Linkage Structure Analysis of Barley and Oat $\beta$-Glucans by High Performance Anion Exchange Chromatography

  • Cereal $\beta$-glucans, linked essentially by mixed $\beta$-(1,4/1,3) glycosidic bonds, were extracted, purified, and structurally identified. Previously chemical structure of barley $\beta$-glucans was characterized from 3 varieties of 'Gang', 'Ohl', and 'Gwangan', and the (1,4)/(1,3) linkage ratio of the $\beta$-glucans was identical. In this study, $\beta$-glucans from 1 barley ('Chal') and 3 oat ('Ohl', 'Samhan', and 'Donghan') varieties were structurally scrutinized, and the linkage pattern of total 7 cereal $\beta$-glucans was compared. The amount of 2 major 3-O-$\beta$-cellobiosyl-D-glucose (DP3) and 3-O-$\beta$-cellotriosyl-D-glucose (DP4) from barley and oat accounted for only 66.6-73.3 and 68.12-81.89% of water-extractable $\beta$-glucan fractions, and the (1,4)/(1,3) linkage ratios of both barley and oat $\beta$-glucans were within very narrow range of 2.27-2.31 and 2.38-2.39, respectively, among the cultivars tested. Structural difference in the cereal $\beta$-glucans was evident when DP3:DP4 ratio in the $\beta$-glucan structure was compared. As a result, this ratio was significantly greater for barley $\beta$-glucan (2.26-2.74) than for oat (1.54-1.66). Chal-B had the greatest DP3 to DP4 ratio among the samples, which in turn reflected the least amount of (1,4)-linkages.

  • $\beta $-1, 4-D-arabinogalactanase isolated from alkalophilic Bacillus sp. HJ-12, approximate Mw 42 kDa, was generally stable in the range of pH 6-10 and below 50$\circ$C and its highest activity was observed at 60$\circ$C with pH 7-9. The isolated $\beta $-1, 4-D-arabinogalactanase specifically hydrolyzed $\beta $-1, 4-galactosyl linkage that is the major structure of soybean arabinogalactan (SAG) but not $\beta $-1, 3-galactosyl linkage of the other polysaccharides. K. was estimated as 0.67 mg/ml by the method of Hanes-Woolf plot. No metals and chemical reagents inhibited the enzyme activity but urea did. The active site of this enzyme assumed to be tryptophan residue. The hydrolysis products from SAG, assayed by gel chromatography, TLC and HPLC, were predominantly galactotetraose (Gal$_{4}$) and triose (Gal$_{3}$) with a small portion. $\beta $-1, 4-D-arabinogalactanase hydrolyzed ONPG as well as SAG, and the degree of hydrolysis of SAG was 15% which is lower than that by the other $\beta $-1, 4-galactanases from different sources. SAG treated with this enzyme resulted in the reduction of specific viscosity up to 70%.

Effect of Temperature on the Extraction of β-Glucan from Different Jeju Barley Varieties

  • The effect of different temperatures (45, 55, 65, and $75^{\circ}C$) on the extraction of ${\beta}$-glucan and the properties of extracted ${\beta}$-glucan were investigated with four different varieties of barley. Jeju naked barley, blue barley, beer barley, and black barley contained 6.85, 5.13, 3.58, and 4.16% of ${\beta}$-glucan, respectively. ${\beta}$-glucan in barley was extracted in the range of 64.88 to 93.84% depending on the extraction temperature and barley variety. The ${\beta}$-glucans in Jeju naked barley, Jeju blue barley, and black barley were optimally extracted at $65^{\circ}C$ for 3 h and Jeju beer barley at $75^{\circ}C$. The extracted ${\beta}$-glucan resolubilized to 43.48-81.73% and the ratio of ${\beta}(1{\rightarrow}3)$ to ${\beta}(1{\rightarrow}4)$ linkage was in the range of 1:3.8-5.8. These results suggest that purification and properties of ${\beta}$-glucan depend not only on the water extraction temperature, but also on the barley variety.

  • 적색효모 세포벽 용해효소의 기질특이성을 밝히기 위해 기질로 사용가능한 다당을 screening하여 Rh.glutinis IFO 0695가 생산하는 mannan을 얻었다. 이 mannan의 구조는 $\beta$-1,3과 $\beta$-1,4 결합이 교대로 연결된 직쇄상의, 중합도 약 650인 수용성의 다당으로 밝혀졌다. 이 다당을 기질로하여 양 효소의 기질특이성을 조사한 결과, Bacillus생산효소는 $\beta$-1. 3, $\beta$-1. 4 mannan을 4당 또는 6당 단위로 가수분해하는 효소로서 그 절단점은 $\beta$-1, 4결합 이었다. 이 $\beta$-1,4 결합을 가수분해하기 위해서는 인접 결합이 $\beta$-1,3결합이어야만 하는 새로운 type의 $\beta$-1,4 mannauase이었다. 한편, Penicillium생산효소는 $\beta$-1, 3, $\beta$-1, 4 mannan의 $\beta$-1, 3결합을 2당 또는 4당의 단위로 가수분해하여 최종적으로는 2당을 생성하는 $\beta$-1, 3 mannanase의 일종이었다.

