그라비올라 잎 추출물의 이화학적 특성 및 항산화 활성 (Antioxidative Effects and Chemical Characteristics of Annona muricata Leaf Extracts)

  • 그라비올라(Annona muricata) 잎으로부터 정제수, 에탄올, 메탄올로 추출하여 수율, 총 페놀성 화합물과 이에 속하는 flavonoid 함량, 미네랄 함량을 측정하고 DPPH free radical scavenging 활성, Cu/Fe 환원력, 간 조직의 microsome 생체막의 지질과산화 억제능 및 linoleic acid를 이용한 지질과산화, ${\beta}$-Carotene 탈색화 정도를 비교검토하였다. 그라비올라 잎의 수율은 수용성 추출물이 가장 높았으며(1.76%), 총 페놀성 화합물 함량은 그라비올라의 메탄올 추출물에서 가장높았으나 페놀성 화합물에 속하는 폴리페놀 함량은 에탄올 추출물에서 가장 높은 결과를 나타내었고, 미네랄 함량은 Ca, K, Mg, Na, Mn의 순으로 높게 측정되었다. DPPH를 이용한 radical 소거능을 측정한 결과, 그라비올라 잎의 에탄올과 메탄올 추출물에서 대조구인 BHT 0.05%와 가장 근접한 활성을 나타내었다. Fe와 Cu의 환원력은 모든 군에서 농도 의존적으로 증가하였고, 그 중 에탄올과 메탄올 추출물에서 Cu/Fe 환원력 활성이 매우 강하였다. 그러나 간 조직을 이용하여 측정하는 지질과산화 억제활성과 TBA법으로 지질과산화를 측정한 결과, 그라비올라 잎의 에탄올 추출물에서 지질과산화 억제능을 보였다. ${\beta}$-Carotene bleaching에서 높은 lipid peroxyl 라디칼 소거 활성이 있음을 확인할 수 있었다. 따라서, 그라비올라 잎은 기능성 식품의 개발을 위한 천연 항산화제 소재로서 활용 될 수 있을 것으로 판단되어진다.

사방오리(Alnus firma) 추출물의 in vitro 항산화 활성 (In Vitro Antioxidant Activity of Alnus firma Extracts)

  • 본 연구는 사방오리 추출 분획물의 항산화 활성을 검증하기 위하여 시행되었다. 사방오리는 메탄올(MeOH) 분획에 가장 높은 총 페놀화합물($452.80{\pm}7.01{\mu}g$ gallic acid equivalents/mg)을 그리고 에틸에세테이트(EA) 분획 에 가장 높은 총 플라보노이드 함량($112.29{\pm}11.14{\mu}g$ rutin equivalents/mg)과 가장 높은 총 항산화 활성($936.23{\pm}0.07{\mu}g$ ${\alpha}$-tocopherol equivalents/mg)을 가지고 있었다. 사방오리의 용매 추출분획물에 대한 항산화 활성은 아스코빅산 등 표준 물질과 비교하여 1,1-diphenyl-2-picrylhydrazyl (DPPH) 라디칼 소거능 측정, 슈퍼옥사이드 음이온 라디칼 소거능 측정, ${\beta}$-carotene-linoleate 에세이, 환원력 측정, ferric thiocyanate법으로 지질과산화 억제력 측정 그리고 금속 킬레이팅능 측정 등 다양한 항산화 활성을 측정하였다. $250{\mu}g/ml$ 농도의 사방오리 EA와 MeOH 분획은 DPPH 라디칼 소거능 측정에서 각각 92.43와 89.20% 항산화 활성을 가지고 있었다. $50{\mu}g/ml$의 EA분획은 72.49% 슈퍼옥사이드 음이온 라디칼 소거능을 보였으며 이는 gallic acid의 소거능(60.88%)보다 조금 높았다. 또한 ${\beta}$-carotene-linoleic acid 시스템에 의한 과산화 억제능 조사에서 0.5과 1 mg/ml 농도의 EA 분획이 각각 73.45와 73.29% 억제능을 보였다. 항산화 활성은 EA 분획 > BuOH 분획 > DCM분획 > HX분획 순 이였다. 이상의 본 연구 결과에서 사방오리는 천연 항산화제제 개발을 위한 용이하게 접근 가능한 잠재적인 자원으로 판단된다.

