$^{13}C$ NMR Analysis for the Characterization of Heme Electronic/Molecular Structure in Horse Myoglobin Cyanide (Myoglobin 시안 단백질에 포함된 Heme에 대한 전자 및 분자구조 규명을 위한 $^{13}C$ NMR분석)

  • The reverse detection heteronuclear multiple quantum coherence, HMQC study of metcyano complex of horse myoglobin(MbCN) has provided the complete assignment of hyperfine shifted resonances of heme carbons attached with proton(s). The application of HMQC experiment to the paramagnetic low-spin MbCN gives clear $^1H$ and $^{13}C$ coherences for the paramagnetic amino acid residues as well as heme side chains, and can be extended to the low-spin paramagnetic hemoprotein derivative for the assignment of natural abundance $^{13}C$ resonances. This assignment strategy can avoid possible ambiguities that may result from the sole utilization of $^1H$ nuclear Overhauser effect for the assignment of heme $^1H$ signals resonating in the diamagnetic region. The resulting 2,4-vinyl ${\alpha}$-carbons and 7-propionate ${\beta}$-carbon follow anomalous anti-Curie behavior, and are indicative of incoplanarity with heme plane. Magnetic/electronic asymmetry of heme induced by proximal histidine(His) makes spread that the hyperfine shifted heme carbon resonances over the range of 250 ppm at $25^{\circ}C$. These heme carbon resonances would be the much more sensitive probe than those of proton resonances in analyzing the nature of heme electronic structure of myoglobin.

  • This paper describes the experiment that determines the ultimate strength of new uniplanar T-joints made of cold-formed square hollow sections. The new T-joint focused on the configuration of a branch member that is oriented 45 degrees to the plane of the truss and welded to the chord member whose web is stiffened with plate. The strength and failure mode are examined using the existing strength formula for the branch-rotated T-joint $(16.7{\leq}2{\gamma}(B/T){\leq}33.3$ and $0.63{\leq}{\beta}(b1/B){\leq}0.7)$. The test result shows that the capacity of the stiffened joint increases with thicker stiffening plate. The failure mode of the specimen $(2{\gamma}=33.3)$ is stiffened with plate changes from M2 (flange failure) to M3 (combined failure). On the other hand, the failure mode of the specimen $(2{\gamma}=16.7)$ is stiffened with plate changes from M1 ( web failure) to M2 (flange failure)

DNA Delivery into Embryogenic Cells of Zoysiagrass(Zoysia japonica Steud.) and Rice(Oryza sativa L.) by Electroporation (Electroporation을 이용한 잔디(Zoysia japonica Steud.) 및 벼(Oryza sativa L.) 배발생세포로의 DNA 도입)

  • To develop simple and efficient transformation methods of monocotyledonous plane, electroporation-mediated delivery of DNA into intact embryogenic cell clumps was investigated in zoysiagrass and rice. Calli of zoysiagrass, induced from 3-week-old immature embryos, were suspension-cultured in MS basic medium supplemented with 1.0 mg/t 2.4-D and used for elechuporation. Calli, derived from immature inflorescences of 20 mm lenth of rice, were also suspension-cultured on N6 basic medium supplemented with 1.0 mg/L 2.4-D. Suspension-cultured embryogenic cell clumps were electroporated in liqid MS medium added with a Plasmid DNA (30 $\mu\textrm{m}$/ml), pGA1074, encoding ${\beta}$-glucuronidiase (GUS). DNA delivery into the cells through cell walls and cell membrane was confirmed by the transient expression of the GUS gene. Cell clumps of zoysiagrass and rice, electroporated with 400 volt at 800 pF capacitance, expressed GUS gene activity at a mean frequency of 25 units (one unit = one clony of blue cells) per 200 ${\mu}\ell$ of packed cell volume. Untreated cells and healed non-embryogenic cells did not exhibit GUS activity These results indicate that electroporation-mediated transformation can use intact embryogenic cells (thus avoiding the use protoplasts) in zoysiagrass and rice.

