Interpretation of Antimicrobial Susceptibility Test According to Resistance Mechanism of ${\beta}$-lactam in Enterobacteriacae (장내세균에서 ${\beta}$-lactam 항균제의 내성기전별 항균제 감수성검사의 해석)

  • It is important to select appropriate antimicrobials for the treatment of infection according to the results of antimicrobial susceptibility tests (ASTs), yet the clinical isolates are sometimes susceptible to antibiotics that are clinically ineffective or this is due to technical error of the ASTs. So, interpretive reading of ASTs is needed and especially for the ${\beta}$-lactams for treating $Enterobacteriacae$. This review describes the interpretive reading of ASTs according to natural antimicrobial resistance and the mechanisms of mechanisms, with giving special attention to the antibiotics phenotypes for $Enterobacteriacae$. Further, as all the diffent tissues have a different antimicrobial concentration for identical antimicrobials, more information is needed on the antimicrobial tissue distribution for the appropriate treatment of infection. Study on the Antitumor Activity of Tripterygium Regelii Sprague (미역줄나무의 항암활성에 관한 연구)

  • Tripterygium regelii has been used as an oriental medicine, especially antiparasitic, anti-inflammatory and detoxifying agents in East asia. During our research to develop new antitumor agents from natural products, MeOH ext. and CH2Cl2 ext. of Tripterygium regelii showed the potent antitumor activity. In order to purify active compounds from Tripterygium regelii, activity-guided fractionation was carried out. Silica gel and RP-18 column chromatography for the active fraction led to the isolation of two compounds and their antitumor activities were studied. Those two compounds didn't show potent antitumor activity against human tumor cell lines. The structure of two compounds were determined by $^1H-NMR$, $^{13}C-NMR$, DEPT, $^1H-^{13}C$ COSY and IR spectrum. Compound I and Compound II were turned out to be Celastrol, and ${\beta}-sitosteryl-3-o-{\beta}-D-glucopyranoside$ respectively.

Anti-inflammaory effects of the MeOH extract of Gentianae Macrophyllae Radix in vivo (진교(秦艽)가 항염 효과에 미치는 영향)

  • Objectives : The present study was examined to evaluate the anti-inflammatory effects of the Gentianae Macrophyllae Radix MeOH extracts (GMR) in vivo. Methods : The effects of GMR on anti-inflammation were measured by production of NO, TNF-$\alpha$ (Tumor Necrosis Factor-alpha) and IL-$1{\beta}$ (Interleukin-$1{\beta}$), IL-6 in Raw 264.7 macrophage cells stimulated with LPS. Results : 1. All concentrations of GMR(0.10 mg/ml) had no significant cytotoxicity in Raw 264.7 cell during the entire experimental period. 2. The level of NO and iNOS in culture medium was dramatically increased by LPS application. However, these increases were dose-dependently(0.03 and 0.10 mg/ml) attenuated by treatment with GMR. 3. All concentrations of GMR significantly inhibited the production of IL-$1{\beta}$ in Raw 264.7 macrophage cells stimulated with LPS. Conclusions : These results provide evidences that therapeutic effect of GMR on heat syndrome, especially due to the acute inflammation, are partly due to the reduction of some of inflammatory factors by inhibiting iNOS and COX-2 through the suppression of $p-I{\kappa}B{\alpha}$. Moreover, it suggests that the mechanism of action of GMR comes from the suppression of inflammatory mediators, such as NO, PGE2 and pro-inflammatory cytokines.

  • The stem of Chrysanthemum morifolium, 'Baekma', were repeatedly extracted with 80% aqueous MeOH and the concentrates was partitioned into ethyl acetate (EtOAc), n-butyl alcohol and $H_2O$ fraction. The repeated silica gel and octadecyl silica gel column chromatographies for the EtOAc fractions led to isolation of two glycosyl glycerides. The chemical structures of the compounds were determined as (2S)-1-O-${\beta}-{\text\tiny{D}}$-galactopyranosyl-2,3-dilinoleoylglycerol (1) and (2S)-1-O-${\beta}-{\text\tiny{D}}$-galactopyranosyl-2,3-dipalmitoylglycerol (2) based on spectroscopic data anlyses including nuclear magnetic resonance, mass sperctrometry, and infrared spectrometry and gas chromatography mass spectrometry.

