농업생태계와 산림생태계 숲 유형에 따른 대형나방 군집 다양성 연구 (The study on diversity of macromoth community in two different forest types from agro and forest ecosystems)

  • 이 연구의 목적은 농업생태계에 존재하는 작고 파편화된 숲과 인근 산림생태계 내 연속된 숲에 서식하는 대형나방의 종 다양성과 종구성의 차이를 알아보는데 있다. 전남 서남부 8개 지역에서 2018과 2019년 6월과 8월 자외선 등 트랩을 이용하여 대형나방을 채집, 분석한 결과 두 숲 유형 간 뚜렷한 차이를 보였으며 두 그룹 간 종 수, 개체수, 종다양성 지수(H') 모두 산림생태계 숲이 농업생태계 숲보다 더 높게 나타났다. 유사도지수를 이용하여 나방 군집간 차이를 알아본 결과 농업생태계 숲에서 베타다양성이 높음을 확인하였다. 또한 종 구성의 변화율(Turnover)과 내포성(Nestedness)을 따로 나누어 분석한 결과 산림생태계는 종 구성 변화율이 큰 반면 농업생태계는 내포성이 비교적 크게 나타나 서식지 유형에 따라 나방 군집 구성의 차이가 다른 것을 확인할 수 있었다.

Characterization of the Microbial Diversity in a Korean Solar Saltern by 16S rRNA Gene Analysis

  • We studied the diversity of the halophilic archaea and bacteria in crystallizer ponds of a Korean solar saltern by analyzing 16S rRNA gene libraries. Although diverse halophilic archaeal lineages were detected, the majority (56%) were affiliated with the uncultured and cultured Halorubrum group. Halophilic archaea that have been frequently observed in solar saltern environments previously, such as Halogeometricum, Halococcus, Haloarcula, and Haloferax, were not detected in our samples. The majority of clones (53%) belonged to the Cytophaga-Flavobacterium-Bacteroides and ${\alpha}-,\;{\gamma}-,\;and\;{\delta}-Proteobacteria$ groups, with 47% of the clones being affiliated with ${\gamma}-Proteobacteria$. We also identified new ${\delta}-Proteobacteria$-related bacteria that have not been observed in hypersaline environments previously. Our data show that the diversity of the halophilic archaea and bacteria in our Korean saltern differs from that of solar salterns found in other geographic locations. We also showed by quantitative real-time PCR analysis that bacteria can form a significant component of the microbial community in solar salterns.

Identification of the bacterial composition in the rockworm gut and biofloc-fed adult gut flora beneficial for integrated multitrophic aquaculture

  • The rockworm gut flora was identified at each growth stage to elucidate the composition of the bacterial community. The source material was Marphysa sanguinea fed regular feed and fed biofloc at the adult stage in parallel. The systematic bacterial community composition was determined based on the next-generation sequencing method, and alpha diversity and beta diversity were conducted to access the species diversity within and between the bacterial communities, respectively. The composition of the gut flora changed considerably as the rockworms developed. The shift in the gut flora was confirmed at the phylum, family, and genera level of the bacterial communities. The Vibrio species associated with high rockworm mortality occupied 7.7% of the gut flora at the larval stage; however, they disappeared in the healthy adult gut. Moreover, different gut flora was observed between adults fed regular feed and those fed biofloc. Specifically in the biofloc-fed adult gut, several immune relevant and water-purifying bacteria were detected. The biofloc-fed adult gut flora could decompose and mineralize organic sediment, and thus be effectively utilized for integrated multitrophic aquaculture. The Venn diagram revealed that only two bacterial species were shared throughout all growth stages, and the biofloc-fed adults exhibited the highest diversity within the bacterial community.

Diversity of the Bambusicolous Fungus Apiospora in Korea: Discovery of New Apiospora Species

  • Many Apiospora species have been isolated from bamboo plants - to date, 34 bambusicolous Apiospora species have been recorded. They are known as saprophytes, endophytes, and plant pathogens. In this study, 242 bambusicolous Apiospora were isolated from various bamboo materials (branches, culms, leaves, roots, and shoots) and examined using DNA sequence similarity based on the internal transcribed spacer, 28S large subunit ribosomal RNA gene, translation elongation factor 1-alpha, and beta-tubulin regions. Nine Apiospora species (Ap. arundinis, Ap. camelliae-sinensis, Ap. hysterina, Ap. lageniformis sp. nov., Ap. paraphaeosperma, Ap. pseudohyphopodii sp. nov., Ap. rasikravindrae, Ap. saccharicola, and Ap. sargassi) were identified via molecular analysis. Moreover, the highest diversity of Apiospora was found in culms, and the most abundant species was Ap. arundinis. Among the nine Apiospora species, two (Ap. hysterina and Ap. paraphaeosperma) were unrecorded in Korea, and the other two species (Ap. lageniformis sp. nov. and Ap. pseudohyphopodii sp. nov.) were potentially novel species. Here, we describe the diversity of bambusicolous Apiospora species in bamboo organs, construct a multi-locus phylogenetic tree, and delineate morphological features of new bambusicolous Apiospora in Korea.

