Effect of Cold Stress on Carotenoids in Kale Leaves (Brassica oleracea) (저온처리가 케일(Brassica oleracea)잎 내 Carotenoid에 미치는 영향)

  • BACKGROUND: Kale (Brassica oleracea) biosynthesizes various phytochemicals including glucosinolates, flavonoids, and carotenoids. Phytochemicals of plants are influenced by light, temperature, carbon dioxide, and growing conditions. Specifically, carotenoids are affected by temperature, light, and oxygen. The aim of this study was to investigate the effect of cold stress (day/night: $25^{\circ}C/20^{\circ}C$, $20^{\circ}C/15^{\circ}C$, $15^{\circ}C/10^{\circ}C$) on carotenoids in kale leaves. METHODS AND RESULTS: Kale was grown in pots for up to 50 days after sowing (DAS) in a greenhouse. For cold acclimation experiments, kale grown in growth chambers for 3 days and was subjected to low temperature for 4 days. The conditions maintained in the growth chambers were as follows: photoperiod, 12/12 h (day/night); light, fluorescent; and relative humidity, 60%. Carotenoid (lutein, ${\alpha}-carotene$, zeaxanthin, ${\beta}-carotene$) contents were analyzed by high-performance liquid chromatography (HPLC). The total carotenoid content gradually increased during cold acclimation for 3 days. When kale was subjected to cold stress, the total carotenoid content was high at $25^{\circ}C/20^{\circ}C$ treatment, but low at $15^{\circ}C/10^{\circ}C$ treatment. The total carotenoid content of kale leaves continuously grown in greenhouse decreased from 50 to 57 DAS (1,418 and 1,160 mgkg-1 dry wt., respectively). The lutein, ${\alpha}-carotene$, and ${\beta}-carotene$ contents were very low and the zeaxanthin contents were very high at $15^{\circ}C/10^{\circ}C$ treatment. When kale was subjected to cold stress, the ratio of individual to the total carotenoid contents of kale leaves was 4553% for -carotene and 210% for zeaxanthin. CONCLUSION: The ${\beta}-carotene$ and zeaxanthin contents in kale leaves indicate their sensitiveness toward cold stress.

Phytochemical contents of agricultural products cultivated by region (재배지역에 따른 농산물의 phytochemical 함량)

  • The purpose of this research is to distinguish the quantitative determination of phytochemicals in various agricultural products and to optimize an HPLC method for the determination of lycopene, lutein, ${\alpha}$-carotene, ${\beta}$-carotene, and cryptoxanthin. Among the different conditions studied, the most suitable ones for our samples were the extraction with hexane/acetone/ethanol (50:25:25, v/v/v), dissolution of the dry extract in tetrahydrofuran/acetonitrile/methanol (15:30:55, v/v/v), injection on a $C_{18}$ column with methanol/acetonitrile (90:10, v/v) + triethylamine $9{\mu}M$ as mobile phase, and ${\lambda}_{detection}$=475 nm. The mean percent recovery for the HPLC method were $120.7{\pm}4.1%$ (lycopene), $89.2{\pm}3.5%$ (lutein), $91.2{\pm}2.9%$ (${\alpha}$-carotene), $99.1{\pm}4.4%$ (${\beta}$-carotene), and $100.0{\pm}5.3%$ (cryptoxanthin). The contents of lutein in the agricultural products were spinach, kiwi, tomato, blueberry, melon, respectively. However, the lycopene contents were the highest in the Black tomato ($56.66{\pm}7.48mg/kg$) and Jangseong tomato ($50.28{\pm}5.42mg/kg$). The concentration of ${\beta}$-carotene in all of the agricultural products ranged from 0.07 mg/kg to 65.03 mg/kg. The quercetin content of the agricultural products increased in the order of blueberry (986.57~1,054.06 mg/100 g), kiwi (44.96~55.09 mg/100 g), hallabong (31.92~35.60 mg/100 g), and tomato (26.38~34.94 mg/100 g). The highest kaempferol content was found in the blueberry (47.79~76.15 mg/100 g) with results in all of the tested samples varying between 6.54~48.11 mg/100 g. The total polyphenol contents of the various agricultural products increased in the blueberry (213.60~229.96 mg/100 g), spinach (112.50~141.67 mg/100 g) and kiwi (46.49~70.44 mg/100 g). The total flavonoid content was the highest in both blueberry and spinach. Vitamin C content was detected in kiwi > hallabong > tomato > blueberry, respectively. The total anthocyanin contents (TAC) was detected in the Damyang blueberry and the imported blueberry.

