EFFECT OF CONCENTRATION OF PHOSPHORIC ACID ON MICROLEAKAGE TO TOOTH STRUCTURE (인산농도에 따른 치질의 미세누출 효과)

  • The purpose of this study was to evaluate the micro leakage of composite resin when various phosphoric acid agents were used to etch the enamel and dentin. In this study, class V cavities were prepared on the buccal surfaces of fourty extracted human molar teeth, and they were randomly assigned into 4 groups with 10 teeth. The cavities of each groups were etched with 10%, 32%, 35% and 37% phosphoric acids for 15 seconds, washed and dried and the cavities were restored with composite resin after application of the adhesive. The specimens were immersed in 2% methylene blue solution for 3 days. And then, the specimens were sectioned buccoligually. Degree of dye penetration at tooth-restoration interfaces was examined by Inverted Metallurgical Microscope at the occlusal and gingival margins. The result were as follows : 1. The degree of microleakage at occlusal and gingival margin in all group was statistically difference among 10% and 35% (P<0.01), 10% and 37% (P<0.01), 32% and 35% (P<0.05) and 32% and 37% (P<0.05) acid concentrations, but was not statistically difference between 10% and 32%, 35% and 37% acid concentrations (P>0.05). The degree of microleakage was showed to dimish with increase of acid concentration. 2. The degree of microleakage at the occlusal margin was statistically significant difference among 10% and 32%,10% and 35%, 10% and 37% acid concentrations(P<0.05). The degree of microleakage among 32% and 35%, 32% and 37% and 32% and 37% acid concentrations was not statistically significant difference. 3. In comparison of microleakage at the gingival margin, the degree of microleakage above 32% acid concentration was not showed less than 10% acid concentration (P>0.05). In comparison of 32%, 35%, 37% acid concentrations, the degree of microleakage at 35% and 37% acid concentrations was showed less than 32% acid concentration(P<0.05).

  • The screening method of varietal resistance on the plant hoppers has generally been evaluated as a reaction of plant after infesting insects. However, feeding amount of insects to the varieties was investigated in this experiment. The new method using isotope $^{32}P$ for rice varietal resistance to plant hoppers was carried out through the following method. Insects tested were caged for a few hours on the plants which had absorbed $^{32}P$ solution in small vials for $24\~48\;hours$. After feeding, insects were killed in the refrigerator with formalin solution, and then were measured by the feeding amount as a count per minute (CPM) with the G.M. Counter. The results obtained were summarized as follows; 1. The apparatus of Type D(Fig. 2) was most effective and the safest among four others. 2. The optimum amount of $H_3PO_4$ solution was found to be $2\~3m1$. 3. Radioactivity of $0.7\mu\;Ci.\;^{32}P$ was sufficient to check varietal difference of feeding amount by the brown planthopper. 4. Radioisotope was found from the body of insects but not in the cuticular layer nymphs cast off.

Relation of pH value to the availability of P32 (인산흡수와 산성도와의 관계)

  • 1. The effect of the pH value on P32 absorption of leaves of three forest trees under the water culture is investigated. 2. The degree of the P32 absorption to the pH value is quite different in each forest tree. 3. The phosphorus contents are proved higher in the leaves of Quercus accutissima(the saw shaped oak) than in the leaves of Pinus densiflora(the red pine) and Lespedeza bicolar (bush clover). 4. Larger contents of phosphorus are found in acidity plots than in alkalinity. The leaves of treated plot of the pH 4 (red pine), of the pH 5.0 (bush clover), and of the pH 5.0-6.5 (saw shaped oak) have a higher nutrient capital as indicated by P32 compared with that of the other pH values. 5. It is also noticed that P32 absorption capacity is decreased with the higher pH values. 6. The content of P of leaves shows the lowest value in the plot of the pH 6.5 (pine), of the pH 7.5(bush clover), and of the pH 9.0(saw shaped oak). It is also noticed that the red pine is to do very well in acid cultivation and then follow bush clover and saw shaped oak in the order.