외래종 배스가 저수지 어류군집에 미치는 영향: 중초산과 북초산 저수지를 대상으로 (Effect of Invasive Species on Reservoir Fish Communities: For Joongchosan and Bukchosan)

  • 2020년 3월부터 10월까지 군산시의 중초산 저수지와 북초산 저수지의 어류상 및 군집분석을 하였다. 조사기간 동안 채집된 어류는 중초산 저수지에서 4목 5과 8종 1,895개체, 북초산 저수지에서 3목 5과 7종 171개체였다. 중초산 저수지의 우점종은 참붕어(661개체, 상대풍부도: 34.7%) 아우점종은 흰줄납줄개(660개체, 상대풍부도: 34.7%), 북초산 저수지는 배스(77개체, 45.0%)와 붕어(60개체, 35.1%) 순으로 나타났다. 군집분석 결과 중초산 저수지는 우점도 0.697, 다양도 1.483, 균등도 0.713, 종 풍부도 0.928, 북초산 저수지는 우점도 0.801, 다양도 1.304, 균등도 0.670, 종 풍부도 1.167을 보였다. 본 연구 결과를 통해 생태계 교란생물인 배스의 영향으로 토착어종의 개체수와 군집의 다양도가 감소한 것을 확인할 수 있었다. 따라서 외래어종의 지속적인 제거와 유입을 막는 관리가 요구된다.

Genomic Sequence Analysis and Organization of BmKαTx11 and BmKαTx15 from Buthus martensii Karsch: Molecular Evolution of α-toxin genes

  • Based on the reported cDNA sequences of $BmK{\alpha}Txs$, the genes encoding toxin $BmK{\alpha}Tx11$ and $BmK{\alpha}Tx15$ were amplified by PCR from the Chinese scorpion Buthus martensii Karsch genomic DNA employing synthetic oligonucleotides. Sequences analysis of nucleotide showed that an intron about 500 bp length interrupts signal peptide coding regions of $BmK{\alpha}Tx11$ and $BmK{\alpha}Tx15$. Using cDNA sequence of $BmK{\alpha}Tx11$ as probe, southern hybridization of BmK genome total DNA was performed. The result indicates that $BmK{\alpha}Tx11$ is multicopy genes or belongs to multiple gene family with high homology genes. The similarity of $BmK{\alpha}$-toxin gene sequences and southern hybridization revealed the evolution trace of $BmK{\alpha}$-toxins: $BmK{\alpha}$-toxin genes evolve from a common progenitor, and the genes diversity is associated with a process of locus duplication and gene divergence.

Cultivable Microbial Diversity in Domestic Bentonites and Their Hydrolytic Enzyme Production

  • We have isolated and identified 72 bacterial strains from four bentonite samples collected at the mining areas located in Gyeongsangbuk-do, Republic of Korea, and measured their hydrolytic enzyme (${\alpha}$-amylase, protease, and cellulase) activities to identify the isolates with industrial-use potential. Most of the isolates belonged to the Bacillaceae, with minor portions being from the Paenibacillaceae, Micrococcaceae, and Bacillales Family XII at the family level. Of the strains isolated, 33 had extracellular ${\alpha}$-amylase activity, 30 strains produced cellulase, and 35 strains produced protease. Strain MBLB1268, having the highest ${\alpha}$-amylase activity, was identified as Bacillus siamensis ($0.38{\pm}0.06U/ml$). Bacillus tequilensis MBLB1223, isolated from Byi33-b, showed the highest cellulase activity ($0.26{\pm} 0.04U/ml$), whereas Bacillus wiedmannii MBLB1197, isolated from Zdb130-b, exhibited the highest protease activity ($54.99{\pm}0.78U/ml$). These findings show that diverse bacteria of the Bacillaceae family adhere to and exist in bentonite and are potential sources of industrially useful hydrolytic enzymes.