Different Cooking Methods for Korean Cabbage and Their Effect on Antioxidant Activity and Carotenoid and Tocopherol Contents (배추의 조리방법에 따른 항산화 활성 및 카로티노이드와 토코페롤 함량 변화)

  • Korean cabbage (Brassica campestris L.ssp.pekinensis) is one of the major cruciferous vegetables. Cruciferous vegetables contain a rare series of secondary metabolites of amino acids called glucosinolates, as well as carotenoids, tocopherol, vitamin C and fibers. This study evaluated the effect of common cooking methods (boiling, microwaving, steaming and frying) on the phytochemical content (lutein, ${\beta}$-carotene, ${\gamma}$-tocopherol, and ${\alpha}$-tocopherol), and total antioxidant capacity of Korean cabbages, determined by DPPH assay and ABTS assay. Boiling caused a decrease in carotenoids, lutein and tocopherols. Microwaving and steaming were relatively good cooking methods for maintaining lutein, ${\beta}$-carotene, ${\gamma}$-tocopherol, and ${\alpha}$-tocopherol. The overall results of this study demonstrate that some domestic cooking procedures, specifically microwave and steaming, increased the bioaccessibility of carotenoids and tocopherol, highlighting the positive role of the nutritional properties of Korean cabbage.

Effects of Erythrosine on the Cholesterol Oxidative Stability in an Aqueous Model System (수용성 모형계에서 콜레스테롤의 산화 안정성에 대한 Erythrosine의 영향)

  • The effects of erythrosine on the oxidative stability of cholesterol in an aqueous model system were studied by depleted headspace oxygen and cholesterol oxidation products (COP). As the oncentration of erythrosine was increased, headspace oxygen depletion, 7-COP and total COP increased during storage at $25^{\circ}C$ for 50 hours under the fluorescent light. As the intensity of fluorescent light was increased, amounts of headspace oxygen depleted and COP formed in an aqueous cholesterol dispersion containing erythrosine also increased. Addition of ${\alpha}-,\;{\delta}-$, mixed-tocopherol and ${\beta}-carotene$ resulted in the enhanced oxidative stability of an aqueous cholesterol dispersion containing erythrosine during the fluorescent light storage.

Influence of Processing on Quality of Carrot Juice (가공공정에 의한 당근쥬스의 품질변화에 관한 연구)

  • This study was conducted to determine the organoleptic and physical properties and carotenoid of commercial canned, frozen and freshly-made carrot juice. Samples were evaluated by sensory panel and measured for viscosity and acidity. For carotene analysis, HPLC of alpha- and beta-carotene, and spectrophotometry of total carotenoid content were used. Sensory evaluation indicated that the canned sample was less acceptable, especially for flavor and texture, than other juices, while forzen juice was considered as acceptable as freshly-made carrot juice. The canned product showed about 10 times higher viscosity and lower acidity than others. Between two kinds of frozen samples, one sample was the same as freshly-made sample for all parameters while the other showed less alpha-carotene content which was 2 times higher than that of canned one. Canned sample contained 70-77% of freshly-made or frozen samples in total carotenoid and beta-carotene content and 24% of freshly-made one in alpha-carotene. These results suggest that freezing process is a good preservation method for carrot juice with respect to sensory evaluation, physical property and carotenoid content.

Pigment Stability of Lavers Porhyra tenera Kjellman during Processing and Storage (김의 가공 저장중에 있어서의 색소의 안정도)