토코페롤 및 카로틴이 정제 잣 지방질의 산화에 미치는 효과 (Effect of Tocopherols and Carotene on the Oxidation of Purified Pinenut Oil in the Model System)

  • The oxidation of purified pinenut oil containing various concentration of tocopherols and β-carotene were studied. α-tocopherol revealed an antioxidant activity at the concentration of lower than 0.05%, however, it showed a prooxidant activity when the concentration was higher than 0.05%. The antioxidant activity of γ-tocopherol was not affected by the concentraitons in the range of 0.01∼0.10% in pinenut oil. γ -Tocopherol resulted in higher antioxidant activity than that of α-tocopherol. β -carotene seemed to be a prooxidant when 0.01% of β-carotene was added. The fatty acids composition of purified pinenut oil have been changed during autoxidation. The concentration of linoleic acid decreased readly while oleic acid seemed to increase. And the concentration of saturated fatty acid has'nt been changed much during autoxidation.

데침시간에 따른 톳의 주요성분 및 항산화 활성의 변화 (Changes of chemical components and antioxidant activities in Hizikia fusiformis(Harvey) OKAMURA with blanching times)

  • This study was performed to investigate the change of chemical components and antioxidant activities in Hizikia fusiformis(Harvey) OKAMURA with blanching times. For chemical composition, the contents of minerals (K, Ca, Na, Mg, Fe, Cu, Mn and Zn), vitamins (vitamin C, ${\beta}$-carotene and ${\alpha}$-tocopherol) and total polyphenol were analyzed. The antioxidant activity was determined through free radicals (DPPH radical, superoxide anion radical, hydroxyl radical and hydrogen peroxide) scavenging activity and linoleic acid peroxidation inhibitory activity. As the blanching time increased, the contents of all minerals, vitamin C and total polyphenol decreased. However ${\beta}$-carotene and ${\beta}$-tocopherol were not affected by the blanching time. In addition antioxidant activities decreased with increasing blanching time.

Assessment of Antioxidant and Phenolic Compound Concentrations as well as Xanthine Oxidase and Tyrosinase Inhibitory Properties of Different Extracts of Pleurotus citrinopileatus Fruiting Bodies

  • Cellular damage caused by reactive oxygen species has been implicated in several diseases, thus establishing a significant role for antioxidants in maintaining human health. Acetone, methanol, and hot water extracts of Pleurotus citrinopileatus were evaluated for their antioxidant activities against ${\beta}$-carotene-linoleic acid and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals, reducing power, ferrous ion-chelating abilities, and xanthine oxidase inhibitory activities. In addition, the tyrosinase inhibitory effects and phenolic compound contents of the extracts were also analyzed. Methanol and acetone extracts of P. citrinopileatus showed stronger inhibition of ${\beta}$-carotene-linoleic acid compared to the hot water extract. Methanol extract (8 mg/mL) showed a significantly high reducing power of 2.92 compared to the other extracts. The hot water extract was more effective than the acetone and methanole extracts for scavenging DPPH radicals. The strongest chelating effect (92.72%) was obtained with 1.0 mg/mL of acetone extract. High performance liquid chromatography analysis detected eight phenolic compounds, including gallic acid, protocatechuic acid, chlorogenic acid, ferulic acid, naringenin, hesperetin, formononetin, and biochanin-A, in an acetonitrile and hydrochloric acid (5 : 1) solvent extract. Xanthine oxidase and tyrosinase inhibitory activities of the acetone, methanol, and hot water extracts increased with increasing concentration. This study suggests that fruiting bodies of P. citrinopileatus can potentially be used as a readily accessible source of natural antioxidants.

Antioxidant Activities and Tyrosinase Inhibitory Effects of Different Extracts from Pleurotus ostreatus Fruiting Bodies

  • We evaluated the antioxidant activity and tyrosinase inhibitory effects of Pleurotus ostreatus fruiting bodies extracted with acetone, methanol, and hot water. The antioxidant activities were tested against $\beta$-carotene-linoleic acid, reducing power, 1,1-diphenyl-2-picrylhydrazyl free radical scavenging activity, and ferrous chelating ability. Furthermore, phenolic acid and flavonoid contents were also analyzed. The methanol extract showed the strongest $\beta$-carotene-linoleic acid inhibition as compared to the other exracts. The acetone extract (8 mg/mL) showed a significantly high reducing power of 1.54 than the other extracts. The acetone extract was more effective than other extracts for scavenging on 1,1-diphenyl-2-picrylhydrazyl radicals. The strongest chelating effect (85.66%) was obtained from the acetone extract at 1.0 mg/mL. The antioxidant activities of the extracts from the P. ostreatus fruiting bodies increased with increasing concentration. A high performance liquid chromatography analysis detected seven phenolic compounds, including gallic acid, protocatechuic acid, chlorogenic acid, naringenin, hesperetin, formononetin, and biochanin-A in an acetonitrile and 0.1 N hydrochloric acid (5 : 1) solvent extract. The total phenolic compound concentration was $188{\mu}g$/g. Tyrosinase inhibition of the acetone, methanol, and hot water P. ostreatus extracts increased with increasing concentration. The results revealed that the methanol extract had good tyrosinase inhibitory ability, whereas the acetone and hot water extracts showed moderate activity at the concentrations tested. The results suggested that P. ostreatus may have potential as a natural antioxidant.

