Quantification of Ceruloplasmin in Wale Rats Exposed to ${\gamma}$-radiation by Enzyme Linked Immunosorbent Assay (ELISA방법에 의한 방사선 피폭 후 흰쥐 혈액 내에서의 ceruloplasmin 정량)

  • Adult male rats were exposed to a whole body with a single dose of 1.0, 2.0, 3.0, 5.0, and 7.0 Gy. The animals were sacrificed 48, 72, 96 and 216 hours following exposure. A competitive enzyme linked immunosorbent assay(ELISA) with antigen immobilized on the solid phase has been developed to measure ceruloplasmin in rat serum and complete dose response curves. Ceruloplasmin was purified from the plasma of turpentine treated male rats. Coating of ceruloplasmin had more effectiveness in 10 mM Tris-HCI, 150 mM sodium chloride, pH 7.4 than in 50 mM carbonate/bicarbonate buffer, pH 9.6. The coating range for ceruloplasmin was $70{\sim}140ng$/well. Levels of ceruloplasmin increased to maximum on the $72{\sim}96$ hours after irradiation. Slope of between response and dose was greatest value 96 hours following irradiation. Normal ceruloplasmin levels were not recorded 216 hours following exposure. In 0.1 Gy irradiated group, levels of ceruloplasmin also increased to maximum on the $72{\sim}96$ hours following irradiation. The concentration of this protein remained significantly different from control value, 196 hours after exposure. Ceruloplasmin was identified as one of the major acute phase protein following irradiation and further studies about gene expression and regulation would be necessary for radiation protection.

Anti-proliferative and Pro-apoptic Effects of Dan-Seon-Tang in Human Leukemia Cells (인체 혈구암세포에 대한 단선탕(丹仙湯) 추출물의 증식억제 및 세포사멸 유도에 관한 연구)

  • Objectives : This study investigated the biochemical mechanisms of anti-proliferative and pro-apoptotic effects of the water extract of Dan-Seon-Tang (DST) in human leukemia U937 cells. Methods : U937 cells were exposed to DST and growth inhibition was measured by MTT assay. Results : Exposure of U937 cells to DST resulted in the growth inhibition in a concentration-dependent manner. This inhibitory effect was associated with morphological changes and apoptotic cell death such as formation of apoptotic bodies, increased populations of apoptotic-sub G1 phase and induction of DNA fragmentation. The induction of apoptotic cell death in U937 cells by DST was associated with up-regulation of death receptor 4 (DR4) and down-regulation of Bid, surviving and cellular inhibition of apoptosis protein-2 (cIAP-2) expression. DST treatment also induced the proteolytic activation of caspase-3, caspase-8 and caspase-9, and a concomitant degradation of caspase-3 substrate proteins such as poly (ADP-ribose) polymerase (PARP), phospholipase (PLC)-${\gamma}1$, ${\beta}$-catenin and DNA fragmentation factor 45/inhibotor of caspase activated DNAse (DFF45/ICAD). Furthermore, apoptotic cell death by DST was significantly inhibited by caspase-3 specific inhibitor z-DEVD-fmk, demonstrating the important role of caspase-3. Conclusions : These findings suggest that herb prescription DST may be a potential chemotherapeutic agent for the control of human leukemia U937 cells; further study is needed to identify the active compounds.

Transformation Behavior of Retained Austenite on Microstructure and Mechanical Properties in Fe-0.7wt%C-2.3wt%Si Steel (Fe-0.7%C-2.3%Si강의 미세조직과 기계적 성질에 미치는 잔류 오스테나이트 변태 거동)

