홀스타인 젖소의 원유내 acetone과 β-hydroxybutyrate acid 함량에 영향을 미치는 환경요인 (Environmental factors influencing acetone and Environmental factors influencing acetone and β-hydroxybutyrate acid contents in raw milk of Holstein dairy cattle)

  • 본 연구는 젖소의 검정기록, 378,086개를 이용하여 케토시스 진단 지시형질로 이용되는 유량내 아세톤과 ${\beta}$-히드록시부틸산의 함량에 영향을 미치는 환경요인을 분석하였다. 형질별 목장, 비유단계, 산차, 착유시간 및 월령에 대한 유의성 검정 결과 모든 요인들에서 고도의 유의성 (p < 0.01)이 나타났으며, 가장 큰 환경요인은 비유단계였다. 1회 착유량, 아세톤과 ${\beta}$-히드록시부틸산의 함량에 대한 월령의 1차 회귀계수는 모두 정 (+)의 관계, 2차 회귀계수는 부 (-)의 관계를 나타내었으며, 비유 2단계 (36~65일) 비유량의 최소자승평균은 19.06kg으로 비유말기에 비하여 6.51kg 높았다. 아세톤과 ${\beta}$-히드록시부틸산의 함량에 대한 최소자승평균은 비유 1단계 (5~35일)에서 각각 0.1929mM/L과 0.0742mM/L로 가장 높았으며, 착유일수에 따라 감소하다가 비유말기 소폭 증가하는 경향을 나타내었다. 1산에 대한 아세톤과 ${\beta}$-히드록시부틸산의 함량의 최소자승평균은 각각 0.1414mM/L과 0.0522mM/L로 2차산 이상의 평균보다 높았으며, 오후착유에 대한 아세톤과 ${\beta}$-히드록시부틸산의 함량의 최소자승평균이 각각 0.1372mM/L과 0.0534mM/L로 오전착유보다 일괄적으로 높게 나타났다.

  • Fed-batch fermentation was used to produce the high concentrations of poly-$\beta $-hydroxybutyrate (PHB) and poly-$\beta $-(hydroxybutyrate-co-hydroxyvalerate) (PHB/V). Specific growth rate ($\mu $), yield of cell from glucose (Y$_{x/s}$) were calculated from the two samples in 3 to 5 hours of interval and they were reflected on the determination of glucose feeding rate to maintain the glucose concentration at around 10 g/l in the culture broth. PHB was accumulated after the nitrogen became limited at 60 g/l of dry cell weight by changing ammonia water to 4N-NaOH solution. As results, the final dry cell weight (DCW) of 170 g/l, PHB of 115 g/l were obtained in 50 hours and the overall productivity was 2.4 g/l$\cdot $h. After PHB accumulation, cosubstrate of glucose and propionic acid (PA) was fed to accumulate PHB/V. But, PA feeding rate was decreased from 3 g/l$\cdot $h to 1 g/l$\cdot $h to prevent PA from accumulating to high level in the broth, which is very inhibitory to the cells. As results, DCW, PHB and PHV were 147.5 g/l, 90 g/l and 8 mole % of hydroxyvalerate, respectively.

Production of Poly($\beta$-hydroxybutyrate-co-$\beta$-hydroxyvalerate) by Two-stage Fed-batch Fermentation of Alcaligenes eutrophus

  • Production of poly($\beta$-hydroxybutyrate-co-$\beta$-hydroxyvalerate)[poly(HB-co-HV) from glucose and propionic acid was studied in a two-stage fed-batch fermentation using Alcaligenes eutrophus NCIMB 11599. When either glucose became sufficient or the feeding rate of propionic acid decreased, production of poly(HB-co-HV) increased but concomitantly resulted in a reduced fraction of HV. During the copolymer accumulation stage, the specific production rate of hydroxyvalerate (HV) increased up to 0.013 (g-HV/g-RCM/h) but it decreased as propionic acid was accumulated. Control of the propionic acid concentration in the medium, therefore, is considered to be one of the most important operating parameters for production of poly(HB-co-HV) with a higher HV fraction. A high titre of poly(HB-co-HV) (85.6 g/I) with HV fraction of 11.4 mol$%$ could be obtained in 50 h by controlling the propionic acid concentration at 1 to 4 g/I.