순천만 칠면초의 근권으로부터 분리된 해양세균의 다양성 및 계통학적 분석 (Diversity and Phylogenetic Analysis of Culturable Marine Bacteria Isolated from Rhizosphere Soils of Suaeda japonica Makino in Suncheon Bay)

  • 순천만 일대 칠면초 군락의 근권 토양에 존재하는 근권 세균의 다양성 분석을 위해 몇 개 지점을 선정한 후 샘플링을 실시하였다. 채취한 토양시료는 marine broth, tryptic soy broth 한천배지를 이용하면서 세균 집락 간 형태학적인 구분을 통해 순수분리 되었다. 분리된 세균의 genomic DNA를 획득한 후, 각각의 16S rDNA 염기서열을 증폭 분석하여 총 29 strain이 부분동정 되었다. 이들의 유연관계 확인을 위해 계통수를 작성한 결과, 이들은 각각 firmicutes문 (44.8%), gamma-proteobacteria group (27.6%), alpha-proteobacteria group (10.3%), bacteriodetes 문(10.3%), actinobacteria 문(6.8%)에 속하였다. 최우점하는 firmicutes 문에서는 Bacillus 속이, 차 우점하는 gamma-proteobacteria group에서는 각각 Marinobacterium, Halomonas, Vibrio 속이 집중 분포하는 것으로 나타났다. 또한 채취 지점별로 몇 가지 척도를 사용하여 다양성 지수를 도출하였을 때, 채취 지점 간 지수의 차이를 보여 전체 순천만 갯벌 중 위치에 따라 상이한 미생물상을 가지는 것으로 추측된다. 분리된 균들 중 일부는 갯벌 특유의 극한환경을 극복하면서 칠면초 근권에서 생존하며 물질순환, 식물생장 촉진 및 병원체 방어작용 유도 등 식물생장에 긍정적 역할을 수행할 것으로 생각된다.

Remarkable Bacterial Diversity in the Tidal Flat Sediment as Revealed by 16S rDNA Analysis

  • A 16S rDNA clone library was generated to investigate the bacterial diversity in tidal flat sediment in Ganghwa Island, Republic of Korea. A total of 103 clones were sequenced and analyzed by comprehensive phylogenetic analyses. No clones were identical to any of known 16S rRNA sequences in public databases. Sequenced clones fell into thirteen lineages of the domain Bacteria: the alpha, beta, gamma, delta, and epsilon Proteobacteria, Actinobacteria, CFB group, Chloroflexi, Acidobacteria, Planctomycetes, Verrucomicrobia, and known uncultured candidate divisions (OP11, BRC1, KSB1, and WS1). Two clones were not associated with any known bacterial divisions. The majority of clones belonged to the gamma and delta Proteobacteria (46.7%). Clones of Actinobacteria were distantly related to known taxa. It is evident from 16S rDNA-based community analysis that the bacterial community in tidal flat sediment is remarkably diverse and unique among other marine environments examined so far.

The Diversity of Culturable Organotrophic Bacteria from Local Solar Salterns

  • We isolated and cultured bacteria inhabiting solar saltern ponds in Taean-Gun, Chungnam Province, Korea. All of the isolated 64 strains were found to be moderately halophilic bacteria, growing in a salt range of 2-20 %, with an optimal concentration of 5% salt. Bacterial diversity among the isolated halophiles was evaluated via RFLP analyses of PCR-amplified 16S rDNAs, followed by phylogenetic analysis of the partial 16S rDNA sequences. The combination of restriction enzyme digestions with HaeIII, CfoI, MspI and RsaI generated 54 distinct patterns. A neighbor-joining tree of the partial 16S rDNA sequences resulted in the division of the 64 strains into 2 major groups, 45 strains of ${\gamma}-Proteobacteria$ (70.3%) and 19 strains of Firmicutes (29.7%). The ${\alpha}-Proteobacteria$ and Cytophaga-Flavobacterium-Bacterioides groups, which were repeatedly found to exist in thalassohaline environments, were not represented in our isolates. The ${\gamma}-Proteobacteria$ group consisted of several subgroups of the Vibrionaceae (37.5%), Pseudoalteromonadaceae (10.9%), Halomonadaceae (7.8%), Alteromonadaceae (7.8%), and Idiomarinaceae (6.3%). Members of Salinivibrio costicola (29.7%) were the most predominant species among all of the isolates, followed by Halobacillus treperi (12.5%). Additionally, three new species candidates were found, based on similarities of the 16S rDNA sequences to those of previously published species.