  • The stability of three major pigments in lavers, namely chlorophylls, carotenoids, and phy-cobilins, during processing and storage was studied. The results of the pigment retention in fresh lavers during storage at low temperatures, of heat lability during heat treatment, and of the pigment loss during storage are summarized as follows: 1. The contents of chlorophyll a, xanthophyll (lutein+zeaxanthin), and carotene ($\alpha-+\beta-carotene$) averaged 1,525, 627, and $409{\mu}g/g$ of dry samples respectively The comparative composition of four carotenoids, lutein, $\beta-carotene$, zeaxanthin, and $\alpha-carotene$, was 35.9, 33.7, 12.2, and $5.8\%$ respectively. 2. In storage of fresh lavers at room ternperature ($20-25^{\circ}C$), 2 to $5^{\circ}C$, and $-15^{\circ}C$, the pigments generally were retained more at 2 to $5^{\circ}C$ than others while carotene only showed more retention at $-15^{\circ}C$ than at 2 to $5^{\circ}C$. Phycocyanin tended to decrease more rapidly than phyco-erythrin at low temperatures. 3. In regard to heat treatment of both dried and wet materials at 40, 60, 80, $100^{\circ}C$ respectively, the pigments were more stable at lower temperatures than at higher temperatures in both cases of materials. Carotenoids were retained more in wet materials than in the sun dried. Xanthophylls showed higher heat stability than carotenes. Phycoerythrin was considered more heat labile than phycocyanin, particularly at higher temperatures. 4. In storage of heat treated lavers for 50 days at room temperature, the retention of pigments appeared differently in accordance with the temperatures of treatment The materials treated at $60^{\circ}C$ showed better relention of pigments than those treated at the other temperatures. Degradation rate of xanthopllylls, carotenes, and phycobilins was decreased in the materials treated at higher temperatures, showing a tendency of great pigment retention even for a long period storage. Chlorophyll il was retained more in sun dried materials whereas xanthophyll and carotene were retained more in the wet. 5. In order to maintain a higher pigment stability during storage, the materials must be treated at $60^{\circ}C$ in a dryer with rapid ventilation until the moisture level reaches about $6.0\%$. In case of treatment at a higher temperature than $60^{\circ}C$, they must be treated for a short period of time to minimize pigment degradation by heat. 6. The lavers treated with the solutions of pH around 7.8 showed a higher retention of chlorophyll a. When treated with the solutions of algal extracts such as agar and alginic acid, uniform layer of laver sheets and better surface gloss of dried layers were obtained.

Estimated Carotenoids Intake in Korean Adults Using Food-frequency Questionnaire: Association with Smoking, Drinking and Other Life-style Factors

  • The aim of this study was to determine the association between, smoking, exercise, sex, and dietary carotenoids ($\alpha$-carotene, $\beta$-carotene, $\beta$-cryptoxanthin, lutein + zeaxanthin, lycopne) intake in Korean middle-aged adults. Food-frequency questionnaire were analyzed from 304 healthy adults (115 men, 189 women) aged 20 - 59. The self-administered questionnaire contained subject s habitual diet and alcohol intake were the previous 3 months. Data on frequency of 102 foods, including vegetables, fruits, beverages and legumes were analyzed. Total dietary carotenoids intake were 27.13 $\pm$ 3.09 mg/d for men and 26.71$\pm$ 2.82 mg/d for women. It was found that smoking had no significant contribution to the dietary intake of carotenoids. Among other lifestyle factors that had significant correlation was the amount of exercise time. The increases in exercise time was associated with increase in carotenoids intake (r= 0.121, p : 0.04). The major contributors of $\alpha$-carotene and $\beta$-carotene were carrots consumed as single-food item or carrot juice. Lutein and Zeaxanthin intake mainly came from spinach and most lycopene intake was derived from tomato products not fresh tomatoes. Persimmon was the major contributor of $\beta$-cryptoxanthin. These findings provide valuable information on understanding the unique pattern of dietary intake of Korean, which might help identify the risks for developing various diseases.

Comparison of Carotenoid Pigments on Korean Dark Sleeper, Odontobutis platycephala and Dark Sleeper, Odontobutis odontobutis interrupta in the Family Eleotridae (구굴무치과에 속하는 동사리와 얼룩동사리의 Carotenoid 색소성분의 비교)