Effect of Dietary Fatty Acids and Vitamin E Supplementation on Antioxidant Vitamin Status of the Second Generation Rat Brain Sections (식이 지방산 및 비타민 E 보충 식이가 흰쥐의 뇌조직 부위별 항산화 비타민 농도에 미치는 영향)

  • Effects of dietary fatty acids and vitamin E on antioxidant vitamin status were studied in rat brain sections. Sources of dietary fat(10t%) were safflower oil(SO) poor in $\omega$3 fatty acid and mixed oil (MO) with computer-adjustd fatty acid ratios(AA/DHA=1.4, $\omega$6/$\omega$3=6.3, P/M/S=1.0/1.5/1, AA=2.%)with (ME) and without(MO) vitamin E(500mg/kg diet). Rats were fed the three kinds of diet from 3-4 wks prior to the conception. At the age of 3 & 9wks of the 2nd generation rat, antioxidant vitamins were measured in frontal cortex(FC), corpus striatum (CS), cerebellum(CB) and hippocampus(HP) using a multiwavelength, reverse phase gradient HPLC system. The levels of antioxidant vitamins converged to the similar value in all groups at 9 wks of age. Retinol, lycopene and cryptoxanthin levels of all experimental groups were found to be the highest in hippocampus at both 3 & 9wks of age. The levels of vitamin E appeared to be higher in the order of HP>CS-CB>FC in MO & ME. Beta-carotene and retinol showed the lowest level in hippocampus of vitamin E supplemented groups, even though vitamin E level tended to be higher in other sections. It seemed that vitamin E has an inhibitory action on the uptake of beta-carotene or acts as a preferred antioxidant to beta-carotene in certain section of the brain. By improving fatty acid balance (AA/DHA = 1.4, $\omega$6/$\omega$3=6.3, P/M/S=1.0/1.5/1, AA = 2%), the levels of vitamin E, retinol, lycopene & beta=carotene tended to be higher in MO than in SO, although crytoxanthin became lower at 3wks of age. In short, dietary fatty acids and vitamin E have different influence on antioxidant vitamin status in different rat brain sections. The higher levels of antioxidant vitamins in hippocampus should be pursued further in relation to behavioral development of rats.

Effects of Antioxidants Supplement in Porcine Sperm Freezing on in vitro Fertilization and the Glutathione and Reactive Oxygen Species Level of Presumptive Zygotes

  • The present study was aimed to determine the effects of green tea extract (GTE) and beta-mercaptoethanol (${\beta}-ME$) supplementation in boar sperm freezing extender on in vitro fertilization (IVF) and reactive oxygen species (ROS) and glutathione (GSH) levels of presumptive zygotes (PZs). Experimental groups were allocated into lactose egg yolk (LEY) without antioxidant (control), GTE (1,000 mg/l in LEY) and ${\beta}-ME$ ($50{\mu}M$ in LEY). In freezing, spermatozoa extended with LEY were cooled to $5^{\circ}C$ for 3 h and then kept at $5^{\circ}C$ for 30 min following dilution with LEY containing 9% glycerol and 1.5% Equex STM. The final sperm concentration was $1{\times}10^8/ml$. Spermatozoa were loaded into straws and frozen in nitrogen vapor for 20 min. For IVF, oocytes were matured in NCSU-23 medium and co-cultured with spermatozoa following thawing at $37^{\circ}C$ for 25 sec. At 12 h following IVF, IVF parameters (sperm penetration and monospermy) were evaluated. In addition, GSH and ROS levels of PZs were determined by Cell Tracker Blue CMF2HC and DCHFDA, respectively. IVF parameters did not show any significant difference among the experimental groups. GSH and ROS levels of PZs were not significantly different between groups. In conclusion, antioxidant supplementation in boar sperm freezing could not influence IVF parameters, ROS and GSH levels of PZs.

