Comparison of Mutant Frequencies Induced by ${\gamma}$-radiation and Pentachlorophenol at hprt Locus in Human T-lymphocytes(I) (인체 T-림프구 hprt 유전자에서 방사선 및 pentachlorophenol에 의한 돌연변이 빈도의 비교(I))

  • In vitro somatic mutation induced by ${\gamma}$-radiation and pentachlorophenol(PCP) which is representative of chemical pollutant was measured at the hypoxanthine-guanine phosphoribosyl transferase(hprt) locus in human T- lymphocytes by a cell cloning assay. Mutant cells were selected by their ability to form a clone in the presence of purine analogue 6-thioguanine. The mutant frequencies by ${\gamma}$-irradiation to a dose of 1.0 Gy, 2.0 Gy and 3.0 Gy were 40%, 400% and 750% higher than those in controls. Significant changes were not observed in mutant frequencies in the 0.2 Gy and 0.5 Gy irradiated groups. When the doses of PCP were 15 ppm, 25 ppm and 50 ppm, the mutant frequencies increased by 30%, 90% and 520%, respectively. No changes were observed in the 10 ppm treated group. Similar types of dose-response relationship were shown in the two different mutagens. Reverse transcriptase/polymerase chain reaction technique(RT/PCR) was needed for the mutation spectrum to discriminate combined exposure to radiation and chemicals.

  • 2011년 동일본 지역에서 발생한 지진으로 인하여 후쿠시마 다이이치에 위치한 원자력 발전소에서 다양한 방사성 물질들이 바다, 하천 그리고 대기와 같은 자연환경 속으로 유출되었다. 방사성 세슘(Cesuim, $Cs^{137}$)은 다양한 방사성 물질들 가운데 반감기(Half-life)가 30.17년으로 가장 긴 물질이다. 방사성 세슘이 환경 생태계로 한번 유출될 경우 긴 반감기과 널리 퍼지는 성질로 인하여 오랜 시간동안 넓은 지역에 막심한 피해를 초래하므로 효과적인 처리방법을 통해 안전하게 처리하는 것이 아주 중요하다. 세슘을 제거하기 위하여 물리적, 화학적, 생물학적 등 다양한 방법들을 통해 연구를 진행하고 있으며, 특히 세슘을 제거하는 아주 효과적인 방법 중 하나인 프러시안 블루(Prussian Blue, PB) 흡착제를 적용하는 방법이 많이 주목받고 있다. 그러나 프러시안 블루는 미세한 분말입자로서 수처리에 사용하게 되면 처리 후 발생되는 슬러지들을 수중으로 부터 분리하기 어려운 한계점을 가지고 있다. 최근 연구에서는 프러시안 블루의 적용 한계점를 극복하기 위하여 자성체(Magnetic substance)를 물리적 지지체로 이용하여 외부 자기장을 통해 수중으로 분리하는 방법들이 연구되고 있다. 자성체란 외부 자기장이 주어지게 되면 입자들 표면에 자성력을 띄는 물질들을 말한다. 본 연구에서는 자성체 종류들 가운데 가장 높은 자성력을 지닌 강자성체(Ferromagnetic Substance)를 물리적 지지체로 하여 산화과정, 실란과정, 합성과정을 거쳐 강자성체 입자의 표면에 프러시안 블루를 합성한 새로운 형태에 합성체를 제조하고, 제조된 합성체를 이용하여 수중에 존재하는 세슘 제거 능력을 평가하였다. 제조된 합성체의 물리적 특성을 분석하기 위하여 SEM, XRD를 이용하여 합성체 입자의 표면 분석을 진행하였다. 합성체의 세슘 제거 능력을 평가하기 위하여 임의 제조된 0.5mg/L의 세슘 농도를 가진 원수 100ml에 제조된 새로운 형태의 합성체 1g을 투입한 뒤 1분간의 반응시간 동안 반응한 이후 잔류 세슘을 측정한 결과 수중의 존재하는 세슘에 대해 99.9%의 세슘 제거율을 기록하였다. 자가분리(Magnetic Seperate)의 원리를 이용하여 수중으로부터 회수율을 측정한 결과, 99%의 합성체 회수율을 얻었다. 실험결과를 통해 외부자기장이 주어지게 되면 수중으로부터 합성체를 대부분 분리하여 회수할 수 있다고 판단된다. 본 연구를 통해 개발된 새로운 형태의 합성체는 수중의 세슘 처리 공정에서 사용자가 직접 접촉하지 않고 세슘제거 및 외부자기장을 통해 수중으로부터 분리가 가능한 합성체라고 판단된다.

An Analysis of Domestic Experimental Results for Soil-to-Crops Transfer Factors of Radionuclides (주요 핵종의 토양-작물체 전이 계수의 국내 실험 결과에 대한 분석)

  • For more realistic assessment of Korean food chain radiation doses due to the operation of nuclear facilities, it is required to use domestically produced data for radionuclide transfer parameters in crop plants. This paper analyzed results of last about 10 year's studies on radionuclide transfer parameters in major crop plants by the Korean Atomic Energy Research Institute, comparing with the published international data, and consequently suggested the proper parameters to use. The trends of transfer parameter shows normal distributions if we have a lot of experimental data, but some radionuclides showed enormous variations with the environment of experimental, crops and soils. These transfer factors can be used to assess realistic radiation doses or to predict the doses in crops for normal operation or accidental release. Some kinds of parameter can be produced as conservatives or fragmentary results because soil-to-plant transfer factors were measured through greenhouse experiments which sometimes showed improper field situations. But these parameters mentioned in this paper can be representative of the status of Korean food chain than that of foreign country.