Incorporation of phosphate into protein and other nitrogenous compounds in Chlorella cells (Chlorella 세포에서의 $^{32}P$-인산의 단백질 및 다른 질소화합물로의 전환)

  • In the process of the incorporation of orthophsphate into protein and other cell constituents, the role of inorganic polyphosphate and RNA-polyphosphate complex and the correlation between them were pursued by analyzing the contents of $^{32}P$ and total P in various fractions of Chlorella cells, which had been uniformly labeled with $^{32}P$ before the inoculation in a normal "cold" medium or P-free medium during the culture. The effects of ionizing radiation and various micronutritional-element deficiencies on the phosphate incorporation into, and biosynthesis of, protein and other introgenus compounds in the cells were also observed. When the uniformly $^{32}P$-labeled algae were grown in a normal "cold" medium the contents of $^{32}$ P in the fractions of protein, DNA and RNA-polyphosphate complex increased, but those in the fraction of acid-insoluble polyphosphate decreased. On the other hand, amount of $^{32}P$in the fraction of RNA was almost unchanged in spite of rapid increase of the total P. In the growing period of $^{32}P$-labeled algae in a P-free medium, amounts of $^{32}P$ in the fractions of DNA, protein and lipid increased, while those in the fractions of RNA-polyphosphate and inorganic polyphosphates decreased. When the algal cells were irradiated with about 70, 000r of gamma-rays before the inoculation in the medium, amounts of phosphate in the fractions of DNA, RNA, nucleotides and protein decreased during the culture, compared with those of the control. However, the phosphate content in the fraction of acid-insoluble polyphosphate of the irradiated cells increased than those of the control. In the growing period of the algae in a Mo-free, medium, amounts of acid-soluble total phosphate and nucleotides of the cells increased, while the amounts of residual protein and RNA decresed compared with those of the normal cells. Amounts of alkali-labile protein and phospholipid of the cells grown in a B-free medium decreased, whereas amount of phosphate in acid-soluble fraction increased compared with the control. In general, the contents of protein and RNA in each microelement deficient cells decreased more or less, compared with those in the normal cells.in the normal cells.

Studies on the Phosphate Metabolism in Chlorella, with Special Reference to Polyphosphate (Chlorella의 인산대사에 관한 연구)

  • Yung Nok Lee (Dept. of Biology, Korea University) : Studies on the phosphate metabolism in Chlorella, with special reference to polyphosphate. Kor. J. Microbiol., Vol.2, No.1, p1-11 (1964). 1. Uniformly $^{32}P$-labeled Chlorella cells which were irradiated with Cobalt-60 gamma-rays of about 70, 000 $\gamma$ dose, were further grown in a standard "cold" medium ("hot".rarw."cold"), and some portions of the algae were taken out at the begining of, and at intervals during the culture, and subjected to analyze the contents of $^{32}P$- and total P in various fractions of the cell materials. Results obtained were compared with those of nonirradiated normal cells. 2. Amounts of phosphate in various fractions of the nonirradiated normal Chlorella cells were measured using uniformly $^{32}P$--labeled cells. Analysis of the $^{32}P$--labeled algal cells showed that the highest value in P-content was the fraction of RNA followed by those of lipid, polyphosphate "C" polyphosphate "B", DNA, nucleotidic labile phosphate compounds, polyphosphate "A" and protein. It was observed that content of total polyphosphates in a single Chlorella cell was almost equal to RNA-P content in the cell, and the amount of RNA-P was almost equal to ten times of DNA-P content. 3. When the $^{32}P$--labeled algae which were irradiated with gamma-rays were grown in a normal "cold" medium, phosphate contents in the fraction of DNA, nucleotidic labile phosphate compounds and protein decreased markedly, while the contents of phosphate in the fractions of polyphosphate "C" and potyphosphate "B" increased in comparison with those of unirradiated normal cells. So, it was considered that the pretreatment of above mentioned dose of gamma-ray inhibited DNA and protein synthesis from polyphosphate in Chlorella cells. 4. Proceeding the culture of $^{32}P$--labeled Chlorella in a "cold" standard medium, whose synthetic activity of DNA and protein from polyphosphate was disturded by gamma-ray irradiation, the amounts of $^{32}P$-in the fraction of polyphosphate "C" increased, in contrast with those of polyphosphate "B" fraction. Optimization of the 32P-postlabeling Assay for Detecting Benzo(a)pyrene-induced DNA Adduct Formation in Zacco platypus

