The aim of this study was to investigate the effects of Cheonghyul-san on the generation of peroxynitrite ($ONOO^-$), nitric oxide (NO) and superoxide anion radical ( ${\cdot}\;O_2^-$), and on the expression of NF-${\kappa}$B-dependent proinflammatory proteins in ob/ob mice. Mice were grouped and treated for 5 weeks as follows. Both the normal lean (C57BL/6J black mice) and control obese (ob/ob mice) groups have received the standard chow. The experimental groups were fed with a diet of chow supplemented with 7.5, 15 and 30 mg Cheonghyul-san per 1 kg of body weight for 14 days. For this study, the fluorescent probes, namely 2',7'-dichlorodihydrofluorescein diacetate (DCFDA), 4,5-diaminofluorescein-2 (DAF-2) and dihydrorhodamine 123 (DHR 123) were used. Western blot was performed using anti-IKK-${\alpha}$, anti-phospho I${\kappa}$B-${\alpha}$, anti-NF-${\kappa}$B (p50, p65), anti-COX-2, anti-iNOS, anti-VCAM-1 antibodies, respectively. Cheonghyul-san prevented $H_2O_2$-induced cell death. Cheonghyul-san inhibited the generation of $ONOO^-$, NO and ${\cdot}\;O_2^-$ in the $H_2O_2$-treated LLC-$PK_1$ cells. The generation of $ONOO^-$, NO and ${\cdot}\;O_2^-$ were inhibited in the Cheonghyul-san-administered ob/ob mice groups. The GSH/GSSG ratio was decreased in the ob/ob mice, whereas the ratio was improved in the Cheonghyul-san-administered groups. Cheonghyul-san inhibited the protein expression levels of phospho-I${\kappa}$B-${\alpha}$, IKK-${\alpha}$, NF-${\kappa}$B (p50, p65), COX-2, iNOS and VCAM-1 genes. These results suggest that Cheonghyul-san is an effective scavenger of $ONOO^-$, ${\cdot}\;O_2^-$ and NO, and has an inhibitory effect on the expression of NF-${\kappa}$B-dependent inflammatory genes in ob/ob mice. Therefore, Cheonghyul-san might be used as a potential therapeutic drug against the diabetes- and obesity-related proinflammatory diseases.
Recently distinguished AOP means technology resolving organic compounds in water to harmless compounds such as $CO_2$ and $H_2O$ by creating OH radical ($OH{\cdot}$) with more powerful oxidation than general oxidants. It has merits which the 2nd pollution is not caused since it uses solar energy, sludge doesn't take place, it can be applied to high-density waste water and it oxidizes non-biodegradable organic compounds more easily. The purpose of the study was to examine about removable characteristics of phenol which was a non-biodegradable organic matter with UV/$O_3$/Catalyst processes which is one out of AOP and to present applicability of photocatalyst and the optimum conditions of treatment. The study regarded initial phenol concentration, initial pH, photocatalyst amount and flow as its conditions. As the results, the test had the highest removable efficiency (92%) when initial phenol concentration was 100 mg/L, initial pH 7, photocatalyst amount 6L and flow 1.5 mg/min. The removable efficiency was increased as much as initial phenol concentration was increased, when initial pH was 7 (neutrality), photocatalyst amount was increased and flow was increased. It was checked that the optimum HRT was 12 hours. Therefore, phenol is enough removable with UV/$O_3$/Catalyst process and its prospect in the future is expected.
