Evaluation of Phytochemical Composition and Antioxidant Capacity in Various Leafy Vegetables

  • Current study investigated phytochemical compositions and antioxidant capacity of Korean leafy vegetables including chajogi, gomchwi, meowi and sseumbagwi. ${\beta}$-carotene, total soluble polyphenol, total flavonoids contents were determined, and antioxidant capacity were evaluated by various methods. ${\beta}$-carotene, total soluble polyphenol, total flavonoids contents in gomchwi were significantly higher (as much as 19 folds) compare to other vegetables (p<0.05). Sseumbagwi extract ($100{\mu}g/mL$) was removed 78.6% of superoxide radicals in xanthin-xanthin oxidase system measured by EPR. Gomchwi showed the highest nitrate scavenging activity as 94.3% at pH 1.2. In SOD-like activity, chajogi, gomchwi, and meowi were evaluated in the range of $12.6{\sim}24.5%$. All samples were revealed to prevent the reaction of ferrous chloride during 9 days, which were comparable ability with 125 mg/100 g of ${\alpha}$-tocopherol. The rich phytochemical contents of gomchwi and sseumbagwi influenced to high antioxidant capacity.

Diet and Colorectal Cancer Risk in Asia - a Systematic Review

  • Diet is one of the major factors that can exert a majorly influence on colorectal cancer risk. This systematic review aimed to find correlations between various diet types, food or nutrients and colorectal cancer risk among Asian populations. Search limitations include dAsian populations residing in Asia, being published from the year 2008 till present, and written in the English language. A total of 16 articles were included in this systematic review. We found that red meats, processed meats, preserved foods, saturated/animal fats, cholesterol, high sugar foods, spicy foods, tubers or refined carbohydrates have been found by most studies to have a positive association with colorectal cancer risk. Inversely, calcium/dairy foods, vitamin D, general vegetable/fruit/fiber consumption, cruciferous vegetables, soy bean/soy products, selenium, vitamins C,E and B12, lycophene, alpha-carotene, beta-carotene, folic acid and many other vitamins and minerals play a protective role against colorectal cancer risk. Associations of fish and seafood consumption with colorectal cancer risk are still inconclusive due to many varying findings, and require further more detailed studies to pinpoint the actual correlation. There is either a positive or no association for total meat consumption or white meats, however their influence is not as strong as with red and processed meats.

품종 및 수확시기에 따른 당근의 Retinol Equivalent 측정 (Determination of Retinol Equivalent of Carrots according to Varieties and Harvest Time)

  • An investigation was made of the effect of carrot variety and harvest time (DAP) on the composition of carotenoids and the Retinol Equivalent value by column chromatography, and of the relation of the total carotenoid content to the Retinol Equivalent by regression analysis. The results are summarized as follows : 1. There were very significant differences of total carotenoid, ${\alpha}-carotene,\;and\;{\beta}-carotene$ contents among carrot varieties and between two harvest times(90 DAP, 99 DAP). Especially, each component of carotenoids in carrots harvested at 99 DAP attained higher concentrations than 90 DAP. 2. Retinol Equivalent value showed the tendency to increase as the numbers of DAP incr The Shindaehyung-Ochon and Hongshim-Ochon varieties had the highest RE. value. 3. In the composition of carotenoids and Retinol Equivalent value, the Shamgae-Ochon variety had the nearest value to the mean of all variety. Therefore, it is most reasonable to use the Shamgae-Ochon variety for the analysis of vitamin A value in carrots. 4. The regression of the totel carotenoid (x) to the Retinol Equivalent (y) was y = 0.074 + 0.12x $(r^2$ = 0.91). So, if total carotenoid content is determined, R.E. value can be predicted by this regression equations, saving time and labor.

옥수수 기능성 물질의 종류와 변이 (Functional Ingredients of Maize and Their Variation)

  • The objective of this report is to review the up to present papers including 79 references about functional ingredients in maize and their variations. Maize is a good source of such antioxidants as ${\alpha}-,\;{\gamma}-tocopherols,\;{\alpha}-,\;{\beta}-carotene,\;{\beta}-cryptoxanthin$, zeaxanthin, quercetin and chlorogenic acid. Unsaponifiable standard extracts of Zea mays L. (ZML) have a beneficial effect on various dental diseases. Flavonoids, flavonoid biosynthetic pathway and their controling genes were also reviewed in this report. Flavonoids are required for successful fertilization in maize pollen, and maysin has a potent antibiotic activities to earworm and fall armyworm in maize silks. Maize silks have been used in traditional medicine for diuretic, vascular diseases and diabetes, etc. and boiling water extracts of maize silk have hemodynamic effects, but their ingredients still remain obscure.

