Purification and Biological Activities of MT 1155 Inducing Morphological Change of Rous Sarcoma Virus-Transformed Normal Rat Kidney Cell (Rous Sarcoma Virus에 의해 형질전환된 NRK 세포의 형태변화를 유도하는 활성물질 MT 1154의 분리와 생물학적 활성)

  • We isolated Actinomycetes strain GMT 1155 and purified the active compound, MT 1155, on the morphological reversion of ts/NRK cell from the isolate. MT 1155 was identified as toyocamycin having antifungal and antitumor activities from physico-chemical properties and UV, IR, $^1H$-NMR, $^13C$-NMR and mass spectrum. MT 1155 showed the morphologically reversional activity on ts/NRK cell and the cytotoxicity on CTLL cell at the final concentrations of 1.7 JlM and 0.2 11M, respectively and its $IC_{50}$ value on protein kinase A enzyme was 2.3 $\mu$M. Also it had strong antifungal activity against several pathogenic fungi but not antibacterial activity. And it did not inhibit both protein kinase C activity and the bleb-formation of K562 cell induced by phorbol esters.

Purification and NMR Studies of RNA Polymerase II C-Terminal Domain Phosphatase 1 Containing Ubiquitin Like Domain

  • RNA polymerase II C-terminal domain phosphatase 1 containing ubiquitin like domain (UBLCP1) has been identified as a regulatory molecule of RNA polymerase II. UBLCP1 consists of ubiquitin like domain (UBL) and phosphatase domain homologous with UDP and CTD phosphatase. UBLCP1 was cloned into the E.coli expression vectors, pET32a and pGEX 4T-1 with TEV protease cleavage site and purified using both affinity and gel-filtration chromatography. Domains of UBLCP1 protein were successfully purified as 7 mg/500 mL (UBLCP1, 36.78 KDa), 32 mg/500 mL (UBL, 9 KDa) and 8 mg/500 mL (phosphatase domain, 25 KDa) yielded in LB medium, respectively. Isotope-labeled samples including triple-labeled ($^2H/^{15}N/^{13}C$) UBLCP1 were also prepared for hetero-nuclear NMR experiments. $^{15}N-^{1}H$ 2D-HSQC spectra of UBLCP1 suggest that both UBL and phosphatase domain are properly folded and structurally independent each other. These data will promise us further structural investigation of UBLCP1 by NMR spectroscopy and/or X-ray crystallography.

  • The root of Astragalus membranaceus is a traditional folk medicine that has been used for many therapeutic purposes in Asia. It reportedly acts as an immunostimulant, tonic, hepatoprotective, diuretic, antidiabetic, analgesic, expectorant, sedative, and anticancer drug. In this study, metabolomic profiling was applied to the roots of A. membranaceus of different ages using NMR coupled with two multivariate statistical analysis methods: such as principal components analysis (PCA) and canonical discriminant analysis (CDA). This allowed various metabolites to be assigned in NMR spectra, including $\gamma$-aminobutyric acid (GABA), aspartic acid, succinic acid, glutamic acid, glutamine, N-acetyl aspartic acid, acetic acid, arginine, alanine, threonine, lactic acid, and valine. The score plot from PCA and also CDA allowed a clear separation between samples according to age.

Isolation of a New Herbicidal Compound Angelicin from Curly Dock (Rumex crispus L.) (소리쟁이(Rumex crispus L.)로부터 신규 살초활성물질 Angelicin의 분리)

  • The objective of this study was to isolate a herbicidally active compound from curly dock (Rumex crispus), a native weed in Korea and to identify its' chemistry. The $GR_{50}$ value of methanol extracts which is determined by a seed bioassay using rapeseed (Brassica napus) seedlings was $935\;{\mu}g\;g^{-1}$. Activity-directed isolation of ethylacetate extract led to the isolation of ECDA fraction with $GR_{50}$ value $53\;{\mu}g\;g^{-1}$. Based on data of GC-MS, $^1H$-NMR and $^{13}C$-NMR, the chemical structure of ECDA was determined as 2H-furo[2,3-H]-[1]-benzopyran-2-one which is known as angelicin. The $GR_{50}$ values of angelicin to barnyardgrass (Echinochloa crus-galli), southern crabgrass (Digitaria ciliaris), and indian jointvetch (Aeschynomene indica) were 426, 66 and $216\;{\mu}g\;g^{-1}$, respectively. Our results suggest that angelicin could be used as a lead compound for the development of new herbicides.

