실험한 결과 4균주에서 $11\beta$-hydroxylation의 활성이 발견되었으며, 그 중 Pellicularia fillamentosark 가장 높은 효소활성을 보였다. 이 균주의 $11\beta$-hydroxylation의 합성을 강력하게 저해하였다. $11\beta$-hydroxylation의 최적 pH 범위는 2.0-8.0으로 광범위하였다. 성장단계 중 균사체 분지시기와 완전하게 성숙한 균사체 시기에 효소합성의 유도가 가장 활발하였으며, 70시간 배양된 균사체가 가장 좋았다. 반면, 포자에는 기질에 의한 $11\beta$-hydroxylation의 유도현상이 없는 것으로 판정되었다.

Effects of Sonication and Vacuum Infiltration on Agrobacterium-Mediated Transformation in Immature Embryos of Korean Wheat Genotypes

  • The effects of sonication and vacuum infiltration on transformation efficiency was investigated by using immature embryos of Korean wheat as explants. Two Agrobacterium tumefaciens strains, KYRT1 and EHA105, carrying pCAMBIA 1305.1 were used. Transformation efficiency was demonstrated by the detection of $\beta-glucu-ronidase$ (GUS) activity. GUS expression showed clear difference among Korean wheat cultivars. Geurumil showed higher GUS expression efficiency $79.1\%$ compared with other cultivars. The effects of the duration of vacuum infiltration and sonication treatment showed a tendency high GUS expression efficiency by their combination. In comparison with other Agrobacterium strains, KYRT1 showed high efficiency in most Korean cultivars.

  • We optimized the biological and physical parameters for DNA delivery into thalli of the red alga Porphyra yezoensis using a particle bombardment device. The efficiency of transformation was determined using the ${\beta}-glucuronidase$ (GUS) assay. The optimal helium pressure, distance of tungsten particle flight, and ratio of DNA to tungsten particles were $23kgf/cm^2$, 8 cm, and $5{\mu}g/mg$ tungsten, respectively. During bombardment, osmotic treatment with a mixture of 0.6 M mannitol and sorbitol increased the efficiency of GUS transformation. After 2 days, the blue color indicating GUS activity was observed using a histochemical assay.

A Study on Transformation of Dynamic DSC Results into Isothermal Data for the Formation Kinetics of a PU Elastomer

  • The present study examines the transformation of dynamic DSC data into the equivalent isothermal data for the formation kinetics of a polyurethane elastomer. The reaction of 2'-dichloro-4,4'-methylenedianiline (MOCA) with a PTMG/TDI-based isocyanate prepolymer was evaluated. DSC measurement was performed in the dynamic scanning mode with several different heating rates to obtain the reaction thermograms. Then, the data was transformed into the isothermal data through a procedure based on Ozawa analysis. The main feature of this procedure was the transformation of $({\alpha}-T)_{\beta}$ curves from dynamic DSC into $({\alpha}-t)_T$ curves using the isoconversional $(t-T)_{\alpha}$ diagram. Validity was discussed for the relationship between the dynamic DSC data and the transformed isothermal results.

  • Cavitation erosion properties of the Cu-9Al-4.5Ni-4.5Fe alloys (Al-bronze) surface-modified by flame quenching process have been investigated. After flame quenching at above $T_{\beta}$, the surfaces of Al-bronze with $\alpha$ + $\textsc{k}$ structure have been changed into the $\alpha$ + $\beta$' martensite phases by the eutectoid reaction of $\alpha$ + $\textsc{k}$\longrightarrow$\beta$ followed by the martensite transformation of $\beta$\longrightarrow$\beta$'. As a result of cavitation test, the measured incubation time and erosion rate of the $\alpha$ + $\beta$' alloy was 1.2 times higher and 1.5 times lower, respectively, compared to those of the conventional $\alpha$ + $\textsc{k}$ alloys, showing a remarkable increase of cavitation resistance with the formation of $\beta$' martensite. This is attributed to a preferential erosion of the $\textsc{k}$ precipitates that show the lowest resistance among the $\alpha$, $\textsc{k}$, $\beta$' phases under cavitation loading.ases under cavitation loading.

  • The effects of the ${\beta}-Si_3N_4$ starting particle size and $\alpha$/$\beta$ phase transformation during sintering process on the microstructure evolution of Yttrium $\alpha$-Sialon ceramics were investigated. As-received ${\beta}-Si_3N_4$ powder (mean particle size: 0.54$\mu$m) and classified ${\beta}-Si_3N_4$ powder(mean particle size: $0.26\mu{m}$) were used as starting powders. With decreasing the starting particle size, the growth of elongated grains was enhanced, which resulted in the whisker -like microstructure with elongated grains. These results were discussed in relation to the two-dimensional nucleation and growth theory. In the specimen heat treated at $1600^{\circ}C$ for 10h before sintering at $1950^{\circ}C$for 1h under 40atm(2-step sintering), the grain size was smaller than of the 1-step sintering at 195$0^{\circ}C$ for 1h. However, bimodal microstructure evolution were not not remarkable in both sample, which is ascribed to the $\alpha$-phase contents existing in ${\beta}-Si_3N_4$ starting powder.

고온가압소결한 탄화규소의 집합조직 (Texture in hot-pressed silicon carbide)

  • $\alpha$ - 및 $\beta$-SiC를 출발원료로 하여 일축성형한 후 $1800^{\circ}C$에서 고온가압법으로 소결한 탄화규소에 나타난 우선방위의 정도를 X-ray pole figure analysis로 조사하였다. $\alpha$-SiC를 출발원료로 사용한 경우에 고온 가압소결 후 약한 집합조직을 보인 반면, $\beta$ - SiC를 출발원료로 사용한 경우 고온가압소결 후 강한 집합조직을 보였으며, 이 경우 ${\beta} {\rightarrow} {\alpha}$ 상변태에 기인한 이중미세구조를 나타내었다. 또한, 집합조직의 강도는 고온가압소결 후 행해진 열처리 조건에 따라서도 변화를 보였다.