잣 지방질의 자동산화에 따른 토코페롤 및 카로티노이드의 변화 (Changes in the Concentration of Tocopherol and Carotenoid in Pinenut Oil during Autoxidation)

  • 잣 지방질을 정제 단계를 달리하여 자동산화 시키면서 토코페롤 및 카로티노이드의 변화를 살펴 보았다. 정제 단계를 거칠수록 산화 안정성이 감소되었으며 총 토코페롤의 함량은 조지방의 경우 55.51mg/100g oil이었으며 이 중 $\alpha$-토코페를 25.48mg, ${\gamma}$-토코페를 23.94mg, $\beta$-토코페를 6.99mg순으로 함유되어 있었으며 $\delta$-토코페롤은 흔적량 존재하였다. 탈검된 지방질 및 탄산된 지방질에서도 $\alpha$-토코페를이 23.85mg, 24.08mg, ${\gamma}$-토코페롤이 19.79mg, 20.04mg, 그리고 $\beta$-토코페롤은 6.12mg, 5.33mg함유되어 있었다. 카로티노이드 중 $\beta$-카로틴은 비 비누화물질 중의 0.63%이었으며 리코펜은 미량 함유되어 있었다. 지방질의 자동산화가 진행되는 동안 카로티노이드와 토코페롤은 감소되었으며, 특히 자동산화의 초기과정에서 항산화제로 작용하여 상당량이 감소된 것으로 여겨졌으며 그 감소속도는 $\alpha$-토코페롤, ${\gamma}$-토코페롤 순이었다.

넙치와 방어의 Carotenoid 색소성분 (Carotenoid Pigments of Flounder and Yellowtail)

  • 천연산 및 양식산 넙치와 방어의 표피 carotenoid를 TLC, column chromatography, UV-visible spectropho-tometer 및 co-TLC로 비교하여 다음과 같은 결과를 얻었다. 넙치의 총 carotenoid함량은 양식산 1.16mg%로서 천연산 1.38mg%보다 낮은 함량치를 보였고, carotenoid조성은 양식산이 lutein, zeaxanthin, tunaxanthin, triol 및 $\alpha$-cryptoxanthin의 순으로 함유하였고, 천연산은 tunaxanthin, zeaxanthin, triol, lutein 및 $\alpha$-cryptoxanthin의 순으로 함유하며, 천연산은 양식산에 비하여 tunaxanthin, triol의 함량이 논은 반면, lutein, zeaxanthin의 함량에서 휠씬 낮은 함량치를 보여 차이가 있었다. 방어의 총 carotenoid함량은 양식산 0.09 mg%로서 천연산 1.08mg%보다 휠씬 낮은 함량치를 보였고, carotenoid조성은 양식산, 천연산 모두 tunax-anthin의 획분이 총 carotenoid의 약 95%로 대부분을 차지 하며, tunaxanthin은 tunaxanthin A, tunaxanthin B 및 tunaxanthin C의 세가지 혼합물로 확인되어 서로 유사하였으나, tunaxanthin A와 tunaxanthin B의 함량에서 다소의 차이를 보였다. 따라서 어류의 총 carotenoid 함량과 carotenoid의 구성성 분은 어종 뿐만 아니라 먹이와 생육조건 등의 서식환경에 의해서도 상당한 차이가 있는 것으로 나타났다.

Glutathione S-transferase (GST) 유전자 다형성에 따른 우리나라 젊은 성인의 항산화 상태, DNA 손상 및 지질 양상 (Antioxidative Status, DNA Damage and Lipid Profiles in Korean Young Adults by Glutathione S-Transferase Polymorphisms)