  • This study was performed as a part of comparative biochemical studies of carotenoid pigment for the fresh water fish. Carotenoids in integument of Korean dark sleeper, Odontobutis platycephala, and dark sleeper, Odontobutis odontobutis interrupta, which are all the Korean native fresh water fish, were separated by thin layer chromatography, column chromatography and HPLC. The separated carotenoid were then reduced and isomerized by NaBH4 and I2 respectively to investigate UV-Vis spectrophotometeric patterns and chracterized by IR, 1H-NMR and Mass spectrum. The content of total carotenoids in the integument of Korean dark sleeper was 3.01mg% in April, but it was increased to 3.74mg% in September at the near of spawning period. The carotenoid isolated in April consisted of $\beta$-carotene(25.6%), lutein(18.5%) and zeaxanthin(12.0%) as major carotenoids and also contained isocryptoxanthin, diatoxanthin, tunaxanthin, cynthiaxanthin, canthaxanthin and $\alpha$-cryptoxanthin as minor carotenoids. Similarly, in September the carotenoid consisted of $\beta$-carotene(16.5%), zeaxanthin(13.7%) and cynthiaxanthin(13.6%) as major carotenoids and also contained lutein, isocryptoxanthin, tunaxanthin, $\alpha$-cryptoxanthin, diatoxanthin and canthaxanthin as minor carotenoids. At the near of spawning period, the content of cynthiaxanthin and $\alpha$-cryptoxanthin were increased. The content of total carotenoids in the integument of spawning period. T도 carotenoid isolated in April and September consisted of $\beta$-carotene(24.9%, 27.5%), zeaxanthin(14.4%, 20.9%) and lutein(12.6%, 11.4%) as major carotenoids and also contained cynthiaxanthin, tunaxanthin, diatoxanthin, isocryptoxanthin, $\alpha$-cryp-toxanthin and canthaxanthin as minor carotenoids. At the near of spawning period, the content of zeaxanthin was increased, indicating that the carotenoid composition were dependent upon their living conditions and their integument colors. Both Korean dark sleeper and dark sleeper contained high amount of cynthiaxanthin and diatoxanthin which are found as rare carotenoids in the other of fresh water fish. It is interes that they also contained tunaxanthin which is a specific carotenoid in marine fishes.

Vitamin A Status of Female University Students (여대생의 비타민 A 영양 상태)

  • The purpose of this study was to diagnose nutritional status of vitamin A in female university students. Total of 59 female students in Korea National University of Education, Home Economics Education department were recruited for this study. Retinol and :t major carotenoids in serum($\beta$-carotene, u-carotene, lycopene and lutein) were analyzed by HPLC. The isocratic separation was performed in a $\mu$ Bondapak$^{TM}$ $C_{18}$ stainless steel column with a solvent system of acetonitrile : methanol=85:15. The results of analysis were as follows: The average retinol concentration of 59 female students was 25.9$\mu\textrm{g}$/100$m\ell$. According to biochemical criteria for the assessment of vitamin A status, 18 of them belong to well-nourished and 6 students belong to adequate status. On the other hand, 8 students, 3 freshmen and 5 sophomore, were in critical vitamin A status. Retinol concentrations of junior and senior students were higher than those of freshmen and sophomore (P<0.001). The average concentrations of serum $\beta$-carotene, lycopene, and lutein were 1.9, 5.4, and 41.4$\mu\textrm{g}$/100$m\ell$, respectively. Serum $\beta$-carotene concentration of senior was significant. higher than those of freshman, sophomore and junior(p<0.001). Lycopene concentration of freshman was significant. lower than those of the other grades and that of senior was sig, higher than those of the other grades(p<0.05). Lutein concentrations of junior and senior were significant. higher than those of freshman and sophomore(p<0.001). The serum $\alpha$-carotene concentrations of the students were too low to analyze.e.

Effect of Lipoxygenase on the Oxidation of Rainbow Trout Lipid in Model system (모델시스템에 있어서 무지개 송어 지방질의 산화에 대한 Lipoxygenase의 영향)

  • The effect of lipoxygenase (LOX) on the oxidation and co-oxidation of lipid fraction was studied in the model system of rainbow trout. For the reaction in model system 1 g of lipid fraction and 50mL of enzyme extract(LOX, 140 unit in 50mL phosphate buffer solution at pH 7, 4)), which were obtained from rainbow trout, were homoginized in the presence of Tween 20 and kept at 23$\circ$C for 3 days. The activity of LOX was decreased to 43% of initial level during the reaction in the model system. The initial composition of rainbow trout lipid was showed to be consisted of trigliceride(TG;82%) and free fatty acid(FFA;0.1%), while this converted to 59% of TG and 20% of FIFA, respectively after reaction in model system. Change of fatty acid composition was also observed and the content of linoleic acid, one of the major fatte acids, was decreased to 13% from 54% in the content of total fatty acids after reaction. The carotenoids in rainbow trout were composed of 0.4% $\alpha$-carotene, 1.6% $\beta$ -carotene, 80% canthaxanthin, 7% lutein and 11% zeaxanthin, thus the canthaxanthin was the major component. This canthaxanthin was the most degraded carotenoid by lipoxygenase catalyzed co-oxidation during the reaction. On the other hand the tocopherol isomers found in the rainbow trout were $\alpha$ and $\beta$ -tocopherol, and $\alpha$-tocopherol had a higher degradation rate by the lipoxygenase catalyzed co-oxidation than of $\beta$-tocopherol in the reaction of model system.