  • This steel has been synthesized integrating concepts from Austempering Ductile Cast Iron (ADI) technology. While ADI has excellent mechanical and physical properties, the Young's modules of ADI is approximately 20% lower than steel. In addition, the presence of graphite nodules in ADI can be sites of crack initiation, where fracture takes place at graphite matrix interface. Because of this limitations of ADI, there has been a growing interest in austempered steels as structural materials in resent years. In this investigation, a new steel with microstructure composed of ferrite and austenite and with simultaneous high tensile strength (1,150 MPa) and high ductility (33%) was developed. The goal of this investigation is to obtain a better understanding of deformation and transformation behaviour in high carbon retained austenite(${\gamma}_{HC}$) and over-saturated ferrite(${\alpha}$) during the plastic deformation. A detailed study of the microstructure of this steel was carried out by means of X-ray diffraction (XRD) and electron back scattering diffraction (EBSD) technic. In this way it was shown that BCC phase (BCC) took up the larger part of the nominal strain whereas the a part of retained austenite responded to the mechanincal load by partial martensite transformation, and misorientation change in the retained austenite after plastic strain could be attributed to the large elongation.

Chemopreventive Actions of Blond and Red-Fleshed Sweet Orange Juice on the Loucy Leukemia Cell Line

  • Background: Red-fleshed sweet orange juice (ROJ) comes from a new variety of citrus cultivated in Brazil that contains high levels of ${\beta}$-carotene and lycopene, and similar amounts of hesperidin (HSP) and nutrients, equivalently to blond orange juice (BOJ). Such bioactive compounds are associated with chemopreventive actions in several cancer cell lines. The purpose of this study was to examine the cytotoxicity, cell cycle, apoptosis, and cytokine secretion after BOJ, ROJ, and HSP treatment of a novel T acute lymphoblastic leukemia cell line, Loucy. Materials and Methods: Loucy cells were incubated for 24-h with BOJ, ROJ, and HSP, and the viability was measured using trypan blue. Cell cycling and apoptosis were assessed by propidium iodide (PI) and annexin V-FITC/PI flow cytometry, respectively. Secretion of cytokines $IL-1{\alpha}$, $IL1-{\beta}$, IL-2, IL-4, IL-6, IL-10, IL-17A, $IFN{\gamma}$, $TNF{\alpha}$, $TGF{\beta}$, $MIP{\alpha}$, and $MIP{\beta}$ was determined by ELISA array. Results: BOJ and ROJ treatments promoted Loucy cell cytotoxicity. Additionally, BOJ induced cell cycle arrest in the G0/G1 phase, and decreased the cell accumulation in the G2/M. ROJ decreased only the G0/G1 fraction, while HSP did not change the cell cycle. BOJ led to apoptosis in a different fashion of ROJ, while the first treatment induced apoptosis by increase of late apoptosis and primary necrotic fractions, the second increased early and late apoptosis, and primary necrotic fraction compared to positive controls. HSP had no effect on apoptosis. IL-6 and IL-10 were abrogated by all treatments. Conclusions: Taking together, these results suggest potential chemopreventive effects of BOJ and ROJ on Loucy cells.

Inhibition of Growth and Induction of Differentiation of SMMC-7721 Human Hepatocellular Carcinoma Cells by Oncostatin M

  • Oncostatin M (OSM) is a multifunctional cellular regulator acting on a wide variety of cells, which has potential roles in the regulation of gene activation, cell survival, proliferation and differentiation. Previous studies have shown that OSM can induce morphological and/or functional differentiation and maturation of many tumor cells. However, the action of OSM on the induction of differentiation of human hepatocellular carcinoma (HCC) has not been reported. Here, we investigated the effects of different concentrations of OSM on human HCC cell line SMMC-7721 growth, proliferation, cell cycling, apoptosis and differentiation in vitro. Cell growth was determined via MTT assay, proliferation by cell cycle analysis, apoptosis by flow cytometry, morphology by transmission electronic microscopy, and cell function by detection of biochemical markers. Our results demonstrated that OSM strongly inhibited the growth of SMMC-7721 cells in a dose-dependent manner, associated with decreased clonogenicity. Cell cycle analysis revealed a decreased proportion of cells in S phase, with arrest at G0/G1. The apotosis rate was increased after OSM treatment compared to the control. These changes were associated with striking changes in cellular morphology, toward a more mature hepatic phenotype, accompanied by significant reduction of the expression of AFP and specific activity of ${\gamma}$-GT, with remarkable increase in secretion of albumin and ALP activity. Taken together, our findings indicate that OSM could induce the differentiation and reduce cell viability of SMMC-7721 cells, suggesting that differentiation therapy with OSM offers the opportunity for therapeutic intervention in HCC.

