방사선조사(放射線照査)에 의한 Prostaglandin E2 및 자연살해세포(自然殺害細胞) 활성도(活性度)의 변화(變化) (Relationships between Radiation-induced Prostaglandin E2 and Natural Killer Cell Activity in Mice)

  • The number of splenic lymphocyte, serum prostaglandin $E_2$ level and natural killer cell activity were assayed after single whole body irradiation of a sublethal dose of $^{60}Co-{\gamma}$ ray to C57BL/6J mice. With a view to knowing the relationships between radiation induced prostaglandin $E_2$ level and the normal natural killer cell activity after natural killer cell-target cell conjugation, The change of normal natural killer cell activity were measured by administration of prostaglandin $E_2$ containing serum from irradiated mice. The results were summarized as follows; 1. The total number of splenic lymphocyte was significantly decreased by irradiation and the number was not affected by indometacin, prostaglandin synthesis inhibitor, treatment. 2. Serum prostaglandin $E_2$ level was increased in irradiated mice, but indometacin treated mice group showed low level of prostaglandin $E_2$. 3. In the case of irradiated mice, natural killer cell activity was not shown any difference between irradiated group and indometacin combined group. But when natural killer cell-target cell conjugations were exposed to the serum of each group during cytotoxic activity assay, whereas the normal natural killer cell activity was significantly decreased by treatment of serum from irradiated mice, the activity was not changed by treatment of indometacin pretreated mice serum. This result indicated that the prostaglandin $E_2$ induced by the radiation inhibited the post-target binding cytolytic process of natural killer activity.

Involvement of Cdc25c in Cell Cycle Alteration of a Radioresistant Lung Cancer Cell Line Established with Fractionated Ionizing Radiation

  • Cancer patients often suffer from local tumor recurrence after radiation therapy. Cell cycling, an intricate sequence of events which guarantees high genomic fidelity, has been suggested to affect DNA damage responses and eventual radioresistant characteristics of cancer cells. Here, we established a radioresistant lung cancer cell line, A549R, by exposing the parental A549 cells to repeated ${\gamma}$-ray irradiation with a total dose of 60 Gy. The radiosensitivity of A549 and A549R was confirmed using colony formation assays. We then focused on examination of the cell cycle distribution between A549 and A549R and found that the proportion of cells in the radioresistant S phase increased, whereas that in the radiosensitive G1 phase decreased. When A549 and A549R cells were exposed to 4 Gy irradiation the total differences in cell cycle redistribution suggested that G2-M cell cycle arrest plays a predominant role in mediating radioresistance. In order to further explore the possible mechanisms behind the cell cycle related radioresistance, we examined the expression of Cdc25 proteins which orchestrate cell cycle transitions. The results showed that expression of Cdc25c increased accompanied by the decrease of Cdc25a and we proposed that the quantity of Cdc25c, rather than activated Cdc25c or Cdc25a, determines the radioresistance of cells.

농산폐기물(農産廢棄物)의 성분분석(成分分析) 및 효소분해(酵素分解)에 관(關)한 연구(硏究) (A Study on the Composition and Enzymatic Hydrolysis of Some Agricultural Waste Products)

  • 주요(主要)한 농산폐기물(農産廢棄物)의 사료적(飼料的) 이용(利用)을 위하여 그들의 성분분석(成分分析)을 실시하였고, 왕겨의 효소적(酵素的) 가수분해(加水分解)를 시도하여 다음과 같은 결과(結果)를 얻었다. 1) 왕겨, 볏짚, 보리짚, 밀짚의 일반성분(一般成分)과 탄수화물(炭水化物) 조성(組成)을 분석(分析)하였다. 2) 왕겨 분해효소(分解酵素) 생산균주(生産菌株)로서 Aspergillus niger를 선정하였고 이를 수세(水洗)밀기울에 배양할 때의 최적조건(最適條件)은 배양일수(培養日數) 3일, 철수량(撤水量) 55%, 철수(撤水) pH는 중성(中性)이었으며 효소추출(酵素抽出)시간은 1시간 정도가 적당하였다. 3) Aspergillus niger에서 생산(生産)한 조효소제(粗酵素劑)의 xylan가수분해율(加水分解率)은 63%, 왕겨 가수분해율(加水分解率)은 7%이었고, 작용(作用) 최적(最適) pH는 xylan 4.0, 왕겨 4.5이었다. 4) 왕겨의 효소분해(酵素分解)는 감마선 조사(照射) $0.5{\sim}5Mrad$에 의하여 풍건상태에서는 일반적으로 억제되었으나 수화상태(水和狀態)에서는 선량(線量)이 높아짐에 따라 그 효과(效果)가 적었다.

