Intestinal Bacterial ${\beta}-Glucuronidase$ Activity of Patients with Colon Cancer

  • The fecal ${\beta}-glucuronidase$ activity of patients with colon cancer and healthy controls were measured to determine the relationship between the fluctuation of intestinal bacterial ${\beta}-glucuronidase$ and colon cancer. The fecal ${\beta}-glucuronidase$ activity of patients with colon cancer was 1.7 times higher than that of the healthy controls. However, when these fecal specimens were sonicated, the enzyme activity of patients with colon cancer was 12.1 times higher than that of the healthy controls. The fecal ${\beta}-glucuronidase$ activity of human Intestinal bacteria was drastically induced by its substrate or the bile secreted after a subcutaneous injection of 1,2-dimethylhydrazine (DMH) and benzo[a]pyrene into rats. DMH-and benzo[a]pyrene-treated biles induced ${\beta}-glucuronidase$ activity in the human intestinal microflora by approximately 1.5- and 2.3-fold, respectively. They also induced ${\beta}-glucuronidase$ in E. coli HGU-3, which is a ${\beta}-glucuronidase$-producing bacterium from the human intestine. D-saccharic acid 1,4-lactone similarly inhibited fecal ${\beta}-glucuronidase$ in several patients with colon cancer in addition to the healthy controls. This suggests that potent ${\beta}-glucuronidase$ activity is a prime factor in the etiology of colon cancer.

${\beta}-Glucuronidase-inhibitory\;Activity$ and Hepatoprotective Effect of Herbal Medicines (생약의 ${\beta}-Glucuronidase$ 저해와 간장보호효과)

  • Inhibitory effect of water and ethyl acetate extract of 60 kinds of herbal medicines was investigated on ${\beta}-glucuronidase$. Among water extract of them, Galla Rhois had the most potent ${\beta}-glucuronidase-inhibitory\;activity$. Termaliae Fructus, Amomi Tsa-ko Fructus and Arecae Semen were also potent inhibitors. Among ethyl acetate extract of them, Galla Rhois had the most potent ${\beta}-glucuronidase-inhibitory\;activity$. Nelumbinis Semen. Ephedrae Radix and Termaliae Fructus were also potent inhibitors. The extract of Galla Rhois had potent hepatoprotective effect on $CCl_4-induced$ hepatotoxicity of rats. These results suggest that the ${\beta}-glucuronidase$ seems to be closely related to the liver injury, which could be prevented by the inhibitor of ${\beta}-glucuronidase$.

Inhibitory Effect of Zizyphi fructus on ${\beta}-Glucuronidase$ and Tryptophanase of Human Intestinal Bacteria (대추의 장내세균 유해효소 ${\beta}-Glucuronidase$와 Tryptophanase 저해효과)

  • In human intestine, more than 100 species of bacteria reside and dietary factors may alter the bacterial flora which produce bacterial enzymatic activities. Especially ${\beta}-glucuronidase$ and tryptophanase activities in colon are closely associated with occurrence of colon cancer. Therefore, the inhibitory effect of traditional herbal food extracts on these intestinal bacterial enzymes are measured. The results of this study showed that Zizyphi fructus and Glycyrrhiziae radix decreased not only ${\beta}-glucuronidase$ and tryptophanase productions of human intestinal bacteria but also inhibited potently ${\beta}-glucuronidase$ and tryptophanase. Among solvent-extracted fraction of tested herbal foods, ether fraction of Glycyrrhiziae radix and ethylacetate fraction of Zizyphi fructus inhibited potently ${\beta}-glucuronidase$ and tryptophanse. Thus, ethylacetate fraction of Zizyphi fructus separated six components by silica gel column chromatography. The component having Rf=0.34 and Rf=0.43 $(developing\;solvent,\;CHCl_3/MeOH\;(3:1))$ shwed the highest inhibitory effect of ${\beta}-glucuronidase$ and tryptophanase among them.

  • The growth kinetics and the production of $\beta$-glucuronidase from transgenic tobacco's suspension culture was investigated in the flask culture and a 2.5 L bubble column reactor. The growth of bubble column reactor was similar to that of flask culture. However, in the bubble column reactor, the production of $\beta$ -glucuronidase reached 2850 U/mg (85-fold higher than that of flask culture). In both case, the production level of $\beta$ -glucuronidase was fluctuated, which was resulted from periodical degradation of the protein. Sucrose is important component in plant culture medium. Twice addition of sucrose in bubble column reactor could not improve cell growth, since other components in a medium were already depleted. However, the addition of sugar decreased cell size, which facilitated the operation of bioreactor. The production of $\beta$ -glucuronidase was continuously increased, however final concentration of $\beta$ -glucuronidase was similar to that without sucrose addition.