  • A transposon Tn5 mutant of Rhodobacter sphaeroides 2.4.1 was isolated for its impaired ability of growth on minimal medium containing ${\beta}$-hydroxybutyric acid as a sole carbon source. The mutant, R. sphaeroides S7 showed approximately 6-fold decrease in ${\beta}$-hydroxybutyrate dehydrogenase activity compared with that of wild type. In R. sphaeroides S7 the Tn5 was located in DNA region corresponding to a 4.2-kb EcoRI DNA fragment of R. sphaeroides 2.4.1 chromosome.

  • The biodegradable characteristics of poly-$\beta$-hydroxybutyrate(PHB) film by fun맥 and soil burial are Investigated. As the results of the American Standards for Testing and Materials(ASTM) method, the you of Aspergillus niger was apparent on the PHB containing plate. This suggests that PHB was utilized as the sole carbon source by Aspergillus niger and ASTM method may have applications as measuring means of biome gradability of polyhydroxyalkanoic acid(PHA). PHB film was studied by monitoring the time-dependant changes in weight loss of PHB film under 30% and relative humidity 80 % during pot-test. As the results of pot-test, PHB film was decomposed about 87 % in 30 days by soul microorganisms. PHB film was more slowly degraded than PHB/HV film.

Effect of the Supplement of Metabolites on Cell Growth and Poly-$\beta$-hydroxybutyrate Biosynthesis of Alcaligenes latus

  • The characteristics of cell growth and poly-$\beta$-hydroxybutyrate biosynthesis of Alcaligenes latus ATCC 29713 were investigated. The PHB accumulation pattern of A. latus followed a growth-associated type where the cell growth and PHB accumulation were carried out simultaneously. Various intermediate compounds such as metabolites involved in the TCA cycle, amino acids, and saturated and unsaturated fatty acids were added to examine their effect on cell growth and PHB accumulation. Citrate, tyrosine, and palmitic acid showed the most significant increase both on cell growth and PHB accumulation. Maximum PHB concentrations were noticeably increased about 1.4 to 1.6 times higher than that of control, corresponding to 5.54, 6.45, and 6.45 g/l for citrate, tyrosine, and palmitic acid, respectively. The stimulatory effects of the supplemented metabolites were analyzed in terms of the increment of enzyme activities related to sugar catabolism and PHB biosynthesis.

분홍색 통성 메탄올 자화세균이 생산하는 Poly-$\beta$-Hydroxybutyrate (Poly-$\beta$-Hydroxybutyrate Produced by Pink-Pigmented Facultative Methylotrophic Bacterium from Methanol)

  • PHB 생산을 위하여 메탄올을 기질로 한 선별배지에서 토양, 하천수, 퇴비 등으로부터 분홍색 색소를 가지는 PHB 축적 facultative methylotroph를 분리하여, 균주의 특성을 검토하였다. 분리균주의 최적 생육조건과 PHB 축적을 위한 배양조건을 조사한 결과 균체의 생육은 메탄올 농도 1.0(v/v), 질소원인$ NH_4C$ 농도 1.0g/l, 즉 C/N ratio 13.2 일때 그리고 pH 7.0과'$30^{\circ}C$에서 가장 좋았으며, PHB는 C/N ratio가 50.8, 즉 메탄올 농도 1.0(v/v )$NH_4CL$ 0.26g/l 일때, 그리고 pH 6.0일 때 건조중량의 약 40까지 축적되었다. 고농도 메탄올에 의한 생육저해를 극복하기 위하여 기질을 간헐적으로 계속 공급해주는 fed-batch 배양을 시도한 결과 균체량은 14g/l, PHB 축척량은 5.5g/l까지 증가시킬 수 있었다. 생산된 PHB를 분리.정제하여 IR과 $^I H-NMR$로 구조를 분석한 결과 3-hydroxybutyric acid 의 homopolymer임이 확인되었다. 또한 균주의 pink-pigment를 추출하여 absorption spectrum를 조사하여 그 특성을 규명하였다.

