  • TEM 이론의 기초가 되는 델타함수, 콘볼루션 적분, 퓨리에 변환에 관한 개념을 소개하고 이에 대한 응용으로 슬릿함수, 현저한 폭을 갖는 2개의 슬릿, 유한 크기의 파동 train, 좁은 슬릿의 주기적인 배열, 임의의 주기 함수, diffraction grating, 회절 격자, 그리고 gaussian 함수에서의 퓨리에 변환에 관한 수학적인 방법의 적용을 소개하였다.

  • 示差熱曲線으로 二酸化망간의 各變態를 分析化學的인 面에서 檢討하였다. 各變態의 轉移溫度는 ${\alpha},{\beta}-MnO_2$인 경우 $580^{\circ}C∼650^{\circ}C$였고, ${\gamma}-MnO_2$인 경우 $405^{\circ}C~605^{\circ}C$, ${\delta}-MnO_2$인 경우 $860^{\circ}C~875^{\circ}C$였으며 吸熱 peak를 나타냈다. 轉移가 完結되는 회복點의 溫度는 試料의 量에 따라서 달랐으나 $T_{max}$點의 溫度는 거의 一定하였다. peak의 모양은 ${\alpha},{\beta}-MnO_2$${\gamma}-MnO_2$는 작고 완만하였으며 ${\delta}-MnO_2$는 크고 예민하였다.

  • Bacillus sphaericus 1593 is pathogenic to the larvae of a number of mosquito species that are known as important vectors for the transmission of certain human and animal diseases. As a preliminary experiment for developing a multfunctional B. sphaericus 1593 as a potent antagonist, we investigated the conditions for the protoplast transformation system of B. sphaericus 1593 using the plasmid pGB215-110$\Delta$B. The protoplast of B. sphaericus 1593 were obtained most efficiency by treating the cells with 500 $\mu$g/ml of lysozyme in the SMM buffer containing 0.5 M sucrose at pH 8.0 and 40$\circ$C for 60 minutes. The cell wall was regenerated on the plate containing 1.2% agar and 0.8 M mannitol. Under the best condition for protoplast formation and regeneration established in the work the highest frequency of transformation was achieved with the 40% PEG (M.W 4,000) treatment for 15 minutes of incubation at 4$\circ$C, and subsequently for 120 minutes incubation at 30$\circ$C for phenotypic expression. The highest transformation efficiency were observed at 1.0 $\mu$g/ml of the final concentration of the plasmid DNA and the plasmids were found to be fairly stable since about 70% of the plasmids were maintained after 8 successive daily transfers onto the fresh medium.

Solvent effect on enzymatic steroid transformation

  • As part of our endeavor to increase the productivity of steroid by enzymatic transformation of corticosteroids, attempts have been made to increase the solubility of steroids by using some organic solvents. When the solubility of steroids is the rate limiting factor in the steroid transformation, it was found that the use of solvents significantly improved the yield. Hydrocorisone as a substrate and 3-ketosteroid .DELTA.$^{1}$ dehydrogense as an immobilized whole cell enzyme were employed as the model system for this study. It was found that the yield of product, prodnisolone, goes through a maximum with an increase in the solvent concentration. At a high solvent concentration, the solvent showed a toxic effect and it causes a decrease in the product yield by the second order inhibition mechanism. Among the solvents evaluated, methanol and ethanol were found to be the best. These alcohols are not only good solvents but also showed minimal toxic effect. Based on the experimental results, it was concluded that the productivity of steroid can be increased by usign well selcted solvents systems for the enzymatic transformation of steroids.

다중 회귀분석법을 이용한 스캐너-모니터간 색보정에 관한 연구 (A study on the color management between scanner and monitor using multiple regression method)

  • 본 연구는 스캔된 이미지의 원본색이 모니터에 그대로 디스플레이 될 수 있게 하는 것을 목적으로 한다. 색보정 과정은 스캐너의 색 특성 묘사를 통한 원본색의 XYZ 추정, 기준 광원의 차이를 고려한 XYZ 변환, XYZ와 RGB간의 색공간 변환으로 구성된다. 특히, XYZ 추정에 있어서 스캐너에 입사되는 빛의 세기와 출력신호간의 비선형적 관계를 두 구간으로 나누어 묘사함으로써 그 정확도를 높였다 EPSON Expression 1680 스캐너를 대상으로 실시한 결과, 228가지 기준색의 기준값과 추정값의 평균 색차는 1.47 $\Delta$ $E_{UV}$ * 이었으며, 36가지의 유채색과 22가지의 무채색으로 구성된 시험색의 평균 색차가 각각 1.51 $\Delta$ $E_{UV}$ * 와 0.90 $\Delta$ $E_{UV}$ * 이었다. 또한 시험색 36가지에 대해 기준값과 추정값으로 부터 산출된 sRGB 모니터 입력신호를 동일 모니터에 디스플레이한 결과, 두 이미지의 색이 동일하게 보였다. sRGB 모니터 입력신호를 동일 모니터에 디스플레이한 결과, 두 이미지의 색이 동일하게 보였다.

