검은비늘버섯 물 추출물 및 유기용매 분획물의 생리활성 효과 (Physiological Activities of Water Extract and Solvent Fractions of an Edible Mushroom, Pholiota adiposa)

  • 검은비늘버섯을 이용하여 심혈관계 질환과 관련된 기능성 식품 개발을 위한 기초자료를 얻기 위해 검은비늘버섯물 추출물과 유기용매 분획물을 이용하여 심혈관계 질환관련 생리활성을 확인하였다. 물 추출물은 57.57%의 항산화활성을 나타냈지만 혈전용해활성, 트롬빈저해활성, ${\alpha}$-glucosidase 저해효과는 매우 낮게 나타났다. 반면 에틸 아세테이트 분획물은 0.70 plasmin unit의 높은 혈전용해 활성과, 87.36%와 46.44%의 높은 트롬빈 저해활성과 ${\alpha}$-glucosidase 저해활성을 나타냈으며, 물 분획물은 48.27%의 항산화활성을 나타냈다. 혈전용해활성이 높은 에틸 아세테이트 분획물은 섬유소원의 $A{\alpha}$$B{\beta}$ chain은 모두 분해시켰지만 ${\gamma}$ chain과는 반응하지 않았으며, $100^{\circ}C$에서 10분간 가열에도 혈전용해 활성에 변화가 없었다. 실험 결과로부터 검은비늘버섯의 에틸아세테이트 분획물은 높은 혈전용해활성과 트롬빈 저해활성과 ${\alpha}$-glucosidase 저해활성을 나타내 심혈관계 질환 관련 치료나 예방을 위한 기능성식품 개발에 이용 가능할 것으로 기대된다.

β-Glucosidase 활성이 있는 균주 Bacillus Strains를 접종해 제조한 Soy Grits 청국장의 품질 특성 (Changes in Biological Qualities of Soy Grits Cheonggukjang by Fermentation with β-Glucosidase-Producing Bacillus Strains)

  • 본 연구는 ${\beta}$-glucosidase 활성이 있는 Bacillus subtilis HJ18-9와 HJ25-8, 두 가지 균주를 혼합한 HJ18-9+HJ25-8의 3가지 균주를 스타터로 접종하여 발효시킨 soy grits 청국장의 품질 특성과 isoflavone의 함량을 측정하였다. 환원당을 유리하는 데 관여하는 ${\alpha}$-amylase 효소 활성의 경우 두 가지 균주를 혼합한 HJ18-9+HJ25-8을 접종해 발효한 시료에서 다른 시료와 비교하였을 때 높은 활성을 보였다. 청국장의 단백질을 분해하여 특유의 구수한 맛 성분을 유리하는 protease 활성의 경우 HJ25-8, HJ18-9+HJ25-8, HJ18-9의 순으로 높은 활성을 나타내었다. 또한 아미노태 질소와 암모니아태질소의 함량은 HJ25-8을 접종해 발효한 SG 청국장에서 높았으며, 청국장 isoflavone 비배당체 함량은 HJ18-9+HJ25-8의 $272.40{\pm}2.04{\mu}g/g$에 비해 HJ18-9와 HJ25-8 접종구에서 $697.03{\pm}9.46$, $683.10{\pm}2.05{\mu}g/g$으로 높았다. ${\beta}$-glucosidase 활성이 있는 두 가지 균의 혼합으로 isoflavone aglycone 함량의 전환율을 더 높일 수 있는 시너지 효과를 기대했으나 단일 균주로 접종하여 발효했을 때 더 높은 aglycone 함량을 얻을 수 있었다. 본 연구를 통해 청국장 제조에 알맞은 균주를 개발 평가하여 청국장 가공품 개발의 기초연구가 되고자 하였다.

  • 버섯이 유산균 증식 효과를 측정한 결과 대부분의 버섯이 B. breve에 대한 증식 효과와 배양배지의 pH 저하 효과가 우수했으며 식용버섯 중에서는 표고버섯, 운지버섯, 영지버섯, 느타리버섯이 우수했다. 사람 및 흰쥐의 장내 세균을 버섯추출물 함유 배지에서 배양시 배지의 pH 저하 효과가 우수한 표고버섯, 운지버섯, 양송이버섯은 장내유산균을 선택적으로 증식시켰다. B. breve 배양시 버섯추출물의 첨가는 균의 성장을 촉진하였으며 ${\beta}-glucosidase$의 효소 활성을 억제하였으며, 이는 배양배지의 pH 저하 효과와 비례하였다. 표고버섯, 영지버섯, 운지버섯은 사람 및 흰쥐의 장내 세균총 효소인 ${\beta}-glucosidase$ 및 tryptophanase 효소 활성을 억제하였으며 pH 저하 효과와 비례하였다.

