Although ureases play important roles in microbial nitrogen metabolism and in the pathogenesis of several human diseases, little is known of the mechanism of metallocenter biosynthesis in this Ni-Containing enzyme. Klebsiella aerogenes urease apo-protein was purified from cells grown in the absence of Ni. The purified apo-enzyme showed the same native molecular weight, charge, and subunit stoichiometry as the holo-enzyme. Chemical modification studies were consistent with histidinyl ligation of Ni. Apo-enzyme could not be activated by simple addition of Ni ions suggesting a requirement for a cellular factor. Deletion analysis showed that four accessory genes (ureD, ureE, ureF, and ureG) are necessary for the functional incorporation of the urease metallocenter. Whereas the $\Delta$ureD, $\Delta$ureF, and $\Delta$ureG mutants are inactive and their ureases lack Ni, the $\Delta$ureE mutants retain partial activity and their ureases possess corresponding lower levels of Ni. UreE and UreG peptides were identified by SDS-polyacrylamide gel comparisons of mutant and wild type cells and by N-terminal sequencing. UreD and UreF peptides, which are synthesized at ve교 low levels, were identified by using in vitro transcription/translation methods. Cotransformation of E. coli cells with the complementing plasmids confirmed that ureD and ureF gene products act in trans. UreE was purified and characterized. immunogold electron microscopic studies were used to localize UreE to the cytoplasm. Equilibrium dialysis studies of purified UreE with $^{63}$ NiC1$_2$ showed that it binds ~6 Ni in a specific manner with a $K_{d}$ of 9.6 $\pm$1.3 $\mu$M. Results from spectroscopic studies demonstrated that Ni ions are ligated by 5 histidinyl residues and a sixth N or O atom, consistent with participation of the polyhistidine tail at the carboxyl termini of the dimeric UreE in Ni binding. With these results and other known features of the urease-related gene products, a model for urease metallocenter biosynthesis is proposed in which UreE binds Ni and acts as a Ni donor to the urease apo-protein while UreG binds ATP and couples its Hydrolysis to the Ni incorporation process.ouples its Hydrolysis to the Ni incorporation process.s.
The active sites of the nickel and iron-containing enzyme, carbon monoxide dehydrogenase (CODH) from clostridium thermoaceticum were investigated using Electron Paramagnetic Resonance (EPR) technique. CODH exhibits several spectral features called NiFeC, $g_{ave}=1.82$, $g_{ave}=1.86$. FCII signals which are originated from different clusters in this enzyme. CODH is know to catalyze two different kinds of reactions - acetyl-CoA synthesis and CO oxidation. The acetyl-CoA synthesis activity can be followed by monitoring CO/acetyl-CoA exchange. The addition of 1,10-phenanthroline (phen) to CODH selectively destroyed the CO/acetyl-CoA exchange activity and eliminated the NiFeC signal completely. CO oxidation activity and other EPR signals were unaffected. Such behavior demonstrates that CODH has two distinct active sites and that the NiFe complex is only responsible for the CO/acctyl-CoA exchange activity. Phen caused the removal of only 30% of Ni in the NiFe complex ($0.3Ni/{\alpha}{\beta}$) as shown by the quantitative metal analysis. The phen-treated CODH could be reactivated fully by incubation In $Ni^{2+}$ solution. Radioactive $^{63}Ni^{2+}$ was used to quantitate the amount of the $Ni^{2+}$ incorporated into phen-treated enzyme and showed that the amount was the same as the removed by the phen treatment. i.e. $0.3Ni/{\alpha}{\beta}$. This indicates that only 30% of NiFe complexes are labile and responsible for the CO/acctyl-CoA exchange activity, the other 70% are non-labile and have no exchange activity. This is the first clear evidence that the NiFe complex is heterogencous and labile and non-labile Ni sites arc interacting differently with substrates and chelating agents like phen.

