Relationship between Molecular Structure Characteristics of Feed Proteins and Protein In vitro Digestibility and Solubility

  • The nutritional value of feed proteins and their utilization by livestock are related not only to the chemical composition but also to the structure of feed proteins, but few studies thus far have investigated the relationship between the structure of feed proteins and their solubility as well as digestibility in monogastric animals. To address this question we analyzed soybean meal, fish meal, corn distiller's dried grains with solubles, corn gluten meal, and feather meal by Fourier transform infrared (FTIR) spectroscopy to determine the protein molecular spectral band characteristics for amides I and II as well as ${\alpha}$-helices and ${\beta}$-sheets and their ratios. Protein solubility and in vitro digestibility were measured with the Kjeldahl method using 0.2% KOH solution and the pepsin-pancreatin two-step enzymatic method, respectively. We found that all measured spectral band intensities (height and area) of feed proteins were correlated with their the in vitro digestibility and solubility ($p{\leq}0.003$); moreover, the relatively quantitative amounts of ${\alpha}$-helices, random coils, and ${\alpha}$-helix to ${\beta}$-sheet ratio in protein secondary structures were positively correlated with protein in vitro digestibility and solubility ($p{\leq}0.004$). On the other hand, the percentage of ${\beta}$-sheet structures was negatively correlated with protein in vitro digestibility (p<0.001) and solubility (p = 0.002). These results demonstrate that the molecular structure characteristics of feed proteins are closely related to their in vitro digestibility at 28 h and solubility. Furthermore, the ${\alpha}$-helix-to-${\beta}$-sheet ratio can be used to predict the nutritional value of feed proteins.

Comparison of Inclusion Complex Formation Capacity of Cyclodextrins with Various Molecules and Characterization of Cyclodextrin-fatty Acid Complex (Cyclodextrin의 Inclusion Complex 형성능과 Fatty Acid와의 Complex 형성조건과 특성)

  • The capacity of inclusion complex formation between ${\alpha}$-, ${\beta}$-, ${\gamma}$-cyclodextrins(CDs) and various compounds, such as pH indicators, biloslalns, glycoside, amino acid, and fatty acids, was compared. Fatty acid was identified as the most suitable ligand for fractionation of CDs in terms of capacity and selectivity. The effects of complex formation conditions, such as, mixing ratio of CD and fatty acid, pH, ionic strength, and temperature, on the capacity of fatty acrid-CD complex was also investigated. The carbon number of fatty acids was identified as the most significant factor determining the capacity and selectivity of inclusion complex formation of CDs. Capric acid(C10) and palmitic acid(C16) showed high specificity for ${\alpha}$- and ${\beta}$-CDs, respectively. Under the optimal conditions, the molar ratio of complex formed was found to be 1.0:2.6 for ${\alpha}$-CD/capric acid and 1.0:1.9 for ${\beta}$-CD/palmitic acid. X-ray diffraction and infrared spectrum of the formed inclusion complex were analyzed. The changes of enthalpy($\Delta$H) of the inclusion complex formation reaction was evaluated by differential scanning calorimetry, showed that the reaction was endothermic.

The Changes of Electroencephalography According to Emotional Stimulus in Sasangin (정서자극이 사상인(四象人)의 뇌파 변화에 미치는 영향)

  • 1. Objectives The objective of this study was to measure the differences of EEG changes accoring to emotional stimulus in Sasangin. 2. Methods 44 healthy volunteers(Soyangin 10, Soeumin 14, Taeumin 20) were participated. Sasang Constitution was diagnosed by Sasang specialist. Volunteers watched horror movie as the emotional stimulus. We recorded EEG data during pre-stimulus rest(120sec), during-stimulus(197sec), rest-stimulus(120sec). Power spectral analysis was done and relitive power and ${\alpha}/{\beta}$ ratio were compared among each groups. 3. Results (1) The mean of relative ${\alpha}$ of Taeumin was significantly decreased according to stimulus compared with Soyagnin. (2) Relative ${\alpha}$ of Soeumin was significantly decreased in T3 area according to stimulus compared with Taeumin. (3) Relative ${\beta}$ of Taeumin group was significantly increased in Fp2 area according to stimulus compared with Soeumin. 4) Relative $high-{\beta}$ of Soyangin group was significantly decreased in F3 by the post-stimulus rest compared with Soeumin. (5) ${\alpha}/{\beta}$ ratio of Soeumin was significantly increased according to stimulus compared with other groups. 4. Conclusions Soyangin showed sensitive changes according to the emotional simulus compared with Soeumin and Taeumin. Above results suggest that the emotional characteristics of Sasangin can be measured by objective methods as by EEG.

