Bacillus sp. JB-99, when grown in a chemically defined medium containing lactose as a carbon source, yielded 3,860 U/ml extracellular $\beta$-mannanase, which was high compared to other examined carbon sources. Among the nitrogen sources, yeast extract enhanced the enzyme activity. The enzyme production was growth-associated. The enzyme was optimally active at $65^{\circ}C$, pH 10, and had a half-life of 190 min at $65^{\circ}C$. N-Bromosuccinamide and $AgNO_3,\;CuSO_4$, and $HgCl_2$ strongly inhibited the enzyme, whereas $Ca^{2+}$ stimulated the enzyme activity. The $\alpha$-galactosidase enzyme production was not found in any of the enzyme assays.
Objective: In this study, two glycosidases (XMosidases), ${\beta}$-xylosidase and ${\beta}$-mannosidase, were investigated on their in vitro hydrolysis activities of feed and on the improvement of growth performance in vivo in weanling pigs. Methods: Enzyme activities of XMosidases in vitro were evaluated in test tubes and simulation of gastric and small intestinal digestion, respectively, in the presence of NSPase. In vivo study was performed in 108 weaned piglets in a 28-d treatment. Pigs were allotted to one of three dietary treatments with six replicate pens in each treatment. The three treatment groups were as follows: i) Control (basal diet); ii) CE (basal diets+CE); iii) CE-Xmosidases (basal diets+ CE+${\beta}$-xylosidase at 800 U/kg and ${\beta}$-mannosidase at 40 U/kg). CE was complex enzymes (amylase, protease, xylanase, and mannanase). Results: In vitro XMosidases displayed significant activities on hydrolysis of corn and soybean meal in the presence of non-starch polysaccharide degrading enzymes (xylanase and ${\beta}$-mannanase). In vitro simulation of gastric and small intestinal digestion by XMosidases showed XMosidases achieved $67.89%{\pm}0.22%$ of dry matter digestibility and $63.12%{\pm}0.21%$ of energy digestibility at $40^{\circ}C$ for 5 hrs. In weanling pigs, additional XMosidases to CE in feed improved average daily gain, feed conversion rate (p<0.05), and apparent total tract digestibility of crude protein (p = 0.01) and dry matter (p = 0.02). XMosidases also altered the gut bacterial diversity and composition by increasing the proportion of beneficial bacteria. Conclusion: Addition of a complex enzyme supplementation (contained xylanase, ${\beta}$-mannanase, protease and amylase), XMosidases (${\beta}$-xylosidase and ${\beta}$-mannosidase) can further improve the growth performance and nutrient digestion of young pigs.

B. longum, B. bifidum, B. adolescentis, B. animalis, B. auglutum, B. breve의 생육활성에 대한 중합도 4와 6의 영향을 검토하기 위하여 modified-MRS배지 상에 탄소원으로 중합도 4와 6를 대체하여 생육활성을 비교한 결과 B. longum에서는 D.P 6 galactomannooligosaccharide를 탄소원으로 대체한 경우 표준 MRS배지와 비교하여 10배의, D.P. 4을 처리한 경우에도 7.6배의 상대 활성을 나타내어 가장 우수한 생육활성을 나타냈었으며, B. bifidum의 경우에서도 D.P 6에서 6.6배, D.P 4에서 4.8배의 활성을 나타내었으며 B. animalis에 있어서는 D.P. 6의 경우 3.8배의 상대활성을 나타내었다. Bifidobacterium 7균주 모두에 대해서 중합도 6의 올리고당이 중합도 4의 올리고당보다 생육활성에 크게 기여하는 것으로 나타났다. 신규 MRS 배지의 제품화와 관련하여 가장 유리한 탄소원 대체소재는 Bacillus sp. 유래 galactosyl mannooligosaccharide 인 ${Gal^3}{M_4}$가 가장 유리한 소재로 결정되어 신규 배지의 scale-up 공정을 구축중에 있다.