  • The structure of the actual detector was computed using the code of the PENELOPE. Using the standard mixed sources (450, 1,000 ml), compare the effectiveness of each energy according to various densities and height of the PENELOPE computer simulation, and calculate the effectiveness of the various environmental specimens and apply them to various environmental specimens to determine the lower limit. The values obtained by the obtained value were obtained by applying the obtained efficiency to the actual environmental specimens and obtaining the lower limit values. The density correction factor is 1.155 g of the density correction factor of $0.4g/cm^3$ (59.54keV), 1.153 (661 keV), $1.06g/cm^3$ 1.064 (1,836.04keV), 1.03, and 1.033. It was confirmed that the radioactivity concentration of environmental samples decreased as the amount of specimen was measured increases, and the MDA value decreased as time measured increases.

Distributions and Pollution History of Heavy Metals in Nakdong Estuary Sediments (낙동강 하구역 퇴적물 중금속의 분포와 오염의 역사 추정)

  • In order to determine the horizontal and vertical distributions of metals and prospect the recent metal pollution history in Nakdong Estuary, we took surface and core sediments. Maximum value of organic matter occurs at the upstream site located 4 km from Nakdong barrage, and the concentration of trace metals (Zn, Cu, and Pb etc.) decrease seaward in the estuary. The sedimentation rates, based on $^{210}$Pb$_{ex}$ and $^{137}$Cs activities, were 0.34 cm/yr in inside of barrage (core 1) and 0.25 cm/yr in Changrim (core 4). Sediment mixing layer does not exist in core 1, where anoxic condition is known to be prevail. The topmost sediment layer of core 4 (<3.5 cm) is severely mixed. At sites 1 and 4, concentrations of Cu slowly increased during the period of 1920-1970, rapidly increased during 1970-1990, and followed by slight decrease after 1990. Zn contents increased in early 1960s and peaked in 1993, and followed by decrease after 1990s. Pb has increased continuously since early 1970s. At the downstream of the barrage, Cu and Zn have increased in the topmost layer. The trend of increase of Cu is evident after 1950 (11 cm in sediment depth). Overall trend of heavy metal concentration clearly indicates the pollution has been increasing after the construction of the barrage.

Characterization of Cement Solidification for Enhancement of Cesium Leaching Resistance (세슘 침출 저항성 증진 시멘트 고화체의 제조 및 특성 평가)

  • Currently, the Korea Atomic Energy Research Institute (KAERI) is planning to build the Ki-Jang Research Reactor (KJRR) in Ki-Jang, Busan. It is important to safely dispose of low-level radioactive waste from the operation of the reactor. The most efficient way to treat radioactive waste is cement solidification. For a radioactive waste disposal facility, cement solidification is performed based on specific waste acceptance criteria such as compressive strength, free-standing water, immersion and leaching tests. Above all, the leaching test is important to final disposal. The leakage of radioactive waste such as $^{137}Cs$ causes not only regional problems but also serious global ones. The cement solidification method is simple, and cheaper than other solidification methods, but has a lower leaching resistance. Thus, this study was focused on the development of cement solidification for an enhancement of cesium leaching resistance. We used Zeolite and Loess to improve the cesium leaching resistance of KJRR cement solidification containing simulated KJRR liquid waste. Based on an SEM-EDS spectrum analysis, we confirmed that Zeolite and Loess successfully isolated KJRR cement solidification. A leaching test was carried out according to the ANS 16.1 test method. The ANS 16.1 test is performed to analyze cesium ion concentration in leachate of KJRR cement for 90 days. Thus, a leaching test was carried out using simulated KJRR liquid waste containing $3000mg{\cdot}L^{-1}$ of cesium for 90 days. KJRR cement solidification with Zeolite and Loess led to cesium leaching resistance values that were 27.90% and 21.08% higher than the control values. In addition, in several tests such as free-standing water, compressive strength, immersion, and leaching tests, all KJRR cement solidification met the waste acceptance or satisfied the waste acceptance criteria for final disposal.

Genes Associated with Radiation Adaptive Response Induced by Low Level Radiation from $^{99m}Tc$ in Human Cell Lines (인체세포주에서 저선량 $^{99m}Tc$에 의해 발현되는 방사선 적응반응에 관련된 유전자에 관한 연구)

  • Purpose: The purpose of this study was to search activated genes that could be related to radiation adaptive response (RAR) induced by low-level radiation from $^{99m}Tc$ in human cell lines. Methods: We used gene discovery array (GDA) and representational difference analysis (RDA) methods. $^{99m}Tc$-pertechnetate was added to $2{\times}106/mL$ NC-37 cells (human lymphoblastic cells) to make concentrations ranging from 148 MBq/mL to 148 Bq/mL by serial 10 fold dilutions. After 44 hours, 2 Gy gamma irradiation was given to them using a Cs-137 cell irradiator. Results: As compared to the control (Con) group to which no $^{99m}Tc$ was added, those cells to which 148 and 14.8 KBq of $^{99m}Tc$ were added showed significantly lower damage to chromosomes, which was evaluated by metaphase analysis. Cells with 148 KBq $^{99m}Tc$ (T148 group) showed most significant protection. Activated genes in the T148 group as compared to Con group were evaluated by GDA and GDA methods. GDA revealed genes of casein kinase 2 (CK2) beta chain, immunoglobulins (lg), human leukocyte antigen (HLA)-B, and two novel genes. Twenty RAR related clones were selected by RDA method. The size of those genes was from 234 to 603 base pairs. Conclusions: RAR was induced by low dose irradiation from $^{99m}Tc$ in NC-37 cell lines. Genes related to the response included CK2, lg, HLA-B in human lymphoblastic cell lines.