The protective effects of sea tangle (Laminaria japonica) extract and fucoidan components on the attack of oxygen radicals in kidney were studied, Sprague-Dawley (SD) male rats (210 $\pm$ 5 g) were with fed experimental diets of Dasi-Ex group (sea tangle extract powder of $4.0\%$ added to control diet), Euco-I, II and III groups (fucoidan powder of 1, 2 and $3\%$, respectively, added to Dasi-Ex group) for 45 days, Hydroxyl radical formations were significantly decreased ($10\~15\%$ and $15\~30\%$) in mitochondria and microsomes of Dasi-Ex and Fuco-I, II, III groups compared with control group. Hydrogen peroxide formations were also significantly decreased ($10\~15\%$) in microsomes of Dasi-Ex and Fuco-I, II, III groups compared with control group. Significant differences in mitochondrial basal oxygen radical (BOR) and microsomal induced oxygen radical (IOR) formations of Dasi-Ex and Fuco-I groups could not be obtained, but mitochondrial BOR and microsomal IOR formations were significantly decreased ($12\~15\%$ and $13\~14\%$) in Fuco-II and III groups compared with control group. BOR formations were significantly decreased ($12\~25\%$) in microsomes of Dasi-Ex and Fuco-I, II, III groups, and IOR formations were also significantly decreased ($10\~15\%$) in mitochondria of Fuco-I, II, III groups compared with control group, Significant differences in mitochondrial Mn-SOD activities of Dasi-Ex group could not be obtained, but mitochondrial Mn-SOD activities were dose-dependently increased by $8\%,\;16\%$ and $36\%$ in Fuco-I, II and III groups compared with control group, Mn-SOD activities an microsome were significantly increased about $20\%$ in Dasi-Ex group, while they were remarkably increased about $40\%$ in Fuco-I, II and III groups compared with control group. lipid peroxide contents were significantly decreased about $15\%$ and $15\~25\%$ in mitochondria and microsomes of Fuco-II and III groups. Membrane fluidities resulted in marked increases ($20\~35\%$ and $17\~24\%$) in mitochondria and microsomes of Dasi-Ex and Fuco-I, II and III groups. These results suggest that administrations of fucoidan added to sea tangle may play a pivotal role in attenuating attack of oxygen radicals in kidney.

White and black garlic pastes were fermented by Leuconostoc mesenteroides and evaluated for its rheological and functional properties such as consistency, antioxidant and angiotensin converting enzyme inhibitory activity. The pH, acidity and solid content of black garlic paste were 4.60, 1.23%, 22.63%, respectively. The viable cell counts and consistency of fermented garlic was decreased by adding higher amounts of black garlic paste. Fermentation of white garlic (40%)/black garlic (10%) showed viable cell counts of $1.6\times10^{11}$, fluid consistency of 9.31 $Pa{\cdot}s^n$. Water and 70% ethanol extract from fermented garlic showed the polyphenol content of 6.29mg/mL and 5.99 mg/mL, respectively. Also, water extract indicated the DPPH radical scavenging effects and angiotensin converting enzyme (ACE) inhibitory activity with $IC_{50}$ 1.03 mg/mL and $IC_{50}$ 68.54 mg/mL, respectively. ACE inhibitory activity was increased with adding black garlic paste. Conversion of sucrose into dextran polymer in fermented garlic was drastically decreased by the addition of black garlic paste, indicating from 85% (0% black garlic) to 20% (20% black garlic) conversion yield. Garlic paste fermented with 10% black/40% white garlic showed the decrease in consistency and viable cell counts during both cold and freezing storages. In particular, consistency of fermented garlic was lower during freezing storage than cold storage, and the viable cell counts was drastically decreased after storage for 2 weeks.

Advanced oxidation process involving $O_3/H_2O_2$ was used to eliminate 1,4-dioxane and to enhance the biodegradability of dioxane-contaminated water. Oxidation process was carried out in a bubble column reactor under different pH and $H_2O_2$ concentrations. The removal efficiencies of 1,4-dioxane were investigated at hydrogen peroxide concentration between 40 and 120 mg/L. At the same pH, removal efficiencies of 1,4-dioxane increased with increasing initial $H_2O_2$ concentration. There was a linear relationship between initial concentration of $H_2O_2$ and the amount of consumed $O_3$. It was observed that the high $H_2O_2$ concentration accelerated the generation of hydroperoxy ions(${HO_2}^-$) and hydroxyl radicals($OH{\cdot}$). Hydrogen peroxide enhanced the decomposition of 1,4-dioxane and the biodegradability of the solution.