  • The characteristics of the rock cleavage of Jurassic Geochang granite were analysed using the parameters from the length and spacing-cumulative frequency diagrams. The evaluation for three planes and three rock cleavages was performed using the 25 parameters such as (1~2) slope angle(${\alpha}^{\circ}$and ${\beta}^{\circ}$), (3) intersection angle(${\alpha}-{\beta}^{\circ}$), (4) exponent difference(${\lambda}_S-{\lambda}_L$), (5~12) length of line(oa, ob, ol, os, ss', ll' and sl') and (13~15) length ratio(ol/os, ss'/ll' and ll'/sl'), (16) mean length((ss'+ll')/2), (17~23) area (${\Delta}oaa^{\prime}$, ${\Delta}obb^{\prime}$, ${\Delta}obb^{\prime}$, ${\Delta}oaa_a^{\prime}$, ${\Delta}obb_a^{\prime}$, ${\Delta}ll^{\prime}s^{\prime}$, ${\Delta}ss^{\prime}l^{\prime}$ and ⏢$ll^{\prime}ss^{\prime}$) and (24~25) area difference(${\Delta}obb^{\prime}-{\Delta}oaa^{\prime}$ and ${\Delta}obb_a^{\prime}-{\Delta}oaa_a^{\prime}$). Firstly, the values of the 11 parameters(group I: No. 1, 3~4, 7, 9~10, 13, 15~16, 20 and 25), the 3 parameters(group II: No. 5, 8 and 17) and the 2 parameters(group III: No. 12 and 22) are in orders of H(hardway) < G(grain) < R(rift), R < G < H and G < H < R, respectively. On the contrary, the values of parameters belonging to the above three groups show reverse orders for three planes. Secondly, the generalized chart for three planes and three rock cleavages were made. From the related chart, the distribution types formed by the two diagrams related to lengths and spacings were derived. The diagrams related to spacings show upward curvature in the chart of rift plane(G1 & H1, R') and hardway(H1 & H2, H). On the contrary, the diagrams related to lengths show downward curvature. These two diagrams take the form of a convex lens in the upper section. Besides, the two diagrams cross each other in the lower section. The overall shape formed by the above two diagrams between three planes($H^{\prime}{\rightarrow}G^{\prime}{\rightarrow}R^{\prime}$) and three rock cleavages($R{\rightarrow}G{\rightarrow}H$) display in reverse order. Lastly, these types of correlation analysis is useful for discriminating three quarrying planes.

The Impact Loads on the Hitch Point of the Tiller-Trailer System (동력경운기의 경사지 견인 및 주행 특성에 관한 연구(제일보)-동력경운기 -트레일계의 힛치점에 작용하는 충격력-)

  • Transporting agricultural products and the other material by the two-wheel-tractor (power-tiler)and trailer system may be one of its most widely used farming functions.The safety and hitching load for all the previaling performing conditions may be the general concern over the operation of the tiller-trailer system. In this study, a mathematical model to determine the static and dynamic forces excerting on the hitch point were developed . Based on the analysis of the model and the field measurements. the limiting hitching load and critical slope were analyzed. The results of the study are summarized as follows ; 1) The limit angle of slope land for the safety steering that two-wheel tractor-single axle trailer system was able to transport agricultural products was the direct angle (${\gamma}$) = 8 ; the cross angle$\beta$) 15 ; and it was decreased in accordance with the increase of carrying load ($W_4). 2) The critical velocity for safe operation in case of running on downward hill road was about 1.08m/sec. 3) The limiting carrying load for the safe steering was W$_4$=600kg. The degree of the safe steering for different braking methods was given in order as follows ; Simulataneous braking the tractor and trailer , braking the trailer only, and braking tractor only. 4) Among the three components of impact loads excerting on the hitch point, the component in the lateral direction ($P_{Vy}$) was near zero in spite of increase of hitching load ($W_4) , while the components in the other two mutually perpedicular directions ($P_{Vx}$ and ($P_{Vz}$) ) had larger values in horizontal plane than those in the slope lands. 5) Moment of forces on the lateral direction (M$y$) had the largest value among the three components of impact moment acting on the hitch point, however all the components were sharply increased in accordance with the increase of hitching loads ($W_4. Three components of the moment were the negative values.

  • The design of osteotomy plane in orthognathic surgery has been developed to diminish the nerve injury. Intraoral Vertico-Sagittal Ramus Osteotomy (IVSRO) is the one of the best way to minimize untoward results, which is designed not to expose the lingula. We evaluated the nerve damage before and after with current perception threshold (CPT) test which is modem and numerically expressible way of nerve damages. Sixty patients underwent IVSRO since 1998 were evaluated. They were divided into 2 groups; one group underwent IVSRO only, and the other underwent IVSRO plus genioplasty. The both groups were evaluated with CPT test 1 week before surgery, and 1, 3 and 6 months after surgery. The CPT test was performed on A-beta, A-delta and C fiber respectively. 111e result showed that the recovery of sensory function of damaged nerve fibers was observed at the period of three to six months after surgery. There was no impairment of nerve function after only the IVSRO . But there were sensory disturbances in cases of additional genioplasty group. We thought that one of major factors on nerve damages were exposure of nerve and traction injury during genioplasty.

Dynamic Stability Analysis of Thick Plates with Varying Thickness and Concentrated Mass on Inhomogeneous Pasternak Foundation (비균질 Pasternak 지반에 놓인 집중질량을 갖는 변단면 후판의 동적안정해석)

  • This paper is to analyze the stability of the thick plate on inhomogeneous Pasternak foundation, with linearly varying thickness and concentrated mass by finite element method. To verify this finite element method, the results of natural frequencies and buckling stresses by the proposed method are compared with the existing solutions. The dynamic instability regions are decided by the dynamic stability analysis of the thick plate on inhomogeneous Pasternak foundation, with linearly varying thickness and concentrated mass. The non-dimensional Winkler foundation parameter is applied as 100, 1000 and non-dimensional shear foundation parameter is applied as 5. The tapered ratios are applied as 0.25 and 1.0, the ratios of concentrated mass to plate mass as 0.25 and 1.0, and the ratio of in-plane force to critical load as 0.4. As the result of numerical analysis of the thick plate on inhomogeneous Pasternak foundation for $u{\times}v=300cm{\times}300cm$ and $a{\times}b=600cm{\times}600cm$, instability areas of the thick plate which has the larger rigidity of inner area are farther from ${\beta}$-axis and narrower than those which has the larger rigidity of outer area.