The Effect of Electroacupuncture at Sobu(HT8) on the EEG and HRV (소부(HT8) 전침이 뇌파(EEG)와 심박변이도(HRV)에 미치는 영향)

  • Objectives : The aim of this study was to examine the effect of electroacupuncture(EA) at an acupoint, HT8(Sobu), on normal humans by using power spectral analysis. We examined the effect on the Heart Rate Variability(HRV), and the balance of the autonomic nervous system. Methods : Thirty-two healthy volunteers participated in this study. EEG(Electroencephalogram) power spectrum exhibits site-specific and state-related differences in specific frequency bands. A thirty-two channel EEG study was carried out on thirty-two subjects(14 males; mean age=23.5 years old, 18 females; mean age=21.5 years old). HRV and EEG were simultaneously recorded before and after acupuncture. Results : In the ${\alpha}$(alpha) band, during the HT8-acupoint treatment, the power values in the ${\alpha}$(alpha) band significantly decreased(p<0.05) at 28 channels. In the ${\beta}$(beta) band significantly decreased(p<0.05) at 26 channels. In ${\delta}$(delta) band significantly decreased(p<0.05) at 18 channels. In ${\theta}$(theta) band significantly decreased(p<0.05) at 20 channels. ${\alpha}/{\beta}$ values were increased at 6 channels and decreased at 10 channels.${\beta}/{\theta}$ values were increased at 10 channels and decreased at 19 channels. Mean-RR(RR-interval), Complexity, RMSSD(Root mean square of successive differences), SDSD(Standard deviations differences between adjacent normal R-R intervals), norm HF showed a significantly increased and mean-HRV, norm LF, LHR(LF/HF Ratio) showed a significantly decreased after HT8-acupoint treatment(p<0.05). Conclusions : These results suggest that EA at the HT8 mostly causes significant changes on alpha(28 channels), beta(26 channels), delta(18 channels), theta(20 channels) bands and mean-HRV, mean-RR, complexity, RMSSD, SDSD, norm LF, norm HF and LHR. If practicing EA at the HT8, it will regulate the function of the cerebral cortex, decrease activity of the sympathetic and increase parasympathetic nervous activity.

Effect of Dietary Starch Level and Kind on the Growth and Body Composition of Juvenile Olive Flounder Paralichthys olivaceus

  • A 7-week feeding trial was conducted to investigate the effect of dietary starch level and kind on the growth and body composition of juvenile olive flounder. Triplicate groups of fish (average weight: 1.5 g) were fed iso-nitrogenous (48% crude protein) and isocaloric (4.8 kcal/g diet) diets containing 15-25% ${\alpha}$-potato starch and 15% ${\beta}$-potato starch. Survival was not affected by dietary starch level and kind. The weight gain of fish fed the diet containing 20% ${\alpha}$-potato starch was significantly higher than that of fish fed the diets containing 15% and 25% ${\alpha}$-potato starch levels. The feed efficiency and protein efficiency ratios of fish fed the diets containing 15% ${\beta}$-potato starch were significantly lower than those of the other groups (P < 0.05). The protein efficiency ratio tended to increase with increasing ${\alpha}$-potato starch. The daily feed intake of fish fed the diet containing 15% ${\beta}$-potato starch was significantly higher than that of the other groups (P < 0.05). The hepatosomatic index, condition factor, and proximate composition of the whole body were not affected by the dietary starch level and kind. These results indicate that up to 20% ${\alpha}$-potato starch could be incorporated into the juvenile flounder diet for optimum growth.

  • The neutron decay constant, ${\alpha}$, and effective delayed neutron fraction, ${\beta}_{eff}$, are important parameters for the control of the dynamic behavior of nuclear reactors. For the heavy water zero power reactor (HWZPR), this document describes the measurements of the neutron decay constant by noise analysis methods, including variance to mean (VTM) ratio and endogenous pulse source (EPS) methods. The measured ${\alpha}$ is successively used to determine the experimental value of the effective delayed neutron fraction as well. According to the experimental results, ${\beta}_{eff}$ of the HWZPR reactor under study is equal to 7.84e-3. This value is finally used to validate the calculation of the effective delayed neutron fraction by the Monte Carlo methods that are discussed in the document. Using the Monte Carlo N-Particle (MCNP)-4C code, a ${\beta}_{eff}$ value of 7.58e-3 was obtained for the reactor under study. Thus, the relative difference between the ${\beta}_{eff}$ values determined experimentally and by Monte Carlo methods was estimated to be < 4%.