DEAE-sephadex ion exchange column chromatography에 의해 Bacillus sp. 유래 h-mannanase의 정제를 수행하여 비활성은 21.57 units/mg, 정제배율은 93.78배를 나타내었다. 최적 온도는 5$0^{\circ}C$, 최적 pH는 6.0이며, 온도 안정성에서는 30∼5$0^{\circ}C$에서는 90%이상의 잔존활성을 나타내었고,70∼8$0^{\circ}C$에서는 30%이하의 잔존활성을 나타내었다. pH 안정성에서는 pH 5.5∼7.0에서 100%의 잔존활성을 나타낸 반면 pH 2.0∼4.0에서는 40%이하로 감소되 었다. 정제효소에 의해 konjac glucomannan을 가수분해하여 1차 activated carbon column chromatography와 2차 sephadex G-25 gel filtration 에 의해 당가수분해물을 분리 회수하여 TLC및 FACE에 의해 주요 당가수분해물은 중합도 5와 7로 확인되었다. B. iongum, B. btfidum, B. infantis, E. adolescentis, B. animalis, B. breve의 생육활성에 대한 중합도5와7의 영향을 검토하기 위하여 modified-MRS배지상에 탄소원으로 중합도 5와 7을 대체하여 생육활성을 비교한 결과 B. longum의 경우 특징적으로 각각 4.67, 5.33배의 상대활성을 나타내어 우수한 생육활성을 나타내었다. 또한 B. breve에 대해서는 중합도 5 glucomannooligosaccharide를 처리시 2.42배의 생육활성을 나타내었으나 B. infantis와 B. adolescentis에 대해서는 중합도 5와 7의 올리고당을 탄소원으로 대체시 오히려 생육활성이 현저히 감소되었다.
A mannanase gene (man26B) was obtained from a sea bacterium, Paenibacillus sp. BME-14, through the constructed genomic library and inverse PCR. The gene of man26B had an open reading frame of 1,428 bp that encoded a peptide of 475- amino acid residues with a calculated molecular mass of 53 kDa. Man26B possessed two domains, a carbohydrate binding module (CBM) belonging to family 6 and a family 26 catalytic domain (CD) of glycosyl hydrolases, which showed the highest homology to Cel44C of P. polymyxa (60% identity). The optimum pH and temperature for enzymatic activity of Man26B were 4.5 and $60^{\circ}C$, respectively. The activity of Man26B was not affected by $Mg^{2+}$ and $Co^{2+}$, but was inhibited by $Hg^{2+},\;Ca^{2+},\;Cu^{2+},\;Mn^{2+},\;K^+,\;Na^+$, and $\beta$-mercaptoethanol, and slightly enhanced by $Pb^{2+}$ and $Zn^{2+}$. EDTA did not affect the activity of Man26B, which indicates that it does not require divalent ions to function. Man26B showed a high specific activity for LBG and konjac glucomannan, with $K_m,\;V_{max}$, and $k_{cat}$ values of 3.80 mg/ml, 91.70 ${\mu}mol$/min/mg protein, and 77.08/s, respectively, being observed when LBG was the substrate. Furthermore, deletion of the CBM6 domain increased the enzyme stability while enabling it to retain 80% and 60% of its initial activity after treatment at $80^{\circ}C$ and $90^{\circ}C$ for 30 min, respectively. This finding will be useful in industrial applications of Man26B, because of the harsh circumstances associated with such processes.

DP 7의 galactosyl mannooligosaccharide와의 각 반응혼합물의 갈변도는 urea의 반응액이 강한 착색을 보였으며 phenylthiourea, thiourea의 반응액의 순으로 착색를 나타내었다. 반응혼합물의 TLC 결과에서 ${\beta}-1,4-mannobiose$와 urea의 반응혼합물 이외에 phenylthiourea, thiourea의 반응액은 새로운 화합물이 생성되었고, $Gal^3Man_4$와 D.P 7 galactosyl manooligosaccharide와의 반응혼합물에서는 모든 반응액에서 새로운 화합물이 출현되었다. 반응혼합물의 환원력은 urea관련화합물 중에서 특히 phenylthiourea, thiourea가 ${\beta}-1,4-mannobiose,\;Gal^3Man_4$, DP 7의 galactosyl manooligosaccharide와의 반응혼합물에서 강한 환원력이 나타났다. Linoleic acid에 대한 반응혼합물의 항산화력 측정에서 ${\beta}-1,4-mannobiose$와 thiourea의 반응혼합물의 항산화력이 Ascorbic acid(AsA)수준의 항산화력을 나타내었고, $Gal^3Man_4$와 thiourea, phenylthiourea의 반응혼합물은 AsA와 유사한 항산화능을 나타내었다. DP 7의 galactosyl manooligosaccharide와 thiourea, phenylthiourea의 반응혼합물의 항산화력은 AsA에 미치지는 못하였다. ${\beta}-1,4-Mannobiose$와 thiourea, phenylthiourea의 반응혼합물이 강한 전자공여능을 나타내고 있고, $Gal^3Man_4$와 phenylthiourea의 반응혼합물 및 D.P 7의 galactosyl manooligosaccharide와 phenylthiourea와의 반응혼합물이 전자공여능을 나타내고 있다.

