FUNGAL EXTRACELLULAR POLYSACCHARIDES INVOLVED IN RECYCLING OF METABOLITES AND OSMOTOLERANCE OF PENICILLIUM FELLUTANUM : APPLICATION OF $^{13}$ C-NMR SPECTROSCOPY FOR THE STUDY ON FUNGAL PHYSIOLOGY AND METABOLISM

  • Penicillium fellutanum produces a phosphorylated, choline-containing extracellular peptido-polysaccharide, peptidophosphogalactomannan (pPxGM) (8). The $\^$13/C-methyl labeled pPxGM ([methyl-$\^$13/C]pPxGM) was prepared from the cultures supplemented with L-[methyl-$\^$13/C]methionine or [2-$\^$13/C]glycine and was used as a probe to monitor the fate of phosphocholine in this polymer. Addition of purified [methyl-$\^$l3/C]pPxGM to growing cultures in low phosphate medium resulted in the disappearance of [methyl-$\^$13/C]phosphocholine and -N,N'-dimethyl-phosphoethanolamine from the added [methyl-$\^$13/C]pPxGM. Two $\^$l3/C-methyl-enriched cytoplasmic solutes, choline-O-sulfate and glycine betaine, were found in mycelial extracts, suggesting that phosphocholine-containing extracellular pPxGM of P.fellutanum is a precursor of intracellular choline-O-sulfate and glycine betaine and thus of phosphatydilcholine (l0). $\^$13/C-Methyl-labeled cells grown in 3 M NaCl-containing medium showed 2.6- and 22-fold more accumulation of $\^$13/C-methyl labeled choline-O-sulfate and glycine betaine, respectively, originated from the extracellular [$\^$13/C-methyl]pPxGM than those grown without added NaCl. The results suggest that, in addition to glycerol and erythritol, glycine betaine and choline-O-sulfate and thus choline are also osmoprotectants and hence that pPxGM is involved in osmotolerance of this fungus (11). Taken collectively, the $\^$l3/C- and $\^$31/P-NMR analyses of cytosolic solute pools and structural modulation of extracellular pPxGM corresponding to environmental stimuli in P. fellutanum, provided evidence that pPxGM is involved in cellular choline metabolism, osmotolerance, and recycling of metabolites.

  • Hyperpolarized (HP) carbon-13 (13C) MRI represents an innovative approach for noninvasive, real-time assessment of dynamic metabolic flux, with potential integration into routine clinical MRI. The use of [1-13C]pyruvate as a probe and its conversion to [1-13C]lactate constitute an extensively explored metabolic pathway. This review comprehensively outlines the establishment of HP 13C-MRI, covering multidisciplinary team collaboration, hardware prerequisites, probe preparation, hyperpolarization techniques, imaging acquisition, and data analysis. This article discusses the clinical applications of HP 13C-MRI across various anatomical domains, including the brain, heart, skeletal muscle, breast, liver, kidney, pancreas, and prostate. Localization of Quantitative Trait Loci for Bone Mineral Density on Chromosome 13 in the Mongolian Population

  • Although the genetic basis for bone mineral density (BMD) has been studied by many groups so far, genes responsible for this complex trait has not been completely revealed. In order to localize quantitative trait loci (QTLs) for BMD variation in Asian population, the study was designed using a group of Mongolian population, a genetically closed population with a homogeneous lifestyle. BMD was measured at the left and right wrists and ankles using DEXA in 1,082 participants from 142 families. Genotyping of 13 polymorphic microsatellite markers on chromosome 13 (average spacing 8-9 cM) and two-point and multipoint linkage analysis were performed. In two-point linkage analysis, we identified two markers, D13S175 (6.03 cM) and D13S265 (68.73 cM) that had LOD scores greater than 1 for left ankle (LOD=2.09, LOD=1.49, respectively). We also found a marker D13S175 (6.03 cM) with a high LOD for left wrist (LOD=1.49) and the markers D13S265 (68.73 cM) and D13S217 (17.21 cM) for the right wrist (LOD= 1.82, LOD= 1.62, respectively). Among these significant marker regions, only two regions at 17 cM (13p11) and 65 cM (13q21) for the right wrist overlapped with major QTLs reported in following multipoint linkage analysis (LOD= 1.7549, LOD=1.4462, respectively). This study provides the possible evidence of the presence of QTLs affecting right wrist BMD in Mongolian populations on 13p11 and 13q21. Modest evidence was also found for genes affecting left ankle and left wrist BMD on 13p13.

바나나의 후숙 밑 저장 온도가 품질에 미치는 영향 (Effects of Ripening and Storage Temperatures on the Quality of Banana)

  • 필리핀산 바나나를 온도(18$^{\circ}C$, 21$^{\circ}C$, $25^{\circ}C$)별로 5일간 후숙하고 13$^{\circ}C$와 및 상온(13-2$0^{\circ}C$)에서 16일간 저장하며 품질 변화를 조사하였다. 후숙 직후 바나나의 색도와 당도는 18$^{\circ}C$와 21$^{\circ}C$ 후숙이 4.2-5.2와 18.7-18.8$^{\circ}$이었고 $25^{\circ}C$ 후숙이 각각 5.5와 20.1$^{\circ}$를 나타냈다. 저장온도 및 후숙 방법에 따른 중량의 변화는 큰 차이 없이 저장 10일경에 약 1%로 증가하였으며, 당도는 20$^{\circ}$ 내외에서 거의 변화가 없거나 약간 증가 후 감소하였으며, 색도는 초기 5부근에서 2-4일 만에 완전 노랑인 7의 상태로 되었고, 혹반점은 정상 및 저온 후숙품이 13$^{\circ}C$에서 4일, 상온에서 2일만에 발생되었다. 변질율은 저온 및 정상 후숙이 13$^{\circ}C$에서 저장 6일 후 2.1%, 상온에서는 4일 후 4.2%로 그 이후부터는 75-100%로 급격히 증가하였으며, 고온 후숙은 13$^{\circ}C$ 저장에서 저장 2일 상온저장에서는 저장 초기 이후부터 변질이 급격히 진전되었다.

