가열처리 및 저장조건에 따른 당근과 시금치퓨레의 Carotenoids 함량변화와 이성질화 형성에 관한 연구 (Changes in Carotenoids Contents in Pureed and Cooked Carrot and Spinach during Storage)

  • Investigations were conducted on the changes in carotenoids content, and quantification of cis-trans-${\beta}$-carotene Isomers in pureed and cooked carrot and spinach during storage. The isomerization and degradation of carotenoids were monitored by high-performance liquid chromatography on a C$\_$30/ reversed-phase column with diode-array detection. The results showed that lutein, ail-trans-${\beta}$-carotene, ${\alpha}$-carotene, 9-cis-${\beta}$-carotene and 13-cis-${\beta}$-carotene were present in carrot and spinach. Zeaxanthin and cryptoxanthin were present in raw spinach. The contents of lutein, zeaxanthin, cryptoxanthin, ${\alpha}$-carotene and all-trans-${\beta}$-carotene in pureed and cooked carrot and spinach decreased with increasing storage period. The 9-cis and 13-cis carotenoid isomers were the major types formed in cooked carrot during storage. Cooking was not found to alter the carotenoid profile of the sample, but increased the total amount of carotenoids compared with pured ones. This increase could be explained that cooking itself increased the extraction efficiency and inactivated the enzymes degradating carotenoids.

취나물류의 Vitamin 및 일반성분 함량분석 (Analysis of Vitamins and General Components in the Leaves of Chwinamul)

  • To produce several high-quality species of Chwinamul (Aster scaber, A. Tataricus, Ligularia fischeri), leaves of the plants grown in different natural habitats were anlayzed. Contents in vitamin C and $\beta$-carotene were generally higher in plants grown in the field. However, Aster scaber picked form Yongwol and A. tataricus from Mt.Taryong contained relatively higher vitamin c and $\beta$ carotene. Crude protein and fat contents varied depending on the growing area. Ca content was markedly higher in the plants grown in the natural havitats.

Antioxidant and Antiaging Assays of Hibiscus sabdariffa Extract and Its Compounds

  • Skin aging is a complex biological process due to intrinsic and extrinsic factors. Free radical oxidative is one of extrinsic factors that induce activation of collagenase, elastase and hyaluronidase. Natural product from plants has been used as antioxidant and antiaging. This study aimed to evaluate antioxidant and antiaging properties of Hibiscus sabdariffa extract (HSE) and its compounds including myricetin, ascorbic acid, and ${\beta}$ carotene. The phytochemical of H. sabdariffa was determined using modified Farnsworth method and presence of phenols, flavonoids and tannins were in moderate content, whereas triterpenoids and alkaloids were in low content. Total phenolic content performed using Folin-Ciocalteu method, was $23.85{\mu}gGAE/mg$. Quantitative analysis of myricetin, ${\beta}-carotene$, and ascorbic acid of HSE was performed with Ultra-High Performance Liquid Chromatography (UHPLC) that shows $78.23{\mu}g/mg$ myricetin, $0.034{\mu}g/mg$ ${\beta}-carotene$, whilst ascorbic acid was not detected. HSE has lower activity on DPPH ($IC_{50}=195.73{\mu}g/mL$) compared to ${\beta}-carotene$, the lowest in ABTS assay ($IC_{50}=74.58{\mu}g/mL$) and low activity in FRAP assay ($46.24{\mu}MFe(II)/{\mu}g\;$) compared to myricetin, ${\beta}-carotene$. Antiaging was measured through inhibitory activity of collagenase, elastase, and hyaluronidase. HSE had weakest collagenase inhibitory activity ($IC_{50}=750.33{\mu}g/mL$), elastase inhibitory activity ($103.83{\mu}g/mL$), hyaluronidase inhibitory activity ($IC_{50}=619.43{\mu}g/mL$) compared to myricetin, ${\beta}-carotene$, and ascorbic acid. HSE contain higher myricetin compared to ${\beta}-carotene$. HSE has moderate antioxidants and lowest antiaging activities. Myricetin is the most active both antioxidant and antiaging activities.