Anaerobic rumen fungi have been regarded as good genetic resources for enzyme production which might be useful for feed supplements, bio-energy production, bio-remediation and other industrial purposes. In this study, an expressed sequence tag (EST) library of the rumen anaerobic fungus Neocallimastix frontalis was constructed and functional genes from the EST library were analyzed to elucidate carbohydrate metabolism of anaerobic fungi. From 10,080 acquired clones, 9,569 clones with average size of 628 bp were selected for analysis. After the assembling process, 1,410 contigs were assembled and 1,369 sequences remained as singletons. 1,192 sequences were matched with proteins in the public data base with known function and 693 of them were matched with proteins isolated from fungi. One hundred and fifty four sequences were classified as genes related with biological process and 328 sequences were classified as genes related with cellular components. Most of the enzymes in the pathway of glucose metabolism were successfully isolated via construction of 10,080 ESTs. Four kinds of hemi-cellulase were isolated such as mannanase, xylose isomerase, xylan esterase, and xylanase. Five $\beta$-glucosidases with at least three different conserved domain structures were isolated. Ten cellulases with at least five different conserved domain structures were isolated. This is the first solid data supporting the expression of a multiple enzyme system in the fungus N. frontalis for polysaccharide hydrolysis.
The experiment was conducted to test the hypothesis that supplementing diets of lactating first parity sows with a mixture of carbohydrases (CS) improves lactation performance and second parity reproductive performance. The CS used in this study contained 7 units/g of $\alpha$-1,6-galactosidase, 22 units/g of $\beta$-1,4-mannanase, $\beta$-1,4-mannosidase and trace amounts of other enzymes. Twenty primiparous sows (Newsham Hybrid) were allotted to either the control group (no CS supplement) or the CS group (0.1% CS supplement) and fed the experimental diets during 21 d lactation period. Sows and nursing pigs were weighed at birth and weekly until weaning. Days of weaning-to-estrus were recorded. Sows had free access to feed and water. Feed intake of sows was measured daily. During the second parity gestation and lactation, all the sows were fed the same gestation and lactation diets and their reproductive performance was measured. During the second parity, there were 14 sows (7 sows per group) remained productive. For the first lactation, maternal body weight loss of the CS group was smaller (p<0.05) than that of the control group. There was no difference in litter weight gain between two groups. Voluntary feed intake of sows did not differ between the two groups. Days of weaning-to-estrus of the CS group were smaller (p<0.05) than those of the control group. In the second parity, there was no difference in the reproductive performance between the two groups. In conclusion, supplementing CS in the diet of lactating sows during the first parity decreased body weight loss and days of weaning-to-estrus of sows. However, these effects of the CS supplementation in the first parity were not successfully carried over to the second parity.
This study was designed to investigate the effects of multi-enzyme on diarrhea and immune responses of weaned pigs. A total 36 weaned pigs ($5.92{\pm}0.48kg\;BW$; 28 d old) were randomly allotted to 2 dietary treatments (3 pigs/pen, 6 replicates/treatment) in a randomized complete block design. The dietary treatments were a typical diet based on corn and soybean meal (CON) and CON with 0.1% multienzyme (Multi; mixture of ${\beta}-mannanase$, xylanase, ${\alpha}-amylase$, protease, ${\beta}-glucanase$, and pectinase). Pigs were fed their respective diets for 6 wk. Frequency of diarrhea, levels of packed cell volume (PCV), white blood cells (WBC), immunoglobulins, cortisol, tumor necrosis $factor-{\alpha}$ ($TNF-{\alpha}$), transforming growth $factor-{\beta}$ ($TGF-{\beta}$), and C-reactive protein (CRP) were measured. Multi group tended to decrease (p<0.1) diarrhea frequency than CON group during 2 wk after weaning. Lower values of PCV on d 3 (p<0.05) and d 7 (p<0.1) were found in Multi group compared with CON group. There were no significant differences on WBC number and immunoglobulin (Ig) M and A between Multi and CON groups. However, Multi group tended to increase (p<0.1) Ig G on d 7 than CON group. Moreover, Multi group showed modulated immune responses, indicated by decreased levels of cortisol (p<0.05) on d 7 and 14, $TNF-{\alpha}$ on d 3 (p<0.05) and d 7 (p<0.10), $TGF-{\beta}$ on d 2 (p<0.05) and d 7 (p<0.10), and CRP (p<0.10) on d 3 and 7 after weaning compared with CON group. Consequently, inclusion of multi-enzyme in diets for weaned pigs improved gut health and modulated immune responses of weaned pigs.