Dual Component Analysis for In Vivo T2* Decay of Hyperpolarized 13C Metabolites

  • Purpose: To investigate the exchange and redistribution of hyperpolarized $^{13}C$ metabolites between different pools by temporally analyzing the relative fraction of dual $T_2{^*}$ components of hyperpolarized $^{13}C$ metabolites. Materials and Methods: A dual exponential decay analysis of $T_2{^*}$ is performed for [1-$^{13}C$] pyruvate and [1-$^{13}C$] lactate using nonspatially resolved dynamic $^{13}C$ MR spectroscopy from mice brains with tumors (n = 3) and without (n = 4) tumors. The values of shorter and longer $T_2{^*}$ components are explored when fitted from averaged spectrum and temporal variations of their fractions. Results: The $T_2{^*}$ values were not significantly different between the tumor and control groups, but the fraction of longer $T_2{^*}$ [1-$^{13}C$] lactate components was more than 10% in the tumor group over that of the controls (P < 0.1). The fraction of shorter $T_2{^*}$ components of [1-$^{13}C$] pyruvate showed an increasing tendency while that of the [1-$^{13}C$] lactate was decreasing over time. The slopes of the changing fraction were steeper for the tumor group than the controls, especially for lactate (P < 0.01). In both pyruvate and lactate, the fraction of the shorter $T_2{^*}$ component was always greater than the longer $T_2{^*}$ component over time. Conclusion: The exchange and redistribution of pyruvate and lactate between different pools was investigated by dual component analysis of the free induction decay signal from hyperpolarized $^{13}C$ experiments. Tumor and control groups showed differences in their fractions rather than the values of longer and shorter $T_2{^*}$ components. Fraction changing dynamics may provide an aspect for extravasation and membrane transport of pyruvate and lactate, and will be useful to determine the appropriate time window for acquisition of hyperpolarized $^{13}C$ images.

  • DOC의 ${\delta}^{13}C$은 수질 오염원 규명에 활용할 수 있는 매우 유용한 지표이다. 본 연구에서는 안정성동위원소비 질량분석기(EA-SIRMS)를 이용하여 DOC의 ${\delta}^{13}C$을 분석할 때 사용하는 두 가지 전처리방법 (과황산칼륨산화-$SrCO_3$침전법과 동결건조법)의 정확도를 비교 평가하였다. 표준물질로는 글루코스(${\delta}^{13}C$; $-12.0{\pm}0.02$‰)와 돈분퇴비 침출액 건조분말(${\delta}^{13}C$; $-23.3{\pm}0.04$‰)을 사용하였다. 과황산칼륨산화-$SrCO_3$ 침전법에서는 용액의 알칼리성에 의해 DIC 제거가 어렵고 공기 중 $CO_2$의 영향에 의해 시료가 오염되어 분석결과의 정확도가 매우 낮았다. 반면, 동결건조법은 산 첨가에 의해 제거가 가능하였고, 측정값과 이론값의 상관관계가 매우 높아 실험조건별로 표준물질을 이용하여 측정값을 이론값에 근접하게 보정할 수 있는 것으로 나타났다. 따라서, EA-SIRMS를 이용하여 DOC의 ${\delta}^{13}C$ 분석시 동결건조법으로 시료를 전처리하면 분석결과의 정확도를 증대시킬 수 있을 것으로 판단된다.

Genetically modified carrot cells acquiring desiccation tolerance

  • To obtain direct evidence for thed involvement of C-AB13, a carrot (Daucus carota L.) homolog of VPI/Ab13, seed-specific transcription factor, in the acquisition of desiccation tolerance carrot non-embryogenic cells (NC) in which the C-AB13 gene was expressed ectopically was prepared. Non-transgenic NC, in which expression of C-AB13 was not detected, did not exhibit desiccation tolerance even after treatment with abscisic acid (ABA). In transgenic NC that expressed C-AB13, embryo-specific ABA-inducible genes (ECP genes) were expressed upon ABA-treatment. Furthermore, the transgenic NC became desiccation-tolerant upon ABA-treatment, but not tolerate desiccation without ABA-treatment. These results provide direct evidence for the involvement of C-AB13 in the ABA-induced acquisition of desiccation tolerance.

하천수 중 미량의 Linear Alkylbenzene Sulfonate의 분석 (Determination of Trace Amount of Linear Alkylbenzene Sulfonates in River Water)

  • 하천수 중 LAS의 각 동족체 및 이성체들의 성분을 BP-5 capillary column을 이용한 GC 및 GC/MS에 의하여 분리 확인 및 정량하였다. 특히 LAS 유도체의 mass chromatogram에 의해 RIC에서는 분리가 되지 않는 $5-C_{11}$$6-C_{11}$$2-C_{12}$, $6-C_{13}$$7-C_{13}$의 LAS 성분을 각각 분리 확인하였다. 중랑천, 진위천 및 한강 하루부에서 채수한 시료 중에서 탄소수 $C_{10}$, $C_{11}$, $C_{12}$$C_{13}$의 LAS 성분을 측정하였으며 동족체 및 이성체의 조성비를 구하였다. 표준품과 하천수 중 LAS 성분의 함량비를 측정 비교함으로써 하천에서 LAS의 생분해 및 동태를 추정할 수 있었다.
