Molecular biologic mechanism of obesity by GGEx18 (경신강지환(輕身降脂丸)18의 분자생물학적인 비만조절 기전에 관한 연구)

  • Objectives : This study was undertaken to verify the modulation mechanism of Gyeongshingangjeehwan18 (GGEx18) in ob/ob male mice. Methods : Eight-week old mice (wild-type C57BL/6J and ob/ob) were used for all experiments. Wild-type C57BL/6J mice were used as lean control and obese ob/ob mice were randomly divided into 5 groups : obese control, GGEx15 (Ephedra sinica Stapf + Rheum palmatum L.), GGEx16 (Ephedra sinica Stapf + Laminaria japonica Aresch), GGEx17 (Rheum palmatum L. + Laminaria japonica Aresch), and GGEx18 (Ephedra sinica Stapf + Laminaria japonica Aresch + Rheum palmatum L.). After mice were treated with several kinds of GGEx for 11 weeks, the mRNA expression of peroxisome proliferator-activated receptor (PPAR) target genes and uncoupling protein (UCP) were measured. In addition, $PPAR{\alpha}$ and $PPAR{\beta}$ transactivation was examined in NMu2Li hepatocytes, C2C12 myocytes, and 3T3-L1 preadipocytes using transient transfection assays. Results : 1. Hepatic $PPAR{\alpha}$ target genes, such as ACOX and VLCAD mRNA levels were significantly increased by GGEx18 compared with obese controls. In skeletal muscle, LCAD mRNA expression was stimulated by GGEx16, GGEx17, and GGEx18, whereas MCAD mRNA expression by GGEx17 and GGEx18. $PPAR{\beta}$ target LPL mRNA levels were also increased by GGEx16, GGEx17, and GGEx18 in skeletal muscle, but adipose LPL mRNA levels were decreased. In addition, GGEx18 upregulated UCP mRNA expression in skeletal muslce. 2. $PPAR{\alpha}$ reporter gene expression was increased by GGEx18 in NMu2Li cells compared with vehicle. $PPAR{\alpha}$ and $PPAR{\beta}$ reporter activities were also increased by all GGEx treatments in C2C12 and 3T3-L1 cells. Conclusions : These results suggest that GGEx can act as $PPAR{\alpha}$ and $PPAR{\beta}$ activators, and that GGEx may regulate obesity by stimulating $PPAR{\alpha}$, $PPAR{\beta}$, and UCP activity. Of the 4 compositions, GGEx18 seems to be most effective in improving obesity and lipid disorders.

The Types of Extended-Spectrum $\beta$-Lactamase (ESBL) Produced by Enteric Bacteria, Klebsiella pneumoniae and Escherichia coli Isolated from Sewage of Wastewater Treatment Plant at Minragdong in Busan, Korea (부산 민락동 오수처리장에서 분리된 장내세균 Klebsiella pneumoniae와 Escherichia coli가 생성한 광범위 베타 락탐(Extended-Spectrum $\beta$-Lactamase, ESBL) 분해효소의 유형)

  • This study was performed to investigate the type of extended-spectrum $\beta$-lactamases (ESBL) produced by bacteria isolated from the sewage of wastewater treatment plant at Minragdong, Suyong-gu in Busan. The facility is located at sushi restaurants and guides its drain water to the wastewater treatment plant at Yonghodong, Nam-gu in Busan. Samples were collected on January, 2009. A total of 19 strains were selected as potential ESBL positive strains through a double disk synergy test. On the basis of the results from biochemical tests including indole, methyl-red, Voges-Proskauer, Simmon's citrate, decarboxylase-dihydrolase and sugar-fermentation tests, the 19 strains were identified with 16 strains of Escherichia coli and 3 strains of Klebsiella pneumoniae. Out of 19 strains, 4 transconjugants against Escherichia coli J53, which is sodium azide resistant recipient strain, were obtained. The plasmids isolated from transconjugants were used for PCR analysis. The type of each extended-spectrum $\beta$-lactamase (ESBL) produced by the strains was determined on the basis of isoelectric focusing analysis and DNA sequencing. The results indicated that the types of ESBL from Klebsiella pneumoniae were SHV-12 (3 strains), and Escherichia coli was SHV-12/TEM-1 (1 strain), respectively.

Resuspension Characteristics of Deposited Fine-Grained Sediments (미세퇴적물(微細堆積物)의 부상특성(浮上特性)에 관한 연구(硏究))

  • A series of experiments on the resuspension of deposited fine-grained sediments were carried out in a recirculating straight flume to investigate the influence of the sediment type, water content and bed shear stress (${\tau}_b$) on the resuspension characteristics of the sediments. The sediments were sampled from Youngkwang coast and Youngdo coast which are located in the western sea and southeastern sea of Korea, respectively. Critical bed shear stress (${\tau}_c$) for resuspension was deduced for each experimental series. For the same sediment, critical bed shear stress for resuspension decreased but suspension mass or rate increased with increasing water content. The resuspension of deposited fine-grained sediments depended strongly on the water content, and the sediment type characterizing the inter-particle bond strength. It has been found that critical bed shear stress for resuspension in the unidirection flow is about 4 times higher than that in the combined wave-current flow, In case of lower bed shear stress, after an initially high suspension, suspension mass approaches a constant value due to the bed hardening with increasing time, but in case of higher bed shear stress, suspension mass increased successively due to the bed softening with time. Initial suspension rate, $E={\alpha}_3({\tau}_b/{\tau}_c-1)^{\beta}$ (where ${\alpha}_3$ and ${\beta}$=empirical constants), was estimated for each experimental series, ${\alpha}_3$ and ${\beta}$ values for the same sediment increased with water content.