  • We have investigated the radial g-r color gradients of early-type galaxies in the Sloan Digital Sky Survey (SDSS) DR6 in the redshift range 0.00$H{\beta}$ absorption-line strengths and/or emission-line ratios that are indicative of the presence of young stellar populations. This implies that most of the residual star formation in early-type galaxies is centrally concentrated. Blue-cored galaxies are predominantly low-velocity dispersion systems. A simple model shows that the observed positive color gradients are visible only for a billion years after a star formation episode for the typical strength of recent star formation. The observed effective radius decreases and the mean surface brightness increases due to this centrally concentrated star formation episode. As a result, the majority of blue-cored galaxies may lie on different regions in the fundamental plane (FP) from red-cored ellipticals. However, the position of the blue-cored galaxies on the FP cannot be solely attributed to recent star formation but requires substantially lower velocity dispersion. We conclude that a low-level of residual star formation persists at the centers of most of low-mass early-type galaxies, whereas massive ones are mostly quiescent systems with metallicity-driven red cores.

Comparative Analysis of Cervical Lateral Mass Screw Insertion among Three Techniques in the Korean Population by Quantitative Measurements with Reformatted 2D CT Scan Images : Clinical Research

  • Objective : Our purpose of this study is to compare insertion angles and screw lengths from Roy-Camille, Magerl, and our designed method for cervical lateral mass screw fixation in the Korean population by quantitative measurement of reformatted two dimensional (2D) computed tomography (CT) images. Methods : We selected thirty Korean patients who were evaluated with thin section CT scans and reconstruction program to obtain reformatted 2D-CT images of the transversal plane passing the cranio-caudal angle using three different techniques. We measured the minimum angle to avoid vertebral artery (VA) injury, the ideal angle and depth for bicortical screwing of cervical lateral mass. Morphometric measurements of the lateral masses from C3-C7 were also taken. Results : In all three techniques, the mean safety angles from the VA were less than 8 degrees and the necessary depth of the screw was about 14 mm for safety to the VA and for the bicortical purchase. In our designed technique, the mean $\beta$ angles of each level from C3 to C7 were 29.0. 29.8. 29.5. 26.3. and 23.9 degrees, respectively. Conclusion : Results of this study and data from the literature indicate that differences may exist between the Korean and Western people in the length and angle for ideal lateral mass screw fixation. In addition, our technique needs further cadaveric and clinical study for safety and efficacy for being performed as alternative method for cervical lateral mass fixation.

COMPUTER SIMULATION OF INTRAMOLECULAR HYDROGEN TRANSFER TO CARBONYL OXYGEN BY A MONTE CARLO METHOD: PHOTOREACTIONS VIA REMOTE PROTON TRANSFER IN BENZOYLBENZOATES

  • The model based on the idea that the p$_y$-orbital of the carbonyl oxygen is responsible to receiving hydrogen was devised for simulation of intramolecular hydrogen transfer. A Monte Carlo method was applied to free rotation of a molecular chain performed by changing the dihedral angles, and a "hit" was defined as the case when the migrating hydrogen comes within the region defined as the p$_y$-orbital and satisfies all the geometrical requirements for abstraction. A set of parameters was employed for defining the region and the requirements; $\tau$ was defined as the angle formed between O...H vector and its projection on the mean plane of the carbonyl group (- 43$\circ$ < $\tau$ < + 43$\circ$), $\Delta$ as the C=O...H angle (90 -15$\circ$ < $\Delta$ < 90 + 15$\circ$), $\theta$ as the O...H - C angle ( 180 - 80$\circ$< 0 < 180 + 80$\circ$), d as the distance from the center of the lobe of the p$_y$-orbital to hydrogen (0 < d < 1.04 ${\AA}$). The minimum value for the distance between carbonyl oxygen (O$_1$) and the migrating hydrogen (H$_i$) and for that between non-bonded atoms except the pair of O$_1$ and H$_i$ were assumed to be 0.52 ${\AA}$ and 1.54 ${\AA}$, respectively. The apphcation of this model to intramolecular $\beta$-, $\gamma$-, $\delta$-, $\epsilon$-, and $\zeta$-hydrogen abstraction in ketones and $\eta$- and $\theta$- proton transfer in oxoesters gave good results reflecting their photochemical behavior. The model was also used for prediction of photoreactivities of 2-(N,N-dibenzylamino)ethyl 2-, 3- and 4-benzoylbenzoate (1a - c). (1a - c).