The treatment of soils and water contaminated with MTBE using the Fenton oxidation was investigated. The effects of dosage of $H_{2}O_{2}$, and Fe$^{2+}$ concentrations, and solution pH on transformation and mineralization in soil were evaluated. Generation of TBA and acetone following Fenton-oxidation of MTBE in water and generation of acetone following Fenton-oxidation of TBA were observed. Therefore TBA and acetone are degradation intermediates of MTBE. There was a large difference of treatment efficiency in Fenton oxidation of MTBE between soil and water system. This may be caused by the complex nature of soil, soil organic matter which can consumed OH $\cdot$ radicals, and interacting with inorganic-soil constituents. The pH of soil was observed to have a significant effect on the chemical oxidation efficient of MTBE in soil The data demonstrated that optimal pH range were pH 3~4 and around 6. The soil batch studies demonstrated that treatment efficiency of MTBE was enhanced by adding additional ferrous salts but Fenton-oxidation occurred in no additional iron which indicated that iron in soil can catalyze the Fenton-oxidation. The most effective parameter of Fentonoxidation was $H_{2}O_{2}$/Fe$^{2+}$ ratio which theocratical ratio is 0.5. The optimal range of this ratio was found to be 0.6~2.3. In evaluating effect of $H_{2}O_{2}$ dosage on treatment efficiency, the increase of $H_{2}O_{2}$ did not always lead to increase of decompositions of MTBE in soil. Fenton oxidation was effective in destroying MTBE in aqueous extracts of contaminated soil and water. Experimental data provided evidence that the Fenton oxidation can effectively remediate MTBE-contaminated water and soil.
Free radicals may attack cells or tissue, leading to chronic diseases, and antioxidant consumption is potentially useful for removing free radicals. Egg proteins may be used as potential sources of antioxidant considering their ability of scavenging free radicals to apply for food or cosmetics industry. In this study, we obtained a natural antioxidant protein from fertilized eggs, which was a dietary supplement in some Asian countries. Meanwhile, antioxidant activities of these proteins were evaluated using different oxidation systems. With increasing incubation time, the antioxidant activity of these proteins increased during 15 d of incubation. The samples on day 15 were performed for isolation of antioxidant protein. The protein, named P4-1 (MW, 45 kDa), was isolated and purified by consecutive chromatographic methods. P4-1 contained 17 amino acids, which was determined by liquid chromatography-mass spectrometry and Amino Acid Analyzer. Moreover, the amino acid sequence was highly similar to that of ovalbumin. Differential scanning calorimetry showed that the denaturation temperature of P4-1 was $57.16^{\circ}C$. Furthermore, P4-1 suggested high oxygen radical-absorbance activity in ${\cdot}OH$ assays, and its antioxidant activity was stable at $30-50^{\circ}C$ in acidic and neutral pH. Thus, these results revealed that P4-1 may be a potential resource as a natural antioxidant.
This study was conducted to determine the optimum condition of extraction and antioxidant activity of ethanol extracts of Epimedium koreanum Nakai based on the icariin quantity. Also, further organic solvent fractions of hexane, chloroform, ethyl acetate and butanol were obtained from the ethanol extract of Epimedium koreanum Nakai at different temperatures. Total ethanol extraction yield of wild grape seed ranged from 11.8% to 39.3% depending on the concentration of icariin as well as different ethanol concentration, extraction temperature and time condition. Among different extraction temperatures and time, the highest extraction yield of 39.3% was obtained at 70% ethanol for 3 hour at $50^{\circ}C$ in the sample containing the 0.596% icariin (KE9412). In the meantime, the strongest free radical scavenging effect $(RC_{50})$ with $18.9{\mu}g/mL$ in the KE9412 sample was observed in 70% ethanol extract of Epimedium koreanum Nakai extracted for 7 hour at $70^{\circ}C$, while $RC_{50}$ with $35.2{\mu}g/mL$ was observed in the KE9405 (0.20% icariin content) sample at same condition. Also, antioxidant activity of ethanol extracts of Epimedium koreanum Nakai increased as icariin concentration increased. Among each fraction obtained from organic solvents, butanol fraction was found to have the strongest $RC_{50}\;(39.2{\mu}g/mL)$ and followed by ethylacetate $(49.0{\mu}g/mL)$, water $(118.8{\mu}g/mL)$, ethylacetate $(119.8{\mu}g/mL)$, and chloroform fraction $(138.5{\mu}g/mL)$ respectively.