Effect of Alcohols Toward the Transphosphatidylation Activity in Phospholipase D Catalyzed Reaction (포스포리파제 D 촉매반응에서 포스파티딜 전달반응 활성에 미치는 알코올의 영향)

  • In the presence of alcohol, phospholipase D (PLD) is known to perform transphosphatidylation activity, during which the overall reaction rate of PLD increased. To elucidate the reaction mechanism of transphosphatidylation further, we investigated rate constants of transphosphatidylation reaction of the purified ${\alpha}$-type PLD from cabbage in the presence of various alcohols. The second-oder rate constants of PLD transphosphatidylation showed a large increase with the primary alcohols examined as expected. In the case of butanol we observed the second-oder rate constant of $33.33{\pm}1.33M^{-1}sec^{-1}$. This second-order rate constant of transphosphatidylation was as 400 times greater as the second-order hydrolysis rate constant of $0.078M^{-1}sec^{-1}$ which was adjusted for the water concentration. A linear free energy relationship between the $pK_a$ of alcohol and transphosphatidylation rate gives a Br${\o}$nsted slope of ${\beta}_{nu}$ = 0.12 ${\pm}$ 0.03. This small ${\beta}_{nu}$ value implicates that the transition state of break down of phosphatidyl-enzyme intermediate (E-P) is likely dissociative. Finally, a reaction mechanism of cabbage PLD is suggested on the basis of our results presented here and the histidine residue known to be located in the active site of cabbage PLD.

Strength Anisotropy through Artificial Weak Plane of Mudstone (인공연약면을 따른 이암의 강도이방성에 관한 연구)

  • The characteristic of induced anisotropy is investigated in this study for the Pohang mudstone involving the cut plane discontinuity. The uniaxial and triaxial compression tests are performed for anisotropic rocks with artificial joint to look into anisotropic strength characteristics. Both the uniaxial compressive strength and triaxial compressive strength show the lowest value at the angle of cut plane, ${\beta}=30^{\circ}$ and the shoulder type of anisotropy is obtained. Anisotropy ratio (Rc) in uniaxial compression measures 9.0, whereas Rc=1.29-1.98 in triaxial compression is appeared. A series of analyses are made with the test results to derive the suitable parameter values when it is applied to the Ramamurthy (1985) failure criterion. The result of uniaxial compression test is analyzed by introducing the n-index into Ramamurthy failure criterion. The result shows that, n=l is suitable for ${\beta}=0^{\circ}{\sim}30^{\circ}$ and n=3 is suitable for ${\beta}=30^{\circ}{\sim}90^{\circ}$. To analyze the result of triaxial compression test by Ramamurthy failure criterion, anisotropy ratio in uniaxial compression test is added to Ramamurthy's equation and material constants are estimated by modified Ramamurthy's equation. When these values are applied back to Ramamurthy failure criterion, the predicted values are well fitted to the test results. And strength anisotropy for failure criteria of Jaeger (1960), McLamore & Gray (1967) and Hoek & Brown (1980) are also investigated.

Phase Transformation During Hot Consolidation and Heat Treatments in Mechanically Alloyed Iron Silicide (기계적 합금화 Iron Silicide의 열간성형 및 열처리에 의한 상변화)

  • An n-type iron$silicide(Fe_{0.98}Co_{0.02}Si_2)$has been produced by mechanical alloying process and consolidated by vacuum hot pressing. Although as-milled powders after 120 hours of milling did not show an alloying progress,${\beta}-FeSi_2$phase transformation was induced by isothermal annealing at$830{\circ}C$for 1 hour, and the fully transformed${\beta}-FeSi_2$phase was obtained after 4 hours of annealing. Near fully dense specimen was obtained after vacuum hot pressing at$ 1100{\circ}C$with a stress of 60MPa. However, as-consolidated iron silicides were consisted of untransformed mixture of ${\Alpha}-Fe_2Si_5$and ${\varepsilon-FeSi$phases. Thus, isothermal annealing has been carried out to induce the transformation to a thermoelectric semiconducting${\beta}-FeSi_2$phase. The condition for${\beta}-FeSi_2$transformation was investigated by utilizing DTA, SEM, and XRD analysis. The phase transformation was shown to be taken place by a vacuum isothermal annealing at$830{\circ}C$and the transformation behaviour was investigated as a function of annealing time. The mechanical properties of${\beta}-FeSi_2$materials before and after isothermal annealing were characterized in this study.