Transformation of Ginseng Saponins to Ginsenoside $Rh_2$ by Acids and Human Intestinal Bacteria Activities of Their Transformants

  • When ginseng water extract was incubated at $60^{\circ}C$ in acidic conditions, its protopanaxadiol ginsenosides were transformed to ginsenoside $Rg_3$ and ${\Delta}^{20}$-ginsenoside $Rg_3$. However, protopanaxadiol glycoside ginsenosides $Rb_1, Rb_2$ and Rc isolated from ginseng were mostly not transformed to ginsenoside $Rg_3$ by the incubation in neutral condition. The transformation of these ginsenosides to ginsenoside $Rg_3$ and ${\Delta}^{20}$-ginsenoside $Rg_3$ was increased by increasing incubation temperature and time in acidic condition: the optimal incubation time and temperature for this transformation was 5 h and $60^{\circ}C$ resepectively. The transformed ginsenoside $Rg_3$ and ${\Delta}^{20}$-ginsenoside $Rg_3$ were metabolized to ginsenoside $Rh_2$ and $\Delta^{20}$--ginsenoside $Rh_2$, respectively, by human fecal microflora. Among the bacteria isolated from human fecal microflora, Bacteroides sp., and Bifidobacterium sp. and Fusobacterium sp. potently transformed ginsenoside $Rg_3$ to ginsenoside $Rh_2$. Acid-treated ginseng (AG) extract, fermented AG extract, ginsenoside $Rh_2$ and protopanaxadiol showed potent cytotoxicity against tumor cell lines. AG extract, fermented AG extract and protopanaxadiol potently inhibited the growth of Helicobacter pylori.

Fe-Mn 합금의 γ→ε 마르텐사이트변태에 필요한 구동력 (Driving Forces for γ→ε Martensitic Transformation of Fe-Mn Alloys)

  • Dilatometric experiment and thermodynamic calculation have been performed to determine $M_s$, $A_s$ and driving forces for ${\gamma}{\rightarrow}{\varepsilon}$ martensitic transformation of Fe-Mn alloys. The transformation temperatures($M_s$, $A_s$, $T_o) were decreased with increasing manganese content and were newly formulated as a function of manganese content. Driving force for ${\gamma}{\rightarrow}{\varepsilon}$ martensitic transformation was increased from -75J/mole to -105J/mole with increasing manganese content from 15wt.% to 25wt.%. Transformation temperature hysteresis($A_s-M_s$) was also increased from 50K to 80K with increasing mangenese content from 15wt.% to 25wt.%. The small driving force(-75J/mole~-105J/mole) and small ${\Delta}T$(50K~80K) for ${\gamma}{\rightarrow}{\varepsilon}$ martensitic transformation indicated that Fe-Mn alloys behave like thermoelastic martensitic alloys : We would like to call them semi-thermoelastic martensitic alloys.

연속주조용 탄소강에서 상변화에 따른 열팽창 및 수축 거동 (Thermal Expansion and Contraction Characteristics of Continuous Casting Carbon Steels)

  • The air gap between the metal and mold, formed by shrinkage during solidification, causes surface and subsurface cracks in the continuous casting process. Molten crack on the surface might also occur due to improper heat transfer between them. In order to compensate the air gap in mold design, the thermal contraction is an essential factor. In this study, the thermal contraction and expansion behaviors were examined from the ($\alpha$ and pearlite)/${\gamma}$ to ${\gamma}$/$\delta$ transformations in continuous casting steels by the commercial dilatometer and the self- assembled dilatometer with laser distance measurement. It was found that the thermal contraction and expansion behaviors were very dependant on the phase transformation of the ${\gamma}$/$\delta$ as well as ($\alpha$ and pearlite)/${\gamma}$. The sudden volume change from $\delta$ to ${\gamma}$ which might cause cracks in the continuous casting process, was observed on cooling just below the melting temperature by the self-assembled dilatometer.

  • 본 연구에서는 탄성문제에 관한 Eshelby의 이론을 응용하여 다수의 개재물이 모상 중에 균일하게 분산하고 탄성적으로 불균질한 복합재료의 거시적 응력-변형관계를 정식화하였다. 정식화의 과정에서, 주위의 구속을 받지 않는 어떤 영역에 응력의 발생을 동반하지 않는 변형률 (transformation strain $\varepsilon_{kl}$), 즉 열팽창 제수를 갖는 물체가 온도변화 ${\Delta}T$ 를 갖는 경우의 열팽창 변형 $\alpha$${\Delta}T$나, 물체가 일정한 소성 변형을 받았을 때의 소성 변형 등을 예로들 수 있는 역학 장을 정의하였다. 본 연구에서 전개한 방법은 선형 탄성론에 기초를 두고 있으며 복합체의 탄성거동만이 아니라 탄성-소성 거동의 해석 또한 가능하게 하였다

저온하에서 LNG저장탱크용 멤브레인재(STS 304강)의 피로균열진전거동 (Fatigue Crack Growth Behavior of Membrane Material for LNG Storage Tank at Low Temperatures)

  • The fatigue crack growth behavior of the cold-rolled STS 304 steel developed for membrane material of LNG storage tank was examined experimentally at 293K, 153K and 111K. The fatigue crack growth rate(do/dN) tends to increase as the stress ratio (R) increases over the testing temperature when compared at the same stress intensity factor range($\Delta$K). The effect of R on do/dN is more explicit at low temperatures than at room temperature. The resistance of fatigue crack growth at low temperatures is higher compared with that at room temperature which is attributed to the extent of strain-induced martensitic transformation at the crack tip. The temperature dependence of fatigue crack growth resistance is gradually vanished with an increase in $\Delta$K which correlates with a decreasing fracture toughness with decreasing temperature. Fractographic examinations reveal that the differences of the fatigue crack growth characteristics between room and low temperature are mainly explained by the crack closure and the strengthening due to the martensitic transformation.