  • Objectives: $^{32}P$-postlabeling assay is the most sensitive method of detecting DNA adduct formation. However, it is limited by a low sample throughput and use of radioisotopes (RI). In this study, we modified it to minimize these limitations and applied it to Z. platypus exposed to Benzo(a)pyrene (BaP) in order to investigate DNA adduct formation (effect biomarker for pollutants) in Z. platypus for assessing risk of waterborne BaP exposure. Methods: DNA hydrolysis was performed only with Micrococcal nuclease (MNase), RI reduction test was performed and the overlapping steps between thin layer chromatography (TLC) and radioisotope high-performance liquid chromatography (RI-HPLC) were omitted. The application of a modified method to Z. platypus exposed to BaP was performed. Results: The results revealed that the amount of RIs used can be reduced roughly 10-fold. Because the analysis time was shortened by 8.5 hours, the sample throughput per hour was increased compared with the previous method. The results of applying modified $^{32}P$-postlabeling assay to Z. platypus, DNA adduct formation in Z. platypus showed dose-dependency with the BaP concentration. Only BPDE-dGMP was detected as a DNA adduct. Conclusion: These results demonstrate that the modified $^{32}P$-postlabeling assay is a suitable method for detecting DNA adduct formation in Z. platypus exposed to waterborne BaP and will be useful in risk assessment of carcinogenic effect in aquatic environment due to BaP.

Studies on the effects of radiation from radioisotopes incorporated into plants (Ⅶ) Effects of internally administered P-32 on the growth of soybeans (작물에 흡수된 방사성 동위원소의 내부조사효과에 관한 연구(제7보) 대두의 생장에 미치는 P-32의 영향)

  • To investigate the radiation effect of P-32 on the soybean plants, the seeds treated with various levels of P-32 solution were sown and cultured in the pots. The growth of the plants and the contents of the macroelements were observed and the following results were obtained. 1) The linear growth of the plants at the early stage seemed to have been promoted by the low-level P-32 treatemnt. At the later growing stage, however, this difference among treatments were less conspicuous. 2) The plants of high level P-32 application showed some growth damaging symptom at the early growing stage. Later this damage was recorded as the time went on and these plants showed even better growth than the control. As a result at the late growing stage, they ensued highest growth. 3) The plants showed in general more growth at the low activity level than at the high-level at the early growing stage. At the late stage, however, the high-level activity promoted more growth than the low-level. 4) At the early growing stage P-32 treatment produced in general significantly more lower than control. At the later stage, however, this difference was not clearly seen. 5) The P-32 treatment seemed to have stimulated earlier florescence and this tendency was more clearly observable eapecially at the high activity level. 6) The weight of the air-dried seeds tended to be increased through P-32 treatment by 10-45%. This tendency was clearly observed especially at the low-level activity. 7) As for the contents of the various macroelements in the leaves, the nitrogen showed significantly larger contents at the middle level(S1) P-32 treatments. The phosphorous contents showed also highest at the middle levels activity and lower both at the high and low-activity levels. The potassium contents was proved, on the contrary, higher at the low-level activity and lower at the high-level. 8) The nitrogen contents in the stems was found significantly higher than control, except at the low-activity level. The phosphorous showed higher contents at the low-activity level and no significant difference at the high-activity level. As for the contents of potassium, calcium and magnesium, three seemed no significant difference among treatments. However, the magnesium showed somewhat higher content at the low-activity level, whereas the calcium was proved high than control. 9) The inorganic contents in the root showed that N and P in the P-32 treated plant were significantly higher than the control and the K-contents was, on the contrary, significantly higher at the control than the rest of the treatments. As for the calcium and magnesium there showed no difference among all treatments.