당근 분말을 첨가한 막걸리의 품질 특성 (Quality Characteristics of Makgeolli added with Carrot Powder)

  • The purpose of this study was to investigate the physicochemical and sensory characteristics and overall preference for makgeolli after conducting fermentation using carrots with various levels of beta carotene and functional materials. The beta - carotene content was highest in 16% carrot powder, and steadily increased as the amount of carrot powder increased. Alcohol content increased with increasing amount of carrot powder. Alcohol content which greatly influences the quality of traditional wine, changes depending on the water soluble solid content and free sugar content during the manufacture of traditional wine. Sensory tests showed that the highest results were in the 12% carrot powder added group. As a result, the content of beta carotene increased with the addition of carrot powder, but the fermentation time was delayed and the sugar content was decreased because of variations in sugar content and color. Therefore, the optimal addition of carrot powder was 12%.

국내산 당근 품종별 carotenoid 함량 분석 (Quantitative Analysis of Carotenoids in Carrot Cultivars Produced in Korea)

  • Carrots (Daucus carota L.) are consumed as an important dietary source of provitamin A including $\beta$-carotene, $\alpha$-carotene and lutein. An HPLC method was applied to determine the content of the carotenoid composition in carrot cutivars cultivated in Korea. HPLC analyses were carried out with five carrot cultivars (Socheon-5-chon, Hongsim-5-chon, Myeongju-5-chon, Seonhongbom-5-chon and Betarich) sown at April, 2007 and six cultivars (Yeoreum-5-chon, Hanyeoreum-5-chon, Sinheukjeon-5-chon, Bibariheukjeon, Manina and Betarich) sown at August of the same year. In general, the former varieties are not used for the sowing at summer because of their bolting (growth of floral axis). The former and the latter carrots were harvested after 110 and 96 days from seeding, respectively, and the carotenoids were extracted with acetone after freeze-drying. The amount of $\alpha$-carotene ($117.7{\sim}205.3\;{\mu}g/g{\cdot}DW$) was similar to that of $\beta$-carotene ($113.1{\sim}189.6\;{\mu}g/g{\cdot}DW$) for the carrot cultivars sown at spring, while the content ($46.2{\sim}71.1\;{\mu}g/g{\cdot}DW$) was about a half of $\beta$-carotene content ($92.5{\sim}140.2\;{\mu}g/g{\cdot}DW$) for the latter cultivars. In addition, the average content of lutein ($25.2\;{\mu}g/g{\cdot}DW$) in the former cultivars was eight times higher than that in the latter cultivars ($3.1\;{\mu}g/g{\cdot}DW$). Among the spring cultivation types, Socheon-5-chon and Myeongju-5-chon showed higher amount of $\alpha$-carotene and $\beta$-carotene, while the higher amount was determined in Yeoreum-5-chon and Sinheukjeon-5-chon among the autumn cultivation types. Validation of the HPLC-DAD method showed good linearity ($r^2$ > 0.997) of the three compounds analyzed in a wide concentration range ($0.025{\sim}20\;{\mu}g/ml$). The R.S.D. values for intra-day and inter-day precision were less than 19.2% and the mean recovery of each compound was 85.4~104.7%.

Effect of Dietary Genetically Modified ${\beta}$-Carotene Biofortified Rice on Immune in Rats

  • This study aims to examine the effect of Genetically Modified ${\beta}$-Carotene Biofortified Rice rice developed by simultaneous expression technology in NAAS on biological immunity. Accordingly, this study added Genetically Modified ${\beta}$-Carotene Biofortified Rice 25, 50% and general rice 50% as control group into diet and provided rats with the prescribed feeds and then measured the contents of immunoglobulin and cytokine in blood. As a result, male and female IgM, IgE, male IgG1, female IgG2a and TNF-a, IL5 and IL12 showed no significant difference; male IgG2a tended to decrease dependently on the combined concentration of Genetically Modified ${\beta}$-Carotene Biofortified Rice; female IgG1 showed significance with control group, but its association with diet was not found. The higher the dietary mixing ratio, the more the male and female IFN-a and female IL-4 contents, regardless of rice variety, and it was found that female IL6 content decreased significantly, but its association with diet was not found. The risk of beta carotene-enriched rice into environment and human body has not been reported yet. The digestion of Genetically Modified ${\beta}$-Carotene Biofortified Rice can be seen as "safe" as this test result showed no big difference between general rice and Genetically Modified ${\beta}$-Carotene Biofortified Rice, and its usability is full of suggestions.