A Study on the Characteristics of Heat Treated ERW Weld Seam and the Technology of Seam Annealing (고장력 강재의 전기저항 용접부 열처리 특성 및 기술에 대한 연구)

  • To fine seam annealer capacity of through thickness seam annealing in terms of through thickness microstructure change with increased toughness and elongation leaving heat trace on it, high strength steel pipes of ERW with different thickness were tested in different seam annealing temperature measured on the outer surface of pipes. Annealing temperature and microstructure of the weld seam were changed through applied seam annealing condition. Toughness and tensile test with hardness and microstructure analysis were done on the annealed weld seam to fine its characteristics as a primary step and annealing characteristics according to different seam annealing condition. Through a study of annealed ERW weld seam characteristics and seam annealing technology, amount of electric power should apply in decreased manner to arranged inductors of annealer in the order of 1st, 2nd, 3rd, so on for proper seam annealing. For example of 15.4mm thick and 610mm outside diameter pipe, applied power for proper seam annealing is 600 -650kw at 1st inductor, 450 - 500kw at 2nd inductor, 200-250 kw at 3rd inductor of annealer during 10 - 12M/minute moving speed of pipe. Also, the penetration depth of heat trace along the thickness direction of weld during seam annealing can be estimated through the equation 17mm/kv$\times$voltage(kv) with the microstructure and hardness analysis of thick weld seam as well as study of seam annealing and comparison of cooling condition to CCT diagram of low carbon high strength steel. From this result, the difference between the technological applicability of full annealing condition based on phase diagram and full penetration of heat trace based on CCT diagram along the thickness of weld seam is discussed.

Induction of Apoptotic Cell Death by Methanol Extract of Houttuynia cordata Thunb. in A549 Human Lung Carcinoma Cells (어성초 메탄올 추출물에 의한 A549 인체 폐암세포 사멸유도에 관한 연구)

  • Houttuynia cordata Thunb, well known as 'E-Sung-Cho' in Korea, is traditional medicinal plant generally used in Oriental medicine therapy. We previously reported that the water extract of H. cordata inhibited cell proliferation and induced apoptosis in human breast carcinoma cells. In the present study, we investigated the biochemical mechanisms of anti-proliferative effects by the methanol extract of H. cordata (MEHC) in human lung carcinoma A549 cells. It was found that MEHC could inhibit the cell growth in a dose-dependent manner, which was associated with morphological change and apoptotic cell death as determined by formation of apoptotic bodies, DNA fragmentation and increased populations of apoptotic-sub G1 phase cells. Apoptosis of A549 cells by MEHC was also connected with a down-regulation of anti-apoptotic Bcl-2 and inhibitor of apoptosis proteins (IAPs) expression. MEHC treatment induced the proteolytic activation of caspase-3, caspase-8 and caspase-9, and a concomitant inhibition of poly(ADP-ribose) polymerase (PARP), ${\beta}$-catenin and phospholipase (PLC)-${\gamma}$1 protein expression. Taken together, these findings provide important new insights into the possible molecular mechanisms of the anti-cancer activity of H. cordata.