Protective Effect of Antler in Experimental Colon Carcinogenesis

  • There are epidemiological evidences that the population with high fecal ${\beta}-glucuronidase$ activity has greater risk of colon cancer than the population with low fecal ${\beta}-glucuronidase$. This relationship was investigated by using the mouse-dimethylhydrazine colon carcinogenesis model and the extract of antler which was a ${\beta}-glucuronidase$ inhibitor. Mice with low fecal ${\beta}-glucuronidase$ activity induced by administration of water and Folch's fraction of antler had significantly fewer aberrant crypts after injections of 1,2-dimethylhydrazine (DMH) than mice treated with DMH alone. The result supports the hypothesis that the inhibitor of ${\beta}-glucuronidase$ such as antler extract can protect an animal against the induction of colon cancer.

  • Lactic acid bacteria (LAB) isolated from kimchi were studied for their ${\beta}$-glucuronidase activity. Among the 156 strains tested, 52 strains utilized glucuronic acid as a carbon source and their intracellular ${\beta}$-glucuronidase activities were significantly higher than their extracellular activities. Leuconostoc mesenteroides KFRI 73007 isolated from turnip kimchi exhibited the highest intracellular ${\beta}$-glucuronidase activity of $0.77{\pm}0.01U/mg$ protein, which was further increased to $1.14{\pm}0.01U/mg$ protein under optimized reaction conditions (pH 7, $37^{\circ}C$). The activity of ${\beta}$-glucuronidase was notably decreased by the addition of divalent cations, and glucuronic acid was the best carbon source to produce ${\beta}$-glucuronidase in Leu. mesenteroides KFRI 73007.

Studies on $\beta$-Glucuronidase Activities in Liver, Stomach and Small Intestinal Tissues of Rabbits Infected with Clonorchis sinensis (간디스토마 감염토끼의 소화기관에 대한 $\beta$-Glucuronidase의 활성치에 관한 연구)

  • The author has studied the $\beta$-glucuronidase activity in several tissues such as liver, stomach and small intestine of the male and female rabbits infected with different doses of metacercariae of Clonorchis sinensis. The metacercariae of Clonorchis sinensis were isolated from Pseudorasbora parva caught in Kim Hae by digestion technic. The experimental animals were sacrificed in the period of 1, 7, 14, 21, 28 and 35th days following the infection. The results obtained were summarized as follows. 1. In the groups infected with 100 metacercariae, $\beta$-glucuronidase activity was slightly increased during the entire periods than control rabbits. It was the highest in the first day with 1.535 and $1.421m{\mu}/g$, 14th days with 2.521 and $2.200m{\mu}/g$, and then lowered by the time, gradually. 2. In the groups infected with 500 metacercariae, $\beta$-glucuronidase activity was highly increased on the first day with 1.535 and $1.856m{\mu}/g$ than that 100 metacercariae groups according to each organs. It was the highest on the 7th day and 14th day. 3. In the groups infected with 1,000 metacercariae, $\beta$-glucuronidase activity was remarkably increased in the first and 14th days according to each organs, and then lowered gradually day by day. 4. $\beta$-glucuronidase activity of all organs was more increased than that of normal organs and the highest activity in the liver with $2.521m{\mu}/g$, intestine(1.612) and stomach (1.581) respectively. 5. $\beta$-glucuronidase activity of rabbits was higher in the female than in the male. On the basis of these results, it was suggested that $\beta$-glucuronidase activity was affected by the duration of infection and by the number of Clonorchis sinensis, according to the organs and sex of the rabbits.

Mice with low facal ${\beta}-glucuronidase$activity induced by consumption of the ether fraction of G lucidum had significantly fewer aberrant crypts(AC) after injections of 1, 2-dimethylhydeazine (DMH) than mice treated with DMH alone. The result supports the hypothesis that the inhibitor such as the ether fraction of G lucidum can protect an animal against the induction of colon cancer.

  • Approximately 1% of intestinal bacteria of human and rats was alkalotolerant. Among these bacteria of human, bacteria producing $\beta$-glucosidase, $\beta$-glucuronidase and sulfotransferase were 40%, 4% and 0%, respectively. Among alkalotolerant intestinal bacteria of rats, bacteria producing, these enzymes were 70%, 8% and 0%, respectively. $\beta$-Glucosidase and $\beta$-glucuronidase of alkalotolerant intestinal bacteria of human and rat were induced by the medium of high pH: these enzymes activities were increased by elevating pH of the medium, but the growths were not changed. The enzyme activities at the medium of pH 7 were about ten-fold higher than those at the medium of pH 6.

  • For the study of human intestinal environments with the intestinal bacteria producing ${\beta}-glucuronidase\;and \;7{\alpha}-dehydroxylase$, genus Clostridium, known as the producer, were isolated from the fecal microflora. Through screening twice for one person, fecal microflora without major bacterial group seemed to be changed, which indicated the microflora would be changeable by the diet factors. With using Neomycin-Nagler selective medium during the screening, 14 Clostridium spp. were isolated and then the harmful enzyme activities were determined. Isolate-11 among them produced strongly ${\beta}-glucuronidase$ and its activity was 0.021 unit/mg Protein. However, the strain producing $7{\alpha}-dehydroxylase$ was not isolated. The Isolate-11 was tentatively identified as Clostridium scatologenes through cultural and physiological characteristic.