  • Veratrylglycerol-${\beta}$-vanillylalcohol ether 결합 (${\beta}$-O-4 linkage)을 갖는 이양체(二量體) 리그닌 모델화합물(化合物)의 합성(合成)하여 각종 리그닌 반응(反應)의 시료(試料)로서 활용(活用)하고자 3,5-dimethoxyacetophenone을 출발물질(出發物質)로 하여 ${\beta}$-ring에 측소(側銷)를 갖는 이양체(二量體) 리그닌 모델화합물(化合物)(I-IV)을 합성(合成)하였고, 융점측정, $^1H(^{13}C)$-NMR 및 Mass 스펙트라에 의하여 그 화학구조(化學構造)를 동정(同定)하였다. 이 합성화합물(合成化合物)중에서 1-(3,4-dimethoxyphenyl)-2-(2'-methoxy-4'-hydroxymethylphenoxy)ropanediol-1,3[IV]는 매우 중요한 리그닌 이양체(二量體) 화합물(化合物)이다. 이러한 리그닌 합성연구(合成硏究)는 기초적(基礎的)인 연구(硏究)의 중요성(重要性) 뿐만아니라 응용면(應用面)에서도 그 활용도(活用度)가 높을 것이다.

  • Spicatoside A and spicatoside B were isolated from Liriopis tuber. Spicatoside A has anticancer activity and it is composed of 1 ${\beta}$-hydroxy diosgenin and trisaccharides. Spicatoside B has the preventive effect of diabetes mellitus and also has 1 ${\beta}$-hydroxy derivatives, which is clevaged the ether linkage of F ring of diosgenin. Therefore, selective synthesis of 1 ${\beta}$-hydroxydiosgenin is required. (omitted)

Inheritence, linkage and Possible Use of Fractured Starch Mutant in Barley (Hordeum Vulga L.)

  • In order to breed barley lines with reduced viscosity of wort, a fractured starch mutant of naked barley cultivar, Nubet, was obtained from the M2 seeds mutated by the diethyl sulfate treatment. Seeds of this fractured starch mutant were opaque and the endosperm consists of angular, irregular and fractured starch. The mutant was caused by single recessive mutation and assigned by the symbol fra. The gene was located on chromosome 4, distal in long arm by linkage recombinations using translocation homozygote lethal test set. The linkage value between the fractured starch mutant and 72-4a, 72-4d were 26.0$\pm$4.9, 34.2$\pm$3.1 percent respectively. In addition to the reduced seed size, fewer kernels per spike and higher tillering ability, lower $\beta$-glucan viscosity and higher lysine content of the grain were associated with this mutant. $\beta$-glucan viscosity of the Nubet grains increased from 3 weeks after anthesis to matury and most of the viscose substances appeared to be stored in the middle of the endosperm tissue. Since the mutant grains showed better milling property as compared to Nubet, it can be used as breeding resources to develope new barley cultivars for maltins and milling purpose.

  • Campylobacter jejuni 그램 음성균의 O-antigen 부위를 구성하는 삼탄당의 반복 단위를 합성하기 위해서 먼저 이들에 포함된 알트로헵토오스의 합성과 ${\alpha}$-알트로피라노시드를 선택적으로 형성시키는 방법을 개발하여야 한다. 본 연구에서는 알알 유도체에 선택적으로 1, 2-trans glycosidation을 시킴으로서 ${\alpha}$-알트로피라노시드 결합을 갖는 이당류를 합성하고자 하였다. 4, 6-O-Benzylidene-D-allal을 DMDO(3, 3-dimethyl diox-irane)와 반응시켜 1, 2-무수당 중간체를 만들고 아릴 알코올과 글루칼 유도체들과 반응시킨 결과 아릴 ${\alpha}$-알트로피라노시드 유도체와 ${\alpha}$-알트로피라노시드 결합을 갖는 이당류를 효과적으로 합성할 수 있었으며 마노오스등에서 간접적으로 ${\alpha}$-알트로피라노시드를 만드는 기존의 방법을 대체할 새로운 방법임을 확인 할 수 있었다.

  • The mode of transglycosylation reaction observed during the action of low-molecular-weight 1,4-$\beta$-D-glucan glucanohydrolase (EC 3.2.1.4) purified from Trichoderma koningii ATCC 26113 was investigated using $^{1}H-NMR $spectroscopy. The H-1 proton resonances were analysed. After reaction of the enzyme with cellotriose, the reaction products were separated by high performance liquid chromatography. H-1 resonances of the products were consisted with those of cellobiose, cellotriose and cellotitraose, respectively. Therefore it was proved that all the reaction products formed by the action of the enzyme on cellooligosaccharides, including transglycosylation products, possess only H-NMR -1,4-glycosidic linkage(s).

$\beta$-glucosidase의 고정화와 효소 반응특성 (Immobilization of $\beta$-glucosidase and properties of Immobilized Enzyme)

  • Aspergillus niger유래의 $\beta$-glucosidase를 (1)glutarald-ehyde를 가교제로 한 chitin과 chitosan담체에 covalent linkage (2)Amberite IRA93과 DEAE-cellulose담체에 succinylation시킨 후 흡착, 그리고 (3)alginarte, polyacry-lamide를 각종 가교제를 이용 entrapment등의 방법으로 고정화 하였다. Glutaraldehyde를 가교제로써 chitosan을 활성화시킨후 $\beta$-glucosidase를 고정화 시켰을때 효소활성 회수율이 31.5%로 가장 높았고, 또한 column형 반응기에서의 15일 경과 후 효소활성 유지도도 69%로서 가장 우수하였다. 또한 succinylation시킨 효소를 Amberite IRA93 담체에 흡착시켰을 때 효소활성 회수율은 24.7%였고 효소 활성유지도는 62%였다. 반면에 entra-pment 방법에 의한 $\beta$-glucosidase의 고정화는 효소의 계속적인 용출로 $\beta$-glucosidase의 고정화에는 적합하지 않았다. Chitosan담체에서의 고정화 최적조건을 조사한 결과 가교제인 glutaraldehyde의 최적농도는 0.4%였고, glutaraldehyde의 최적 반응 pH는 4.8이었다. 또한 column형 반응기를 이용하여 cellobiose로부터 glucose로의 전환율을 조사하여 고정화 효소의 효용성을 검토하였다.