  • Buckwheat seeds produced in previous year (1965) in an experimental pot culture in which nine levels of P-32 ranging from $1.4{\times}10-4 to 3.3{\times}104$\mu$c/pot(as of 27 July 1965)$, with the same specific activity, had been applied to the corresponding pots respectively, were used this year(1996) in water and soil culture as well as in germination test to investigate the feature and extent of possible residual effects of P-32 incorporated upon germination and plant growth, and the following results were obtained: 1. Under the given experimental conditions both stimulative and inhibitory effects of radiation were observed. 2. The germination rate of the seeds was lower at the higher level of P-32 aplication ranging from $3.0{\times}103 to 3.3{\times}104$\mu$c/pot$ and higher at 0.2 $\mu$c P/pot than the control. 3. Among the seeds produced at the higher level of application about 80% was failed to germinate, owing to the radiation injury. The remaining 80% was failed to germinate, owing to the radiation injury. The remaining 80% survived the damaging effect and showed vigorous growth and increased yield. The latter group of seeds thus proved themselves to be more radioresistant than the former. 4. The survived seeds produced later more straw and root on dry weight basis. The higher the level of P-32 applied, the stronger the stimulative effect showed in vegetative growth. 5. No radiation effect on linear growth of the plants was observed in the soil culture. 6. The dry weight of straw produced showed little difference at the moderate range compared with that of control in the soil culture. At high level of application, i.e. over $\mu$c P32/pot, however, the production was increased by 12-37% of control. 7. As for the dry weight of root harvested, the P-32 treatment over 24$\mu$c P32/pot produced 82-155% more than the control, whereas little difference was observed under 2.2$\mu$c P32/pot. 8. The seed production increased in general by the P-32 treatments. Particularly at the moderate level of application the rate of increase amounted to 70% of the control. 9. Those individual plants which survived damaging effects of radiation at the germinating stage shwoed remarkable stimulative effects both in vegetative and in reproductive stage of growth.

  • In order to evaluate the technique of P-32 labelling method for screening lines of rice to whitebacked planthopper, the relationship between the amount of ingestion and feeding preference of insects were observed with the resistant and susceptible lines where characteristics were predetermined by comparing their feeding preference and antibiosis method. The feeding preference of the insects was significantly correlated with the amount of P-32 ingestion. It was more manifest in adults than nymphs of the whitebacked planthopper. The density of nymph and radioactivity of P-32 were high in the susceptible line. This suggested that deforming the ingestion amount of P-32 in the insects seemed to be useful technique for accurate screening. For this technique, the uniform labelling of P-32 on the rice seedings was prerequisite and the uniformity was increased by triming roots and leaves in the length and number.

Prevalence of GII.4 Sydney 2012 and Recombinant GII.3P[12] Noroviruses Associated with Acute Gastroenteritis in Hospitalized Children in Thailand, 2015-2017

  • Norovirus (NoV) is an important pathogen causing acute gastroenteritis worldwide. The purpose of the present study was the molecular characterization of NoV. A total of 408 stool specimens collected from hospitalized children associated with acute gastroenteritis in Chiang Rai, Thailand, 2015-2017 were investigated for the presence of NoVs by RT-PCR. NoV GII was detected in 32 samples (7.8%). Five distinct genotypes were identified, including GII.4 (13/32, 40.6%), GII.3 (11/32, 34.3%), GII.17 (4/32, 12.5%), GII.2 (2/32, 6.3%), and GII.14 (2/32, 6.3%). NoV infection occurred mostly in young children under 3 years of age (31/32, 96.9%) and showed the main peak in summer months from March to April (18/32, 56.3%). Phylogenetic analysis revealed that all 13 GII.4 strains clustered with GII.4 Sydney 2012 variant. Representative GII.3 strains were analyzed as a recombinant GII.3P[12] strain. Several amino acid differences were found in the antigenic epitopes and antibody binding sites of the VP1 capsid of the GII.3P[12]. Homology modeling of the P domain of the GII.3P[12] strain demonstrated that 10/13 amino acid differences were predicted to be located on the surface-exposed area of the capsid structure. These amino acid changes might affect the infectivity and the antigenicity of the recombinant GII.3P[12]. The prevalence of GII.4 Sydney 2012 and recombinant GII.3P[12] strains indicates the genetic diversity of circulating NoVs in Thailand, emphazing the importance of continuous surveillance to mornitor newly emerging NoV strains in the future.