  • Two furanosesquiterpenes, (5'E)-5-(2',6'-dimethylocta-5',7'-dienyl) furan-3-carboxylic acid (1) and (1'E,5'E)-5-(2',6'-dimethylocta-l',5',7'-trienyl) furan-3-carboxylic acid (2), were isolated from soft coral Sinularia lochmodes collected from Palikir pass at Pohnpei Micronesia, June, 1990 in Hawaii. Their structures were elucidated by $^1H$, $^{13}C$ NMR, Homo-COSY, $^1H$-$^{13}C$ (1 bond) Heteronuclear Multiple Quantum Coherence Spectroscopy (HMQC), $^1H$-$^{13}C$ (2 and 3 bond) Heteronuclear Multiple Bond Coherence Spectroscopy (HMBC), Electron Impact Mass Spectroscopy (EI-ms), and Infrared Spectroscopy (IR).

  • Antibiotic HS-2 was purified from the culture broth of Streptomyces plicatosporus which was isolated from soil, by solvent extraction, silica gel column chromatography and gel filtration. Through the analysis of UV, $^{1}$H-NMR, $^{13}$C-NMR spectrum, HS-2 was identified as rubiginone B$_{2}$. It was confirmed that HS-2 enhanced the cytotoxicity of colchicine against multidrug-resistant tumor cells.

New Cryptand Complexes of Lanthanides(Ⅲ) and Dioxouranium(Ⅵ) Nitrates

  • The following new cryptand 221 complexes of lanthanides(Ⅲ) and dioxouranium(Ⅵ) nitrate have been synthesized: $(Ln(C_{16}H_{32}N_2O_5)(H_2O)_2(NO_3)_3\ and \((UO_2)_2(C_{16}H_{32}N_2O_5)(H_2O)_4(NO_3)_4$. These complexes have been identified by elemental analysis, moisture titration, conductivity measurements and various spectroscopic techniques. The proton and carbon-13 NMR as well as calorimetric measurements were used to study the interaction of cryptand 221 with La(Ⅲ), Pr(Ⅲ ), Ho(Ⅲ) and $UO_2(Ⅱ)$ ions in nonaqueous solvents. The bands of metal-oxygen atoms, metal-nitrogen atoms and O-U-O in the IR spectra shift upon complexation to lower frequencies, and the vibrational spectra ({\delta}NMN$) of metal-amide complexes in the crystalline state exhibit lattice vibrations below 300 $cm^{-1}$. The NMR spectra of the lanthanides(Ⅲ) and dioxouranium(Ⅵ) nitrate complexes in nonaqueous solvents are quite different, indicating that the ligand exists in different conformation, and also the $^1H$ and $^{13}C-NMR$ studies indicated that the nitrogen atom of the ring has greater affinity to metal ions than does the oxygen atom, and the planalities of the ring are lost by complexation with metal ions. Calorimetric measurements show that cryptand 221 forms more stable complexes with $La^{3+}$ and $Pr^{3+}$ ions than with $UO^{22+}$ ion, and $La^{3+}/Pr^{3+}$ and $UO^{22+}/Pr^{3+}$ selectivity depends on the solvents. These changes on the stabilities are dependent on the basicity of the ligand and the size of the metal ions. The absorption band (230-260 nm) of the complex which arises from the direct interaction of macrocyclic donor atoms with the metal ion is due to n-{\delta}*$ transition and also that (640-675 nm) of $UO^{22+}$-cryptand 221 complex, which arises from interaction between two-dioxouranium(Ⅵ) ions in being out of cavity of the ligand ring is due to d-d* transition.