방사선 조사에 의한 감귤의 β-Carotene, 멸치의 비타민 D3 및 쇠고기의 α-Tocopherol의 함량변화 (Effects of Gamma Irradiation on the Content of β-Carotene in Jeju Orange, Vitamin D3 in Anchovy and α-Tocopherol in Beef)

  • 방사선 조사식품의 지용성 비타민에 대한 안정성을 확인하기 위하여 지용성 비타민 중 $\beta-carotene$, 비타민 $D_3$, $\alpha-tocopherol$을 택하여 한국인의 상용 식품이면서 각각의 급원식품인 귤, 멸치, 쇠고기에 대해 방사선조사 전후의 해당 비타민 함량을 분석하였다. 귤의 $\beta-carotene$의 함량은 조사구가 비조사구에 비해 약간 증가하는 경향을 보였으나 유의적이지는 않았다. 멸치의 비타민 $D_3$ 함량은 조사선량에 비례하여 5kGy 조사 시 $109.30\%$, 10kGy 조사 시 $125.25\%$로 증가하였다. (p<0.05). 소고기의 $\alpha-tocopherol$ 함량은 고지방 부위인 등심육의 경우 조사선량에 따라 큰 폭으로 감소되어 비조사구에 비해 10kGy 조사구에서 $84\%$의 감소율을 보였으나, 지방함량이 비교적 적은 사태육의 경우 조사구 모두에서 비조사구 대비 약 $13\%$의 감소율을 보였다. 순수 용액의 비타민 A, 비타민 $D_3,\;\alpha-tocopherol$은 조사선량에 비례하여 함량이 감소하는 것으로 나타났으며 특히, 비타민 $D_3$는 지용성 비타민 중 조사에 가장 안정한 비타민으로 나타났고, $\alpha-tocopherol$의 경우는 10kGy 처리구에서 전량 감소를 보여 지용성 비타민 중 조사에 가장 취약한 비타민으로 나타났다. 마이크로웨이브 처리구나 가열 처리구에서는 이들 지용성 비타민들의 손실이 거의 없었다. 이사의 결과로부터 10kGy이내의 방사선 조사 시, 귤의 $\beta-carotene$ 함량에는 영향을 미치는 않는 것으로 보이며, 비타민 $D_3$의 경우는 적어도 10kGy까지는 매우 안정함을 보여주었고 지방함량이 높은 등심육의 경우 $\alpha-tocopherol$의 상당한 분해가 일어남을 알 수 있었다.

미세조류 Dunaliella bardawil에서 오존과 과산화수소에 의한 ${\beta}$-carotene의 축적과 식용기름을 이용한 추출 (Induction of ${\beta}$-carotene by Ozone and Hydrogen Peroxide and Extraction Using Vegetable Oil from Microalga Dunaliella bardawil)

  • Dunaliella bardawil의 적절한 생장과${\beta}$-carotene의 효과적인 축적 조건을 확립하기 위하여 염 농도, pH, 광량, 교반속도, 오존처리, 과산화수소 첨가 등의 영향을 비교하였고, 식용 식물성 기름을 이용한 ${\beta}$-carotene의 추출조건을 비교하였다. D. bardawil의 비생장 속도는 1N NaCl, pH 8.0 광량 80${\mu}$$E/\m^2/s$ , 70rpm 으로 배양 시에 0.168/hr로 최대를 나타냈다. 흰색 형광등을 250${\mu}$$E/\m^2/s$ 로 조사한 경우 ${\beta}$-carotene isomer 축적은 cis isomer가 trans isomer에 비해 높은 증가율을 보였다. ${\beta}$-carotene의 축적을 위하여 50${\mu}$M 과산화수소를 배양 2일 경과 후에 첨가한 경우 대조군의 축적율이 150${\mu}$g/ml인데 비해 324${\mu}$g/ml로, 200ppb로 2시간 오존 처리했을 때 약 1.8배 증가한 270${\mu}$g/ml이었다. 식용기름에 의한 37${\circ}C$에서의 추출효율은 올리브유, 참기름, 미강유, 옥수수유, 대두유의 순서로 나타났으며, 올리브유를 수층 부피의 9배로 첨가하고 50${\circ}C$에서 1시간 동안 sonication한 결과 96.95의 추출효율을 나타내었다.