Factors Influencing Diabetes Educational Needs in Patients with Diabetes Mellitus (당뇨병환자의 교육요구도 영향요인)

  • This study was carried out to identify the perceived educational needs of diabetes mellitus type 2(DM 2) patients and the factors influencing their diabetes educational needs. The descriptive and correlation study design was used. The data was collected from January 2, 2012 to February 29 10, 2012 and a total of 346 diabetes patients responded in a structured instrument regarding their characteristics, diabetes knowledge and needs for diabetes education. The data was analyzed using ANOVA, t-test, Scheffe test, Pearson correlation, and stepwise multiple regression. The mean score of the diabetes educational needs was 3.03 (range: 1.09-4.00). Of the factors significantly influencing the diabetes educational needs, age (${\beta}$=-.25, p<.001), diabetes education participation of the family (${\beta}$=.21, p=.003), and self blood sugar test (${\beta}$=-.13, p=.045) explained the 11% variance of the diabetes educational needs. These three factors need to be considered when implementing diabetes education programs for diabetes management.

Characterization of the nar Promoter of Escherichia coli to use as an inducible promoter in Wild-type host Agrobacterium.tumefaciens

  • In this study, the nar promoter of E. coli was characterized to see whether the nar promoter cloned onto pBBR122 can be used as an expression promoter of gram negative microbes. For this purpose, a plasmid with lacZ gene expressing ${\beta}-galactosidase$ instead of the structural genes of nar operon in a gram negative host strain(Agrobacterium.tumefaciens) was used to simplify an assay of induction of the nar promoter. The following effects were investigated to find optimal conditions: methods of inducing the nar promoter, optimal nitrate concentration, maximally inducing the nar promoter, the amount of expressed ${\beta}-galactosidase$ and induction ratio(specific ${\beta}-galactosidase$ activity after maximal induction/specific ${\beta}-galactosidase$ activity before induction). The following results were obtained from the experiments: the growth of Agrobacterium with E.coli nar promoter was not much affected by nitrate concentration in the shake-flask; induction of nar promoter was optimal when Agrobacterium was grown in the presence of 1% nitrate ion at the beginning of culture and when overnight culture was completely grown in the shake-flask before being transferred to other shake-flask; the amount of ${\beta}-galactosidase$ per cell and per medium volume was maximal when Agrobacterium was grown under aerobic condition to $OD_{600}$ of 1.7; then the nar promoter was induced under microaerobic and anaerobic condition made by lowering dissolved oxygen level(DO). After 2-3h of induction in the YEP medium selected as a main culture medium, the specific ${\beta}-galactosidase$ activity became about 17,000 Miller units in the fermentor cluture.

Induced monoterpene and lignin production in mechanically stressed and fungal elicited cultured Cupressus lusitanica cells

  • Cultured Cupressus lusitanica cells induced by various stresses are thought to produce different complexes of defense chemicals to optimize defense. To compare the induced products of two stimulations, we investigated the emission of monoterpenes, biosynthesis of ${\beta}-thujaplicin$, and accumulation of lignin in mechanically stressed and fungal elicited cultured C. lusitanica cells. Both mechanical stress and fungal elicitor caused emission of qualitatively similar monoterpene blends indicating de novo biosynthesis of these compounds after stimulation, while mechanical stress alone is sufficient to induce fungal elicitor-related monoterpene emission. Sabinene and limonene were the dominant compounds over the time course in both volatile blends. Although the emitted volatile blends were qualitatively similar, the time course and the relative ratios of the constituents of the volatile blends differed with the type of stimulation. While fungal elicited cells produced significant amounts of ${\beta}-thujaplicin$ over the 5-day time course, no ${\beta}-thujaplicin$ was observed in the mechanically stressed cells. The production of ${\beta}-thujaplicin$ was the main dissimilarity of the induced products of these two treatments, suggesting that synthesis of ${\beta}-thujaplicin$ is not a general response to all types of stresses, but is a specific response and serves as a strong toxic compound against already invaded fungus. Significantly higher amounts of lignin accumulations were observed in the fungal elicited and mechanically stressed cells on the 5th day after induction. Based on these results, we suggest the composition of induced products was dependent on the method of stimulation.

Tight junction protein 1 is regulated by transforming growth factor-β and contributes to cell motility in NSCLC cells

  • Tight junction protein 1 (TJP1), a component of tight junction, has been reported to play a role in protein networks as an adaptor protein, and TJP1 expression is altered during tumor development. Here, we found that TJP1 expression was increased at the RNA and protein levels in TGF-${\beta}$-stimulated lung cancer cells, A549. SB431542, a type-I TGF-${\beta}$ receptor inhibitor, as well as SB203580, a p38 kinase inhibitor, significantly abrogated the effect of TGF-${\beta}$ on TJP1 expression. Diphenyleneiodonium, an NADPH oxidase inhibitor, also attenuated TJP1 expression in response to TGF-${\beta}$ in lung cancer cells. When TJP1 expression was reduced by shRNA lentiviral particles in A549 cells (A549-sh TJP1), wound healing was much lower than in cells infected with control viral particles. Taken together, these data suggest that TGF-${\beta}$ enhances TJP1 expression, which may play a role beyond structural support in tight junctions during cancer development.