Nickel (Ni) electroplating was implemented by using a metal Ni powder in order to establish a $^{63}Ni$ plating condition on the PN junction semiconductor needed for production of beta-voltaic battery. PN junction semiconductors with a Ni seed layer of 500 and $1000{\AA}$ were coated with Ni at current density from 10 to $50mA\;cm^{-2}$. The surface roughness and average grain size of Ni deposits were investigated by XRD and SEM techniques. The roughness of Ni deposit was increased as the current density was increased, and decreased as the thickness of Ni seed layer was increased. The results showed that the optimum surface shape was obtained at a current density of $10mA\;cm^{-2}$ in seed layer with thickness of $500{\AA}$, $20mA\;cm^{-2}$ of $1000{\AA}$. Also, pure Ni deposit was well coated on a PN junction semiconductor without any oxide forms. Using the line width of (111) in XRD peak, the average grain size of the Ni thick firm was measured. The results showed that the average grain size was increased as the thickness of seed layer was increased.
The species of four heavy metals (Cr, Cu, Ni and Pb) associated with sediments (viz exchangeable, carbonates, reducible, organic and residual fractions) were determined with respect to the particle sizes and depths at four locations of the lower Kumho river. In the exchangeable fraction, 3.7~19.52% of Ni and 2.8~14.81% of Pb were found, and in the carbonates fraction 2.12~19.43% of Ni and 1.39~15.42% of Pb were found. The reducible fraction retained about 8.66~44.93% of Cr, 0.41~9.79% of Cu, 17.38~35.74% of Ni and 9.5~44.89% of Pb. In the organic fraction about 0~21.06% of Cr, 2.95~35.74% of Cu, 0~14.66% of Ni and 0~10.65% of Pb were found. The residual fraction retained about 52.6~83.53% of Cr, 63.86~86.39% of Cu, 39.66~66.16% of Ni and 39.97~71.75% of Pb. The order of release or mobility of heavy metals was Ni > Pb > Cr > Cu. Mobile fraction of heavy metals by particle sizes (1.0~0.5 mm and 0.5~0.25 mm) was found to be higher in particle sizes 1.0~0.5 mm than that of 0.5~0.25 mm. The release or mobility of heavy metals by depths (0~5 cm, 5~10 cm and 10~15 cm) was found to be higher in the upper sediments than in the lower sediments, except Cu.
We have performed $^{63,65}$Cu nuclear quadrupole resonance (NQR) measurements on Zn and Ni doped YBa$_2Cu_3O_7$ (YBa$_2Cu_{3-x}M_xO_7$, M=Zn or Ni, x = 0.00 ${\sim}$ 0.09). Doping effects are markedly different in relaxation rates as well as in superconducting transition temperatures. Both the spin-lattice and the spin-spin relaxation rates decrease for Zn doped YBCO. However, those increase for Ni doped YBCO. This contrast in local electronic dynamics provides a clear microscopic evidence that Zn forms no local moment, while Ni develops a local moment. Consequently, the antiferromagnetic spin fluctuation is suppressed by Zn doping whereas it is preserved by Ni doping.

A reforming catalyst for hydrogen production from ammonia is being studied. Non-novel metal based Ni catalysts for use in ammonia reforming processes are being developed. In this study, the ammonia reforming characteristics according to Ni content of the alumina pellet supported catalyst in the mid-temperature region were investigated under different space velocity. 20 Ni and 3,000 h-1 showed the best catalytic activity with ammonia conversion of 63% among all conditions.

Effect of nickel powder to added to the hydrogen absorbing alloy electrode of $MmNi_{4.5}-xCoxMn_{0.3}Al_{0.2}$ system alloy was investigated. The addition of nickel powder was effective for the improvement of discharging characteristic. It was found that the discharge capacity was 310mAhig when the alloy negative electrode was mixed $MmNi_{3.75}CO_{0.75}Mn_{0.3}Al_{0.2}$ and nickel powder with a mix of one to three. Still another, we have investigated thermodynamic stability of hydrogen in the alloy negative electrode. As a result, enthalpy of hydrogen and hydrogen equilibrium pressure in the alloy negative electrode were a suitable value to easy hydrogen absorption-desorption.
In this study, the effect of mixing condition of raw material powders possessing various particle size and particle size distribution on thermite reaction efficiency was investigated. When fine raw powders are used, rather the reaction yield tends to decrease due to agglomeration. In contrast, coarse raw powders make deteriorate the contact area between raw material powders containing Al reducing agent. To ensure the optimal thermite reaction efficiency, it is required to optimize a mixture condition of raw material powders prior to thermite reaction. From the current experiment, the maximum thermite reaction efficiency is 77%, which came from Nb2O5 + NiO +Al mixtures with size distribution from 9.25 to 22.63 ㎛.
반도체에 이온화 방사선을 조사시키면 소자 내부에서 전자-전공쌍(Electron-Hole Pair, EHP) 생성현상이 나타난나. 이 발성 전하는 방사선원에 따라 반영구적인 전원으로 활용이 가능하다. 본 논문에서는 반감기가 100여년인 Ni-63 베타 방사성 능위원소와 PN 접합형 반도체 소자를 사용한 방사선 전지를 개발하기 위한 연구의 일환으로 효율적인 방사광 변환 법과 전류 발생방법 및 전지회로 구현에 대해서 고찰하였다. 또한 Ni-63 방사선의 베타선 스펙트럼을 계산하고 이에 해당하는 광입력 특성에 대한 실리콘 PN 접합 다이오드의 반응특성을 태양광 시뮬레이션용 소프트웨어를 사용하여 검증하였다.
Thirty sediment samples of the <63${\mu}{\textrm}{m}$ fraction collected from the southwestern coast of Korea were analysed for their heavy metal (Fe, Mn, Cr, Co, Cu, Ni, Zn and Pb) concentration. The results show that sediment texture plays a controlling role on the total metal concentrations and their spatial distribution. A single lM HCl extraction procedure was used in order to assess the environmental risk of heavy metals in bottom sediments. The non-residual fraction was the most abundant pool for Mn and Pb in most samples, which means that this metals are highly avaliable in these sediments. Cr, Ni, Fe, Co, Zn and Cu were mainly associated with the residual fraction, suggesting that their concentrations are controlled significantly by transport processes with the fine particles as carriers from diffuse pollution source. Concentration enrichment ratios(CER) were calculated from the non-residual contents and their values allowed us to classify the sediments according to their environmental risk.