  • Oxidative stress leads to the induction of cellular oxidative damage, which may cause adverse modifications of DNA, proteins, and lipids. The production of reactive species during oxidative stress contributes to the pathogenesis of many diseases. Antioxidant defenses can neutralize reactive oxygen species and protect against oxidative damage. The aim of this study was to assess the antioxidant status and the degree of DNA damage in Korean young adults using glutathione s-transferase (GST) polymorphisms. The GSTM1 and GSTT1 genotypes were characterized in 245 healthy young adults by smoking status, and their oxidative DNA damage in lymphocytes and antioxidant status were assessed by GST genotype. General characteristics were investigated by simple questionnaire. From the blood of the subjects, GST genotypes; degree of DNA damage in lymphocytes; the erythrocyte activities of superoxide dismutase, catalase, and glutathione peroxidase; plasma concentrations of total peroxyl radical-trapping potential (TRAP), vitamin C, ${\alpha}$- and ${\gamma}$-tocopherol, ${\alpha}$- and ${\beta}$-carotene and cryptoxanthin, as well as plasma lipid profiles, conjugated diene (CD), GOT, and GPT were analyzed. Of the 245 subjects studied, 23.2% were GSTM1 wild genotypes and 33.4% were GSTT1 wild genotype. No difference in erythrocyte activities of superoxide dismutase, catalase, or glutathione peroxidase, and the plasma TRAP level, CD, GOT, and GPT levels were observed between smokers and non-smokers categorized by GSTM1 or GSTT1 genotype. Plasma levels of ${\alpha}$- and ${\gamma}$-tocopherol increased significantly in smokers with the GSTT1 wild genotype (p < 0.05); however, plasma level of ${\alpha}$-carotene decreased significantly in non-smokers with the GSTM1 wild genotype (p < 0.05). DNA damage assessed by the Comet assay was significantly higher in non-smokers with the GSTM1 genotype; whereas DNA damage was significantly lower in non-smokers with the GSTT1 genotype. Total cholesterol and LDL cholesterol levels were significantly higher in non-smokers with the GSTT1 genotype than those with the GSTT1 wild genotype (p < 0.05). In conclusion, the GSTM1 genotype or the GSTT1 wild genotype in non-smokers aggravated their antioxidant status through DNA damage of lymphocytes; however, the GSTT1 wild type in non-smokers had normal plasma total cholesterol and LDL-cholesterol levels. This finding confirms that GST polymorphisms could be an important determinant of antioxidant status and plasma lipid profiles in non-smoking young adults. Further study is necessary to clarify the antioxidant status and/or lipid profiles of smokers with the GST polymorphism and to conduct a study with significantly more subjects.

Protective Effect of Artificially Enhanced Level of L-Ascorbic Acid against Water Deficit-Induced Oxidative Stress in Rice Seedlings

  • Effects of the enhanced level of L-ascorbic acid (AA) on the water deficit-induced oxidative damage were studied in rice (Oryza sativa L.) seedlings. The seedlings sprayed with 20 to 80 mM L-galactono-${\gamma}$-lactone (GL), a putative precursor of AA, showed 2 to 5-fold higher levels of AA compared with controls. Pretreatment of the seedlings with GL prior to water stress imposition caused virtually no effect on dehydration of tissues during water deficit but substantially mitigated oxidative injury, as accessed by 2-thiobarbituric acid-reactive substances, ${\alpha}$-tocopherol, chlorophylls and ${\beta}$-carotene. Proline accumulation during water stress was also significantly lowered in the treated seedlings. In a complementary experiment, AA retarded photodegradation of ${\alpha}$-tocopherol in isolated thylakoids far more efficiently than glutathione. GL in itself did not show any noticeable reactivity toward ${\alpha}$-tocopheroxyl radical. The results demonstrate the antioxidative function of AA in rice seedlings encountering water-limited environments, suggesting a critical role of AA as a defense against oxidative stress in plants.

녹차, 우롱차 및 홍차의 일반성분, 아미노산, 비타민류, 카테킨류 및 알카로이드류의 성분분석에 관한 연구 (A Study on the Content of General Compounds, Amino Acid, Vitamins, Catechins, Alkaloids in Green, Oolong and Black Tea)