Effect of High Frequency Heat Treatment on the Microstructure and Wear Properties of Ni based Self Fluxing Composite Coating Layer Manufactured by HVOF Spray Process (High Velocity Oxygen Fuel 공정으로 제조된 Ni 계 자용성 복합 코팅 소재의 미세조직과 마모 특성에 미치는 고주파 열처리의 영향)

  • In this study, the formation, microstructure, and wear properties of Colmonoy 88 (Ni-17W-15Cr-3B-4Si wt.%) + Stellite 1 (Co-32Cr-17W wt.%) coating layers fabricated by high-velocity oxygen fuel (HVOF) spraying are investigated. Colmonoy 88 and Stellite 1 powders were mixed at a ratio of 1:0 and 5:5 vol.%. HVOF sprayed self-fluxing composite coating layers were fabricated using the mixed powder feedstocks. The microstructures and wear properties of the composite coating layers are controlled via a high-frequency heat treatment. The two coating layers are composed of ${\gamma}-Ni$, $Ni_3B$, $W_2B$, and $Cr_{23}C_6$ phases. Co peaks are detected after the addition of Stellite 1 powder. Moreover, the WCrB2 hard phase is detected in all coating layers after the high-frequency heat treatment. Porosities were changed from 0.44% (Colmonoy 88) to 3.89% (Colmonoy 88 + ST#1) as the content of Stellite 1 powder increased. And porosity is denoted as 0.3% or less by inducing high-frequency heat treatment. The wear results confirm that the wear property significantly improves after the high-frequency heat treatment, because of the presence of well-controlled defects in the coating layers. The wear surfaces of the coated layers are observed and a wear mechanism for the Ni-based self-fluxing composite coating layers is proposed.

Deformation and Failure Behavior during Thermo-Mechanical Fatigue of a Nickel-Based Single Crystal Superalloy (열기계적 피로에 따른 단결정 니켈기 초내열합금의 변형 및 파괴거동)

  • The out-of-phase thermo-mechanical fatigue (OP TMF) in a <001> oriented single crystal nickel-based superalloy CMSX-4 has been studied. OP TMF life was less than a half of low cycle fatigue(LCF) life in spite of a small hysteresis loop area of OP TMF compared to that of LCF. The failure was caused by the initiation of a crack at the oxide-layered surface followed by its planar growth along the <100> ${\gamma}$ channel in both LCF and OP TMF. However, deformation twins appeared near the major crack of OP TMF. The multiple groups of parallel twin plates on {111} planes provided a preferential path for crack propagation, which caused a significant decrease in OP TMF life. Additionally, the analysis on the surface crack morphology revealed that the tensile strain at the minimum temperature of OP TMF was found to accelerate the crack propagation.

Preparation and Characterization of Silicone Hydrogel Lens Containing Poly(ethylene glycol) (PEG를 포함한 실리콘 수화젤 렌즈의 제조 및 특성)

  • Silicone hydrogels incorporated with poly(ethylene glycol)(PEG) were prepared and characterized to evaluate the effects of PEG on contact lenses. The silicone hydrogels were copolymerized with methacryloxypropyl tris(trimethylsiloxy) silane (TRIS), methyl methacrylate (MMA), N,N-dimethyl acrylamide (DMA) and PEG-containing monomers such as poly(ethylene glycol) methyl ether methacrylate (PEG- MEM). The silicone hydrogels were characterized using Fourier transform infrared spectroscopy (FT-IR), electron spectroscopy of chemical analysis (ESCA), and scanning electron microscopy (SEM). Water absorbance, water contact angle and light transmittance of the silicone hydrogels were evaluated. The experiments of protein adsorption were also carried out to evaluate the protein adsorption in tears. The peak intensity of C-O bond was increased by the incorporation of PEG-containing monomers and thus PEG incorporation into silicone hydrogels could be confirmed. Phase separation was not shown by the SEM observation of the cross-section of silicone hydrogels. Water absorbancy was increased, while water contact angle and light transmittance were decreased with increasing incorporation of the PEG-containing monomers. The absorption of proteins in tears, albumin, lysozyme and $\gamma$-globulin, on the surface of silicone hydrogels was decreased with increasing incorporation of the PEG-containing monomers.