모델시스템에 있어서 무지개 송어 지방질의 산화에 대한 Lipoxygenase의 영향 (Effect of Lipoxygenase on the Oxidation of Rainbow Trout Lipid in Model system)

  • The effect of lipoxygenase (LOX) on the oxidation and co-oxidation of lipid fraction was studied in the model system of rainbow trout. For the reaction in model system 1 g of lipid fraction and 50mL of enzyme extract(LOX, 140 unit in 50mL phosphate buffer solution at pH 7, 4)), which were obtained from rainbow trout, were homoginized in the presence of Tween 20 and kept at 23$\circ$C for 3 days. The activity of LOX was decreased to 43% of initial level during the reaction in the model system. The initial composition of rainbow trout lipid was showed to be consisted of trigliceride(TG;82%) and free fatty acid(FFA;0.1%), while this converted to 59% of TG and 20% of FIFA, respectively after reaction in model system. Change of fatty acid composition was also observed and the content of linoleic acid, one of the major fatte acids, was decreased to 13% from 54% in the content of total fatty acids after reaction. The carotenoids in rainbow trout were composed of 0.4% $\alpha$-carotene, 1.6% $\beta$ -carotene, 80% canthaxanthin, 7% lutein and 11% zeaxanthin, thus the canthaxanthin was the major component. This canthaxanthin was the most degraded carotenoid by lipoxygenase catalyzed co-oxidation during the reaction. On the other hand the tocopherol isomers found in the rainbow trout were $\alpha$ and $\beta$ -tocopherol, and $\alpha$-tocopherol had a higher degradation rate by the lipoxygenase catalyzed co-oxidation than of $\beta$-tocopherol in the reaction of model system.

미나리과 산채의 아질산염 소거능 (Nitrite Scavenging Effect of Umbelliferaeceae)

  • 미나리과 산채인 참나물, 신선초, 방풍의 조단백은 22.99~31.00%, 조지방은 3.51~5.08%였으며, $\beta$-carotene은 5.40~19.81 mg%, ascorbic acid는 미량 함유하였고, Selenium은 모든 시료에서 검출되지 않았다. 총 페놀 함량은 617.87~875.77 mg%를 나타내었으며, 이 중 방풍이 가장 높은 함량을 보였다. 참나물, 신선초, 방풍의 지방산 조성은 전 지방산 중 linoleic acid와 linolenic acid가 66.46~77.44%를 차지하였다. 아질산염 소거능은 참나물, 신선초, 방풍 모두에서 메탄을 가용성 획분이 수용성 획분에 비하여 높은 소거능을 보였고, pH 1.2〉 4.2〉6.0 순으로 소거능이 높았다. 방풍의 메탄을 가용성 획분의 아질산염 소거능이 pH 1.2에서 95.3%로 가장 높았으며, 다음이 참나물, 신선초 순이었다. 총 페놀과 아질산염 소거능은 높은 상관관계를 나타내었다.

  • This study was performed to investigate the change of biologically functional compounds and antioxidant activities in Ecklonia cava with blanching times. As biologically functional compounds, the contents of minerals(K, Ca, Na, Mg, Fe, Cu, Mn and Zn), vitamins($vitamin\;C,\;{\beta}-carotene\;and\;{\alpha}-tocopherol$) and total polyphenol were analyzed. And antioxidant activity was determined through free radicals(DPPH radical, superoxide anion radical, hydroxyl radical and hydrogen peroxide) scavenging activity and linoleic acid peroxidation inhibitory activity. As the blanching time increased, the contents of minerals, vitamin C, ${\beta}-carotene$ and total polyphenol were decreased however ${\alpha}-tocopherol$ was not affected by blanching time, and antioxidant activities were decreased with blanching time.

Gingerol의 산화방지 효과 (The Antioxidant Activity of Gingerol)

  • 생강으로부터 분리한 gingerol의 산화방지 효과에 대하여 시험하였다. 생강의 에탄올 추출물로부터 핵산-에테르 혼합용매를 이용하여 crude gingerol을 얻어 TLC에 의하여 gingerol을 분리.정제하였다. 분리한 gingerol은 발색반응, 산탈수반응, UV, IR 및 NMR 스펙트로메트리등을 거처 6- 및 10-gingerol임을 동정하였다. Gingerol은 리노레산-${\beta}$-카로틴-물의 메멀죤에 0.02%(w/v)농도로 첨가되었을 때 대조구에서보다 ${\beta}$-카로틴을 크게 안정화시켰다. 그러나 BHA 및 BHT보다는 그 효과가 약간 낮았다.