Determination of Isoprenyl and Lavandulyl Positions of Flavonoids from Sophora flavescens by NMR Experiment

  • All fifteen flavonoids (1-15) have been isolated from the roots of Sophora flavescens (Leguminosae) as active principles of the cytotoxic property toward human tumor cell lines such as A549, SK-OV-3, SK-Mel-2, XF498 and HCT15, in vitro. By means of spectral analyses, particularlyby the aid of various two dimensional NMR experiments, all $^1H-NMR$ ad $^{13}C$ -NMR signals of 1-15 were completely assigned, and thus the structures of 1-15 were established unambiguously.

NMR Studies of Metal-binding Luteinizing Hormone Releasing Hormone

  • Functions of the luteinizing hormone releasing hormone (LHRH) and its induced release by divalent metal ions have received great attention because this neurotransmitter subsequently regulates the secretion of luteinizing hormone (LH). Metal-LHRH complexes were synthesized by addition of various Cu(II),Ni(II),Zn(II) ions into LHRH in order to understand how the induced release of LHRH is possible. The degree of complexation was monitored by $^1H$, $^{13}C$-NMR chemical shifts, and final products were identified by Mass spectrometry. Solutionstate structure determination of Zn(II)-LHRH out of metal-complexes was accomplished by using NMR and NMR-based distance geometry (DG). Interproton distance information from nuclear Overhauser effect spectroscopy was utilized for structure determination. Structure obtained in this study has a cyclic conformation exhibiting a specific ${\alpha}$-helical turn with residue numbers His[2]-Leu[7] out of 10 amino acids. Comparison of chemical shifts and EPR studies of Ni(II),Cu(II)-LHRH complexes exhibit that these metal complexes have 4-coordination geometry.

A Study on the Chemical Composition and Structure of Sludge, Compost and Charcoal (폐수처리 슬럿지와 퇴비 및 목탄의 화학적 특성과 구조에 관한 연구)

  • To understand the chemical structure of sewer sludge in comparison with commercial compost and charcoal used as a soil improver, it was carried out to analyse their ash contents and metal ions, and to elucidate the chemical structure of their residuals after a sequential treatment of alcohol-benzene(1:2) extraction in Soxhlet, 3% HCl reflux and 79% H₂SO₄ hydrolysis, using CHNS analyzer and solid C-13 NMR spectrometer. The results obtained were as follows: 1. Ash content of sludge was about 46% that is higher than those of compost (17%) and charcoal (4%). 2. The residual of sludge after a sequential treatment of HCl and H₂SO₄ hydrolyses had high ash content about 23%, too. 3. The sludge seems to be suitable to the soil improver because the content of heavy metal ions in sludge was near the compost and below the organic fertilizer standard. 4. Elemental composition of sludge residual after HCl-H₂SO₄ hydrolyes was C/sub 56/H/sub 91/O/sub 12/N₂S = (C/sub 6/H/sub 10/O/sub 5/)/sub 7/(C/sub 6/H₄)/sub 7/C₂H/sub 43/O₂N₂S, similar to C/sub 103/H/sub 122/O/sub 33/N/sub 6/S = (C/sub 6/H/sub 10/O/sub 5/)/sub 6/(C/sub 6/H₄)/sub 10/C/sub 7/H/sub 22/O₃N/sub 6/S of compost. 5. The sludge residual had proved to have both considerable aliphatic and aromatic groups, but the compost residual to have mainly aliphatic groups and the charcoal to have mainly aromatic groups, through the peak analysis of solid C-13 NMR charts. 6. So, the sewer sludge is proved to have a considerable amount of aromaticity like in woody biomass containing lignin.