발아억제 선량의 전리에너지가 마늘의 성분변화에 미치는 영향 (Chemical Changes in Garlic Bulbs Resulting from Ionizing Energy Treatment at Sprout-Inhibition Dose)

  • 마늘의 품질보존을 위한 수단으로서 전리에너지인 장마선의 이응이 가능해짐에 따라, 발아억제 선량인 0.1kGy 처리와 저장조건$(3{\pm}1^{\circ}C,\;75{\sim}85%\;R.H.;\;12{\pm}5^{\circ}C,\;75{\sim}85%\;R.H.)$이 마늘의 몇가지 성분변화에 미치는 영향을 조사하였다. 0.1kGy의 감마선 조사는 시료의 수분함량에 거의 영향을 미치지 않았으나 실온저장 10개월 후의 대조시료는 조사시료에 비해 수분함량이 유의적으로 감소되었다(P<0.05). 발아억제 처리는 저장중 시료의 총당 함량의 변화를 완만하게 하였으며, 저온저장 8개월 후 대조시료의 유리당 함량은 조사시료에 비해 유의적으로 증가되었다. 발아억제 선량의 감마선 처리는 시료의 아스코르브산과 지방산 참량에 거의 영향을 주지 않았으나 아미노산 조성중 글루탐산과 아스파르트산 함량에는 영향을 미치는 것으로 나타났다.

양성자 및 감마선처리에 의한 유채 내염성 변이계통 선발 (Selection of Salt Tolerant Rapeseed (Brassica napus L.) Mutant Lines Induced by Irradiation of Proton Ion Beams and Gamma Ray)

  • This study was carried out to develop salt tolerant varities of rapeseed (Brassica napus L.) which can be grown in the high salty reclaimed land. The seeds of three varieties 'Naehan', 'Tammi', and 'Halla' were treated by proton ion beams and gamma rays with 0 to 2,000 Gy. For the selection of salt tolerant lines, emergence and survival rate, and growth characteristics of $M_2$ to $M_4$ generations were investigated in the Saemangeum reclaimed fields with the different salt concentrations. The lines with potential salt tolerance were selected in the $M_4$ generation and tested indoor for their growth characteristics. There was no significant changes in the soil pH for $M_4$ generation during growth period. However, soil EC was higher in early spring than sowing period (mid October). In $M_4$ generation test, the seeds of original and selected line showed high rates of emergence and survival, as determined one month after sowing. After wintering, however, the original varieties showed the significant reduction in the survival rate, while the selected lines showed a higher survival rate and good growth, leading to the completion of their life cycle. Consequently we selected 9 lines from $M_4$ generation with better performance in growth and yield. Soil EC was $2.8{\sim}4.3dS{\cdot}m^{-1}$ during $M_4$ generation growth period. The laboratory test of the lines selected from $M_4$ generation was made for their salt tolerance potential. The selected lines showed higher chlorophyll and proline contents than the original varieties. There was also no significant difference in the emergence rate of seed between the original and selected varieties. In 200 mM natural sea salt, the $N{\gamma}600-21-1-641$ line derived from 'Naehan' was the highest in growth rate, leaf chlorophyll and proline contents. $T{\gamma}800-20-2-461$ line derived from 'Tammi' didn't show significant difference in growth rate compared to original variety in 200 mM and withered in 250 mM like other lines as time passed. $H{\gamma}200-7-1-740$ line showed similar growth and chlorophyll content compared to its original variety.