  • The repeated silica gel colum chromatographies of n-BuOH fraction obtained from MeOH extracts of Eucommia ulmoides leaves led to isolation of a flavonoid-glycoside inhibiting monoamine oxidase B activity. Its chemical structure was determined to be $3-O-[{\beta}-D-glucopyranosyl\;(1{\rightarrow}2)\;{\beta}-D-xylopyranosyl]$ quercetin through interpretation of spectral data and adaptation of acid hydrolysis. The $IC_{50}$ value of this compound in rat brain mitochondrial MAO-B inhibitory activity was evaluated to be $8.05\;{\mu}mol/l$.

Anti-Inflammatory Activity of Compounds from the Whole Plant of Patrinia saniculaefolia

  • An in vitro bioassay-guide revealed that the methanol (MeOH) extract of the whole plant of Patrinia saniculaefolia (Valerianaceae) showed cyclooxygenase-2 (COX-2) and 5-lipoxygenase (5-LOX) dual inhibitory activity by assessing their effects on the production of prostaglandin $D_2$ ($PGD_2$) and leukotriene $C_4$ ($LTC_4$) in mouse bone marrow-derived mast cells (BMMCs). Phytochemical study of the MeOH extract of this plant led to the isolation of twelve compounds; ${\beta}$-farnesene (1), squalene (2), nardostachin (3), patridoid I (4), patridoid II (5), patridoid II-A (6), oleanolic acid (7), oleanonic acid (8), 23-hydroxyursolic acid (9), oleanolic acid 3-O-${\alpha}$-L-arabinopyranoside (10), oleanolic acid 3-O-${\beta}$-D-glucopyranoside (11), oleanolic acid 3-O-[${\beta}$-D-xylopyranosyl-(1${\rightarrow}$3)-${\beta}$-D-(6-O-butyl)glucuronopyranoside] (12). Among the compounds, 4 and 5 strongly inhibited both the COX-2-dependent $PGD_2$ generation with $IC_{50}$ values of 8.7 and 13.6 ${\mu}M$, respectively, and the generation of $LTC_4$ in the 5-LOX dependent phase with $IC_{50}$ values of 41.7 and 46.9 ${\mu}M$, respectively, which suggest that the anti-inflammatory activity of P. saniculaefolia might occur in part via the inhibition of both $PGD_2$ and $LTC_4$ generation by 4 and 5.

The repeated octadecyl $SiO_2$ and silica gel ($SiO_2$) column chromatographies for the EtOAc and nbutyl alcohol fractions led to isolation of two ergosterol peroxides. There chemical structures were determined as ($3{\beta}$,$5{\alpha}$,$8{\alpha}$,22E)-5,8-Epidioxyergosta-6,22-dien-3-ol (ergosterol peroxide) (1) and 3-O-${\beta}$-D-glucopyranosyl ergosterol peroxide (2) based on spectroscopic data analyses including nuclear magnetic resonance, infrared spectrometry, and mass spectrometry (MS). Compounds 1 and 2 were for the first time isolated from S. crispa in this study.

The Measurement of National Standard ${\beta}$-Rays Energy Spectrum (기준 베타선장의 에너지 스펙트럼 측정)

  • In the present study, we measured the pure beta particle energy spectra of $^{147}Pm$, $^{85}Kr$, $^{90}Sr+^{90}Y$ radionuclide sources. We confirmed the residual maximum energies of KRISS sources meet the requirement of ISO 6980 and calculated mass collision stopping power ratio, which is essential for absolute measurement of absorbed dose from the reference ${\beta}$-rays. The residual maximum energies of KRISS $^{147}Pm$, $^{85}Kr$, $^{90}Sr+^{90}Y$ sources are 0.14, 0.57 and 0.93 MeV, respectively and the mass collision stopping power ratios are 1.123, 1.120 and 1.109, respectively.