Evaluation of glucosidases of Aspergillus niger strain comparing with other glucosidases in transformation of ginsenoside Rb1 to ginsenosides Rg3

  • The transformation of ginsenoside Rb1 into a specific minor ginsenoside using Aspergillus niger KCCM 11239, as well as the identification of the transformed products and the pathway via thin layer chromatography and high performance liquid chromatography were evaluated to develop a new biologically active material. The conversion of ginsenoside Rb1 generated Rd, Rg3, Rh2, and compound K although the reaction rates were low due to the low concentration. In enzymatic conversion, all of the ginsenoside Rb1 was converted to ginsenoside Rd and ginsenoside Rg3 after 24 h of incubation. The crude enzyme (b-glucosidase) from A. niger KCCM 11239 hydrolyzed the ${\beta}$-($1{\rightarrow}6$)-glucosidic linkage at the C-20 of ginsenoside Rb1 to generate ginsenoside Rd and ginsenoside Rg3. Our experimental demonstration showing that A. niger KCCM 11239 produces the ginsenoside-hydrolyzing b-glucosidase reflects the feasibility of developing a specific bioconversion process to obtain active minor ginsenosides.

Production of Cellulase and Xylanase by Aspergillus niger KKS

  • A fungal strain capable of producing extracellular cellulase was isolated from farmland. It was identified as Aspergillus niger, and named Aspergillus niger KKS. Production of cellulase and xylanase by the A. niger KKS was studied through a shake-flask culture. The effects of culture conditions such as inoculum size, temperature, pH, and medium composition on the cellulase and xylanase production were examined. The optimum temperature and pH for the enzyme production were $30^{\circ}C$ and pH 7.0, respectively. The optimized medium was composed of 2.0% (w/v) rice straw, 0.5% (w/v) proteose peptone, 0.5% (w/v) $KH_2 PO_4$, 0.05% (w/v) yeast extract, 0.01% (w/v) $CoSO_4 \cdot 7H_2O$, and 0.05% (w/v) $CuSO$_4$\cdot 5H_2O$. When the strain was incubated with the optimized medium, it gave the activities of endoglucanase, $\beta$-glucosidase, $\beta$-xylosidase, xylanase were 3.80, 4.20, 4.00, 80.0 (IU/mL), respectively. Filter paper and cotton activities were 0.68 and 0.045 (IU/mL), respectively. The results of this study show that A. niger KKS is a potential organism with a wide spectrum of enzyme activities, such as those of $\beta$-glucosidase, $\beta$-xylosidase, and xylanase.

효소처리한 도공지의 인쇄적성에 관한 연구 (The Study of the Printability Coated Paper by the Enzyme Treatment)

  • We investigated the effects of the enzyme treatment of fiber for printability of coated paper. The results could summarized as follows, 1. The samples with enzyme treatment (average 87.8%) have higher ink gloss than the blank (85.6%). Printed density showed same trends with the ink gloss. However, the ${\beta}$-Glucosidase treated sample showed the lowest printed density (2.14 %) due to the lowest thickness of the ink layer, which is influenced by coated weight and surface smoothness. 2. The samples whose base paper was Xylanase and ${\beta}$-Glucosidase treated showed higher surface strength (4.2% and 4.0%, respectively) than the blank while the samples with the Hernicellulase and ${\beta}$-Glucuronidase treatment showed lower surface strength (3.2%and 3.7%, respectively) due to the influence of air permeability. 3. Hemicellulase and ${\beta}$-Glucuronidase treated base paper, which have relatively low air permeability, showed better ink repellence (4.3 and 4.4 %, respectively) than the blank (3.8 %). 4. The blank and the Xylanase treated base paper showed high set-off, which is the last category of printability.

Complete genome sequence of Lactococcus taiwanensis strain K_LL004, encoding hydrolytic enzymes of plant polysaccharides isolated from grasshopper (Oxya chinensis sinuosa)

  • The Lactococcus taiwanensis strain K_LL004 was isolated from the gut of a grasshopper (Oxya chinensis sinuosa) collected from local farm in Korea. L. taiwanensis strain K_LL004 is the functional probiotic candidate with an ability to hydrolyse plant polysaccharides. The complete genome of the L. taiwanensis strain K_LL004 contains one circular chromosome (1,995,099 bp) with a guanine + cytosine (GC) content of 38.8%. Moreover, 1,929 Protein-coding sequence, 19 rRNA genes, and 62 tRNA genes were identified based on results of annotation. L. taiwanensis strain K_LL004 has a gene, which encodes hydrolytic enzymes such as beta-glucosidase and beta-xylosidase, that hydrolyzes plant polysaccharides.

Amygdalin의 in Vitro 분해에 관한 연구 (A Study on the Decomposition of Amygdalin Using an In Vitro Assay)

  • Amygdalin is a cyanogenic glycoside which is commonly found in almonds, bamboo shoots, and apri-cot kernels, and peach kernels. Amygdalin was first hydrolysed into prunasin, then degraded into cyanohydrin by sequential two-stage mechanism. The objective of this study was to examine the amygdalin decomposition and cyanide formation at various in vitro conditions, including acid, enzyme and anaerobic microbes (AM) in human feces (HF). In acid hydrolysis mimicking gastric environment, amygdalin was degraded to cyanide up to 0.2% in specific pH. In contrast, enzyme assay showed higher cyanide generation either by ${\beta}$-glucosidase, or by incubation with microbe. In conclusion, we are convinced of cyanide generation are occurred mainly by microbiological activities of the gut flora up to 41.53%. After ingestion with some staff, the degree and site of degradation in an organism is a key parst of regulatory decision making of that staff.