Synthesis of Silicon Nitride from Kimcheon Quartzite (김천규석으로부터 질화규소의 합성)

  • Silicon nitride powders were prepared by the simultaneous reduction and nitridation from powder mixtures of Kim cheon quartzite and carbon (graphite or carbon black) at1400$^{\circ}C$ for 10 hours in nitrogen atmosphere. The effects of the reaction variables on the yield of products and on the ${\alpha}$/${\beta}$ ratio were examined. The average particle size, density, and the ${\alpha}$/${\beta}$ ratio of the obtained si3N4 were 1.0$\mu\textrm{m}$, 3.10g/㎤ and 90/10, respectively. It was found that the Si3N4 powders obtained in this work were comparable to the foreign commercial products.

Effects of Auditory Stimuli Using Pop Music on EEG (대중음악이 뇌파에 미치는 영향)

  • The purpose of this study is to analyze the effects of the auditory stimulation using pop music on EEG. The subjects in the study were eight healthy university students; five men and three women, age group of 22-28 years old. EEGs were measured for one minute at 19 channels, according to the international 10-20 system method, after both pop musics which the subject likes and not were provided with 100% and 50% volume of standard value, respectively, and the ratio of $\beta$/$\alpha$ was obtained. As a result, when the subjects heard their favorite music, the values of $\beta$/$\alpha$ ratio at the Right-Parietal.Temporal.Occipital lob(T6, O2) showed to be lower than stable state in the situation with 50% volume of standard value.

Inhibitory Effect of Amygdalin on Lipopolysaccharide-Inducible TNF-$\alpha$ and IL-l$\beta$ mRNA Expression and Carrageenan-Induced Rat Arthritis

  • Amygdalin is a cyanogenic glycoside plant compound found in the seeds of rosaceous stone fruits. We evaluated the anti-inflammatory and analgesic activities of amygdalin, using an in vitro lipopolysaccharide (LPS)-induced cell line and a rat model with carrageenan-induced ankle arthritis. One mM amygdalin significantly inhibited the expression of TNF-$\alpha$ and IL-l$\beta$ mRNAs in LPS-treated RAW 264.7 cells. Amygdalin (0.005, 0.05, and 0.1 mg/kg) was intramuscularly injected immediately after the induction of carrageenan-induced arthritic pain in rats, and the anti-arthritic effect of amygdalin was assessed by measuring the weight distribution ratio of the bearing forces of both feet and the ankle circumference, and by analyzing the expression levels of three molecular markers of pain and inflammation (c-Fos, TNF-$\alpha$, and IL-l$\beta$) in the spinal cord. The hyperalgesia of the arthritic ankle was alleviated most significantly by the injection of 0.005 mg/kg amygdalin. At this dosage, the expressions of c-Fos, TNF-$\alpha$, and IL-l$\beta$ in the spinal cord were significantly inhibited. However, at dosage greater than 0.005 mg/kg, the pain-relieving effect of amygdalin was not observed. Thus, amygdalin treatment effectively alleviated responses to LPS-treatment in RAW 264.7 cells and carrageenan-induced arthritis in rats, and may serve as an analgesic for relieving inflammatory pain.

A Study on Synthesis of High Purity $\beta$-SiC Fine Powders from Ethyl Silicate(III) Effect of Additives (Ethyl Silicate를 이용한 고순도 $\beta$-SiC 미분말 합성에 관한 연구(III) 첨가제의 영향)

  • The particle size of synthesized SiC powders was decreased with increasing carbon content when the mixture of carbon and silica was carbonized at 1, 45$0^{\circ}C$ after hydrolysis of the mixture with the ranges of 3.1 to 3.5 in the mole ratio of Carbon/Alkoxide. The reacted fraction of $\beta$-SiC nearly had nothing to do with the mole ratio of Carbon/Alkoxide. When the reaction was made by adding 0.5wt% additives in the composition of 3.1 in the mole ratio of carbon/alkoxide, the additives decreased the yield of $\beta$-SiC and its sequence was Ba2O3>B>Fe>Al>Al2O3>Si. The effect of additives promoted the transformation of $\beta$-SiC to $\alpha$-SiC form and shwoed the increasing tendency of lattice constant. The two colors of $\beta$-SiC powder came out : one was the black grey with addition of Al, Al2O3 and B the other the light grey with addition of Fe, B2O3 and Si.