균형 생육조건하에서 Actinobacillus sp. EL-9에 의한 Poly-$\beta$-Hydroxybutyrate의 생산

  • Microorganisms growing alcoholic distillery wastes were isolated from soil. Of them, the selected strain EL-9 had a capability of accumulating poly-$\beta$-hydroxybutyrate (PHB) when grown in alcoholic distillery wastes. The strain EL-9 was identified as a genus Actinobacillus based on the morphological, cultural, and biochemical characteristics. The strain EL-9 was named temporarily as Actino- bacillus sp. EL-9. The optimal temperature and pH for cell growth of Actinobacillus sp. EL-9 were 30$\circ$C and 7.0, respectively. The optimal medium compositions for cell growth comprised glucose 2%, NH$_{4}$NO$_{3}$ 0.15%, Na$_{2}$HP0$_{4}$-12H$_{2}$O 0.25%, KH$_{2}$PO$_{4}$ 0.25%, MgSO$_{4}$4-7H$_{2}$O 0.15%, FeSO$_{4}$-7H$_{2}$O 0.005%, CaCl$_{2}$-2H$_{2}$O 0.003% and trace element solution 5m/l. To investigate the optimal conditions for PHB production, two-stage culture technique was used. The result showed that the optimal conditions for PHB production are identical those for cell growth. When propionic acid was added as a cosubstrate, PHB/HV copolymer was produced.

주정증류 폐액을 이용한 Actinobacillus sp. EL-9로부터 Poly-$\beta$-Hydroxybutyrate의 생산 및 폐약의 처리

  • Alcoholic distillery wastes are utilized as dual purposes to produce PHB in lower production cost and to reduce the amount of waste to be treated. In this study, various attempts were made to increase PHB production under various conditions by Actinobacillus sp. EL-9 in a shaker culture. The addition of glucose, NH$_{4}$NO$_{3}$ to alcoholic distillery wastes slightly promoted cell mass and PHB production. Enzyme hydrolysis of alcoholic distillery wastes increased the production of PHB than that of untreated waste and acid hydrolysis treatment. The PHB weight in alcoholic distillery wastes was 1.91 g/l. Fermentation process of PHB production reduced the amount of COD value up to 54%, which reduced organic loading rate and capacity of activated sludge system.

  • 케토시스 질병의 지표 형질인 milk ${\beta}$-hydroxybutyratc acid (BHBA), milk acetone, 그리고 사료효율의 지표형질 에너지보정유량 및 젖소의 주요 경제 형질인 산유량에 대한 유전력과 이들 간의 유전적 관계를 통하여 젖소의 케토시스 저항성 증진 가능성을 확인하기 위하여 본 연구를 수행하였다. 국내 젖소 14,397두에서 수집된 산유능력 검정자료 75,072건의 기록과 혈통기록 44,954두의 기록을 다형질 임의회귀 검정일 선형 모형을 이용하여 분석하였다. 케토시스 지표형질들의 유전력은 범위 0.06 ~ 0.15, 평균 0.08 (${\pm}0.03$)로 추정되었으며, 케토시스 지표형질들 간에 표현형 상관은 범위 0.73 ~ 0.90, 평균 0.78 (${\pm}0.04$), 유전 상관은 0.93 ~ 0.98 평균은 0.97 (${\pm}0.01$), milk BHBA, milk acetone과 산유량의 표현형 상관은 범위 -0.18 ~ -0.05 평균 -0.08 (${\pm}0.027$), 범위 -0.05 ~ -0.23, 평균 -0.1 (${\pm}0.041$), 유전상관은 -0.55 ~ 0.05, 평균 -0.16 (${\pm}0.18$), -0.62 ~ -0.04, 평균 -0.24 (${\pm}0.17$)로 각각 추정이 되었다. mill BHBA, milk acetone과 ECM의 표현형 상관관계는 0.056 (${\pm}0.023$), -0.04 (${\pm}0.029$), 유전적 관계는 평균 0.023 (${\pm}0.08$), 0.09 (${\pm}0.067$)로 각각 추정되었다. 따라서 산유능력 검정자료 수집 시 부가적으로 수집되는 케톤체를 이용하면 케토시스 저항성 증진을 할 수 있을 것으로 사료된다.