Functionalization of polyethylene by graft copolymerization for separation processes

  • Incorporation of polar functional moieties into polyethylene (PE) film has been achieved by graft copolymerization of polar monomers such as methacrylic acid (MAAc) and acrylamide (AAm) on to PE film, preirradiated with ${\gamma}$-rays from $^{60}Co$ source, using benzoyl peroxide (BPO) as initiator in aqueous medium. Percentage of grafting of MAAc and AAm was determined as a function of irradiation dose, monomer and initiator concentration, temperature, reaction time and amount of water. Maximum percentage of grafting of MAAc (1453%) and AAm (21.28%) was obtained at [MAAc] = $235.3{\times}10^{-2}$ mol/L, [AAm] = $23.4{\times}10^{-2}$ mol/L, [BPO] = $5.5{\times}10^{-2}$ mol/L and $16.5{\times}10^{-2}$ mol/L at $80^{\circ}C$, $90^{\circ}C$ in 180 min and 90 min respectively. The grafted PE films were characterized by FTIR, Thermogravimetric analysis (TGA) Scanning Electron Micrography (SEM) and X-ray diffraction methods. Some selective properties of grafted films such as swelling behavior, ion and metal uptake have been carried out. The biodegradation studies of the grafted PE films have also been investigated. The grafted films developed superior swelling behavior with maximum swelling (480%) in water as compared to pristine PE (13.55%), better thermal stability and ion and metal uptake studies showed promising results that can be effectively used for desalination of brackish water and separation of metals from the industrial effluents.

Thermoluminescence 측정에 의한 감자와 마늘의 방사선 조사유무 확인 (Detection of Irradiated Potato and Garlic by Thermoluminescence Measurement)

  • 발아억제 선량범위인 0.15 kGy와 0.30 kGy의 감마선과 전자선이 조사된 감자와 마늘을 대상으로 thermoluminescence (TL) 측정에 의해 방사선 조사유무를 확인하였다. 감자와 마늘로부터mineral을 추출하여 TL 측정을 함으로써 방사선 조사선량과 TL intensity 간의 높은 상관관계를 확인하였다. 방사선 조사되지 않은 시료들은 특이한 TL glowcurve를 나타내지 않았으나 방사선 조사된 시료들은 $200{\sim}260^{\circ}C$부근에서 명확한 TL glowcurve를 나타내었다. 시료의 TL intensity는 조사선량에 따라 증가하였으나 같은 조사선량에서도 방사선 조사선원에 따라 시료의 TL intensity는 일정하지 않았다. 그러나 이상의 결과에서 볼 때 발아억제를 위한 감자와 마늘의 방사선 조사는 TL glowcurve의 측정으로써 조사유무의 확인이 가능하였다.

Line Image Correction of the Positron Camera in the Secondary Beam Course of HIMAC

  • A positron camera, consisting of a pair of Anger-type scintillation detectors, has been developed for verifying the ranges of irradiation beams in heavy-ion radiotherapy. Images obtained by a centroid calculation of photomultiplier outputs exhibit a distortion near the edge of the crystal plane in an Anger-type scintillation detector. The images of a $\^$68/Ge line source were detected and look-up tables were prepared for the position correction parameters. Asymmetry of the position distribution detected by the positron camera was prevented with this correction. As a result, a linear position response and a position resolution of 8.6 mm were obtained over a wide measurement field.

  • Food allergy is defined as adverse reactions toward food mediated by aberrant immune mechanisms. Cow's milk allergy is one of the most common food allergies in childhood. This allergy is normally outgrown in the first year of life, however 15% of allergic children remain allergic. Cow's milk allergy seem to be associated with casein (${\alpha}_{s1}$-CN), ${\beta}$-lactoglobulin and whey protein. In addition to this, many other milk proteins are antigenic and capable of inducing immune responses. Various food processing affects the stability, structure and intermolecular interactions of cow milk proteins, as a result reduction the allergenic capacity. Heating, hydrolysis, chemical, proteolytic and other processes such as gamma-ray irradiation, high pressure, using probiotics treatments of milk to obtain hypoallergenic milk have been developed to reduce allergic reactions.

은나노 입자가 첨착된 활성탄의 항균특성에 관한 연구 (Study of Anti-bacterial Properties for Impregnated Activated Carbon by Silver Nano-particles)

  • 본 연구에서는 질산은 용액을 감마선 조사에 의하여 은나노 입자를 제조한 후, 이것을 활성탄과 혼합하여 은/활성탄 복합체를 제조하여 대장균에 대한 항균특성을 조사하였다. 제조된 은/활성탄 복합체의 특성은 주사전자현미경, X-선 회절법 그리고 원자흡수분광법에 의해 알아보았다. 은/활성탄 복합체의 대장균에 대한 억제농도는 0.387 ppm으로 나타났으며 대장균에 대한 사멸농도는 1.017 ppm이었다. 이 결과로 은/활성탄 복합체의 대장균에 대한 우수한 항균효과를 확인할 수 있었다.