Tocopherol and Carotenoid Contents of Selected Korean Cooked Combination Foods Consumed by Young Korean Children

  • To more accurately estimate vitamin A and vitamin E intake of Koreans, food composition databases of the tocopherol and carotenoid contents of Korean foods are needed. In this study, the tocopherol $(\alpha-,\;\gamma-,\;and\;\delta-)$ and carotenoid ($\alpha-carotene,\;\beta-carotene,\;\beta-ctyptoxanthin$, lutein, and zeaxanthin) contents of 12 Korean cooked combination foods commonly consumed by children in Kwangju, Republic of Korea, were determined using reversed-phase HPLC. All samples were obtained from 3 different households in Kwangju during summer, 2005. All cooked foods in this study had detectable quantities of $\alpha-tocopherol,\;\gamma-tocopherol$ except for shoegogimugook, and $\delta-tocopherol$ except for myulchibokkeum Doejigogibokkeum had the highest $\alpha-tocopherol$ content (0.64 mg/l00 g edible portion), and $\gamma-tocopherol\;and\;\delta-tocopherol$ contents of gimbab were the highest among the foods (1.01 and 0.26 mg/l00 g edible portion). $\beta-carotene$ was found in all food samples. Gimbab had the highest contents of $\alpha-carotene,\;\beta-carotene$, and lutein among Korean cooked combination foods (158.3, 266.6, and $375.4{\mu}g/100g$ edible portion). Miyukgook contained only $\beta-carotene$ ($2.5{\mu}g/100g$ edible portion). Considerable sample-to-sample variability in tocopherol and carotenoid compositions were observed in several Korean cooked combination foods included in this study. Some of these carotenoids $(\alpha-carotene,\;\beta-carotene,\;and\;\beta-cryptoxanthin)$ are vitamin A precursors. The findings of this study may be valuable for use in Korean databases as well as nutrient consumption research for vitamin A and vitamin E.

β-Carotene, Cucumisin Content and Fruit Morphology of Melon (Cucumis melo L.) Germplasm Collections

  • Melon fruits are a year-old plant in the family Cucurbitae and are the most cultivated fruit in tropical countries. Melon flesh is an important source of anti-cancer, antioxidant effects, attracting attention as a functional food. We investigated the morphological properties, β-carotene content, and cucumisin activity of seeds of 58 melon genetic resources. Melon resources have shown various morphological properties. Melons grouped by morphological properties were the five groups. β-carotene content varied between 82.34 mg/kg, 86.75 mg/kg, 25.56 mg/kg, 86.25 mg/kg, and 54.65 mg/kg. Between β-carotene, cucumisin activity and other quantitative fruit morphological properties, the color of the pulp and the firmness of the pulp had a significant amount of correlation between the β-carotene content. However, cucumisin activity and β-carotene content had significant negative correlation, and the color of the fruit and shape of the fruit had significant negative correlation with β-carotene content and activity of cucumisin. Our study vindicated that high diversity in melon morphological characters of genetic sources that provide beneficial baseline data for the future and in the activity of β-carotene and cucumisin, and these results could predict indicators of β-carotene content by the length of leaf, the width of fruit, the length of joint, and the length of seed, and collectively the morphological properties of melons could help predict indicators of β-carotene content and help develop functional sarcoma and farmhouse cultivation.