  • 항산화효과, 항돌연변이효과, 콜레스테롤농도 상승억제작용이 우수한 것으로 알려진 차추출물의 주요성분을 분리 정량하기 위하여 각 발효정도에 따른 분류 및 녹차의 채엽시기별에 따라 녹차 1번차 녹차2번차 녹차 3번차, 우롱차 및 홍차 등에 대한 일반성분, 아미노산, Vit.C, Vit. E, ${\beta}-carotene$, 카테킨류, 카페인, 데오브로마인 및 데오필린 등을 분석하고 특히 항산화성분인 카테킨류의 항산화효과 및 항돌연변이 효과의 연구에 기초자료로 이용하고자 하였다. 녹차 I, II, III, 우롱차 및 홍차의 일반 성분중 조지방함량은 각각 1.1, 2.5, 4.9, 0.8 및 1.2% 이었으며, 총 아미노산함량은 녹차I>녹차II>녹차III의 순이었고 대부분의 아미노산은 차 성분의 유일한 아미노산인 theanine이 차지하였다. 녹차 I, II, III, 우롱차 및 홍차에 함유된 Vit. C함량은 각각 101.6, 87.5, 95.9, 99.1 및 108.0 mg%이었으며, ${\beta}-carotene$ 함량은 각각 270, 268, 481, 80 및 181 ppm이었고, Vit. E 함량은 ${\alpha}-tocopherol$이 대부분을 차지하였으며 ${\beta}-$${\delta}-tocopherol$은 모두 검출되지 않았다. 한편, 총카테킨류의 함량은 각각 10.5, 10.4, 7.2, 8.4 및 1.8%로 총카테킨류중 EGCG의 함량이 가장 높았다. 카테킨류 중 EGC의 함량은 녹차I>녹차III>녹차II>우롱차>홍차의 순으로 녹차I이 가장 높았으며, EGCG와 ECG의 함량은 우롱차>녹차I>녹차II>녹차III>홍차의 순으로 우롱차가 가장 높았다. 또한 GA함량은 0.01, 0.02, 0.05, 0.13 및 0.31%로 홍차에 가장 많은 것으로 나타났으며, 카페인 및 데오브로마인 함량도 홍차의 경우가 가장 높았으나 데오필린 함량은 녹차I이 가장 높게 나타났다.

$^1O_2$으로 유도된 사람 적혈구의 광용혈에 있어서 대두의 아이소플라본인 제니스테인의 보호작용 (Protective Effect of Isoflavone, Genistein from Soybean on Singlet Oxygen Induced Photohemolysis of Human Erythrocytes)

  • 자외선에 노출된 피부에서 생성된 활성산소종에 의한 산화적 스트레스는 세포 구성 성분들을 손상시키고 궁극적으로는 피부 광노화 및 발암을 야기시킬 수 있다. 이 연구에서는 대두(Glycine max MERRILL)의 성분인 genistein(4',5,7-trihydroxyisoflavone)을 포함하는 몇 가지 천연 황산화제들에 대하여 로즈벵갈로 증감된 사람 적혈구의 광용혈에 대한 보호효과를 조사하였다. Genistein$(10-100\;{\mu}M)$은 농도-의존적으로 광용혈을 억제하였다. 그리고 지질 과산화 연쇄반응의 차단제인 ${\alpha}$-tocopherol보다도 더 효과적이었다. 그러나, Genitein의 배당체인 genistin, 소용성 항산화제인 L-ascorbate, 철-킬레이트제인 sodium phytate는 광용혈에 대하여 보호 효과를 나타내지 않았다. 오히려 보다 높은 농도$(500\;{\mu}M)$에서는 L-ascorbate와 sodium phytate 모두 광용혈을 촉진시켰다. 반면, $^1O_2$ 소광제로 알려진, ${\alpha}$-carotene 3,3'-diol은 현저한 보호 효과를 보여주었다. 이 결과는 로즈벵갈로 증감된 광용혈에 있어서 $^1O_2$이 중요함을 가리킨다. Luminol-의존성 화학발광법을 이용하여, $Fe^{3+}-EDTA/H_2O_2$ 계에서 생성된 ROS에 대한 몇가지 천연 항산화제들의 ROS 소거활성을 측정하였다. 활성 산소 소거활성$(OSC_{50})$의 크기는 sodium phytate > L-ascorbate>${\alpha}$-tocopherol > genistein > genistin순이었다. Gnistein, genistin, ${\alpha}$-tocopherol, L-ascorbate 및 sodium phytate의 활성산소 소거활성$(OSC_{50})$은 각각 41.0, 109.0, 9.0, 5.2, 및 $0.56{\mu}M$이었다. Free radical 소거활성$(FSC_{50})$의 크기는 L-ascorbate > ${\alpha}$-tocopherol > genistein > genistin순으로 나타났다. 이상의 결과들은 genistein이 $^1O_2$ 혹은 다른 ROS를 소거함으로써 그리고 ROS에 대항하여 세포막을 보호함으로써 생체계, 특히 태양 자외선에 노출된 피부에서 항산화제로서 작용할 수 있음을 가리킨다.