The objectives of this study are summarized with two. The one is to remove of $COD_{Cr}$, color and turbidity with coagulation and ozonation in nonbiodegradable landfill leachate, the other is to compare of water qualities with pre-ozonation and post-ozonation. The results are summarized as follows ; 1) 90 minutes ozonation with $75mgO_3/min$($4.5gO_3/hr$) was conducted at pH 4,7, 10 to remove $COD_{Cr}$. Removal efficiencies were investigated with 48.2%, 52.6%, and 62.3% respectively. As increasing pH, $COD_{Cr}$ removal efficiencies were increased, it was considered that hydroxyl radical($OH{\cdot}$) which strongly oxdize and nonselectively react with organic compounds, was rapidly produced by ozone self-decomposition at high pH. 2) Alum, ferric chloride and ferrous sulfate were used for coagulation as inorganic coagulant. Ferric chloride was investigated with optimal coagulant, and it removed $COD_{Cr}$ about 12.0% at pH5 and dosage of $2,000m{\ell}/{\ell}$. Cation(C-101P), anion(A-601P) and nonion(SC-050) were tested to remove organic pollutants in landfill leachate. Cation(C-101P) was investigated with the most effective organic coagulant, and removal efficiency of $COD_{Cr}$ was 19.8% at pH5 and dosage of $100m{\ell}/{\ell}$. 3) Color and turbidity were removed up to 88.6%, 97.0% at pH10, when contacted 90 minutes with ozone, respectively. These removal efficiencies were much higher than those of $COD_{Cr}$ and $COD_{Mn}$. It was considered that ozone could oxdize the triggering materials of color and turbidity selectively and preferentially. 4) $COD_{Cr}$, color and turbidity were more effectively removed with pre-ozonation than post-ozonation about 8%, 3.5% and 1% respectively. These results were well corresponed with other's studies that pre-ozonation will increase the effect of coagulation.
The present study investigated anti-oxidative and anti-inflammatory activity of glycoprotein isolated from Morus Indica Linne (MIL glycoprotein). We found that MIL glycoprotein has a molecular weight of 32 kD and consists of carbohydrate (40.03%) and protein (59.97%), and that it has a strong scavenging activity against 1,1-diphenyl-2-picrylhydrazyl (DPPH), hydroxyl radical $({\cdot}OH)$, and superoxide anion $(O_2{\cdot}\;^-)$ radicals. In addition, MIL glycoprotein had a stable character and an optimal DPPH radical scavenging activity in the alkaline and neutral pH solution, and up to at 105. However, the results indicated that it has a minimal scavenging activity in the metal ionic solution ($Ca^{2+}$, $Mn^{2+}$, and $Mg^{2+}$) in the presence of EDTA. In addition, we further investigated whether MIL glycoprotein scavenges oxygen radicals and blocks inflammation-related signals in the bisphenol A (BPA)-stimulated Raw 264.7 cells. The results in this study showed that it has a character to scavenge the productions of reactive oxygen species (ROS) and nitric oxide (NO) dose-dependently. Also it blocked the activities of inflammation-related signals such as nuclear factor-kappa B ($NF-{\kappa}B$) and inducible nitric oxide synthase (iNOS). For example, it had an inhibitory effect on the activation of $NF-{\kappa}B$ (p50) and iNOS proteins at 200 ${\mu}g/mL$ MIL glycoprotein. Here, we speculate that MIL glycoprotein is one of natural antioxidants and of modulators of the BPA-induced inflammation.
